Effects of temperature and transfer from seawater to freshwater on blood microrheology in Pacific salmon.

Blood of Pacific salmon was studied with particular interest in red blood cell (RBC) deformability in relation to migration. Blood samples were taken via cardiac puncture or chronic cannula placed in the dorsal aorta and heparinized. As an index of RBC deformability the mean passage time of single RBCs through micropores of 8 micron in diameter and 10 micron in length was determined under a pressure difference of 10 cmH2O. Despite about 100 mOsmol/l difference in plasma osmolality, there was no marked difference in RBC passage time between fish in seawater and those well acclimatized to freshwater. However, it seemed probable that a transient decrease in RBC passage time, i.e., an increase in RBC deformability, occurred immediately following transfer from seawater to freshwater. Plasma osmolality decreased to about 300 mOsmol/l within 1 hr after the transfer and showed no fluctuations thereafter. The temperature dependence of RBC deformability was much smaller in comparison with those previously observed in yellowtail and carp; salmon RBCs were still highly deformable even at 5 degrees C, a possible temperature of cold river water.

Elevated serum alpha-hydroxybutyrate dehydrogenase in patients with ovarian carcinoma.

To differentiate between ovarian carcinoma and benign ovarian tumor, we compared alpha-hydroxybutyrate dehydrogenase (HBD), lactate dehydrogenase (LDH) and its isoenzyme activities in serum in 115 patients with ovarian carcinoma and benign ovarian tumor. Both serum HBD activity and total LDH activity in ovarian cancer patients were significantly (p less than 0.001) higher than that in patients with benign ovarian tumor. From analysis of serum LDH isoenzymes, a significant decrease in LDH-1, and significant increases in LDH-4 and LDH-5 in ovarian cancer patients were observed, when compared with those in patients with benign ovarian tumor. When the ovarian cancer mass could be removed surgically, a significant decrease in the HBD was observed, suggesting that the HBD might fairly reflect the tumor burden. Serum HBD activities in a half of ovarian cancer patients were higher than the mean +2 SD of those in benign control subjects.

The effect of cimetidine on natural killer activity of peripheral blood lymphocytes of patients with ovarian carcinoma.

The effect of cimetidine on the natural killer (NK) activity of peripheral blood lymphocytes (PBL) of 32 patients with ovarian carcinoma was studied before and after surgery or chemotherapy. There was no significant difference in the NK activity between normal healthy women and patients who had been disease-free for more than a year after initial surgery, while those in patients with a large residual tumor after surgery were profoundly suppressed. Cimetidine stimulated the NK activity in the disease-free patients in vivo or in vitro. In addition, cimetidine augmented the response to phytohemagglutinin of PBL from the disease-free patients. The NK activity of PBL of patients with a large residual tumor under chemotherapy consisting of cisplatinum, adriamycin and cyclophosphamide, "CAP," was about half of that before initiation of chemotherapy. The inhibition of the NK activity by chemotherapy was abrogated by in vitro exposure of the PBL to cimetidine.

Demonstration of structural polymorphism among MB3 light chains by two-dimensional gel electrophoresis.

The heavy and light chain subunits of MB3 molecules were isolated from KT2 (DKT2, DR4, MB3 homozygous), ER (Dw4, DR4, MB3 homozygous), JMe (Dw5, DR5, MB3 homozygous), EBV-Sh (DSh, DRw6.2, MB3 homozygous), and EBV-Ky (DKy, DRw9, MB3 homozygous) cells and were compared with one another by two-dimensional gel electrophoresis. The MB3 light chains from KT2, ER, and EBV-Ky cells were clearly different in terms of their isoelectric points, whereas those from ER, JMe, and EBV-Sh cells were indistinguishable. No differences in charge or m.w. were noted for the MB3 heavy chains from the five cell lines. Thus, three out of the five MB3-positive, D/DR-disparate cell lines were found to express structurally distinct MB3 molecules, demonstrating that MB3 is a public serologic specificity shared by at least three structurally distinct MB (human I-A-like) molecules. Because the DR light chain subunits isolated from EBV-Wa, KT2, ER, JMe, EBV-Sh, and EBV-Ky cells differed from one another in their isoelectric points, the DR light chains were apparently more polymorphic than the MB3 light chains.

Effects of cimetidine on tumor growth and immune function in nude mice bearing human ovarian carcinoma.

The effects of cimetidine on the growth of a human ovarian cancer cell line inoculated into BALB/c nude mice were examined. The cell line, designated "KK," was derived from a cystadenocarcinoma of the ovary. The passage number was about 40, and its tumorigenicity in nude mice was 100% even when 10(5) cells were inoculated. About 2 weeks after inoculation, the KK cells formed palpable tumors, and the tumor volume reached 2.29 cm3 on day 36. Conversely, the nude mice given cimetidine (100 mg/kg/day) orally with drinking water had about one-third the tumor volume (0.81 cm3) of that in untreated nude mice on day 36. The natural killer activity against the YAC-1 (a T-cell lymphoma) cell line in spleen cells of the nude mice challenged with human xenogeneic tumor (KK cell line) was not affected by treatment with cimetidine while inhibiting the tumor growth. The capacity to lyse the KK cells did not exist in the spleen cells of nude mice challenged with the KK cell xenograft and not treated with cimetidine. The cimetidine-treated spleen cells acquired the capacity to lyse the KK cells on day 14. Thereafter, the capacity was maintained at the same level as long as cimetidine was administered, whereas that in untreated nude mice remained undetectable.

BCGFII activity on activated B cells of a purified murine T cell-replacing factor (TRF) from a T cell hybridoma (B151K12).

Experiments were performed to examine a growth-promoting activity on B cells or B leukemic cells of T cell-replacing factor (TRF) produced by a murine T cell hybridoma (B151K12) which constitutively produces TRF. The cellfree supernatant (CFS) from B151K12 cells (B151-CFS) could induce terminal differentiation of pre-activated B cells or in vivo passaged chronic B leukemia cells, BCL1, into immunoglobulin-secreting cells, while it did not exert a nominal lymphokine activity such as BCGFI (now known as BSFpl), IL 2, or gamma-interferon. However, it promoted [3H]thymidine uptake of dextran sulfate (DXS)-stimulated normal B cells and in vivo passaged BCL1 cells, suggesting that it also has BCGFII activity. We tried extensively to purify and to separate the TRF active molecule from the BCGFII active molecule by using many types of purification procedures. The purification scheme consisted of ammonium sulfate precipitation, DEAE-cellulose chromatography, Blue-Sepharose chromatography, hydroxylapatite chromatography, and gel permeation with fast protein liquid chromatography (FPLC). It was revealed that the BCGFII active molecule was hardly separable from the TRF during the entire purification procedure. The TRF as well as BCGFII active materials were glycoprotein with an apparent m.w. of 50 to 60 Kd on gel permeation chromatography and 18 Kd on SDS-PAGE under reducing conditions. The BCGFII active materials were hardly separable from the TRF active one, even after a reverse-phase FPLC, in which both BCGFII and TRF activities were recovered in the fractions eluted at 44 to 48% acetonitrile in 0.1% trifluoroacetic acid (TFA). Furthermore, the absorption of TRF and BCGFII active materials by using BCL1 cells removed not only TRF but also BCGFII activity. Moreover, B cell-specific monoclonal antibody (9T1), which can preferentially block TRF-dependent plaque-forming cell responses, also inhibited the expression of BCGFII activity to BCL1 cells. Taking all of the results together, we conclude that the TRF from B151K12 cells promotes growth of appropriately activated, such as DXS-stimulated normal cells and BCL1 tumor cells. These results suggest that B151-TRF may act on B cells as B cell growth and differentiation factors.

[A rare case of subarachnoideal pneumocephalus after ear douche therapy].

No detailed reports on pneumocephalus caused by any factors other than head trauma, and their courses accompanied with this disease have been so far available. We recently experienced a case of pneumocephalus complicated with severe clouding of consciousness, ocular deviation, and unilateral spatial neglect with the results being reported hereinafter. A 54-year-old man had often received ear douche therapy due to chronic sinusitis and tubal obstruction about 3 years before without any history of head trauma. On Jan. 6, 1984, sudden clouding of consciousness accompanying stiffness of the left arm occurred immediately after ear douche, and then he was transferred to our center. At admission, semicomatose, bilateral ocular deviation to the right, and left hemiparesis were observed. Plain skull X-ray films showed a retention of air in the frontal and temporal regions, while CT scan revealed air retention on the bilateral frontal region, bilateral temporal tip and suprasellar cistern. However, no abnormal findings were detected in the brain. Consciousness and hemiparesis recovered on the next day of hospitalization, however, the left hemispatial neglect still remained. This symptom was still observed on the 3rd day but disappeared by the 4th day of hospitalization. For clarifying its cause, cerebral angiography, CT scan and electroencephalography were then performed. CT scan revealed no anomalies in the brain, while cerebral angiography showed a cerebral circulation pattern in favor of the right internal carotid artery.(ABSTRACT TRUNCATED AT 250 WORDS)

[A case favorably progressed after operation for hypertensive intracerebral hemorrhage showed extravasation on angiogram occurred in chronic alcoholism].

It is said that the prognosis is generally unfavorable in patients with hypertensive intracerebral hemorrhage showed extravasation on angiogram. Recently, we experienced a case whose prognosis was eventful after the removal of hematoma. So we reported this case and discussed between alcoholism and intracerebral hemorrhage accompanied with extravasation on angiogram in this paper. A 59-year-old male was transferred to our emergency center because of right hemiplegia and mild clouding of consciousness at around nine in the evening on December 12, 1983. At the time of admission, his neurological state was classified into grade II, exhibiting the right putamenal hemorrhage and pyramidal destruction type of hematoma by CT scan with 58 ml of hematoma volume. The right carotid angiography was immediately performed and confirmed the hematoma being of lateral type. At that time, extravasation proximal to the lateral lenticulostriate artery was noted. Repeated CT scan revealed the enlarged hematoma (105 ml) accompanying with ventricular hemorrhage. At the completion of these examinations, the neurological grade was III. The hematoma was surgically removed after 4 hours following the onset of cerebral hemorrhage. His postoperative course was very favorable. Although acute hydrocephalus appeared later on, it was cured by ventricular drainage. The patient become possible to walk with a helper by 1 month after operation. CT scan obtained 1 month after operation revealed a remarkable brain atrophy, which was probably derived from chronic alcoholism.(ABSTRACT TRUNCATED AT 250 WORDS)

[Effect of treatment with synthetic eel-calcitonin derivative in patients suffering pain due to bone metastases].

Synthetic Calcitonin Derivative obtained from eel (eel-calcitonin) was given to 15 patients suffering from pain due to bone metastases of malignant tumors. The analgesic effect of synthetic eel-calcitonin derivative was evaluated through pain and narcotic score with the Ridit method. With regard to pain score, significant analgesic effect was observed in the first week (p less than 0.001), and after a month there was also a decrease in the narcotic score (p less than 0.05). However, changes in serum Ca, P, ALP values, and the findings of bone scintigrams and roentgenography, rarely occurred following the treatment. The toxicities of eel-calcitonin which were observed in this series were as follows; nausea and vomiting (33%), flush (11%), eruption (6%). These findings suggest that synthetic eel-calcitonin derivative is a very useful agent for pain due to bone metastases.

Establishment and characterization of a new human cultured cell line from a prolactin-secreting pituitary adenoma.

A new human cell line was established from a prolactin (PRL) secreting pituitary adenoma. This cell line, designated as HPA, initially produced and secreted PRL, but the ability was decreased with increasing passage number. After about 30 passages in vitro, these cells had a short doubling time (14 h) and a low plating efficiency (9%). When a minimum of 10(5) cells was injected per mouse, virtually all athymic nude mice developed a slow growing, nonmetastasizing tumor at the injection site about 30 days after injection. PRL production by the HPA cells after Day 150 was demonstrated by immunocytochemistry as well as radioimmunoassay. In addition, cimetidine (10(-4) M) had a significant stimulatory effect on PRL secretion by 4-day-cultured HPA cells. When the HPA cells were incubated in the presence of 5.0 and 10.0 nM bromocriptine, the proliferation rate was inhibited to 53.4 and 25.1% of untreated controls, respectively. On the other hand, the same concentrations of bromocriptine did not affect the proliferation rate of YK cells derived from human immature teratoma of the ovary. In addition, bromocriptine inhibited significantly the growth rate of xenotransplanted HPA but not YK cells. These results suggest that bromocriptine inhibits specifically the proliferation of HPA cells.

[A case of convexity meningioma associated with chronic subdural hematoma].

There are 5 reported cases of meningioma complicated by chronic subdural hematoma so far as we reviewed. Considering frequency of occurrence, association of meningioma and chronic subdural hematoma is very rare. Recently, we experienced with a case of convexity menigioma complicated by chronic subdural hematoma. The patient was a 61-year-old woman who complained of headache and numbness of the left hand. She received hysteromyomectomy 28 years before but had no past history of trauma of the head. She suffered from frequent profound pain in both orbits for one year, and she consulted a neighboring practitioner due to occurrence of numbness of the left hand and malaise. Then, CT scan revealed abnormalities, so that she was referred to our center. On admission, neurological oxamination revealed no abnormalities but plain X-ray film of the skull showed a round destruction in the right parieral bone. CT scan showed a round high density area of 3 cm in diameter in the right parietal region, this is markedly enhanced with contrast media. There is a crescent-shaped low density area extending forward from the high density area. The body of the right lateral ventricle is totally collapsed. Selective arteriography of the right external carotid revealed tumor stain in the parietal region. Therefore, under the diagnosis of association of chronic subdural hematoma and convexity meningioma, craniotomy of the right frontal, parietal and temporal regions was carried out and a meningioma of 10 g, and a subdural hematoma with its capsule touching the maningioma were entirely extirpated.(ABSTRACT TRUNCATED AT 250 WORDS)
