Expression of inhibin alpha-subunit gene during mouse gametogenesis.

Mammalian gametogenesis is regulated through complex interactions between germ and somatic cells. To investigate the mechanism underlying the differentiation of functional gametes, some genes specifically expressed during gametogenesis have been isolated and characterized. In a search for further examples of such genes, we have isolated from a newborn mouse testis cDNA library, a clone corresponding to mouse inhibin alpha-subunit. Although it is known that the inhibin alpha-subunit molecule is abundantly produced in ovarian follicle and in testicular Sertoli cells, the spatial and temporal patterns of expression of this gene remain to be elucidated. In this study, the patterns of expression of inhibin alpha-subunit mRNA during mouse gametogenesis were examined by RNA blot, cytoplasmic dot and in situ hybridization techniques. In the testis, the concentration of inhibin alpha-subunit mRNA increased from about 16 dpc (days post coitum), peaked at birth and then gradually decreased, paralleling testicular development. Inhibin alpha-subunit mRNA was localized in Sertoli cells of wild type as well as W/Wv testes. In adult testis, mRNA was restricted to the perinuclear cytoplasm of Sertoli cells. Inhibin alpha-subunit mRNA was expressed in follicle cells of adult ovary more abundantly than in adult testis. Analysis of expression during folliculogenesis showed that the accumulation of this mRNA began in preantrum follicles and the level of expression reached a maximum in Graafian follicles.

Induction of tumoricidal activated macrophages by a liposome-encapsulated glycolipid, trehalose 2,3,6'-trimycolate from Gordona aurantiaca.

A mycolic acid-containing glycolipid, trehalose 2,3,6'-trimycolate, prepared from a non-pathogenic acid-fast bacterium Gordona aurantiaca, was shown to induce strong tumoricidal activity in peritoneal exudate cells by intravenous or intraperitoneal injection of liposome-encapsulated preparations. The mycolic acid derivative containing a high proportion of unsaturated fatty acids rendered macrophages cytotoxic against syngeneic mastocytoma cells in the absence of endotoxin, for over 14 days after the injection. The macrophages were ascertained to be at low intracellular levels of a lysosomal enzyme beta-galactosidase and an ectoenzyme alkaline phosphodiesterase, a specific pattern as previously described for "primed macrophages". However the culture supernatants of the peritoneal exudate cells were not cytotoxic.

Inhibitory effect of glutamic acid and aspartic acid on calcium oxalate crystal formation.

The effects of Glu and Asp on calcium stone formation was evaluated in three experiments. Studies using mixed suspension, mixed product removal crystallization and scanning electron microscopy showed that Glu and Asp inhibited the nucleation rate, growth rate and suspension density (crystal mass produced) in proportion to the concentration. The main amino acids in calcium oxalate stones and calcium phosphate stones were Glu and Asp. However, the main amino acids in uric acid stones were glycine and urea, and there were no specific amino acids in struvite stones. The activity of urinary GOT and GPT, which convert Asp and alanine, respectively, to Glu in normal subjects was significantly greater than in calcium stone formation.

Messenger RNA levels of guanine nucleotide-binding proteins are reduced in the ventricle of cardiomyopathic hamsters.

The expression of guanine nucleotide-binding protein (G-protein) genes (Gs alpha, Go alpha, Gi alpha 1, Gi alpha 2, and Gi alpha 3) was examined in the ventricle of cardiomyopathic Syrian hamsters of the Bio14.6 strain (10-35 weeks old). Northern blot analysis of total cellular RNA revealed that all G-protein genes except Gi alpha 1 were expressed in the ventricle of Syrian hamsters. Gs alpha and Gi alpha 2 genes were abundantly expressed. The expression levels of the Gs alpha and Gi alpha 2 messenger RNAs in Bio14.6 ventricles were lower than the levels in ventricles of the F1B hamster strain; the abundance of Go alpha and Gi alpha 3 messenger RNAs did not change markedly. Moreover, the messenger RNA levels of Gs alpha and Gi alpha 2 decreased as the stage of cardiomyopathy progressed. Since G-proteins are linked to adenylate cyclase, these alterations of G-protein messenger RNA levels may be related to reduced contractility of cardiomyopathic heart.

Molecular mechanism of cardiac hypertrophy.

Pressure overload induces cardiac hypertrophy and reexpression of contractile protein isogenes. To ascertain the molecular mechanism of these events, we examined the expression of cellular oncogenes and the early change in the translational activity of specific cardiac mRNA by two-dimensional gel electrophoresis of in vitro translational products. Pressure overload increased the expression levels of c-fos, c-myc, and c-Ha-ras genes. The relative predominance of 8 species out of over 400 translational products was increased by pressure overload while that of 2 translational products was decreased. We cloned four pressure-overload-responsive cDNA clones by differential dot blot hybridization. The expression pattern of each cDNA clone in the pressure-overloaded hearts was similar to that in fetal hearts. To examine whether mechanical stimuli directly induce specific gene expression in the heart, we cultured rat neonatal cardiocytes in elastic silicone dishes and stretched these adherent cells. Myocytes stretching stimulated amino acid uptake and expression of the c-fos gene, which was blocked by protein kinase C inhibitors. These results suggest that there are some early responsive genes in cardiac hypertrophy and that mechanical loading directly stimulates gene expression possibly via protein kinase C activation.

[Gamma-ray spectra of 201Tl-radiopharmaceuticals with a scintillation camera--crosstalk of contaminating nuclides of 200Tl and/or 202Tl onto 201Tl-photopeaks].

In cardiac imaging with 201Tl, the collimator for low energy high resolution is generally used, and also the energy window, which is set on the spectral display of a pulse height analyzer of a scintillation camera, is chosen 70 +/- 12.5 keV. The purpose of this study is to discuss those conditions in 201Tl imaging with the scintillation camera. Two types of collimators for HR (high resolution) and ME (medium energy) were used in this experiment, and we measured the pulse height spectra of 201TlCl radiopharmaceuticals in air and in a cuboid phantom, connecting a multi-channel pulse height analyzer to the scintillation camera. As a result of measuring of the pulse height spectra, two different energies of gamma rays which are not supposed to emit from 201Tl nuclide were observed, and we also identified the presence of a small amount of 202Tl (with 439 keV) and/or 200Tl (with 368 keV) from their half-life measurements. Thus, the use of the HR-collimator with 201Tl imaging is not suitable, because the shielding effects of its septa is poor to 439 keV gamma-rays, and the scattered radiation produced by the Compton interaction contributes to the principal photopeak on the pulse height spectrum. Here, we recommend the use ME-collimator instead of the HR-one, and of the window width of 76 +/- 25 keV for increasing the count rate.

[A study of the auditory responses to intra-uterine sound in the first few months after birth].

The auditory responses of infants until 3 months after birth were examined using the doll "Nenkororin" which contains a tape recorder of the intrauterine sound (IUS). The purpose of this study was to assess the applicability of these responses to screening of infants with severe hearing impairment. The results are summarized as follows. 1. The IUS with 1/3-octaval audiofrequency of 400Hz-4kHz was presented to 152 neonates. Positive responses showing alert inactivity were obtained in 148 infants, but negative responses in 2 neonates. 2 infants were undetermined. The neonates showed negative responses had severe hearing impairment with comparison of ABR. ABR was also carried out in 69 of the 152 infants. The results of ABR well agreed with those of the IUS test. 2. A typical reaction of neonates who stopped crying and assumed a state of alert inactivity in responses to the IUS was demonstrated until approximately one month after birth. At the end of the first month after birth, the threshold of this reaction grew smaller than that of the one-week period after birth. The latent time of this reaction also tended to become shorter. 3. The infants mostly failed to respond to the 1/3-octaval noise consisting of 0.5, 1.0, 2.0, 4.0kHz extracted from the IUS. From the experiment using high and low pass noise components extracted from the IUS. High response were obtained at low frequency band components below 1kHz. 4. Public health nurses without experience of auditory test were requested to compare the reaction by the IUS test and an auditory reflex test using tongue-clicking, whistling and paper-crumpling sounds. The IUS test was found to be easier for evaluation of the reactions than the other method of test. From the above results it is concluded that the auditory response to the IUS was useful for screening of severe hearing impairment in neonates until one after birth.

[Clinical studies on thirty-two cases of childhood urolithiasis].

We evaluated 32 patients with urinary calculi under 16 years of age over the past 14 years since the founding of the Department of Urology, Kinki University School of Medicine. They comprised 0.8% of the total urolithiasis patients. They consisted of 18 boys and 14 girls with a male-to-female ratio of 1.29. The average age was 8.7 years for boys and 10.4 years for girls without any marked peak. The most frequently chief complaint was hematuria which was present in 15 cases (47%). Although pyuria was seen in seven cases (22%), urinary bacterial culture was positive only in 5. The underlying diseases could be diagnosed in 13 cases (41%), of which eight cases (62%) developed metabolic disorder. The sites of calculi were determined in 29 cases (91%), of which 28 had stones in the upper urinary tract. Surgical treatment was performed on 16 cases in 17 sessions. Ureterolithotomy was done in as many as five cases, followed by pyeloplasty in four cases. Nephrectomy was performed in only one case. There was only one case which had been treated with extracorporeal shock-wave lithotripsy (ESWL). The composition of calculi was found in 21 cases (66%), the majority or 13 cases (57%) of which had calcium-containing stones. Nevertheless, there was no case of hypercalciuria. When compared to the previous reports in Japan, it was worthy of note that calculi in the upper urinary tract and calcium-containing stones had higher incidences. It is expected that more patients will be treated with ESWL in the future.(ABSTRACT TRUNCATED AT 250 WORDS)

[Calcium urolithiasis and bone change].

Bone mineral contents of calcium urolithiasis patients (105 males and 52 females) were measured by the microdensitometry (MD) method, and the patients were divided into the MD normal group and the MD abnormal group. The patients were also divided into the group (21 males and 3 females) treated with thiazides for 1 year or more and the nontreated group to examine various factors in blood and urine. [Nontreated group] The rate of MD abnormality was higher in younger males. The rate tended to increase with age in females. Alkaline phosphatase values were significantly higher in MD abnormal group males than in MD normal group males. Urinary calcium excretion and PTH values were significantly higher in MD abnormal group females than in MD normal group females. Comparison of hypercalciuria and normocalciuria revealed no significant difference between the MD normal rate and the MD abnormal rate. Comparison of single of stone formers and recurrent stone formers also revealed no significant difference between the MD normal rate and the MD abnormal rate. [Treated group] PTH and alkaline phosphatase values were significantly higher in the treated group than in the nontreated group. Alkaline phosphatase values were significantly higher in the MD abnormal group. From the viewpoint of stone recurrence prevention, the monitoring of bones where the majority of calcium in the body is present is considered important besides behavior of calcium in blood and urine.

Ceramic interspinous block (CISB) assisted anterior interbody fusion.

Ceramic interspinous block (CISB) assisted anterior fusion system was developed. The system is recommended for patients with symptomatic disc degeneration and destructive lesions of the lumbar spine. One hundred and six patients who underwent CISB-assisted anterior fusion by one doctor (H.T.) from 1981 to 1987 were followed-up 2-6 years after the operation. A comparative study of the results between the CISB-assisted group and the CISB-unassisted anterior fusion group, performed by the same doctor during 1979-1981 for deranged disc, was also done. Satisfactory relief of back and leg pains, marked ADL recovery and early bone union were obtained in all of the CISB-assisted anterior fusion cases. The union rate was 98.9% in the CISB-assisted anterior fusion group, whereas in the CISB-unassisted group, it averaged 79%. The efficacy of CISB for anterior interbody fusion was also verified using a computer simulation. CISB implementation appears to create equalization of stress distribution on the graft-vertebra interface.

Effects of transforming growth factor-beta 1 on murine clonal osteoblastic cells (MC3T3-E1).

Transforming growth factor (TGF)-beta family is considered to be an important local factor that greatly regulates bone metabolism. However, the effects of this polypeptide on osteoblasts have been divergent under various experimental conditions. Moreover, three forms of TGF-beta have been recently described. Therefore, we reexamined the effects of TGF-beta 1 on clonal murine osteoblastic MC3T3-E1 cells. TGF-beta 1 dose- and time-dependently depressed alkaline phosphatase activity in the cells supported by low concentration of serum. On the contrary, in the same range of concentrations, TGF-beta 1 stimulated DNA synthesis in the cells. These effects of TGF-beta 1 were observed in the cells cultured in the media without or with a high concentration of serum. These effects of TGF-beta 1 are not mediated by endogenous production of prostaglandin, since the basal level of prostaglandin E2 was very low and rather inhibited by TGF-beta 1; and, further, indomethacin did not modify the effects of TGF-beta 1 on the cells under the present conditions. Furthermore, TGF-beta 1 greatly stimulated not only type I but also type III collagen production. Hydroxyurea completely blocked the stimulation of DNA synthesis by TGF-beta 1, but partially inhibited the collagen synthesis, suggesting that the TGF-beta 1-stimulated collagen synthesis is at least in part linked to the proliferation. However, the stimulation of collagen synthesis by TGF-beta 1 was greater than that of DNA synthesis, and further, the amount of hydroxyproline in the cell was evidently augmented by TGF-beta 1. Our data presented here thus suggest that TGF-beta 1 may act on preosteoblasts to increase the number of osteoblasts and the amount of bone organic matrix.

[A case of metastatic ureteral tumor].

We report a case of metastatic ureteral tumor resulting from gastric cancer in a 56-year-old female. She had undergone distal gastrectomy for gastric cancer in our hospital 3 years earlier, on the histological diagnosis of poorly differentiated adenocarcinoma with absolute curative resection. In March, 1987, she visited our hospital complaining of microscopic hematuria and lumbago. Intravenous pyelography and left retrograde pyelography revealed the stenotic change of the left ureter and hydronephrosis. Endoscopic ureteral biopsy was performed, and the histological diagnosis was an inflammatory change of the ureter. But the hydronephrosis increased, so partial ureterectomy was performed. The histological examination confirmed adenocarcinoma in the left ureter resulting from gastric cancer. From the 340th postoperative day, she complained of general fatigue and vomiting, and gastroscopy revealed recurrent gastric cancer.

Immunoscintigraphy of human tumors transplanted in nude mice with radiolabeled anti-ras p21 monoclonal antibodies.

Anti-ras p21 monoclonal antibody (RASK-3) was used for immunoscintigraphy of human cancer cell lines in nude mice. Iodine-125-labeled RASK-3 was injected into nude mice with either human colon cancers (FCC-1 or BM-314) or lung cancer (KNS-62). Clear images were obtained in all three cancers 7 days after the injection of antibody. No localization of 125I-labeled control monoclonal antibody was observed. The ratio of tissue/blood radioactivity and % ID/g in the tumor were significantly higher than other organs by Day 8. The specific localization index examined by 131I-RASK-3 and 125I-control monoclonal antibody was also higher in the tumor than in other tissues. In the in vitro study, binding of RASK-3 to tumor cells increased significantly by treatment of cells with either lysolecithin or periodate-lysine-paraformaldehyde, which confirmed the intracellular localization of ras p21. The mechanism by which anti-ras p21 antibodies accumulate in tumor sites could be the necrotic changes in tumor cells or changes in membrane permeability of non-necrotic cells. These results provide a strong rationale for the utilization of ras p21 as a target antigen in the imaging of a variety of human cancers.

[Autopsy cases of hereditary ataxia pathologically diagnosed as the Japanese type of Joseph disease--cliniconeuropathological findings].

Two cases (father and son) clinically diagnosed as Menzel type of heredoataxia were examined pathologically, and found nonspecific, multisystem degenerations of central nervous system spreading from brain stem to cerebellum and spinal cord. Common neuropathological findings of both cases were marked neuronal loss, demyelination and cellular and fibrous gliosis in pallidum (esp. internal segment), body of Luys, substantia nigra, dentate nucleus, red nucleus, cranial and spinal motor nuclei, spinocerebellar tracts and Clarke's column. There were no abnormalities in cerebral cortex, caudate nucleus, putamen, thalamus and inferior olive nucleus. These findings had enough similarity to that of Japanese type of Joseph disease. The son who died 36 years old had showed clinically hyperreflexia, cerebellar ataxia, dystonic extrapyramidal movement, muscle atrophy and bulging eyes. Dominant pathological findings compared to the father case were neuronal loss of substantia nigra and red nucleus, fibrous gliosis of superior peduncles and grumose degeneration of dentate nucleus. The father case died 76 years old had showed hyporeflexia, cerebellar ataxia and no bulging eyes clinically. The pathologica findings of pallidum, cranial and spinal motor nuclei, dorsal column and spinocerebellar tracts were more dominant than the other reported case. Dementia, mental retardation, epileptic episodes were not found in this family pedigree. Pathological changes of pallidonigral, pallidoluysian, dentatorubral and spinal cord which were common in Japanese type were less found in original type of Joseph disease in Europe. Authors discussed this difference in relation with progressive pallidumatrophy, dentato-rubro-pallido-luisian atrophy and hereditary spastic ataxia. According to the classification of clinical feature of Joseph disease in Europe, the father case was classified as type III (Machado phenotype) and the son case was as type I (Joseph phenotype). Reported cases of type III and familial autopsy cases of Joseph disease were so rare in Japan, authors reported on their clinicopathological findings.

[A clinical study of 76 cases in carcinoma of the colon and rectum with metastasis to the peri-aortic lymph nodes. With particular reference to the skip metastasis in carcinoma of the sigmoid colon to the peri-aortic lymph nodes].

The therapeutic result by tumorous site were evaluated in 76 cases of carcinoma of the colon and rectum with the confirmed presence of metastasis to the peri-aortic lymph nodes. The results revealed that significant good out-comes were noted in the relatively non-curative cases of carcinoma of the sigmoid colon, with particularly good prognosis being found in six cases with skip metastasis. There was a 26.1% (6/23) incidence of the skip metastasis in carcinoma of the sigmoid colon with the peri-aortic lymph nodes metastasis. We confirmed two lymphatic routes of the skip metastasis; one route was from the lymph nodes near the tumor to the para-aortic lymph nodes directly, and the other was from the lymph nodes near the tumor to the lymph nodes at the aortic diffract directly and finally to the para-aortic lymph nodes. In the all six cases, the peri-aortic lymph nodes metastasis occurred only in inferior site of the inferior mesenteric artery. The average number of the peri-aortic lymph nodes metastasis were as few as 1.7 in these cases. The prognosis was favorable with three of the six cases scoring a three-year survival and included a five-year survival case.

[Transplantation of gynecologic tumor in nude mice].

Seventy-four cases of gynecologic tumor; 29 of the endometrium, 20 of the ovary, 9 of the trophoblastic, 7 of the cervix, 2 of the fallopian tube and one of the vulva, and 6 of malignant ascites, were transplanted into nude mice. 1. Overall success rate at the trial of primary transplantation revealed 35.1% or 26 cases out of 74. Trophoblastic neoplasia appeared the highest in the rate among each tumors, showing 55.5% of 9 trials. 2. Out of 26 successful cases of the primary trials, 14 or 53.8% are serially transplantable. These consisted of 7 endometrial, 2 ovarian, 3 choriocarcinoma and 2 cervical carcinomas. 3. The serially transplantable tumors are reproducible of the morphology of each original tumor. 4. The doubling times of the tumor size in choriocarcinoma are evenly short although life span of the tumor-bearing mice varies from each other. Endometrial and ovarian carcinomas spend the time longer than choriocarcinoma. The doubling times differ from each case of endometrial carcinoma with the same histological grade. 5. Choriocarcinoma lines preserve hCG secreting ability. Tumor size of the choriocarcinoma originated from the kidney correlates well with its serum hCG titer. These lines of gynecologic malignancy serially transplantable in nude mice will be useful for further cancer research.

Amino acids in urine and plasma of urolithiasis patients.

Urine and plasma amino acids in healthy males and patients with calcium stones, were analyzed for thirty-four amino acids. There was no difference in urinary excretion of thirty-two amino acids between the two groups. Plasma values of taurine, aspartic acid, hydroxyproline, glutamic acid, glycine, alanine, cystathionine, ornithine and lysine were significantly higher in controls than in stone formers. Of these amino acids, aspartic acid, glutamic acid, glycine, alanine and lysine are the main amino acids of matrix protein in renal stones. The levels of aspartic acid, glutamic acid, glycine, alanine and lysine in the serum of normal individuals were higher than those in plasma. On the basis of the findings, these amino acids may form soluble complexes with calcium and heparin in stone formers. It is also possible that some amino acids act as promoters or inhibitors in the pathogenesis of calcium oxalate stones.

Magnesium-to-calcium ratio in tap water, and its relationship to geological features and the incidence of calcium-containing urinary stones.

We examined the relationship among magnesium and calcium content in tap water, the geological features and urinary stone incidence in Japan. The magnesium-to-calcium ratio in tap water correlated negatively with the incidence of urolithiasis. There was no correlation between calcium and magnesium concentration in tap water and urinary stone incidence. Geological features in Japan were classified into 5 groups. The magnesium-to-calcium ratio in the basalt areas was higher than in the other areas, while ratio in the granite areas was low. In the sedimentary rock areas calcium and magnesium concentrations were high; the magnesium-to-calcium ratio in these areas was between those of the basalt and granite areas. The limestone areas had a much higher calcium concentration. The incidence of urinary stones in the sedimentary rock and basalt areas was lower than that of the granite areas, while that in the limestone areas was the highest. Thus, the incidence of urinary stone is related to the magnesium-to-calcium ratio in tap water and the geological area.
