Relaxation of depolarized guinea pig taenia caecum induced by some antispasmodics.

Taenia isolated from the guinea pig caecum were used for the experiments. No inhibitory response of the depolarized taenia to isoprenaline was observed during the significant increase of cyclic AMP level. These observations suggest that the total tissue level of cyclic AMP is not an important determinant of relaxation in the depolarized taenia. Antispasmodics, such as papaverine, benactyzine and Aspaminol (1,1-diphenyl-3-piperidino-butanol hydrochloride) relaxed the depolarized taenia, while the depolarized taenia was not relaxed by concentrations of dibutyryl cyclic AMP sufficient to relax the polarized taenia. Ca uptake by the depolarized taenia was inhibited by papaverine, benactyzine and Aspaminol but not by isoprenaline and dibutyryl cyclic AMP. These results indicate that relaxation of the depolarized taenia induced by the antispasmodics used was mainly due to inhibition of Ca uptake.

[Changes in the response to drugs of the rat stomach after a chronic vagotomy and a sympathectomy (author's transl)].

Effects of cholinergic agonists and serotonin were examined on the chronically denervated stomach fundus. We developed a new method of adrenergic denervation (sympathectomy), which was confirmed chemically, histochemically and functionally. Vagotomy was performed by the usual method. The sensitivity of the longitudinal muscle of the fundus to cholinergic agonists and serotonin was not modified by chronic sympathectomy or vagotomy when determined from the ED50 of the dose-response curve. The maximum contractile response of vagotomized fundus to each cholinergic agonist and to serotonin was significantly decreased, whereas only a slight decrease was observed in sympathectomized fundus. Ca2+-contracture in Ca2+-free isotonic K+ Ringer's solution was markedly decreased in chronically vagotomized or sympathectomized fundus. Efflux of 45Ca in Ca2+-free isotonic K+ Ringer's solution was significantly lowered by vagotomy but only slightly by sympathectomy. No significant change in ATP content was observed in the vagotomized or sympathectomized fundus.

Influence of surgical sympathectomy on gastric secretion and ulceration in rats.

The influences of sympathectomy on gastrointestinal mucosa, gastric secretion, acute or chronic gastric ulcers were studied in rats. Uncer ether anesthesia, sympathectomy was performed by surgical removal of the celiac ganglion. Surgical sympathectomy per se produced no pathological changes in the gastrointestinal tract as determined by macroscopical observation 3, 10 or 20 days after operation. The volume of gastric juice and pepsin output were not influenced by the sympathectomy but gastric acid output was significantly increased in pylorus-ligated rats. The sympathectomy worsened the stress- and the indomethacin-induced ulcer and delayed the healing of chronic gastric ulcers a little but not significantly, and had no deteriorative influence on the reserpine-induced ulcers. In contrast, Shay ulcers, aspirin- or secrotonin-induced ulcers were significantly aggravated by sympathectomy. The loss of H+ ions and gain of Na+ ions in the gastric juice of pylorus-ligated and aspirin-treated rats were not affected by sympathectomy.

Regional differences in the effects of denervation, cocaine and chronic reserpine administration on the responses of the rat vas deferens to norepinephrine and acetylcholine.

The prostatic half and epididymal half of the rat vas deferens was found to show the difference with respect to both the sensitivity (expressed as the geometric mean ED50) and maximal response to norepinephrine and acetylcholine. The epididymal half was more sensitive to both drugs than the prostatic half; the maximal response to norepinephrine was greater in the epididymal half than in the prostatic half. Effects of denervation, 10-6 M cocaine and chronic administration of reserpine (1 mg/kg/day for 5 days) on the responses to the drugs were also found to be qualitatively and/or quantitatively different between the two halves. In the epididymal half, all these procedures produced supersensitivity to acetylcholine as well as to norepinephrine. Whereas, in the prostatic half, supersensitivity to only norepinephrine was observed; the responses to acetylcholine were more or less depressed, accompanied by little change in the sensitivity, by these procedures. Degree of supersensitivity (ED50 control/ED50 treated) to norepinephrine produced by denervation was greater in the prostatic half than in the epididymal half. However, in the case of cocaine or reserpine, there was little difference in the degrees of the two halves. In both halves, the maximal response to norepinephrine was increased by denervation and cocaine, but not by reserpine. The magnitude of such increase in the prostatic half was greater and less, after denervation and cocaine, than that in the epididymal half, respectively. Only cocaine increased the maximal response of the epididymal half to acetylcholine. These results are discussed in relation to those previously obtained using whole vas deferens.

Synthesis and adrenergic beta-blocking activity of some 1,3-benzodioxole derivatives.

A series of 1,3-benzodioxole derivatives was synthesized. We found four compounds (2,3,10 and 11 in Table IV) to have about the same order of beta-blocking activity as that of sotalol. In addition, it is of interest that some of the compounds (2-4) were found to have hypotensive activites, although they were about one-tenth of that of hydralazine. Sotalol did not produce any change in blood pressure, and propranolol raised the blood pressure.

Potentiation of antitussive effect of codeine by some 1-dimethoxyphenyl-3-alkylaminobutanols in guinea pigs.

Newly synthesized 1-(2',5'-dimethoxyphenyl)-1-n-butyl-3-diethylaminobutanol (compd. 4) and its analogs enhanced the antitussive effect of codeine and morphine as tested on the cough induced by mechanical stimulation of the trachea in guinea pigs. This effect was illustrated to be a potentiation on the Gaddum's diagram. The following parameters were affected little or to a small extent: 1. analgesic effect of codeine and morphine in guinea pigs and mice, 2. duration of anesthesia induced by hexobarbital in mice, 3. respiratory depression caused by codeine in guinea pigs, and 4. LD50 of codeine in guinea pigs and mice. Explorations of the mechanism of potentiating action suggested some peripheral mechanism, but the exact one remained to be elucidated.
