Identification of a pathogenic isolate-specific 30,000-Mr antigen of Entamoeba histolytica by using a monoclonal antibody.

A monoclonal antibody (MAb) produced against trophozoites of Entamoeba histolytica strain HM-1:IMSS, reacted with all of 42 isolates and 4 clones showing pathogenic zymodeme (Z) patterns, i.e., Z-II, Z-II alpha-, Z-II (glucose phosphate isomerase: gamma +), Z-VII, Z-VII (glucose phosphate isomerase: alpha lack, gamma +), Z-XI, Z-XIV, and Z-XIX, regardless of culture conditions, geographical origins, or host symptoms in an indirect fluorescence antibody test. In contrast, the MAb failed to react with 14 isolates possessing nonpathogenic zymodemes Z-I and Z-VIII and did not react with other enteric protozoan parasites, such as E. histolytica-like Laredo, Entamoeba hartmanni, Entamoeba coli, Endolimax nana, Dientamoeba fragilis, Trichomonas hominis, and Giardia lamblia. Western immunoblotting analysis showed that the molecular weight of the antigenic component recognized by the MAb was exclusively 30,000 in pathogenic isolates of different zymodemes. These results suggest that the 30,000-molecular-weight antigen is a marker of pathogenic isolates and that the indirect fluorescent-antibody test with the MAb is useful for the accurate discrimination of pathogenic amebae.

Amebiasis in institutions for the mentally retarded in Kanagawa Prefecture, Japan.

A parasitologic survey of 620 mentally retarded patients, institutionalized in five different facilities in Kanagawa Prefecture, revealed a high incidence (12.6%) of infection with Entamoeba histolytica. A concomitant serologic survey, by the indirect fluorescent antibody test, gave a much higher incidence (26.5%). Moreover, most zymodeme patterns of the amebae isolated from infected individuals were of a pathogenic type (Zymodeme II). Our findings demonstrate that the mentally retarded in Japan, as in the United States, still are plagued by a high rate of amebic infection.

Effects of berberine, a plant alkaloid, on the growth of anaerobic protozoa in axenic culture.

Extracts obtained by organic solvents from the root of Coptis teeta in Myanmar (Burma), were tested for growth inhibitory activity against Giardia lamblia, Trichomonas vaginalis and Entamoeba histolytica in axenic culture. All extracts had anti-protozoal activity with the methanol extract, in particular, being effective against all 3 parasites. By chromatographic analysis, the extracts were shown to contain berberine compounds and other alkaloids. Comparing the inhibitory effects of the methanol extract with berberine sulfate, a berberine salt previously shown to possess inhibitory activity, it was observed that the crude extract was more effective than the salt. This suggests that the greater inhibitory activity of the crude extract may be due to the cumulative contributions of its contained berberines and alkaloids. Based on these findings, it appears that the berberine compounds may be useful as chemotherapeutic agents against the 3 parasites tested.

[Risk factors influencing lymph nodes metastasis in lung cancer with stage I, II or IIIA].

The purpose of this study was to assess the risk factors involved in the intrapulmonary, hilar and mediastinal lymph nodes metastases in seventy-eight patients with stage I, II or IIIA lung cancer postoperatively, which were resected from 1978 to 1988. In the histological type, the incidence of the mediastinal lymph nodes metastases in adenocarcinoma was higher than that in other types, such as squamous cell carcinoma and large cel carcinoma. In addition, the incidence of mediastinal lymph nodes metastases in the papillary type was significantly higher than that in the tubular type (p less than 0.05). The incidence of mediastinal lymph nodes metastases increased as invasion into the lymphatic duct and/or vessel was demonstrated (p less than 0.01, p less than 0.05). The proximal type, in which the cancer spread to the secondary segmental bronchus, metastasized to the hilar lymph nodes more frequently than the distal type, in which the cancer was located in the bronchus distal to the third segmental one. Although there was no significant relationship between the site of the cancer and the incidence of the metastatic lymph nodes, the hilar and superior mediastinal lymph nodes (#1-4, 3a, 3p) metastases were demonstrated regardless of the lobe in which the cancer was located. The primary tumor located in the left lower lobe of the lung tended to metastasize to the inferior mediastinal lymph nodes (#8, 9). Twenty-five out of 33 patients with the lymph nodes metastases had hilar metastatic lymph nodes. However, the mediastinal lymph nodes metastases were proved in 5 patients without any intrapulmonary and hilar lymph nodes metastases. No relationship between the histological differentiation, size of tumor, pT factor and the incidence of lymph nodes metastases was found.

[Aortic resection for lung cancer with use of biomedicus centrifugal pump--a case report].

A 56-year-old woman with lung cancer that is invading the descending aorta, a part of the left atrial wall, and the esophagus was treated. The treatment included a pneumonectomy, resections of the descending aorta, resections of part of the left atrial wall and resections of the thoracic esophagus. For the resection of aorta and for its reconstruction, partial left heart bypass from the left atrium to the femoral artery by a Bio pump was used. The Bio pump proved to be simple, safe and effective, and capable of protecting the peripheral viscera from ischemia during the descending aortic cross clamping.

Protection of Toxoplasma gondii-infected mice by stearylamine-bearing liposomes.

The cytotoxic activity of stearylamine-bearing liposomes against Toxoplasma gondii (RH strain) was examined. When tachyzoites were treated in vitro with liposomes consisting of 20 mol% stearylamine and 80 mol% phosphatidylcholine (130 micrograms/ml total lipids), more than 95% of the parasites were killed within 90 min. Intraperitoneal injection of 10 mg of 30 mol% stearylamine/70 mol% phosphatidylcholine-liposomes in mice shortly before or after T. gondii challenge afforded protection from death for more than 30 days to 70-80% of the treated mice, whereas all untreated mice succumbed within 9 days. The liposome-injected mice that survived remained symptom-free and behaved normally.

Introduction and expression of the human insulin gene in adult rat liver.

Liposomes associated with gangliosides and loaded with the human insulin gene were incubated with HVJ (Sendai virus) and red blood cell membranes containing non-histone chromosomal protein, high mobility group 1 (HMG-1), or bovine serum albumin (BSA) to form vesicle complexes. These vesicle complexes were then injected into the portal vein of adult rats. The insulin gene co-introduced with HMG-1 was transported into the nuclei of liver cells much more efficiently than the gene co-introduced with BSA. Moreover, the amount of transcript of the insulin gene co-introduced with HMG-1 was more than 10 times greater than that of the gene co-introduced with BSA. Similar amounts of DNA and RNA of human insulin were found in the liver within 7 days after injection of the gene, and then the amounts of both decreased rapidly. The level of human insulin in rat serum gradually increased to a maximum (20-30 microunits/ml) after 7 or 8 days and then decreased rapidly.

Increased expression of DNA cointroduced with nuclear protein in adult rat liver.

DNA and nuclear proteins were transferred into cells simultaneously at more than 95% efficiency by means of vesicle complexes. The DNA was rapidly transported into the nuclei of cultured cells, and its expression reached a maximum within 6 to 8 hours after its introduction. Moreover, when the plasmid DNA and nuclear protein were cointroduced into nondividing cells in rat liver by injection into the portal veins of adult rats, the plasmid DNA was carried into liver cell nuclei efficiently by nuclear protein. The expression of the DNA in adult rat liver, on introduction of the DNA with nuclear protein, was more than five times as great as with nonnuclear protein.

Secretion of Mr 46,000 protein from human hepatoma cells treated with tumor promotors is regulated by c-myc gene expression.

Previously an Mr 46,000 protein (named p46) was shown to be induced in the culture medium of human hepatoblastoma cells, Huh-6 Cl-5, treated with 12-O-tetradecanoyl phorbol-13-acetate (TPA). For further characterization of p46, Huh-7 Cl-4, another line of well differentiated liver cells, was incubated in the presence and absence of TPA, and proteins were labeled with [35S]methionine, and the proteins secreted into the medium were analyzed by one- and two-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis. A protein of Mr 46,000 was observed in the culture medium of Huh-7 Cl-4 cells treated by TPA or plated at low density, but only slightly in the culture medium of Huh-7 Cl-4 cells plated at high density. As these results suggested that the expression of p46 was regulated by a factor that was activated in the growth state and by TPA treatment, the effect of introduction of the competence gene c-myc into Huh-7 Cl-4 cells was examined. All 37 independent transfected colonies obtained showed enhanced expression of p46. As a control, the c-Ha-ras gene was introduced into Huh-7 Cl-4 cells and shown not to enhance expression of p46. These results strongly suggested that the expression of p46 was regulated by the competence gene c-myc.

Blood LH level and induced ovulation after hCG and PMSG treatment in ovarian quiescent cattle.

Ovarian quiescent cattle bearing follicle with palpable size were treated with single intramuscular injection of 750-6,000 IU of human chorionic gonadotrophin (hCG) in 13 cases and 1,000-2,000 IU of pregnant mare serum gonadotrophin (PMSG) in 5 cases. Changes of blood luteinizing hormone (LH) level, estrus and ovulation after the treatments were examined. After the hCG treatment LH level became slightly high from 0.2-0.6 ng/ml of pre-treatment to 0.3-1.9 ng/ml of post-treatment and maintained the level up to ovulation without the ovulatory LH surge. Ovulation was induced about 36 hr after the treatment in 12 cases. The ovulations were all silent ovulations. After the PMSG treatment LH level became slightly high from 0.6 ng/ml of pre-treatment to 1.3 ng/ml of post-treatment and the level lasted until the ovulatory LH surge. The ovulatory LH surge occurred about 39 hr after the PMSG treatment in 4 cases with a peak of about 32 ng/ml. Ovulation was induced about 74 hr after the treatment in all 5 cases. Four cases showed estrus but one in which the LH surge could not be confirmed did silent estrus preceding the induced ovulations. It was demonstrated that hCG induced ovulation without the LH surge but PMSG induced the ovulatory LH surge and the subsequent ovulation in ovarian quiescent cattle.

An outbreak of amebiasis in an institution for the mentally retarded in Japan.

The results of an epidemiological survey in a 190-patient institution for mentally retarded were reported. Twenty percent of the patients had either cysts or trophozoites of Entamoeba histolytica in their stools, and 38% were positive serologically. The amebic outbreak revealed a sex-independent but age-dependent distribution; younger patients had more serious symptoms in cases invasive amebiasis. A high prevalence of amebic infection was found in the heavily retarded patients, and the positive cases tended to concentrate in certain training classes. Further demographic analysis suggests that the amebic infection was possibly caused by abnormal behavior of heavily retarded patients.

[The effect of pulsatile perfusion on aortocoronary bypass grafting].

To evaluate the effect of pulsatile perfusion on myocardial protection and peripheral circulation, 23 patients undergoing aortocoronary bypass grafting were studied by dividing two groups; one with pulsatile perfusion technique (11 patients) and the other with non-pulsatile (12 patients). The cardiac functional assessments were made on CI, LVSWI, PCWP, CVP at 3, 6, 12 and 24 hours after the operation and the enzyme (CPK, CPK-MB, LDH, GOT) sampling were performed on 1, 2, 4 and 7 days after the operation. The difference between esophageal and rectal temperature, and urine output during cardiopulmonary bypass, indicating the peripheral circulation, were also measured. From these clinical results, it is concluded that pulsatile perfusion method is to be beneficial in myocardial protection and peripheral circulation during operation of aortocoronary bypass grafting.

Serodiagnosis of Chagas' disease using monoclonal antibody against Trypanosoma cruzi-specific Mr 25,000 antigen.

A mouse monoclonal antibody TCF87, prepared previously, was reactive with Trypanosoma cruzi-specific Mr 25,000 antigen regardless of strain. The Mr 25,000 antigen was recognized by all sera from chagasic patients living in different areas of South America, when examined by Western immunoblotting analysis. Although many antigens of T. cruzi epimastigotes were also recognized by sera from patients with leishmaniasis, the Mr 25,000 antigen of T. cruzi did not react with leishmaniasic sera. These results indicate that Mr 25,000 antigen recognized by TCF87 is valuable as a diagnostic antigen for Chagas' disease. When a competition enzyme-linked immunosorbent assay using TCF87 was carried out, all sera from Chagas' disease patients showed positive inhibition. By contrast, all patients with leishmaniasis or other parasitic diseases were scored as seronegative. The present study suggests that competition enzyme-linked immunosorbent assay using monoclonal antibody against the Mr 25,000 antigen of T. cruzi will be useful for serodiagnosis of Chagas' disease in areas where leishmaniasis is co-endemic.

Lipid analysis of Giardia lamblia and its culture medium.

The neutral lipid and phospholipid composition of Giardia lamblia and its culture medium were analysed by a thin-layer chromatography flame-ionized detection system. Both lipid compositions of the parasite differed from that of the culture medium. Sterol was found to be the major neutral lipid, and phosphatidylcholine, phosphatidylethanolamine and sphingomyerin were also present. Fatty acid composition of G. lamblia and its culture medium were also analysed by gas liquid chromatography. Oleic and palmitic acid were the major fatty acids in the total lipid of the organism. The influence of porcine and bovine bile extracts on the lipid composition of G. lamblia was studied. The addition of bile to the medium caused no change in lipid composition of the parasite. Lipid composition of the culture medium was studied both before and following growth of the parasite, and it became evident that consumption of phosphatidylcholine occurred in the growth medium supplemented with bile extract. These results indicate that the presence of bile extract may possibly accelerate consumption by G. lamblia of lipid in the environment.
