Exercise-induced change of nasal resistance in asthmatic children.

Exercise-induced change of nasal resistance and forced expiratory volume in 1 second (FEV1.0) were studied in 30 asthmatic children and seven normal children. Exercise-induced asthma (EIA) was diagnosed in 19 (63%) of the 30 asthmatic patients. Unilateral complete nasal blockage after exercise (exercise-induced nasal obstruction [EINO]) was found in nine (30%) of the 30 asthmatic patients. A marked decrease in nasal resistance took place immediately or 4 minutes after exercise in all cases. EIA is most severe 5 minutes after exercise, and EINO took place 14 or 19 minutes after exercise. The seven normal children had neither EIA nor EINO. The pathophysiologic relationship between EIA and EINO is discussed.

Biochemical and allergenic properties of the house dust mite extract, Dermatophagoides pteronyssinus.

A further study has been made on the house dust mite extract, Dermatophagoides pteronyssinus, with emphasis on gel-filtrated fraction 2 (F2). The crude mite extract showed at least nine discs on polyacrylamide disc electrophoresis and contained less than 0.25% sialic acid and less than 0.5 mM hexosamine and no detectable uronic acid. From gel filtration of the crude extract a No 2 fraction (F2) with allergenic activity showed at least five components on SDS disc electrophoresis covering a molecular weight range of between 15,000 and 70,000. The major allergenic activity of F2 dissolved in pH 7-8 and 4-5 on an isoelectric focusing column. Affinity chromatography and lectins showed that allergenic activity did not relate to structures of N-acetyl-D-glucosamine or N-acetyl-D-galactosamine. Allergenic activity of the crude extract was no affected by peptic digestion and the mite digest prepared by trypsin and pronase showed a similar fractionation and activity profile as crude extract.

Determination of catecholamines by radioenzymatic assay using ion-pair liquid chromatography.

A simple radioenzymatic method for the simultaneous determination of norepinephrine, epinephrine and dopamine in human serum was developed. catecholamines were converted to normetanephrine (NMN), metanephrine (MN), 3-methoxytyramine (3-MT) and their 4-O-methyl isomers by catechol O-methyl transferase with S-adenosylmethionine-(3H-methyl) ([3H]SAM). After addition of unlabelled NMN, MN and 3-MT as carriers, the methyl derivatives were isolated from the reaction mixture by passing through a small boric acid gel column and separated from each other by ion-pair liquid chromatography. Interference by tailing of radioactivity of [3H] SAM and its degradation products eluted earlier was minimized by the use of a precolumn and a branched flow path.

Mite fauna and fungal flora in house dust from homes of asthmatic children.

Mite fauna and fungal flora in the house dust from homes of asthmatic children with positive and negative skin test to house dust allergen and non-asthmatic controls were examined. There was no conspicuous difference in mite species distribution among the three groups. Pyroglyphid mites dominate the mite fauna in house dust more than half of which being Dermatophagoides: D. pteronyssinus and D. farinae. There was no statistically significant difference in numbers between the two species and either species could dominate depending on the conditions of the individual houses. The average number of acarina in 0.5 g of fine dust did not differ statistically among the three groups; however, mite number per square meter floor differed between patients with positive skin test and negative skin test. The results suggest that house-cleaning might influence the possible sensitization of children. The genetic distribution of mould fungi in house dust was largely similar to that of airborne fungi. The average number of fungal colonies detected in 0.5 g of dust did not differ statistically among the three groups. Wallemia with its minute spores may cause sensitization but has so far been insufficiently investigated.

Studies on the physicochemical properties of house dust mite (Dermatophagoides pteronyssinus) allergens involved in reaginic reaction.

The physicochemical nature of the allergenic component of Dermatophagoides pteronyssinus extract was studied by gel filtration, ion exchange chromatography, isoelectrofocusing, and polyacrylamide gel electrophoresis. Fractions were assayed for allergenic activity by the direct radioallergosorbent test (RAST) (dilution method), RAST inhibition (50% inhibition method) and human skin testing. Fractions were further studied by preparative polyacrylamide gel electrophoresis and the band showing a flow rate value of 0.35 was the most potent in allergenic activity. Fractionation by isoelectrofocusing yielded three factions with allergenic activity of pI = 4.8, PI = 7.4 and pI = 10.0 Sephadex G-100 gel filtration revealed that allergenic activity was present in fractions with molecular weight ranging from 17,000-45,000 and discrete ion exchange chromatography evidenced three fractions with very high allergenic activity. In the present study we propose that the allergenic material of D.P is composed of 3 physicochemically different substances whose molecular weight lie in the 17,000 to 45,000 range.

Clinical significance of the house dust mite (Dermatophagoides pteronyssinus) in asthmatic children in Japan.

Clinical studies were conducted to evaluate the role of the house dust mite. Dermatophagoides pteronyssinus, as an allergen in children's asthma in Japan. Skin testing and Prausnitz-Küstner type skin testing showed good correlations between the allergenicity of house dust and the mite. A positive Prausnitz-Küstner type skin reaction to house dust was neutralized by a mite extract. Radioallergosorbent test showed good correlation between house dust and the mite (r=0.687). The mite exhibited in vitro inhibition of radioallergosorbent test to house dust. The inhalation provocation test resulted in an acute respiratory attack in one of the patients. These results suggest that the house dust mite is an important allergen in asthmatic children in Japan.

Liquid chromatographic determination of barbaloin (aloin) in foods.

A simple and rapid liquid chromatographic method is described for the determination of barbaloin (aloin, 10-D-glucopyranosyl-1,8-dihydroxy-3-(hydroxymethyl)-9(10H)-anthraceno ne) in foods. Barbaloin is extracted with water from foods containing aloe and the extract is cleaned up on a disposable cartridge by using methanol-water (55 + 45) as eluant. The eluted barbaloin is separated by liquid chromatography on a YMC A-302 column with methanol-water (50 + 50) mobile phase, and detected at 293 nm. Recoveries of barbaloin added to foods at the levels of 0.05 and 0.50 mg/g were 94.4-100%. Assay results for commercial food samples indicated that the present method is applicable to a variety of foods supplemented with aloe.
