Expression of cloned rpoB gene of Escherichia coli: a genetic system for the isolation of dominant negative mutations and overproduction of defective beta subunit of RNA polymerase.

The rifampin resistance rifD18 allele of rpoB, carried on the expression plasmid pXT7 beta, is controlled by a strong bacteriophage T7 late promoter and two weak Escherichia coli promoters. Depending on the host strain, pXT7 beta specifies different levels of Rifr beta subunit, providing a system for the isolation, maintenance, and overexpression of dominant lethal alleles of rpoB. In rpoB+ hosts, pXT7 beta confers the Rifr phenotype on the Rifs host. Negative rpoB mutations in the plasmid DNA can thus be scored by screening transformants for Rifs. In an rpoB(Am) supD(Ts) host in which chromosomal rpoB expression is decreased as the temperature goes up, some of the negative plasmid-borne rpoB mutations displayed a dominant phenotype. In a host harboring inducible T7 RNA polymerase, the defective beta subunits could be overexpressed independently of the E. coli transcriptional machinery. With this system, we isolated several negative rpoB mutations induced in vitro by hydroxylamine. Seven of the mutant rpoB alleles, when overexpressed, were found to specify normal-size beta polypeptides. Two of them displayed the dominant lethal phenotype in the rpoB(Am) supD(Ts) background. We also constructed a mutation (rpoB1800) in which 24 carboxy-terminal amino acids were substituted with a random 19-amino-acid sequence. The nonfunctional rpoB1800 beta polypeptide was isolated and assembled in vitro into the core enzyme molecule.

Ventricular tachyarrhythmias during cesarean section after ritodrine therapy: interaction with anesthetics.

This case illustrates that patients receiving ritodrine for preterm labor may risk interactions between the residual betamimetic effects of ritodrine and the effects of anesthetics during cesarean section. Such interactions may result in serious cardiovascular complications even after cessation of an infusion of ritodrine. Preoperative assessment should focus on cardiovascular status and serum potassium level. Delaying induction of anesthesia should be considered whenever possible. Careful fluid administration and cautious use of titrated doses of ephedrine are advised. After delivery of the infant, there should be no contraindication to the use of an alpha-adrenergic vasopressor such as phenylephrine to treat hypotensive patients with tachycardia.

Effects of quipazine on blood pressure in pithed rats.

The effects of quipazine on blood pressure in pithed rats and the mechanism of the pressor response were studied. Quipazine and 5 HT produced an abrupt increase in blood pressure. The pressor responses were reduced by phentolamine and phenoxybenzamine. The noradrenaline depletion induced by pretreatment with reserpine significantly reduced the pressor effects of quipazine. Methysergide, at a dose which did not alter pressor responses to noradrenaline, reduced the increase in blood pressure induced by quipazine and 5 HT. These results suggest that quipazine activates 5-hydroxytryptaminergic receptors stimulating the release of noradrenaline. The effect observed in reserpinized rats can be explained by the interconnection of 5-hydroxytryptamine receptors and alpha-adrenoceptors.

Novel beneficial effects of defibrotide, an endothelium protecting agent, following ischemia and reperfusion in the isolated perfused rat heart.

Infiltrating polymorphonuclear leukocytes (PMNs) have been implicated as key mediators of ischemia/reperfusion injury of the heart. These toxic effects are due to PMN and endothelial cell interactions. This microvascular dysfunction results in an impairment of the coronary circulation which enhances myocardial damage. The effect of defibrotide was examined in a neutrophil dependent isolated perfused rat heart model of ischemia (I) (20 min) and reperfusion (R) (45 min). Administration of defibrotide (200 micrograms/kg) to I/R hearts in the presence of PMNs preserved coronary flow and protected against cardiac contractile dysfunction (p < 0.001) in comparison to those I/R hearts perfused with PMNs but receiving only vehicle. Defibrotide also significantly decreased PMN accumulation in the ischemic myocardium as evidenced by an attenuation in myeloperoxidase activity (p < 0.001). Defibrotide exerted a significant cardioprotection in PMN mediated I/R injury of rat heart. The mechanism appears to be related to inhibition of PMN-endothelium interaction and eventual PMN infiltration into the ischemic myocardium.
