Analysis of the biological properties and use of comparative genomic hybridization to locate chromosomal aberrations in the human testicular seminoma cell line JKT-1 and its highly metastatic cell line JKT-HM.

OBJECTIVE: To examine the biological properties, and utility of comparative genomic hybridization (CGH) to locate chromosomal aberrations, in a new human testicular seminoma cell line, JKT-1, and a highly metastatic cell line, JKT-HM, developed as an animal model of spontaneous metastasis. MATERIALS AND METHODS: JKT-HM was isolated by transplanting cells of JKT-1 into the dorsal skin of nude mice, using methods developed previously. The biological properties of JKT-1 and JKT-HM cells were examined using assays of cell proliferation and in vitro tumour cell invasion, and the DNA index (by flow cytometry). CGH was used to analyse chromosomal aberrations and to detect chromosomal regions causing metastasis in the testicular seminoma cell lines. RESULTS: The JKT-1 and JKT-HM cells showed no difference in morphology or cell proliferation. After transplanting JKT-HM into mice, the cells metastasized to the lung and lymph nodes in all five mice by 50 days. The in vitro tumour cell invasion and animal assays suggested potential invasion and metastasis of JKT-HM. The DNA index was 1.48 for JKT-1 and 1.50 for JKT-HM. CGH analysis revealed various chromosomal aberrations undetected by examining their karyotypes, e.g. loss of 18p, Yq and gain of Xq, and the technique has potential to detect genetic aberrations related to metastasis in this model. CONCLUSIONS: The two cell lines JKT-1 and JKT-HM, and the metastatic animal model used, were useful in studying human testicular seminoma and the metastatic behaviour of cancer. In addition, CGH was useful for analysing the chromosomes of two different cell lines derived from the same parent line.

Characterization of antithrombotic activity of lumbrokinase-immobilized polyurethane valves in the total artificial heart.

Thirty ng/mm2 lumbrokinase, a potent fibrinolytic enzyme, was immobilized in a Korean type total artificial heart (KORTAH) valve by photoreaction; polyallylamine was used as a photoreactive linker. Lumbrokinase-immobilized polyurethane valves were then fitted to the total artificial hearts of 3 healthy 50 kg lambs. In the control lamb, the valves were untreated; in one other, only valves on the right were treated; and in the remaining animal, only those on the left. Implants were in place for up to 3 days, and cardiac output was 5 L/min. To facilitate thrombus formation, low doses of heparin were administered. In the control lamb, thrombi was observed only in the inlet parts of the valves. In the other 2 experiments, thrombi formed in untreated control valves but not in lumbrokinase treated valves. The grade of thrombus formation in untreated valves was 1.06+/-1.37 versus 0+/-0 in the treated part by one-sided Student's t-test (p < 0.1). After implantation, fibrinolytic activity was only observed in treated valves by fibrin plate methods. The proteolytic activity of the treated valves was 3 times higher than that of untreated valves using the azocasein method. These data show that lumbrokinase treated polyurethane valves lead to decreased thrombus formation in vivo and that their biocompatibility is therefore greater than that of untreated valves.

Polymorphism of heat-shock protein gene HSP70-2 in Crohn disease: possible genetic marker for two forms of Crohn disease.

BACKGROUND: Previous reports have shown that heat-shock protein 70 (HSP70) may be associated with Crohn disease. However, genetic analysis of the HSP70 gene in Crohn disease has not been done. The aim of this study is to investigate whether HSP70-2 gene polymorphism is involved in susceptibility to Crohn disease in the Japanese population and whether it correlates with clinical manifestation of the disease. METHODS: A total of 108 genetically unrelated patients with Crohn disease and 108 healthy controls were typed for HSP70-2 PstI polymorphism by restriction fragment length polymorphism analysis (alleles A and B). Patients with Crohn disease were classified into two types: either perforating or non-perforating. RESULTS: Genotype and allele frequency did not differ between patients and controls. In patients with Crohn disease, allele A frequency was significantly higher in the non-perforating than in the perforating type (P = 0.02). When patients with Crohn disease of more than 6.7 years' duration were assessed, the differences in genotype and allele frequency between the two groups became more significant (P < 0.001 and P < 0.001, respectively). CONCLUSIONS: These data suggest that HSP70-2 gene polymorphic allele A is a possible genetic marker of less severe clinical phenotype in Japanese patients with Crohn disease.

Double-contrast radiographic assessment of lupus-associated enteropathy.

PURPOSE: To describe the double-contrast radiographic features of lupus-associated enteropathy. MATERIALS AND METHODS: Six patients with systemic lupus erythematosus involving the small bowel were assessed by double-contrast radiography of the duodenum and small intestine, with reference to clinical manifestations and jejunoscopic findings. RESULTS: Lupus-associated enteropathy could be categorized into two types: acute onset enteritis in four patients and protein-losing enteropathy with hyperlipidaemia in two patients. The former group presented with irregular thickening and spiculation in the folds of the multiple segments of the duodenum to the terminal ileum, and they were frequently accompanied by thumbprinting, suggestive of ischaemic change. The latter group was characterized by mildly thickened folds with multiple submucosal nodules in the upper portion of the jejunum. In one patient from this group, jejunal biopsy demonstrated lymphangiectasia. Both groups were successfully treated by high-dose prednisolone. Follow-up radiography in the former group showed a complete improvement within 2-7 weeks, whereas radiographic abnormalities in the latter remained even after 2 months. CONCLUSIONS: Lupus-associated enteropathy cases may be divisible into two types; an acute ischaemic enteritis type and a protein-losing enteropathy type, each presenting distinct radiographic features.

Establishment and characterization of a new human testicular seminoma cell line, JKT-1.

BACKGROUND: A new human testicular cancer cell line (JKT-1) was established, successfully transplanted into nude mice, and has been maintained for over 2 years. We examined the biological characteristics of JKT-1 cells. METHODS: The original material for JKT-1 was derived from a primary lesion of a left testicular seminoma (pure, typical-type) from a 40-year-old male. The tumor tissue was minced and cultured according to the explant culture method. The cells grew as a monolayer with a doubling time of 28.5 hours. RESULTS: JKT-1 produced neither alpha-fetoprotein or beta-human chorionic gonadotropin, but showed strong immunoreactivities for vimentin and placental alkaline phosphatase. A chromosomal analysis revealed a modal number of 66 with loss of the Y chromosome. Morphologically, JKT-1 cells have a pleomorphic polygonal shape, an increase in the nuclear/cytoplasm ratio, and poor development of organelle and desmosome-like cell-cell junctions. JKT-1 cells were subcutaneously transplanted into the backs of 6-week-old nude mice, and grew classical seminomatous tissue. CONCLUSION: This report profiled a seminoma cell line established for both in vitro and in vivo experimental systems. Future studies are planned to investigate germ cells using this seminoma line.

[Transrectal ultrasound guided transperineal systematic prostate biopsy].

We retrospectively reviewed the files of patients who had transperineally undergone a systematic six sextant prostate biopsy under guidance of transrectal ultrasound in longitudinal transsection by using Biopsy needle with the spring-roaded Biopsy gun. Of the 442 patients with suspicious glands visiting our clinic between May in 1992 and February in 1998, 486 biopsies were taken and pathological diagnosis was made to be prostate cancer in 153 (34.6%) patients, benign prostate hyperplasia in 270 and prostatitis in 19. Positive predictive values obtained in this series were calculated to be 49.4% in digital rectal examination (DRE), 55.0% in transrectal ultrasonogram (TRUS), 47.8% in prostate-specific antigen (PSA) level 4 ng/mL or greater and 83.5% if a patient had positive DRE and TRUS and PSA level 4 ng/mL or more. Relationship between number of positive cores and clinical stage of the patients was found (p < 0.0001). No complications relating to the biopsy were observed. This transrectal ultrasound guided transperineal systematic six sextant prostate biopsy was thought to be one of useful tools of the biopsy for detecting prostate cancer.

Platelet noradrenaline content as an integrated measure of variations in plasma noradrenaline.

To determine whether or not platelet noradrenaline (NA) content reflects a long-term variation of plasma NA concentration, the following experiments were carried out. During a 2-hour intravenous infusion of NA in rabbits, plasma NA concentration increased rapidly to an almost consistent level 1 h later, but platelet NA content increased linearly. One hour after the infusion, the plasma NA concentration returned to normal level, while platelet NA content decreased little and only returned to the original level about 80 h later. By the 24-hour cold exposure stimulation (4 +/- 0.5 degrees C), platelet NA content and urinary NA excretion increased significantly in rats. There was a significant correlation between the urinary NA excretion and platelet NA content. In a patient with pheochromocytoma, the high plasma NA concentration returned to nearly normal levels a few hours after removal of the tumor, platelet NA content returned to normal levels 4 days after surgery. These results indicate that the measurement of platelet NA content serves as a better index for obtaining an average variation of NA concentration and sympathetic nerve activity. The measurement of platelet NA content also provides a reliable means of diagnosing pheochromocytoma.

Blood pressure and sympathetic activity following responses to aerobic exercise in patients with essential hypertension.

Fourteen untreated patients with essential hypertension (EH) were subjected to mild aerobic exercise with the intensity at 50% of the maximum oxygen uptake (VO2max) twice a week for 12 weeks, as compared with 14 normotensive subjects (NT). Blood pressure and norepinephrine (NE) in plasma, platelet and urine were measured as the indices of sympathetic activity. In EH, a significant fall in both systolic and diastolic blood pressure was observed. In addition, exercise training produced a significant decrease both urinary and platelet NE, but not in plasma NE. Neither significant changes in blood pressure nor NE in plasma, platelet and urine were observed in NT. These results suggest that an inhibition of the sympathetic nervous system might be related to the blood pressure fall in physical exercise training.

Rapidly and infiltratively growing Crohn's carcinoma of the small bowel: serial radiologic findings and a review of the literature.

We carried out a retrospective evaluation of serial changes in the small bowel radiographs of a patient with small bowel cancer accompanied by long-standing Crohn's disease. During the 8 months before diagnosis, marked morphological changes were noted. A solitary and irregular protrusion, and rapidly growing stricture under careful medical management of the underlying disease may indicate the development of cancer.

Erectile function after brachytherapy with external beam radiation for prostate cancer.

The effect of therapeutic modalities on sexual potency is an important consideration for patients choosing a treatment for prostate cancer. We assessed erectile function after iridium-192 (1r-192) high-dose rate (HDR) brachytherapy with external beam radiation therapy (EBRT), and examined the efficacy of sildenafil after this treatment. Forty-two prostate cancer patients (T1c to T3bN0M0) were treated with 22Gy HDR brachytherapy with 36.8Gy EBRT without neoadjuvant hormone therapy. Erectile function was assessed using a 5-item version of the International Index of Erectile Function questionnaire (IIEF-5), pre, 3 and 12 months after treatment, Potency was defined as an IIEF-5 score > or = 11. Ten patients with potency before HDR brachytherapy with EBRT with or without neoadjuvant hormone therapy requested Sildenafil 3 months after treatment. The mean IIEF-5 score of all patients was 10.5 +/- 8.5, 4.5 +/- 5.3 (p < 0.001), and 3.8 +/- 4.7 (p < 0.001), pre, 3 and 12 months after treatment, respectively. Seventeen (40.4%) patients were potent before treatment. The mean IIEF-5 score of those patients was 15.8 +/- 3.2, 9.6 +/- 5.1 (p = 0.04), and 11.3 +/- 6.1 (p = 0.06), pre, 3 and 12 months after treatment, respectively. Ten of 17 (58.8%) patients maintained their potency 12 months after treatment. In 10 patients with potency before treatment who were treated with sildenafil, the mean IIEF-5 score increased from 6.2 +/- 3.5 at 3 months to 13.6 +/- 5.1 (p < 0.001) at 12 months after treatment. Eight of 10 (80%) patients treated with sildenafil had recovered 12 months after treatment. HDR brachytherapy with EBRT can be performed with favorable results for maintaining potency.

Production and characterization of monoclonal antibodies specific to atrazine group compounds: effects of coating ligand structure on the variation of sensitivity and specificity.

Hybridoma cells were prepared by immunizing mice with carboxylic derivatives of atrazine conjugate to bovine serum albumin. After the screening of culture supernatant of hybridomas, five cell lines producing monoclonal antibodies were established and 1.8-5.3 ml of ascitic fluid per mouse was obtained from each cell line. The protein A affinity purification yielded 0.35-0.65 mg per ml of ascitic fluid from each cell line. The characterization studies in terms of sensitivity and specificity indicate that MAb 2F9 and MAb 4B9 showed the best responses with atrazine and its group of ametryne and cyanazine, using microtiter plate coated with simazine derivative of 6-amino hexanoic acid; no cross-reactivity was shown with simazine and cyanuric chloride.
