Two adenoid cystic carcinomas, one of the nasal cavity, the other a bronchial tumor, were cytogenetically analyzed. The former had a t(6;9)(q21-22;p 13-21) as the sole karyotypic abnormality. The latter had two related abnormal clones, resulting in the mosaic karyotype 46,XY,t(9;17)(p13;p13)/46,Y,t(X;6)(p22;q23),t(9;17)(p13;p13). The karyotypic profiles of the two cases, the only respiratory tract adenoid cystic carcinomas that have been cytogenetically characterized, differ little from those of previously reported adenoid cystic carcinomas of the major salivary glands, underscoring the fundamental biologic similarity among these tumors even when they develop from different structures and in different anatomical sites and organs. Because in the second case the t(9;17) obviously must have preceded the t(X;6), we conclude that both tumors had rearrangement of 9p13 as the primary cytogenetic change. The data thus add to the evidence that 6q changes are frequent, albeit at least sometimes secondary, aberrations in malignant salivary gland tumors. A subset of adenoid cystic carcinomas instead have rearrangement of 9p as the primary, and presumably pathogenetically essential, abnormality.
Cox and Bürk (Eur. J. Biochem., 1991) reported the partial characterization of Milk Growth Factor (MGF) which stimulated the migration of fibroblasts. We have fractionated the partially purified sample by RP-HPLC and obtained the separation of two peaks of activity. The two active components were isolated as pure MGF-a and MGF-b by RP-HPLC and preparative SDS-PAGE. The purified MGF-a, consisting of a single band by gel electrophoresis and a single peak on an HPLC reversed-phase C-4 column, has the same specific activity as TGF-beta 2 in the fibroblast migration assay. MGF-a was digested by endoprotease Asp-N and the cleaved peptides were analyzed by Edman degradation and plasma desorption mass spectrometry (PDMS). The whole sequence of MGF-a determined by automated sequenator and PDMS of S-pyridylethylated protein and selected fragments was found to be identical to that of TGF-beta 2. MGF-b protein mixture separated by SDS-PAGE was electrophoretically transferred onto a Biometra Glassybond membrane, and the blotted MGF-b protein was directly sequenced on an automated sequenator. The identified 29 amino acids sequence of MGF-b was identical to the amino-terminal sequence of TGF-beta 1. Our study demonstrates that MGF is composed of both TGF-beta 1 and TGF-beta 2. TGF-beta 2 (85%) is the predominant form.

1. Adult male Wistar rats were injected with streptozotocin (STZ: 55 mg/kg) for inducing diabetes. Then blood and atria for RNA extraction were withdrawn from rats treated 3 and 11 weeks previously with STZ respectively. Atrial total RNA were extracted with cold phenol method. The ANP mRNA contents were determined using Dot blot hybridization technique with alpha-32-P-labelled r-prepro ANP cDNA probe. 2. Plasma glucose was increased and plasma immunoreactive insulin was lowered in rats at 3 and 11 weeks after injection of STZ. ANP gene expression in diabetic rats was depressed. ANP mRNA contents in rats treated 3 and 11 weeks with STZ were 86.4% and 31.7% of that of control rats. 3. Three weeks after treatment of STZ, the rats were gastrically perfused with FOC (Fish Oil Compound) (0.355 ml/kg) once a day successively until 11 weeks. This treatment induces lower blood pressure in rats. ANP gene expression in FOC group was apparently recovered which had been decreased because of the effect of diabetes mellitus.
The ocular hypotensive effects of medetomidine, a relatively selective alpha 2-agonist, and its analogs were tested in rabbits and cats and their inhibition of adenylate cyclase in the isolated bovine ciliary process was also studied. It was found that topical unilateral administration of medetomidine (0.5-2.0%) to the normotensive rabbits produced a dose-dependent bilateral decrease in IOP with peak reduction in IOP at 2 hr in the treated eye and 1 hr in the untreated eye. A dose-dependent mydriasis was also observed in the treated eye. The dose-response curves of medetomidine and its analogs showed that the ranked order of intrinsic activity at lowering IOP was medetomidine greater than or equal to MPV-1440 greater than detomidine greater than MPV-1441 and MPV-305 BII. At concentrations lower than those used in rabbits, topical application of medetomidine to the normotensive cats lowered IOP in both treated and untreated eyes. Medetomidine and detomidine caused a dose-dependent inhibition of isoproterenol-stimulated adenylate cyclase activity. Detomidine was found to be a partial agonist producing about 43% of maximum inhibition obtained by medetomidine. The IOP efficacy of these alpha 2-agonists paralleled their effects on adenylate cyclase activity. The results demonstrated that these imidazoline derivatives are effective ocular hypotensive agents which may be useful in understanding the contribution of alpha 2-receptors to the regulation of IOP.

The multistage theory of carcinogenesis specifies that cells progress to cancer through a series of discrete, irreversible genetic alterations, but data on radiation-induced cancer incidence in rat skin suggests that an intermediate repairable alteration may occur. Data are presented on cancer induction in rat skin exposed to the following radiations: 1. an electron beam (LET = 0.34 kev/mu), 2. a neon ion beam (LET = 45 kev/mu) and 3. an argon ion beam (LET = 125 kev/mu). The latter 2 beams were generated by the Bevalac at the Lawrence Berkeley Laboratory, Berkeley, CA. About 6.0 cm2 of skin was irradiated per rat. The rats were observed every 6 weeks for at least 78 weeks and tumors were scored at first occurrence. Several histological types of cancer, including squamous and basal cell carcinomas, were induced. The total cancer yield was fitted by the quadratic equation, and the equation parameters were estimated by linear regression for each type of radiation. Analysis of the DNA from the electron-induced carcinomas indicated that K-ras and/or c-myc oncogenes were activated in all tumors tested. In situ hybridization indicated that the cancers contain subpopulations of cells with differing amounts of c-myc and H-ras amplification. The results are consistent with the idea that ionizing radiation produces stable, carcinogenetically relevant lesions via 2 repairable events at low LET and via a non-repairable, linked event pathway at high LET; either pathway may advance the cell by 1 stage in the multistage model.
From 1977 to 1987, 1033 cases of unstable tibio-fibular fractures were treated with an external fixator designed by the authors. Among them, 232 involved open fractures. All of the patients were followed up for 4 to 90 months (average 20 months). The duration of bed confinement averaged 8.3 days, and the mean time necessary for clinical bone union was 54 days. Anatomical or nearly anatomical bone apposition was effected in 878 cases (85.0%), functional apposition in 141 cases (13.6%) and malunion in 14 cases (1.4%). The overall functional results were as follows: excellent in 769 cases (74.4%), good in 218 cases (21.1%), fair in 32 cases (3.1%), and poor in 14 cases (1.4%). The design of the device and its indications are also discussed.
The monoclonal antibody against bone morphogenetic protein (BMP-McAb) was first used for demonstration of bone morphogenetic protein (BMP) in 13 patients with osteosarcoma. Using avidin-biotin complex method (ABC), we demonstrated that BMP mainly existed in the tumor cell cytoplasm and tumorous osteoblast with positive staining in 10 out of 13 osteosarcoma patients. Using this staining method, we can not only differentiate osteosarcoma from fibrosarcoma (all are negative) and other non-osteogenic tumors, but also further classify osteosarcoma according to the BMP content and distribution by means of quantitative histological analysis. The BMP quantity of osteosarcoma with the patients' clinical situation will be useful in clinical diagnosis, treatment and prognosis. The relationship between BMP and the formation of the tumorous bone, and the relation between BMP and the process of osteosarcoma are discussed.
Cerebral vasospasm (V.S.P.) is a major complication of subarachnoid hemorrhage. In order to investigate this cerebral vasospasm, an intracisternal injection of autologous blood and topical application around the internal carotid artery was performed in the dog model. Thirty dogs were divided into five groups and a cerebral angiography was performed on D0, D2, D5, D7 and D9. In comparison with the control group (group 1), the angiography in group 2 (12 dogs) systematically exhibited severe V.S.P. In group 3 (intracisternal injection of venous blood: 5 dogs), the angiography exhibited no vasospasm at all. In group 4, a left frontoparietal flap was opened, allowing us to deposit blood around the carotid bifurcation. In these cases, a severe narrowing of cortical arteries was demonstrated. In group 5 (intracisternal injection of heparinated blood), no vasospasm was observed. These findings suggest that the vasospasm involves the circle of Willis' as well as the cortical arteries.

1. Effects of sodium loading and dehydration on ANP gene expression were investigated in rats. 2. ANP-mRNA was determined using Northern blot and dot blot hybridization technique with alpha-32P-labeled r-preproANP-cDNA probe. Salt loading increased the ANP-mRNA content in atria. Correlation with ANP-mRNA content and the plasma sodium concentration was established. 3. Deprivation of water for 2 and 4 days increased ANP-mRNA 2.1- and 1.6-fold, respectively. 4. These results demonstrated that water-salt balance affects the ANP-gene expression.
Numerous studies have differentiated schizophrenic patients and normal controls in electroencephalography (EEG) spectral patterns recorded at rest. We replicated the resting EEG spectral differences between these groups and observed significant differences in periodic photic stimuli on the EEG spectra. Drug-free schizophrenic male patients (n = 8, mean age = 23.9) and normal male controls (n = 11, mean age = 24.3) were studied. Eighty seconds of EEG were collected from each subject for each of four experimental conditions: one resting and three photic-driving conditions (2.38, 4.54, and 8.33 Hz). Eye movement and other movement artifacts were minimized by use of an automatic amplitude threshold filter. Although large eye movements could be excluded as confounding factors, the filter could not for certain exclude small eye movements. Subjects were instructed to keep their eyes closed throughout. A significant difference was found between the groups both at rest and following photic stimulation in EEG activity. This result was characterized by increased delta activity and decreased alpha activity in schizophrenic patients at rest. The EEG activity following the photic driving also differentiated the groups. Schizophrenic patients had decreased sensitivity to the photic stimulation in the alpha range for spectra derived from both fundamental and harmonic analysis.
The monoclonal antibody against bovine bone morphogenetic protein (bBMP-McAb) was first used for demonstration of bone morphogenetic protein (BMP) in osteosarcoma. The avidin-biotin complex method (ABC) demonstrated that of the 18 osteosarcomas, 15 stained positive, while all 6 fibrosarcomas were negative. The results showed that BMP mainly exists in the tumor cell plasma and some tumor-like bone tissues. Using this staining method, we can not only differentiate osteosarcoma from fibrosarcoma and other non-bone-derived tumors, but also classify osteosarcoma according to the content and distribution of BMP and the patient's clinical situation, thus providing a scientific basis for clinical treatment.
The Chinese translation of the Wechsler Intelligence Scale for Children--Revised (WISC-RC) was administered to 660 children (ages 6 through 16 yr.) in the city of Shanghai. The obtained norms represent children's intelligence levels in big cities where the economic and cultural development is advanced. The norms are reported as "Scaled Score Equivalents of Raw Scores" for each age group and as "IQ Equivalents of Sums of Scaled Scores." The reliability and validity of the norms indicate that the WISC-R is suitable for use with school-age children in China. The difference between the results for our Shanghai sample (WISC-RCs) and a USA sample (WISC-R) is also discussed.
Aging for 5 years, from 50 to 55 years of age, influenced the various markers in a normotensive group but a similar correlation could not be seen in a hypertensive group. This suggests an impaired control of physiological homeostasis in hypertensive subjects. An increase in body mass index (BMI) is an important predictor in understanding the pathogenesis of hypertension, although the mechanism is uncertain. A reduction in BMI may reduce the risk of high blood pressure.
Disodium cromproxate (SCP) is an anti-allergic drug. SCP 80, 160, 320 mumol/L markedly increased spontaneous beating rate of cultured myocardial cells, especially SCP 160 mumol/L. SCP 320, 640 mumol/L increased the amplitude, maximal rate of rise and prolonged the duration of the action potentials of the cultured myocardial cells of neonatal rats. The maximal diastolic potential was not affected by SCP.
