We used TIANLONGXIFENG granulae to treat the patients with GANYANGHUAFENGZHENG of cerebral infarction, and mobilely observed the contents of plasma norepinephrine, epinephrine, cortisol, thromboxane B2 and serum triiodothyroidoglobulin. The results showed that with the improvement of curative effect and the decrease of symptom scores, the contents of parameters mentioned above changed accordingly. It is suggested that these parameters may have value for diagnosis.
OBJECTIVE: To explore the role of granulocyte-macrophage colony-stimulating factor (GM-CSF) in eosinophil inflammation of asthma airway. METHODS: Guinea pig models of asthma were established with aerosolized ovalbumin. A group of asthmatic guinea pigs were treated with injection of triptolide, density of eosinophil infiltration in airway tissues was observed under microscope and expression of GM-CSF mRNA in airway tissues was detected with dot hybridization by Dig-labeled cDNA probe. RESULTS: Expression of GM-CSF mRNA in asthmatic animals were higher markedly than that in the triptolide-treated and the control groups. However, there was no statistically differences of density of eosinophils infiltration between the asthma and the triptolide-treated groups. CONCLUSIONS: GM-CSF might participate in eosinophil inflammation in airway of asthmatic animals and triptolide might be of potential value in anti-inflammation for asthma.
OBJECTIVE: To identify the astigmatism changes after excimer laser photorefractive keratectomy (PRK) and the visual influence of astigmatism. METHODS: 109 myopic eyes followed up for more than 1 year were analyzed by using videokeratography, and their visual acuities were examined. Before the operation, the astigmatic errors with cycloplegic refractive examination were -1.00(-)-2.00 D. RESULTS: The position of astigmatism axis was basically consistent with that in corneal topography, but the astigmatism diopter was of some difference between the cycloplegic examination and topographic analysis. With-the-rule astigmatism was in 61% of the eyes, asymmetrical was 33% and against-the-rule was 6% in topographic analysis. Only was a little change in the position of astigmatism axis after PRK observed. Some changes of postoperative astigmatism diopter were seen in different periods. It was increased in the postoperative 10 days or 1 month, and afterwards it became gradually decreased and stable in 6 months to 1 year. The postoperative residual astigmatism was low in degree, that did not affect the visual acuity a great deal. The actual corrected diopter was within the anticipated corrected diopter range of +/- 1.00 D, accounting for 97%. CONCLUSION: By using spherical equivalent method of calculation in PRK, the refractive correction of the operation in cases with myopia and low degree of astigmatism (< -2.00 D) can be satisfactory.
The levels of plasma norepinephrine (NE) and epinephrine (E) were determined by HPLC in 220 patients with various TCM liver syndromes, and in 96 healthy subjects as controls. The plasma NE and E contents were higher in patients with liver excess syndromes, including Ganqi Yujie (GQYJ [symbol: see text]) syndrome, Ganyang Shangkang (GYSK [symbol: see text]) syndrome, Ganhuo Shangyan (GHSY [symbol: see text]) syndrome, and Ganyang Huafeng (GYHF [symbol: see text]) syndrome; while they were lower in patients with liver deficiency syndromes, including Ganqixu (GQX [symbol: see text]) syndrome, Ganxuexu (GXX [symbol: see text]) syndrome, and Ganyinxu (GYX [symbol: see text]) syndrome. Futhermore, when Ganshen Yinxu (GSYX [symbol: see text]) syndrome was turned into GYSK and then into GYHF, the plasma NE and E contents increased in order of precedence; while when GQYJ was turned into Ganyu Pixu (GYPX [symbol: see text]) syndrome, both NE and E contents decreased. The results suggest that the plasma NE and E contents are reliable objective parameters for the study of pathophysiological basis of the liver excess and liver deficiency syndromes, and the liver-kidney and liver-spleen interrelated syndromes.

OBJECTIVE: To investigate whether antisense oligodeoxynucletides(ODNs) of VEGF can become a kind of drugs for antitumor cancer treatment. METHODS: With a tumor model on the cornea of the mouse the inhibition of angiogenesis and tumor growth by 5 ODNs with different modifications and sequences was determined. RESULTS: ODNs with partial phosphorothioate modification, or with hairpin structure could effectively inhibit tumor-induced angiogenesis, but the linear ODN could not. The use of 2 ODNs at different sites gave the best results. CONCLUSION: VEGF antisense ODNs can hopefully become new drug with antitumor and antiangiogenesis activities.
AIM: To statistic the usage of hypnotics and sedatives in outpatient and emergency clinics prescriptions in one day. METHOD: Check all prescriptions on April 29, 1987 and November 13, 1995 in order to know the quantity, kind and distribution of hypnotics and sedatives in different outpatient departments. RESULT: There were 1,319 prescriptions on April 29, 1987. Among them, hypnotics and sedatives were 122(9.24%); in comparing with 2,065 prescriptions on November 13, 1995, hypnotics and sedatives were 141 (6.82%). Benzodiazepine used more than 80% in all prescriptions. CONCLUSION: The possibility of long-term low-dosage using benzodiazepine is present. It must strengthen the control and education to avoid the drug-abuse.
To determine the relationship between apoptosis and carcinogenesis of oral mucosa, the distribution patterns of apoptosis in normal (N), inflammation (IF), dysplasia (DYS) and squamous cell carcinoma (SCC) of oral mucosa were evaluated by in situ end-labeling method. Results showed that the apoptosis levels in N and IF were relatively lower and stable, whereas the apoptosis of DYS was the highest one in all observed groups. The apoptosis of DYS and SCC were increased obviously higher than N and IF. The results indicated that apoptosis played an important role in premalignant lesion and malignancy of oral mucosa.
Effect and mechanism of simulated weightlessness (SWL) in humans and rabbits erythrocyte deformation were studied. Erythrocyte deformation and membrane fluidity in humans and rabbits, and erythrocyte morphology and hemorreology indices in control and HDT rabbits were measured. The results were a decrease in erythrocyte deformation and membrane fluidity in humans and rabbits during SWL, a significant increase in abnormal erythrocyte, blood viscosity, hematocrit, fibrinogen, and red blood cell aggregation index in HDT rabbits. These results show that the changes in erythrocyte shape, increase of erythrocyte internal viscosity and changes in erythrocyte visco-elasticity may be causing the decrease of erythrocyte deformation in SWL humans and rabbits.
Much effort in TLC has been devoted to optimization methods of solvent system in order to get best separation of components. Various optimization methods have been established, but the universality of some methods was poor. They were only applicable to samples of known components. This paper reports a complex method. When the complexity of the solvent system was determined, uniform design was used to arrange experiments, the data obtained from scanning chromatogram were input computer to get regression equation. The precision of the regression equation should be judged at first. If the precision was proper, the regression equation could be input the program of the complex method to obtain the composition of the solvent system. Optimization results were dichloroethane-benzene-methanol (4.81:4.42:0.78) for cortical hormone; toluene-ethanol-dichloroethane-trichloromethane (2.30:1.46:3.32:2.92) for sulfa drugs and trichloromethane-methanol-ethyl acetate (3.30:3.29:3.41) for ginkgo biloba. Then the verification experiment was made. The verifying test showed that the experimental results are identical with the calculated ones.
The results show that middle shear specific viscosity and reductive viscosity (P<0.05), low shear specific viscosity and reductive viscosity of whole blood (P<0.01) decreased significantly in experimental group after 9-day of pulse magnetic fields, comparing with control group, high shear specific viscosity, low shear reductive viscosity (P<0.05) and middle and low shear specific viscosity (P<0.01) of whole blood in experimental group were lower after exposed than before exposed to pulse magnetic fields. The results suggest that the pulse magnetic fields may reduce specific and reductive viscosity of whole blood in human body.
The increase in nuclear cytoplasmic (N/C) ratio is one of the features of cellular atypia which is used in the histopathological assessment of premalignant lesions of the oral mucosa. Since this feature is readily quantifiable using morphometry, we have analysed both N/C and nuclear volume densities in basal and spinous cells from human cheek lesions with and without malignant potential in order to ascertain the validity of this parameter as a predictor. Using a strictly standardised sampling procedure, measurements of cellular and nuclear areas of basal and spinous cells from normal and pathological human cheek mucosa were made on haematoxylin and eosin-stained sections using a VIDAS image analyser. Cases examined comprised fibrous hyperplasia (FH), traumatic inflammation (IF), benign hyperkeratosis (HK), lichen planus (LI), leukoplakia with dysplasia (DYS), squamous cell papilloma (PP), dysplastic epithelium from the edges adjacent to invasive carcinoma (CE) and islands from invasive squamous cell carcinoma (CI). In basal cells, N/C ratios and nuclear volume densities were lower than values obtained for the normal controls. In spinous cells, these parameters were elevated in the potentially premalignant lesions (DYS, CE) as well as in CI but values were similarly elevated in FH, IF, HK and PP, lesions which appear to have no malignant potential. The N/C ratio is of no value as a predictor of malignant potential in basal or spinous cells from cheek lesions. The putative increase in N/C which has been previously described qualitatively is probably due to increased nuclear hyperchromatism, which may provide an illusory increase in relative nuclear size at the expense of the cytoplasm.
Basic fibroblast growth factor (FGF-2) plays an important role in myocardial growth and development and in particular cardiac myocyte proliferation. FGF-2 exerts its effects by binding to cell surface receptors (FGFR-1) of the tyrosine kinase family. We have detected the presence of both long and short isoforms of FGFR-1 in embryonic and adult mouse heart. In this report, we have examined the ability of long and short FGFR-1 isoforms to signal a mitogenic response. Assessment of RNA from rat myoblast H9c2 cells by reverse transcriptase-polymerase chain reaction and RNA blotting revealed that they were deficient in transcripts corresponding to long and short FGFR-1 species. Hybrid genes containing the cDNAs coding for long and short FGFR-1 isoforms directed by the myosin light chain-2 promoter and simian virus 40 enhancer sequences, were used to transiently transfect H9c2 cells. Total tyrosine phosphorylation was increased 2.0 and 2.6 fold in H9c2 cells transfected with the long and short FGFR-1 isoforms, respectively, compared to 'control' transfected H9c2 cells. This was accompanied by a 2.1 and 2.0 fold increase in DNA synthesis, as measured by tritiated thymidine incorporation, in H9c2 cells expressing the long and short FGFR-1 isoforms, respectively. To assess effects on proliferation, H9c2 cells were stably transfected with the myosin light chain-2/FGFR-1 cDNA genes. The rate of proliferation was increased 1.6 and 3.1 fold in H9c2 cells stably expressing the long and short FGFR-1 isoforms, respectively, compared to 'control' H9c2 cells. In contrast to non transfected H9c2 cells, treatment of H9c2 cells stably expressing long FGFR-1 with FGF-2 for 24 h resulted in a slight increase (1.3 fold, p < 0.02) in cell number. However, a greater response (1.5 fold, p < 0.0005) was observed with H9c2 cells stably expressing short FGFR-1 after treatment with FGF-2. These results suggest that both long and short FGFR-1 isoforms are capable of signalling a mitogenic response.

Data from 206 pairs of nuclear family members were analyzed to study the influence of genetic factors on the occurrence of chronic obstructive pulmonary disease (COPD) and its familial aggregation. Results showed that occurrence of COPD appeared in family aggregation. Risk of occurrence of COPD in the relatives (father, mother, brothers and sisters) of the proband with COPD increased by 2.07 times, as compared with relatives of the controls. Significant differences of the risk of COPD in fathers, brothers and sisters between proband and controls were observed, but not in mothers. It suggests relatives of the proband are more susceptible to COPD than those of the controls, and genetic factor may contribute to pathogenesis of COPD.
Plasma norepinephrine (NE) epinephrine (E) concentrations in patients with five types of Gan syndromes and relative syndromes were determined. The healthy individuals served as control. The contents of NE and E in Ganyang Huafeng Zheng (5975.8 +/- 305.6 pmol.L-1, 2227.7 +/- 12.57 pmol.L-1) were significantly increased; Ganxue Xu Zheng (1100.4 +/- 105.5, 451.2 +/- 70.9 pmol.L-1) were decreased, and lower than the normal controls. NE and E contents in both Ganyang Shangkang Zheng (3962.6 +/- 354.4, 1225.6 +/- 163.5 pmol.L-1) and Ganhuo Shangyan Zheng (3398.1 +/- 303.8, 1134.4 +/- 146.9 pmol.L-1) were increased remarkably, but there was no difference between them. Plasma NE, E concentrations can be considered as one of the comprehensive laboratory parameters in diagnosis of the four above mentioned Gan syndromes.
Little is known about what determines the nuclear matrix or how its reorganization is regulated during mitosis. In this study we report on a monoclonal antibody, mAb2A, which identifies a novel nuclear structure in Drosophila embryos which forms a diffuse meshwork at interphase but which undergoes a striking reorganization into a spindle-like structure during pro- and metaphase. Double labelings with alpha-tubulin and mAb2A antibodies demonstrate that the microtubules of the mitotic apparatus co-localize with this mAb2A labeled structure during metaphase, suggesting it may serve a role in microtubule spindle assembly and/or function during nuclear division. That the mAb2A-labeled nuclear structure is essential for cell division and/or maintenance of nuclear integrity was directly demonstrated by microinjection of mAb2A into early syncytial embryos which resulted in a disintegration of nuclear morphology and perturbation of mitosis.
