Topographic analysis of EEG photic driving in patients with schizophrenia following clozapine treatment.

Reduced EEG photic driving has been found to be diagnostically sensitive and specific for schizophrenia. Thirty-one patients with schizophrenia were tested in this study to identify the typical and atypical neuroleptic effects on the photic driving. Compared with the placebo, clozapine significantly enhanced the photically driven EEG in theta and low alpha frequency band, while haloperidol did not have the same effect. These changes with clozapine appeared to be symmetrical and located primarily in the frontal, central and mid-parietal areas but not in the lateral parietal, temporal and occipital regions. Results were consistent with previous findings and suggested that the atypical EEG profile of clozapine might be associated with 5-HT2 antagonistic property.

[Effects of static magnetic fields on free radical metabolism of human body].

The effects of static magnetic fields at 150 to 155 mT on the activities of superoxide dismutase (SOD) and the contents of maglonydiadehyde (MDA) in the serum and the activities of glutathione peroxidase (GSH-Px) in the whole blood were observed in the healthy volunteers. The results showed that the activities of SOD and GSH-Px were significantly increased, and the content of MDA was significantly decreased in short term at the observed magnitude of static magnetic fields. The results suggest that the function of the free radical metabolism of human body could be improved by static magnetic fields at that magnitude.

[Genetic epidemiological study of asthma in general].

With theory and method of genetic epidemiology, data of 641 nuclear families on asthma were analysed, and the results showed that the history of parents asthma was one of risk factors about offsprings asthma, and the risk was 4.0-8.0 times higher (P < 0.01), compared with the one of offsprings of parents with no asthma. The effect of parents with asthma on the risk of sons was the same as one of daugters. The result declared that the airway responsiveness of parents did not relate to the one of offsprings. The results suggest that the genetic factors play an important role in the asthma.

[Morphological studies of myocardial fibrosis in different types of Keshan disease].

OBJECTIVE: To clarify the localization, distribution, content and type of collagen in the myocardium of patients with Keshan disease (KD). METHODS: Picrosirium red staining, polarization microscopy and image analysis were used to study autopsy specimens from 335 KD patients. RESULTS: A loose network of collagen and a higher ratio of type III collagen were found in the myocardial scar tissues of acute KD patients. In the myocardial scar tissues from chronic KD patients, the collagen fibers were dense, the ratio of type I collagen was higher, the ratio of type I to type III is increased in chronic KD and decreased in acute KD. The amount of collagen in non-necrotic regions was significantly higher than that of the controls. CONCLUSION: The degeneration and necrosis of myocytes in KD is associated with disorders of collagen metabolism and with structural modelling of collagen.

[The influence of vascular endothelial growth factor and its receptor on the growth of laryngeal cancer cell].

The function of VEGF in laryngeal carcinoma was studied by observing either the expressions of VEGF and its receptor flk-1 in laryngeal carcinoma, or the effect of anti-VEGF antibody and anti-VEGF receptor antibody on the growth of human laryngeal cancer cell line HEP-2 in vitro. The expression of VEGF and flt in the specimen of laryngeal carcinoma were determined by standardized immunohistochemistry (Elite ABC method). Anti-VEGF antibody and anti-VEGF receptor antibody were also added at various concentrations to the culture medium, respectively, and the growth of HEP-2 was measured by MTT assay. The immunohistochemical staining showed that the expressions of VEGF and flt were detected both in carcinoma cells and endothelial cells. The growth of HEP-2 cell line in vitro was suppressed when adding various concentration of anti-VEGF antibody and anti-VEGF receptor antibody to the culture medium. The laryngeal cancer cells can secret VEGF and there exists VEGF-binding site on laryngeal cancer cell. Both blockading VEGF with anti-VEGF antibody and blockading VEGF-binding site with anti-VEGF receptor antibody could inhibit HEP-2 growth.

[A study of the correlations between asthma and airway responsiveness].

OBJECTIVE: To evaluate the effect of airway responsiveness on asthma and to study the causes of asthma. METHODS: The 641 nuclear families were confirmed by indicator cases with asthma in three counties. Investigation and methacholine challenge test were conducted for all relatives of the nuclear families. The data were by EPI5.01 and the odds ratio (OR) was employed to evaluate the correlation between asthma and airway responsiveness. RESULTS: 72 percent of persons with asthma history showed airway hyperresponsiveness, but only 70 per cent of persons with airway hyperresponsiveness suffered asthma. The risk for asthma in relatives with positive airway responsiveness was 5.8 times higher than those with negative airway responsiveness (P > 0.01) among the nuclear families. CONCLUSION: Airway hyperresponsiveness is one of the risk factors of asthma.

[Myopic regression after photorefractive keratectomy].

OBJECTIVE: To investigate myopic regression after excimer laser photorefractive keratectomy (PRK) in patients with myopia. METHODS: 103 eyes with regression (>or= -1.00 D) found in the follow-up for more than 1 year were included in this study. The preoperative spherical equivalent was -4.00 - -14.50 D (-8.72 +/- 2.65 D). Patients were divided into two groups: (1) moderate myopia (-4.00 - -7.90 D) and (2) high myopia (>or= -8.00 D). RESULTS: A significant positive correlation was found between regression and the extent of attempted correction, the rate of regression in high myopia group was 6.93% and that in moderate group, 1.17%. The rate of regression in older age group was higher than that in younger cases. (P < 0.01). Relatively severe haze under the corneal epithelium often accompanies the regression, and decrease of the dosage of topical corticosteroid application or its withdrawal because of the development of corticosteroid ocular hypertention could lead to the regression. The stability of the refraction is not related to the difference in sex. The regression is often accompanied by the increase in K value. CONCLUSION: The regression is significantly related to the extent of attempted correction, patients' age, corneal haze and topical application of corticosteroid.

[Relationship between the mRNA transcriptions of bFGF and KGF in the carcinogenesis of oral mucosa].

The aim of the study was to determine the expressions and effects of KGF and bFGF of FGF superfamily in oral mucosa. The mRNA transcriptional levels of bFGF and KGF in dysplasia (DYS) and squamous cell carcinoma (SCC) of buccal mucosa were observed and evaluated by in situ hybridization method. The results showed that bFGF and KGF mRNA were expressed by the mesenchymal cells in both DYS and SCC groups, but not in any of the epithelial cells. The positive reactions were located in fibroblasts, endothelial cells and some of the chronic inflammatory cells. There is an increasing mRNA transcriptional level in SCC than in DYS. The results indicated that bFGF and KGF may not only enhance the neovascularization and formation of stroma by autocrine and paracrine action, but also involved in the growth of neoplastic cells.

FISH characterization of head and neck carcinomas reveals that amplification of band 11q13 is associated with deletion of distal 11q.

In order to characterize homogeneously staining regions (HSR) and other 11q13 rearrangements identified cytogenetically, we performed fluorescence in situ hybridization (FISH) using a CCND1 cosmid and five YAC clones spanning chromosomal bands 11q13-14 on metaphase cells from 14 primary and one metastatic head and neck carcinomas. At the cytogenetic level, a total of 17 HSR were detected in ten cases: five were in derivative chromosomes 11 in band 11q13, and 12 were located in other derivative chromosomes. Other forms of 11q13 rearrangements were observed in five cases, whereas two cases had normal chromosomes 11. FISH analysis demonstrated that all HSR but two were derived from the 11q13 band. The size of the amplicon varied from case to case, but the amplification always included the region covered by YAC 55G7, which contains the CCND1 locus. The amplification of CCND1 was confirmed by use of a CCND1 cosmid. We also showed that most of the cases (9 of 11) with 11q13 amplification had lost material from distal 11q. The breakpoints were mapped by FISH and were shown to cluster to the region between YACs 55G7 and 749G2. We conclude that loss of gene(s) in distal 11q may be as important as amplification of genes in 11q13 for the biological aggressiveness of head and neck carcinomas.

Purification, cDNA cloning and molecular characteristic of a fibrinolytic enzyme from the venom of Agkistrodon acutus.

A nonglycoprotein-like fibrinolytic enzyme ((FIB-I) was purified from the crude venom of Agkistrodon acutus by CM-Sepharose CL-6B and DEAE-Sepharose CL-6B ion exchange chromatography and then by FPLC through Superose 12 gel filtration. Its molecular weight is about 23 kDa and isoelectric point is near 6.0. It not only has fibrinolytic and caseinolytic activity, but also can hydrolyze BAEE. The local hemorrhagic activity was found in mice after the subcutaneous injection of this enzyme. EDTA can inhibit its fibrinolytic activity completely, but PMSF and arrowhead proteinase inhibitor have no such obvious inhibitory effect, thus implying that FIB-I is a metalloproteinase. The N-terminal ten amino acid residues 'STEFQRYMEI' of FIB-I was elucidated. A full-length cDNA gene of this enzyme was cloned by using RT-PCR from the total RNA extracted from the snake venom gland and FIB-I was expressed in E. coli. Having analyzed the sequence, we found that it had a typical zinc-chelating characteristic as 'HEXXHXXGXXHD.'

Single cell multiple biomarker analysis in archival breast fine-needle aspiration specimens: quantitative fluorescence image analysis of DNA content, p53, and G-actin as breast cancer biomarkers.

Fine-needle aspiration (FNA) is a sensitive and cost-effective method for evaluating breast lesions. However, the diagnosis of early premalignant lesions is less reliable by FNA because of a lack of distinctive cytological features. Accurately defining the risk of such lesions at the individual level may have significant impact in breast cancer prevention and management. The main objective of this preliminary study was to develop a method to study multiple biomarkers on archival FNA slides using quantitative fluorescence image analysis (QFIA). Biomarkers p53, G-actin, and DNA content were labeled with an immunofluorescence technique and measured by QFIA simultaneously on a single cell basis. QFIA allows the labeling and measurement procedures to be carried out in situ, without the need to remove cells from the slide while preserving the morphology of the cells. FNA slides from 72 incident patients were obtained for this study. Fifty-six cases had an adequate number of cells for the actual analysis (25 benign breast lesions, 14 proliferative breast diseases with nuclear atypia, and 17 malignant lesions). The DNA content (> or = 5c) and G-actin (average gray mean, > 90) were positive in 81% and 88% of malignant lesions, respectively. These were significantly higher than the corresponding positive rates in benign lesions (7% and 15%, respectively; P <0.01 for both). None of the benign cases were positive for G-actin and DNA simultaneously, and none of the malignant cases were negative for G-actin and DNA together. p53 was positive in 33% of malignant lesions and 8% of benign lesions (P >0.05). Our study demonstrates the feasibility of evaluating multiple biomarkers by QFIA on archival FNA-fixed specimens. The G-actin and DNA content assayed by QFIA may be potential intermediate end point markers for breast cancer individual risk assessment.

The human integrin beta3 gene is 63 kb and contains a 5'-UTR sequence regulating expression.

The human blood platelet fibrinogen receptor, integrin alphaIIbbeta3 (glycoprotein IIb-IIIa) is an archetypal member of the integrin family of adhesive molecules and is the only integrin encoded by genes physically linked in the genome. Because studies on the normal and abnormal expression of any gene require a thorough understanding of its organization, the initial goals of the current study were to determine the size and complete the genomic organization for the beta3 gene. We now report the isolation of the entire beta3 gene in a single P1 plasmid and for the first time have linked the first and second exons on a contiguous fragment of DNA. Using pulsed-field gel analysis, we determined the full size of the beta3 gene to be 63 kb and show a large (16.7 kb) first intron; based on this information, we propose a uniform numbering system for the beta3 exons. We have completed the 5' genomic structure and generated a long-range restriction map. The promoter and the 5' end of the first intron were found to have approximately 50% sequence identity with a region of the avian beta3 gene known to possess functional transcriptional activity. Analysis of three different homologous regions led to the identification of a sequence in the 5'-UTR of the human gene, CCGCGGGAGG, which shares 90% identity with the avian gene and which bound nuclear proteins in DNaseI and electrophoretic mobility shift assay studies. Mutating this sequence caused a 2.6-fold reduction in reporter gene activity. In these studies we have (1) determined the full length and 5' organization of the beta3 gene, (2) identified a large region of homology between the 5' regions of the avian and human genes, and (3) identified a sequence in the 5'-UTR that augments gene expression. Knowing the genomic structure of beta3 has permitted the uncovering of new mechanisms of mutagenesis causing Glanzmann thrombasthenia (Jin et al, J Clin Invest 98:1745, 1996), and our findings will be valuable for such genetic analyses as well as for studies on the transcriptional regulation of beta3 and other integrin genes.
