Defective transcriptional activation by diverse VP16 mutants associated with a common inability to form open promoter complexes.

Three different types of VP16 mutants were assayed in vitro. These included deletion of the C-terminal activation subdomain and alterations in either the acidic or non-acidic components of the minimal activation domain. The mutant GAL4-VP16 proteins were found to be transcriptionally defective using a HeLa cell nuclear extract. In all three cases the loss of transcription activity was accompanied by a commensurate loss in ability to form open transcription complexes. The comparison implies that the diverse components of GAL4-VP16 activate transcription by the common facilitation of steps required for open complex formation. The results further imply that open complex formation may be a common target for mammalian transcriptional activation, as known previously to be the case in bacterial systems.

Long-term effect of omega-3 fatty acid supplementation in active rheumatoid arthritis. A 12-month, double-blind, controlled study.

OBJECTIVE: To study the long-term effects of supplementation with omega-3 fatty acids (omega 3) in patients with active rheumatoid arthritis. METHODS: Ninety patients were enrolled in a 12-month, double-blind, randomized study comparing daily supplementations with either 2.6 gm of omega 3, or 1.3 gm of omega 3 + 3 gm of olive oil, or 6 gm of olive oil. RESULTS: Significant improvement in the patient's global evaluation and in the physician's assessment of pain was observed only in those taking 2.6 gm/day of omega 3. The proportions of patients who improved and of those who were able to reduce their concomitant antirheumatic medications were significantly greater with 2.6 gm/day of omega 3. CONCLUSION: Daily supplementation with 2.6 gm of omega 3 results in significant clinical benefit and may reduce the need for concomitant antirheumatic medication.

Selective inability of LPS-activated B lymphocytes to produce antibodies against autologous and allogeneic MHC class I antigens.

Lipopolysaccharide (LPS)-activated B lymphocytes have been found to produce autoantibodies of a variety of specificities. We found that LPS-induced autoantibodies did not include antibodies against self H-2 class I antigens, suggesting a selective tolerance induction against some, but not other, autoantigens. However, LPS-activated B lymphocytes not only failed to produce antibodies against autologous H-2 class I antigens, but also against allogeneic H-2 class I antigens, suggesting a selective regulation of B cells to synthesize antibodies directed against autologous and allogeneic major histocompatibility complex antigens.

Triterpenoid saponins from the root of Sideroxylon cubense.

Two triterpenoid saponins were isolated from the root of Sideroxylon cubense and their structures were established as 3-O-beta-D-glucopyranosyl-28-O-[alpha-L-rhamnopyranosyl(1-->3)- beta-D-xylopyranosyl(1-->4)-alpha-L-rhamnopyranosyl(1-->2) beta-D-xylopyranosyl] protobassic acid and 3-O-beta-D-glucopyranosyl protobassic acid. The first, designed as sideroxyloside A, is a new natural product.

Mapping of a gene for hypergammaglobulinemia to the distal region on chromosome 4 in NZB mice and its contribution to systemic lupus erythematosus in (NZB x NZW)F1 mice.

IgM class hypergammaglobulinemia develops spontaneously in the early life of NZB mice. To determine the chromosomal location of the NZB gene(s) linked to this hypergammaglobulinemia and possible contribution to systemic lupus erythematosus (SLE)-like autoimmune disease observed in (NZB x NZW)F1 hybrid mice, we examined (NZB x NZW)F1 x NZW backcross mice, using a mapping technique based on polymorphism in simple nucleotide sequence repeats. In light of our previous data, we directed attention to chromosome 4. A single NZB locus or tightly linked group of loci on the distal region (70-90 cM distal from the centromere), theoretically mapped to 14.3 cM distal from marker locus D4Mit48, had the strongest association with IgM hypergammaglobulinemia. The backcross progeny heterozygous for this locus on chromosome 4, in association with heterozygosity of the MHC haplotype (H-2d from NZB/H-2z from NZW) on chromosome 17, showed significantly higher serum levels of IgG anti-DNA antibodies and more severe lupus nephritis than did the progeny heterozygous for either one of the two loci or homozygous for both loci on chromosomes 4 and 17. The NZB gene in the distal region of chromosome 4 is likely to be responsible for terminal maturation of B cells and, in concert with the effect of H-2 heterozygosity, may predispose to the full expression of SLE in (NZB x NZW)F1 mice.

Genetic study of indirect inguinal hernia.

We performed a genetic analysis of 280 families with congenital indirect inguinal hernia ascertained in Shandong province. The multifactorial threshold model and segregation analysis were applied to these families to investigate the mode of inheritance of congenital indirect inguinal hernia. Our results indicate that congenital indirect inguinal hernia is not compatible with a multifactorial threshold model, and the frequent vertical transmission and high segregation ratio suggest autosomal dominant inheritance with incomplete penetrance and sex influence. Through further pedigree analysis of the multiple case families with at least two closely related affected members, we noted preferential paternal transmission of the disease gene, which might suggest the role of genomic imprinting in the aetiology of this condition.

MCP-1-stimulated monocyte attachment to laminin is mediated by beta 2-integrins.

Migration of monocytes to sites of inflammation involves a series of attachments and detachments to extracellular matrix proteins. We examined the capacity of a chemokine, monocyte chemoattractant protein-1 (MCP-1), to regulate attachment of human monocytes to laminin, collagen I, collagen IV, or fibronectin. MCP-1 increased monocyte attachment to laminin in a dose- and time-dependent manner and stimulated a lesser increase to the other matrix proteins. Function-blocking monoclonal antibodies (MAbs) to the integrin beta 2-subunit (CD18), including Fab' fragments and alpha M (CD11b) blocked > 70% of attachment, whereas MAbs to the beta 1-integrin subunit reduced attachment by < 30%. This suggests that the CD11b/CD18 integrin is the predominant molecule involved in adhesion of MCP-1-stimulated monocytes to laminin. The association of CD11b with F-actin illustrated by confocal microscopy further supports this concept. In contrast, when monocytes were stimulated with the beta 1-stimulatory MAb TS2/16, monocyte adhesion to laminin occurred through beta 1-integrins. Thus MCP-1 can stimulate monocyte attachment to laminin, and this process is mediated through beta 2-integrins, principally CD11b/CD18.

The effect of gap depth on the perception of whether a gap is crossable.

Four experiments were performed in order to examine the effect of gap depth on human observers' perception of whether or not a gap is crossable. Experiments 1 and 2 showed that as the gap's depth increased, observers tended to increasingly underestimate the maximum width of a gap they could step across. Experiments 3 and 4 clarified this finding: The observed covariation of perceived gap crossability and gap depth depended on the observer's direction of gaze, rather than on the physical depth of the gap. The optical relations to which observers might be attending are discussed, as well as the possibility that cognitive-affective processes might have contributed to observers' underestimation of their actual capabilities.

[Single channel analysis on calcium channel blockade action of panaxadiol and panaxatriol saponins on cultured rat ventricular myocytes].

Wistar rat ventricular myocytes were isolated. Panaxadiol saponins 1500 micrograms.ml-1, panaxatriol saponins 300 micrograms.ml-1, verapamil 37.5 micrograms.ml-1, or BAY k 8644 5 mumol.L-1 were added into the bath solution separately. The single channel activities of L, T, and B type calcium channels were recorded before and after the administration, using voltage patch-clamp technique in cell-attached configuration. The calcium channel blockade effect of these 2 groups of ginsenosides was authenticated verified. The mechanism existed in the decrease in both the open time and the open-state probability of the calcium channel.

[Facial nerve surgery with increasing innervation of correct target].

Aberrant regeneration of facial nerve has a higher propensity to innervate the wrong target. This study states that transection of these nonsomatic motor nerve will increase the innervation of correct target, which is cataloged as 'transection group', and preservation of nonsomatic motor nerve as 'untransection group'. According to the House grading system for reporting recovery of the facial nerve, in incomplete lesions (Bell's palsy, Hunt's syndrome, trauma etc) treated with nerve total or subtotal decompression (101 cases), transection group produced better voluntary functional movement (average grade 1.66 +/- 0.74) than untransection group (average grade 2.81 +/- 0.87). In complete lesions (trauma, tumor and chemical destruction etc) treated with nerve grafting (65 cases), transection group obtained better improvement (average grade 2.63 +/- 0.73) than untransection group (average grade 3.65 +/- 0.78).

[Regulation on beta-END in tumor-bearing mice by moxibustion on Guanyuan point].

The experimental results demonstrated that moxibustion on Guanyuan point could promote the hyperplasia of the pituitary and the adrenal gland which showed atrophy in control group. Also, the moxibustion treatment stimulated the secretion of beta-END from the pituitary and the adrenal gland, increased the level of serum beta-END significantly and kept the high level for quite a long time, that was advantageous for beta-END to carry out the immunomodulation. The data further suggested that moxibustion treatment do not cause the instant release but probably constant release of the beta-END.

[Case-control study on intrauterine growth retardation and vitamin nutritional status in late pregnancy].

Ninety-seven cases of pregnant women with intrauterine growth retardation (IUGR) and 133 controls were investigated on their dietary intake and blood vitamin levels in late pregnancy. Results showed insufficient calorie and protein intake, deficiency in calcium and vitamin B2, and low intake of vitamins A and B1 in pregnant women with IUGR. Their serum level of vitamin E was only half as that in controls (P < 0.01). Mothers' serum level of vitamin E correlated positively to several indicators reflecting infant growth with multiple stepwise regression analysis. There was no significant difference of activity of erythrocytic transketonas activity (TPP%) and activity coefficient (AC) of glutathione reductase between the two groups. It suggested pregnant women in both groups suffered from deficiency in thiamine and riboflavin (TPP% > 16%, AC > 1.2).
