Chemokines, a family of chemotactic cytokines.

Chemokines are low-molecular-weight proteins that stimulate recruitment of leukocytes. They are secondary pro-inflammatory mediators that are induced by primary pro-inflammatory mediators such as interleukin-1 (IL-1) or tumor necrosis factor (TNF). The physiologic importance of this family of mediators is derived from their specificity. Unlike the classic leukocyte chemo-attractants, which have little specificity, members of the chemokine family induce recruitment of well-defined leukocyte subsets. Thus, chemokine expression can account for the presence of different types of leukocytes observed in various normal or pathologic states. There are two major chemokine sub-families based upon the position of cysteine residues, i.e., CXC and CC. All members of the CXC chemokine sub-family have an intervening amino acid between the first two cysteines; members of the CC chemokine sub-family have two adjacent cysteines. As a general rule (with some notable exceptions), members of the CXC chemokines are chemotactic for neutrophils, and CC chemokines are chemotactic for monocytes and a small sub-set of lymphocytes. This review discusses the potential role of chemokines in inflammation and focuses on the two best-characterized chemokines, monocyte chemoattractant protein-1 (MCP-1), a CC chemokine, and interleukin-8 (IL-8), a member of the CXC chemokine sub-family.

Ossification processes and perichondral ossification groove of Ranvier: a morphological study in developing human calcaneus and talus.

In this histologic-radiologic investigation of 32 feet of 16 fetuses and newborns ranging in age from 15 to 44 weeks, two types of ossification, i.e., endochondral ossification through primary ossification center and intramembraneous ossification through periosteal bone formation (PBF), were found to coexist in both calcaneus and talus. In addition, Ranvier's grooves (RG), or perichondral ossification groove, also was observed as a shallow, saucer-like or semicircular structure. As with PBFs, there are three and two RGs in the calcaneus and talus, respectively. RG and PBF appeared in time order and were located regularly in the concave areas of the adult calcaneus and talus. The findings support the hypothesis that one function of PBF and RG is to limit the growth of the calcaneus and talus. Thus, they are responsible for the irregular contour of the calcaneus and talus.

Fetal gallbladder growth and development during gestation.

This study sought to establish normal dimensions of the fetal gallbladder throughout pregnancy. The gallbladders in 300 normal fetuses between 15 and 40 weeks of gestation were measured prospectively for length, anteroposterior dimension, and transverse dimension. The gallbladder length, anteroposterior diameter, and transverse diameter have a linear relationship with gestational age between 15 weeks and 30 weeks of gestation, after which a plateau is observed. Calculated gallbladder area, volume, and sagittal perimeter have similar rates of growth in utero. The normative dimensions we have established for the fetal gallbladder throughout pregnancy can be expected to serve as a basis against which aberrant growth can be evaluated prenatally.

[Some biochemical indexes in white rabbit's blood affected by acute high intensity microwave].

Irradiation of white rabbits by 10, 50, 100 and 200 mW/cm2 microwave respectively can cause the disorder of protein metabolism, the abnormality of blood sugar, and the change of the activity of serum alpha-hydroxybutyrate dehydrogenase, lactate dehydrogenase, glutamic oxalacetic transaminase, glutamic pyruvic transaminase, acid phosphatase ect. These changes can be used as indexes in the evaluation of the effect of acute high intensity microwave exposure. The effect on the organism mainly depends on the intensity of exposure provided the dose of microwave remains the same.

[Quantitation of Ras p21 oncogene and DNA content of gastrointestinal smooth muscle tumors and prognostic significance].

Expression of ras p21 oncogene and DNA content of paraffin-embedded tissues from 55 smooth muscle tumors of the gastrointestinal tract were analysed simultaneously by using immunofluorescence and flow cytometry. All of 14 leiomyomas were found to be DNA diploid and lower-expression of ras p21. Four cases of DNA aneuploidy (33%) and 9 cases of ras p21 overexpression (75%) were found in 12 potential malignant smooth muscle tumors, while all 29 cases of leiomyosarcomas were aneuploidy (100%) (P < 0.005), but the rate of ras p21 overexpression was not increased continuously (72%) (P < 0.005). 24 of 33 aneuploid tumors (73%) were found to be ras p21 overexpression, while in 22 diploid tumors, there were only 6 cases (27%) (P < 0.005). The outcome of the patients with ras p21 lower-expressed diploid tumor was excellent, compared to the patients with ras p21 overexpressed aneuploid tumor it was worst (P < 0.005). It was suggested that DNA aneuploidy and ras p21 overexpression could be regarded as the mark of malignancy. The overexpression of ras p21 oncogene was always presented in the early stage of malignant smooth muscle tumors, when the cell proliferation was active but the DNA content was still normal, and stable in the whole procss of the tumor progression. ras p21 expression and DNA content could supply a deficiency each other in the diagnosis and could be used as objective parameters in distinguishing malignancy from benign and predicting the prognosis of the patients with smooth muscle tumors of the gastrointestinal tract.

[Influence of the 2nd and 3rd grade abstracts from scorpion venom of Buthus martensii Karsch on action potentials of fast response myocardiocytes].

AIM: To screen the active components of sodium channel blockade action from Buthus martensii Karsch (BMK) scorpion venom. METHODS: Myocardiocytes of mice were cultured. Action potentials of fast response myocardiocytes were recorded. The effects of 22 contents of BMK scorpion venom in a concentration of 3 mg.L-1 were tested and compared with that of TTX 2.5 mg.L-1, nimodipine (Nim) 3 mg.L-1 and BaCl2 24.4 mg.L-1. RESULTS: BMK-1 and other 19 contents significantly decreased depolarization parameters Vmax, APA, OS, and MDP, which was similar to that of TTX and different from that of Nim and BaCl2. CONCLUSION: The variations in the action potentials of fast response myocardiolcytes indicated that 20 contents of BMK scorpion venom had sodium channel blockade action.

[Idiopathic acute renal failure in nephrotic syndrome--a report of 10 cases].

Ten cases of idiopathic acute renal failure (IARF) in idiopathic nephrotic syndrome (NS) were reported. Heavy proteinuria and severe edema were the main clinical manifestations in these cases. Sudden oliguria, decrease of urinary osmolarity and increase of blood urea nitrogen and creatinine occurred without any difinite cause. Pathological examination showed normal or near normal glomeruli, diffuse interstitial edema and patchy necrosis of the tubular cells. The renal function in all the patients recovered after therapy with diuretics, prednisone, etc. It is shown that IARF in idiopathic NS commonly occurred in patients with normal or near normal glomeruli, for example, minimal change disease (6/10 cases), mild mesangial proliferative glomerulonephritis (4/10 cass). The incidence of IARF in idiopathic NS was 4.1% (10/245 cases), the IARF was mostly reversible.

[Sonographic diagnosis of ruptured corpus luteum with hemoperitoneum].

This article reports sonographic characteristics of a ruptured corpus luteum with hemoperitoneum in 44 cases. Sonographic findings were classified and compared with laparotomy and pathology. From sonographic imaging, it was found that the ruptured corpus luteum cyst was pathognomonic. There was a good correlation between ultrasound findings and surgical results when bleeding was over 100 ml. The sonographic value in diagnosis of ruptured corpus luteum with hemoperitoneum is discussed in detail.

[Ultrasonic diagnosis of abnormalities of abdominal wall in fetuses].

Twelve cases of abnormal abdominal walls in fetuses are reported in this paper. Among them, there were 4 cases of umbilical hernia, 4 cases of a defective abdominal wall, 3 cases of diaphragmatic hernia and 1 case of a defective thoracic and abdominal wall. From the features of the sonographic picture, the reasons for misdiagnosis are discussed. From our analysis, the main points of differential diagnosis and the earliest time for making diagnosis are proposed. The diagnostic rate could be improved obviously by systemic and careful examination which is important for deciding the model and time of deliveries.

[The level of soluble interleukin 2 receptor in esophageal carcinoma patients before and after radiotherapy].

The serum levels of soluble interleukin 2 receptor (sIL-2R) before and after radiotherapy in 57 esophageal carcinoma patients and 20 healthy controls were determined with an enzyme-linked immunosorbent assay. The objectives of this study were to determine the value of serum SIL-2R level as an index of tumor burden, and its predictive value in response to therapy and prognosis. The data showed that the serum sIL-2R level in esophageal carcinoma patients was significantly higher than that of healthy controls (P < 0.001). The elevated serum sIL-2R levels correlated with the length of the esophageal cancer. The serum levels of sIL-2R were significantly reduced at the end of radiotherapy (P < 0.001). Moreover, the serum level after radiotherapy was not different from that of healthy controls (P > 0.05). But, the reduced levels were not related to the immediate response to radiotherapy. These results seem to suggest that monitoring serum sIL-2R levels has clinical and prognostic significance in patients with esophageal carcinoma.

[Calcium channel blockade and anti-free-radical actions of panaxadiol saponins Rb1, Rb2, Rb3, Rc, and Rd].

AIM: To identify the calcium channel blockade and anti-free-radical actions of panaxadiol saponins Rb1, Rb2, Rb3, Rc, and Rd. METHODS: On ventricular myocardiocytes of Wistar rats, single channel activities of L, T, and B type Ca2+ channels were recorded with the cell-attached configuration of patch-clamp technic, and free radical contents were measured with electron spin resenance method. RESULTS: Rb1, Rb2, Rb3, and Rc 200 mumol.L-1 shortened the open times, prolonged the close times, and reduced the openstate probabilities of calcium channels and 30 mumol.L-1 antagonized the increase of free radical content induced by xanthine 0.42 mmol.L-1-xanthine oxidase 5.3 nmol.L-1, but Rd in the same dose behaved none of the effects. CONCLUSION: Rb1, Rb2, Rb3, and Rc had both the calcium channel blockade and anti-free-radical actions.

[Silicone oil removal in 31 cases].

Silicone oil removal was performed on 32 eyes of 31 cases after silicone oil injection for an average of 13.9 months. The reasons for the silicone oil removal are the complications such as cataract, glaucoma, emulsification of the silicone oil and so on. Complete retinal reattachment without any complications was found in 6 eyes of the 32 cases. Retinal reattachment remained in place in 24 eyes after the removal, and 24 eyes achieved postoperative visual acuities > or = 0.02. After the removal, in 7 of 8 eyes with complication, retinal detachment reoccurred. The significance and the timing of the silicone oil removal were discussed in this paper.

Factors related to long-term effects of thymectomy on myasthenia gravis.

The authors report the results of long-term follow-up after thymectomy on 92 cases with myasthenia gravis (MG), including 22 cases of thymomatous MG. These patients were followed up for 6-73 months with an average of 23 months. The surgically effective rate was 88.0% and the late mortality, 6.5%. The sex of the patients and the durations of disease were not obviously related to the prognosis, however the effect of thymectomy was much better in patients aged 10-39 years than in other age groups. The modified Osserman's clinical classification, acetylcholine receptor (AchR) antibodies and citric acid extract (CAE) antibodies titers in plasma and pathological patterns of the surgical specimens of thymus including immunohistochemical observation play an important role among various factors related to the long-term effects.

Treatment of biliary colic by water injection in the region of Qimen, Riyue, and Juque points.

Water injection was used in the treatment of 56 cases of biliary colic. The method was based on the writings of Zhang Zhongjing about the role of the Qimen point in treating diseases and clinical practice in this respect, water injection being performed at the most sensitive spot in the region defined by the lines joining the Qimen (Liv 14), Riyue (GB 24), and Juque (Ren 14) points. Pain disappeared in 32 (57.1%) of the 56 cases treated, and the total effective rate was 96.4%, showing a significantly better result compared with that of the control group treated with Western medicine (P < 0.05).

Quantitation of P53 protein expression in gastrointestinal smooth muscle tumors. Clinicopathological correlation and prognostic significance.

Quantitative analysis of P53 protein expression was performed on paraffin-embedded tissues from 55 smooth muscle tumors of the gastrointestinal tract, using immunofluorescence and flow cytometry. No positive expression was found in normal smooth muscle tissues of the gastrointestinal tract. Over-expression of P53 gene was found in a significantly higher proportion in leiomyosarcomas (90%) and potentially malignant smooth muscle tumors (75%) as compared to leiomyomas (14%) (P < 0.005). The quantitation of P53 expression was found to be progressively enhanced in the sequence from leiomyoma through potentially malignant smooth muscle tumor to leiomyosarcoma (P < 0.005). It was markedly over-expressed when the mitotic counts ranged from one to more than one per 10 high power fields (P < 0.005) or the mild cytologic atypia was found (P < 0.005). The five-year survival rate was significantly higher in patients with low-expression of P53 than in those with over-expression of P53 (P < 0.005). It was suggested that P53 over-expression might be associated with the transformation of leiomyoma into leiomyosarcoma and could be used as an objective parameter in distinguishing the malignant from the benign and predicting the prognosis of patients with smooth muscle tumors of the gastrointestinal tract.

Characteristics of an established retinoblastoma cell line HXO-Rb44.

PURPOSE: To study the retinoblastoma cell culture and to establish a new retinoblastoma cell line. METHODS: 22 retinoblastomas were cultured by using the method of single cell suspension. Characteristics of the cultured cells were studied in the following programs: tumor cell morphology in vitro, electron microscopic, growth curve, cloning in soft agar, immunohistochemistry, karyotype and tumorigenicity. RESULTS: 22 retinoblastomas were cultured successfully in vitro, only a continued cell line HXO-Rb44 was established (more than 3 years). The characteristics of this cell line are that it grew as a suspension of round cell in graps like clusters in vitro, its population doubling time was 44 hours, and it could be cloned in soft agar. Histopathologic and ultrastructured pictures showed the characteristics of Rb. HXO-Rb44 cell was positive to NSE and negative to GFAP in immunohistochemical staining. A subcutaneous injection of HXO-Rb44 cells produced a retinoblastoma in BALB/C athumic nude mice. CONCLUSIONS: HXO-Rb44 has the characteristics of retinoblastoma and is a new retinoblastoma cell line. It is a useful material for study this tumor both in basic and clinical fields.

[Immunohistochemical analysis of nm23 gene product/nucleoside-diphosphate kinase expression in human lung carcinoma].

It was proposed that nm23 gene may function as a suppressor gene for tumor metastasis. Using an antibody to NDPK, levels of NDPK/nm23 expression in lung carcinomas were examined immunohistochemically, 45 of 58 squamous carcinomas and 26 of 36 adenocarcinomas showed strong immunoreactivity for NDPK/nm23 in most of cancer cells within tumor tissues. On some normal bronchial epithelium and serous glands, there also show weak expression. No relationship was found between NDPK/nm23 expression and clinicopathological parameters including tumor grade, size, nodal involvement and stage but the increases in NDPK/nm23 expression were stronger in advanced stages of squamous cell carcinomas. In conclusion, our results indicate that the increased NDPK/nm23 expression in the analysed carcinomas is not consistent with the proposed metastasis-suppressor function, but the nm23 gene may be implicated in the mechanism of carcinogenic process and tumor progression.
