[Regulation of inflammatory responses by MAPK signal transduction pathways].

Mitogen activated protein kinase (MAPK), which is one of the important signal transduction systems in organisms, is involved in many cellular processes, such as cell growth, development, division, differentiation, death and coordination of cellular functions, and etc. Four subfamilies of MAP kinases, i e ERK, JNK/SAPK, p38/RK and ERK5/BMK1, have been identified and cloned in mammalian cells These MAP kinases are activated by many proinflammatory stimuli and play an important role in the pathogenesis and development of inflammation. In this article recent advances in the study of the mechanisms underlying activation of MAPKs in infection and inflammation and the molecular basis of specific inhibitors for MAPKs are reviewed, in special reference with the perspective prevention and treatment of inflammation by these kinases.

[Effect of oxidatively modified high density lipoprotein on transcriptional expression of proto c-fos and PDGF receptor genes in cultured human arterial smooth muscle cell].

The proliferation of arterial smooth muscle cell (SMC) plays an important role in the process of atherosclero-genesis. Using the establishment of primary culture and sub-culture method for human arterial SMC, we observed the effects of native HDL (N-HDL) and oxidized HDL of proto c-fos and PDGF receptor genes transcriptional expression in cultured SMC. The results showed that N-HDL had no effect on the expression of proto c-fos and PDGF receptor genes in the cultured human SMC, that OX-HDL had no effect on the expression of PDGF receptor gene in the cultured human SMC, and that OX-HDL promoted the expression of protoc-fos gene significantly (P < 0.01). These results suggest that the OX-HDL's atherogenic role is closely related to its stimulating effect on proto c-fos gene expression of SMC.

[The relationship of cellular DNA content with clinical stage and biological features of colorectal cancer].

The purpose of this study was to explore the relationship of cellular DNA content with clinical stage and biological features of cancer. Flow cytometry was performed on fresh specimens from 86 patients from 1997 to 1998. Forty-five (53.3%) specimens were found to contain cells with abnormal DNA (DNA nondiploidy). Although none of the sex, age, site, differentiation variables correlated with flow cytometric DNA ploidy, nondiploidy was associated with Dukes' stage and lymph node metastasis. Duke's A stage tumors were more frequently diploid than were more advanced tumors, but no difference between Duke's B, C and D stages were observed. These findings suggest that ploidy is associated with some pathological factors.

[Expressions of apoptosis-associated proteins in prostate carcinoma and their clinicopathological significance].

This study was made to explore the expressions of P53, P16, Bc1-2 and Bax protein in prostate carcinoma and their relations to histopathological grading, clinical staging and prognosis of this tumor. Immunohistochemical staining method was used to assess the expression of P53, P16, Bc1-2 and Bax protein in 51 cases of prostate carcinoma. The positive rates of P53, P16, Bc1-2 and Bax proteins were 31.37%, 54.90%, 21.57% and 74.51% respectively. The expression of P53 and Bc1-2 showed positive correlation with the histopathological grading and clinical staging of this tumor (P < 0.05). However, the expression of P16 and Bax showed negative correlation with the grading and staging of the tumor (P < 0.05). The expression of P53 protein correlated with the survival rate of these patients. There was a relation between P53 and Bc1-2, and a relation between Bc1-2 and Bax also (P < 0.05). These findings suggest that expressions of P53, P16, Bc1-2 and Bax may be helpful to the grading, staging and prognostic evaluation of prostate carcinoma.

[Quantification analysis of vascular endothelial cells in human solid tumor].

Angiogenesis is essential for the growth and metastasis of solid tumors. Intratumoral microvessel count, which represents a measure of tumor angiogenesis, has been associated with prognosis of patients with a variety of malignancies. But till now, it is still a problem to quantitate the angiogenesis of tumors exactly. In this study, the endothelial cells in 20 solid tumors were conjugated with anti-CD34 monoclonal antibodies labelled with red-fluorescent substances, and were observed by fluorescence microscopy and quantitated by flow cytometry. The results of flow cytometry were analysed by t-test. The correlation was analysed between percentage of flow cytometry and vessel counts (light microscopy, x 200) in each tumor which was stained with anti-CD34 monoclonal antibodies and anti-VIII factor related antigen polyclonal antibodies by using immunohistochemistry. The results showed there was significant difference between the treatment group and control group in flow cytometry, and a significant correlation between the endothelial cells percentage by flow cytometry and the vessel counts by immunohistochemistry was observed, although the average vessel counts were approximately three times with CD34 staining as much as with VIII factor related antigen staining. These data indicate that flow cytometry as a novel method can quantitate angiogenesis of tumors exactly and quickly.

[Dynamic study of hepatocellular pathological change and uptake rate of 125I-insulin during experimental hepatocarcinogenesis].

This study was designed to observe the hepatocellular pathological change and uptake rate of 125I-insulin during experimental hepatocarcinogenesis in rats and clarify the possible mechanism of increasing uptake of 125I-insulin. 80 SD rats were divided into 2 groups, control and experimental groups. All of the rats were given common feed, and the rats of experimental group were given extra diethylnitrosamine (DENA) 70 mg/kg each week. At 6th, 11th, 15th and 20th week after start of the experiment, 10 control and 10 experimental rats were killed 1 hour post administration of 125I-insulin by tail vein. Blood, liver, lung, spleen, kidney, muscle and bone were collected; the radioactivity was measured and calculated %ID/g. The data of 2 groups were compared and examined with t-test. All of livers were pathologically examined. The results showed all livers of control group were normal. At 6th week, the surfaces of experimental rats' livers were coarse. Hepatocellular hyperplasia was observed, 125I-insulin-uptake rate was 1.86 time as much as that of control group. At 11th week, the liver's colour became lighter than that of control group. Hyperplasia and hepatocirrhosis were observed, 125I-insulin-uptake rate was 1.50 time as much as that of control group. At 15th week, hyperplastic nodules were observed in all experimental rats' livers. Hepatomacellulae were observed in 6 rat livers. 125I-insulin-uptake rate was 1.56 time as much as that of control group. At 20th week, the livers became enlarged out of shape. There were a lot of big or small greyish white nodules in all livers. Necrosis, liquefaction and hemorrhage were observed. Hepatomacellulae were observed in all of experimental rats livers. 125I-insulin-uptake rate was 1.46 time as much as that of control group. The differences of 125I-insulin-uptake rate between experimental and control groups were significant. These results demonstrated that the liver ability of uptaking 125I-insulin increased which mainly took place during the period of hepatocellular hyperplasia, and the hepatomacellulae kept this characteristic.

[Gastro-pyloro-duodenal motor cooperation in patients with dyspepsia].

OBJECTIVE: To investigate the role of the midgastric transverse band(MTB) and gastro-pyloro-duodenal motility in functional dyspepsia (FD) and diabetic dyspepsia (DD). METHODS: Healthy subjects, patients with FD and DD participated in the study. Of them, 65 subjects received radionuclide gastric emptying, 48 complex ultrasonograph, and 35 gastro-pyloro-duodenal manometry. RESULTS: (1) MTB was (7.01 +/- 1.31), (3.39 +/- 0.21) cm in FD and DD, which was significantly wider than that in HS group [(2.42 +/- 0.16) cm, P < 0.01, P < 0.001]. (2) The half gastric emptying time (t 1/2) in FD and DD were prolonged, gastric remained rate at 120 min increased, the gastric emptying velocity reduced when compared with HS. (3) The gastric occlusive and non-occlusive peristaltic contractions could be clearly shown under ultrasonograph. The percentage of emptying circle > 10 seconds was reduced in FD and DD. (4) There was decreased number of migrating motor complex phase III in FD and DD, when compared with HS. CONCLUSIONS: MTB is an area with physiological function, which affects intra-gastric food distribution. Patients with FD and DD showed delayed gastric emptying associated with decreased gastric motor activity and antro-pyloro-duodenal in-cooperative function. There may be different pathogenesis in FD and DD. Further study needs to be performed.

[Effect of Tripterygium Wilfordii Hook T4 monomer on proliferation and interleukin-6 production of synovial fibroblasts of patients with rheumatoid arthritis].

OBJECTIVE: Tripterygium Wilfordii Hook multi-glycosides T2 has been wildly used in China in treatment of RA. T4 was abstracted from T2 and was reported much more efficient in anti-inflammatory and immune suppression than T2. This study was to investigate the effect Tripterygium Wilfordii Hook T4 monomer on proliferation and interleukin-6 production of synovial fibroblasts of patients with rheumatoid arthritis. METHODS: Synovium was obtained from patients with rheumatoid arthritis undergoing synovectomies or joint replacement. Cultures of synovial fibroblasts were established. After 3 generations, cultured synovial fibroblasts were stimulated with IL-1. Then 1.5 ng/ml, 5 ng/ml and 15 ng/ml T4 were added, and synovial fibroblasts were cultured in the presence of T4 for 48 hours. Cell proliferation was assayed using MTT method. IL-6 level of supernatant was measured by ELISA. RESULTS: Proliferation of synovial fibroblasts was inhibited by T4. The proliferation inhibition effect of T4 was dose dependent and inhibition rate was 5.18%, 10.95% and 21.37%, respectively. And T4 had no effect on IL-6 production by IL-1 stimulated synovial fibroblasts. CONCLUSIONS: T4 might control the disease activity of RA by inhibiting the proliferation of synoviocyte. And T4 might not influence the concentration of IL-6 in synovial fluid, as a central effect, since IL-6 has protective effect on articular cartilage.

[In-situ FTIR spectroscopic studies of electro oxidation of ethanol in alkaline media at a nm-Pt/GC electrode].

The electro-oxidation of ethanol on a nm-Pt/GC electrode in alkaline solutions was investigated by using cyclic Voltammetry and in-situ FTIR spectroscopy. The results demonstrated that the main product of ethanol oxidation was CH3COO-, only small quantity of CH3CHO was determined simultaneously. In contrast with the dual path reaction mechanism for the oxidation of ethanol in acid media, the oxidation of ethanol in alkaline media was revealed via the intermediate process of dissociative adsorption.

[In situ FTIR studies of CO adsorption on palladium loading zeolite electrode].

Adsorption of CO on palladium loading zeolite electrode supported on Pt or GC substrate was studied by in situ MSFTIR spectroscopy. The results demonstrated that the geometry of faujasite supercages favors the formation of bridge-bonded CO species. The enhanced IR adsorption of CO adsorbed on Pd clusters located in the supercages of zeolite has been observed for the first time, which is a new phenomenon and differs from the abnormal infrared effects discovered by our group and the surface enhanced infrared absorption phenomenon reported in the literature.

[Photoluminescence characteristics of mixed aggregates in Langmuir-Blodgett multilayers].

Effects of the mixed aggregates of different active molecules and UV illumination on photoluminescence(PL) spectra of Langmuir-Blodgett multilayers were investigated. The strong interaction between hemicyanine and stilbazolium salt molecules in the mixed films caused a blue or red shift of the fluorescence band with respect to that of interleaving hemicyanine or stilbazolium salt/arachidic acid multilayers. UV illumination would dissociate the aggregates even more destroy the molecular structure, leading to a significant reduction of fluorescence intensity.

Micro CT and Micro MR imaging of 3D architecture of animal skeleton.

Quantitative assessment of three-dimensional (3D) trabecular structural characteristics may improve our ability to understand the pathophysiology of osteoporosis, to test the efficacy of pharmaceutical intervention, and to estimate bone biomechanical properties. Considerable progress has been made in advanced imaging techniques for noninvasive and/or nondestructive assessment of 3D trabecular structure and connectivity. Micro computed tomography (microCT) has been used to measure 3D trabecular bone structure in rats, both in vivo and in vitro. It can directly quantify 3D trabecular bone structure such as trabecular volume, trabecular thickness, number, separation, structure model index, degree of anisotropy, and connectivity, in a model-independent manner. We have used microCT to study ovariectomy (OVX) induced osteopenia in rats and its treatment with agents such as estrogen, and sodium fluoride. We have demonstrated that 3D microCT can quantify mouse trabecular and cortical bone structure with an isotropic resolution of 9 microm(3). It is also useful for studying osteoporosis in mice and in phenotypes of transgenic mice or gene knockout mice. MicroCT can be used to quantify osteogenesis in mouse Ilizarov leg lengthening procedures, to quantify osteoconduction in a rat cranial defect model, and to quantify cortical bone porosity. Recently, microCT using high intensity and tight collimation synchrotron radiation to achieve spatial resolution of 1-2 microm has provided the capability to assess additional features such as resorption cavities. Unlike microCT, micro magnetic resonance imaging (IMRI) is nonionizing. Recently, the ability of microMRI to assess osteoporosis in animal models has been explored. Using a small, high-efficiency coil in a high-field imager, microMRI can give resolutions sufficient to discriminate individual trabeculae. We have shown that, with appropriate settings, it is possible to image trabecular bone in rats in vivo and in vitro. In our study of OVX rats, analysis of microMR images can demonstrate differences in rat trabecular bone that are not detected by DXA measurements. In a rabbit OA model, with the OA induced by meniscectomy or anterior cruciate ligament transection, MRI shows decreased cartilage thickness, subchondral osteosclerosis and osteophytes, while radiographs can only show subchondral osteosclerosis and osteophytes could not be found. Advanced imaging methods are able to measure 3D trabecular structure and connectivity in arbitrary orientations in a highly automated, objective, non-user-specific manner, allowing greater numbers of samples for unbiased comparisons between controls and the disordered or treated. They can be utilized on a large sample leading to fewer sampling errors. They are non-destructive allowing multiple tests such as biomechanical testing and chemical analysis on the same sample; and non-invasive permitting longitudinal studies and reducing the number of animals needed.

A role for GATA-4/5/6 in the regulation of Nkx2.5 expression with implications for patterning of the precardiac field.

Interactions between the key regulatory genes of the cardiogenic pathway, including those from the GATA and Nkx2 transcription factor families, are not well defined. Treating neurula-stage Xenopus embryos with retinoic acid (RA) causes a specific block in cardiomyocyte development that correlates with a progressive reduction in the region of the presumptive heart-forming region expressing Nkx2.5. In contrast, RA does not block expression of the GATA-4/5/6 genes, which are transcribed normally in an overlapping pattern with Nkx2.5 throughout cardiogenesis. Instead, GATA-4/5/6 transcription levels are increased, including an expansion of the expression domain corresponding to lateral plate mesoderm that is part of the early heart field, but that normally is progressively restricted in its ability to contribute to the myocardium. GATA-dependent regulatory sequences of the Nkx2.5 gene that implicate GATA-4/5/6 as upstream positive regulators were described recently. However, our experiments also indicate that GATA factors might normally antagonize transcription of Nkx2.5. To test this hypothesis we generated a dominant negative isoform of GATA-4 (SRG4) capable of inhibiting transcription of GATA-dependent target genes. Ectopic expression of SRG4 results in a transient expansion of the Nkx2.5 transcript pattern, indicating that a normal function of GATA factors is to limit the boundary of the Nkx2.5 expression domain to the most anterior ventral region of the heart field. Regulatory mechanisms altered by excess RA must function normally to limit GATA-4/5/6 expression levels, to define the region of Nkx2.5 expression and regulate myocardial differentiation.

Induction of tumor suppression and glandular differentiation of A549 lung carcinoma cells by dominant-negative IGF-I receptor.

Overexpression or activation of insulin-like growth factor I receptor (IGF-IR) has been observed in many human cancers including breast, lung, colon and gastric carcinomas. We demonstrate that inhibition of the endogenous insulin-like growth factor I receptor by stable expression of a dominant-negative IGF-IR represses the transforming activity in vitro and tumorigenicity of human lung carcinoma cells A549 in vivo. The suppression of tumorigenicity in nude mice is correlated with the induction of glandular differentiation. In addition, functional inhibition of the endogenous receptor dramatically increases the sensitivity of A549 cells to a variety of apoptotic signals including UV irradiation and proteasome inhibitors. These effects are due to the formation of a stable heterocomplex of the dominant-negative receptor with the endogenous wild type receptor which reduces the kinase activity of the latter by twofold. Thus, inhibition of the IGF-IR signaling pathway not only suppresses tumorigenicity but also enhances sensitivity to apoptosis-inducing agents. Antagonizing IGF-IR signaling by promoting tumor differentiation and enhancing sensitivity to apoptotic death are potential cancer therapeutic approaches.
