Effects of sofalcone on necrotizing agents-induced gastric lesions and on endogenous prostaglandins in rats stomachs.

Attempts were made to investigate the effect of 2'-carboxymethoxy-4,4'-bis(3-methyl-2-butenyloxy) chalcone (sofalcone) on necrotizing agents-induced gastric lesions and on prostaglandin E2 (PGE2)-like activity of the gastric tissue in rats. Sofalcone, 100 mg/kg i.p. and 300 mg/kg p.o., markedly suppressed 0.6 N HCl- or 100% EtOH-induced gastric lesions. Sofalcone, 100 mg/kg i.p., also significantly suppressed 0.2N NaOH-induced gastric lesions. Sofalcone suppressed 0.6 N HCl-induced gastric lesions with both oral and intraperitoneal routes, and the effect was particularly marked at 60 min. A dose of 100 mg/kg i.p. showed suppression lasting for up to 300 min. 0.6 N HCl-induced gastric lesions were significantly aggravated by indomethacin treatment (10 mg/kg s.c.). Oral sofalcone (300 mg/kg) significantly suppressed the aggravation of gastric lesions by indomethacin given before and after sofalcone, but the i.p. (100 mg/kg) did not show significant suppression in the case of pretreatment with indomethacin. PGs-like activity in the gastric tissue was increased in both of the fundus and the antrum by the administration of sofalcone without any dose-dependency. The increase was continuous and lasted for 6 h in the fundus of the stomach.

[Curative limb saving operation (curative wide resection)].

The recent results of surgical treatment of soft tissue sarcomas have dramatically improved. The result of the group study of soft tissue sarcomas, which was supported by the Ministry of Health and Welfare (1962-1976) showed that the local recurrence rate was 50%, and the metastatic rate was 60%. In contrast with this result, the recent results of our curative wide resection showed the local recurrence rate of 10%, and the metastatic rate of 30%. These procedures are based on the clinico-pathological findings which we studied. Curative wide resection is a method of radical wide resection, similar to radical local resection (reported by Enneking) and radical local excision (reported by Stenar). Our procedures are the least aggressive of the three radical wide resections, but our results are the same as the other wide resection methods. To obtain good results with these procedures, accurate preoperative diagnosis and skillful surgery are necessary.

Polyamine inhibition of gastric ulceration and secretion in rats.

The effect of polyamines on gastric ulceration and secretion in rats was studied. Stress-induced gastric ulceration and ulceration in pylorus-ligated rats were inhibited by subcutaneous or oral administration of spermine; spermidine's inhibitory effect was somewhat less. Histamine-induced ulceration was also inhibited by the subcutaneous injection of spermine. In addition, the daily oral administration of spermine for 10 days was therapeutic against an acetic acid-induced ulcer (chronic ulcer). Gastric secretion in pylorus-ligated rats and in rats with fistulae and stimulated by histamine injection was decreased by the subcutaneous injection of spermine.

Myofibrillogenesis in vitro as seen with the scanning electron microscope.

The present study describes an experimental approach whereby myofibrillogenesis and the three-dimensional arrangement of myofibrils present within cultured skeletal muscle cells can be examined using the scanning electron microscope. This procedure uses cells that have been cultured on gold-coated coverslips, and treated with Triton X-100 to extract the cell membrane and the soluble cytoplasm. Subsequent electroconductive staining by treatment with thiocarbohydrazide and osmium allows the myofibrils to be visualized. The images of myofibrils in various states of development observed by this method generally accords to those previously reported by transmission electron microscopy. Cell elongation and adhesion to the substrate causes mechanical stress from different directions which meet at branchings of the cultured myotubes. Many myofibrils are observed to run in the direction of the inferred stress lines.

Effect of 2'-carboxymethoxy-4,4'-bis(3-methyl-2-butenyloxy)chalcone (SU-88) on gastric local blood flow.

Effect of 2'-carboxymethoxy-4,4'-bis(3-methyl-2-butenyloxy)chalcone (SU-88), a new anti-ulcer drug, on gastric tissue blood flow was investigated by using an inhaled hydrogen gas clearance method in rats. As a result, following the intravenous infusions of 10,20 and 30 mg/kg/h of SU-88, the gastric blood flow increased with an increase rate of 38.1, 70.5 and 61.7% as compared with the control value, respectively. Following the intraperitoneal administrations of 50 and 100 mg/kg of SU-88, the gastric blood flow increased by 59.8 and 51.2%, respectively, immediately after the administration.

A Japanese lower limb prosthesis with a fore-joint foot and turntable.

Amputees wearing conventional prostheses have often found difficulty when conforming with the conventions of Japanese social life, since the prosthesis is not well adapted to walking in bare feet or sitting cross-legged. The authors described a modified prosthesis with a spring-loaded rotation mechanism that can be initiated unobtrusively and a modified foot piece in which the ankle motion is transmitted via a lever arm to a joint placed more anteriorly than usual. Nearly 250 patients have been fitted with the prosthesis and 40 have undergone laboratory gait analysis. The results indicate a walking cycle and foot pressure pattern more closely resembling the normal, than that obtained by other prosthetic systems.

The etiology of caffeine-induced aggravation of gastric lesions in rats exposed to restraint plus water-immersion stress.

Caffeine augmented formation of gastric lesions in rats exposed to restraint plus water-immersion stress for 3 h, together with inhibition of gastric secretion and gastric motility. The etiology of the lesion aggravation by caffeine was studied by measuring several parameters which were relevant to the mucosal defensive factors. Caffeine increased the susceptibility of gastric mucosa to erosive action of acid when simultaneously combined with the stress procedure. Injection of caffeine via an intraperitoneal route, but not via an intraarterial route, caused a decrease in gastric tissue blood flow when no marked fall in body temperature was seen. This indicates that the caffeine-induced decrease in gastric tissue blood flow is not due to its direct action on the gastric mucosal vessels. Hexosamine content of gastric wall tissues, however, was not significantly changed in caffeine-pretreated rats at 3 h after stress. These results suggest that caffeine aggravates stress-induced gastric lesions through some mechanism in reducing mucosal blood flow which is responsible for liability of mucosal autodigestion.

[Effects of the anti-ulcer drug SU-88 on the gastric blood flow and the cardiovascular system (author's transl)].

Effects of a newly synthetized anti-ulcer drug, 2'Carboxymethoxy-4,4'-bis(3-methyl-2-butenloxy)chalcone (SU-88), on gastric blood flow and the cardiovascular system were investigated. Intravenously administered SU-88 dose-dependently increased the blood flow of gastric tissues in rats. In anethetized dogs, i.a. administered SU-88 also increased the short gastric and the femoral arterial blood flows. SU-88 (i mg/kg, 3 mg/kg and 10 mg/kg i.v.) caused a mild fall in systemic blood pressure, an increase in heart rate, and a stimulation of respiration; however, these effects were only transient and recovered to the basal level within few minutes. No notable changes were observed in the electrocardiogram except for a shortening of the R-R interval accompanied by an increase in heart rate. The blood flow of the common carotid, the vertebral, the coeliac, the superipor mesenteric, and the femoral arteries in dogs were all increased by i.v. administration of SU-88, and among them an increase in the coeliac arterial blood flow was prominent. SU-88 increased the perfusates in isolated rabbit ear and relaxed the contraction of isolated rabbit aorta induced by noradrenaline, but showed no influences on the amplitude, heart rate, and coronary flow of isolated rabbit heart. The hypotensive effect of SU-88 was not abolished by i.v. administered atropine, diphendydramine, and propranolol and not influenced by vagotomy. Intraverterbrally administered SU-88increased the vertebral blood flow, but showed no influence on systemic blood pressure. Furthermore, SU-88 showed no influence on the rise in blood pressure by both common carotid occlusions. The increase in the femoral and the short gastric arterial blood flow induced by i.a. administration of SU-88 was not inhibited by atropine, diphenhydramine, and propranolol and not influenced by pretreatment with aminophylline. These results suggest that the hypotensive and vasodilative effects of SU-88 are not mediated by the central nervous system, autonomic nervous system, histamine and the action of adenosine augmentation and further experiments are necessary for precise explanations of its mechanism.
