[A study of prostatic tissue levels of cefoperazone].

Prostatic tissue levels of Cefoperazone (CPZ) were studied after the administration of 3 g of CPZ, in 16 patients with prostatic hypertrophy who underwent retropubic prostatectomy. The CPZ concentration in the surgical capsule at 60 min. after the administration of CPZ was about 2 times higher than that in the gland. As compared with the histological type of removed gland, the levels in the glandular hyperplasia were slightly higher than that in the fibromuscular and mixed type hyperplasia, but the difference was not statistically significant. CPZ was found to be effective for the treatment of post-operative and bacterial prostatitis, since the CPZ levels of prostatic tissue were considerably high and above the minimal inhibitory concentration level for most bacteria.

[Clinical studies of bestatin on genitourinary cancer].

Bestatin was administered to 20 patients with urogenital tumors. The therapeutic results showed 12 surviving patients and 8 dead patients (including 7 due to cancer and 1 due to a cerebrovascular disorder). One patient has survived for 5 years since bestatin treatment of pulmonary metastasis of renal cancer, and one other patient achieved 5-year survival in spite of systemic metastases of testicular tumor. It is noteworthy that in earlier times these patients could have been expected to experience sudden relapse and aggravation, instead of the long-term survival recorded in this bestatin trial. Studies were conducted on the immunological parameters of the patients before and after the use of bestatin, and it was found that these parameters showed improvement as a result of the treatment. Even though bestatin was ingested for long periods of time by these patients, there was almost no development of adverse reactions to the treatment.

Effects of FPL-52694, a new mast cell stabilizer, on gastric secretion and various acute gastric lesions in rats.

Oral FPL-52694 [5-(2-hydroxypropoxy)-8-propyl-4-oxo-4H-1-benzopyran-2-carboxylic acid Na], a new mast cell stabilizer, dose-dependently inhibited gastric acid secretion but increased the volume and pepsin output in pylorus-ligated rats. Intraduodenal FPL-52694 significantly inhibited all of the volume, acidity, acid output and pepsin output. Concerning the acidity, oral administration of the agent showed much more potent inhibition than intraduodenal administration. Oral FPL-52694 markedly inhibited the development of pylorus-ligated ulcers, water-immersion stress- and aspirin-induced gastric erosions and moderately inhibited the formation of reserpine-induced gastric erosions in rats. Intraduodenal FPL-52694 also inhibited pylorus-ligated ulcers whereas it had no effect on aspirin-induced gastric erosions. Histamine-induced gastric erosions were not affected by oral FPL-52694. These effects of FPL-52694 were almost the same as those of cimetidine, except that cimetidine tended to inhibit histamine-induced gastric erosions. Although the precise mechanism of action of FPL-52694 remains unknown, oral FPL-52694 appears to be a promising agent for the treatment of peptic ulcers.

Effect of thromboxane synthetase inhibitors (OKY-046, OKY-1580) on the action of bronchoactive agents in guinea pig tracheal strips and on arachidonate metabolism in guinea pig lung lobes.

We investigated the effect of the thromboxane synthetase inhibitors, OKY-046 and OKY-1580, on the action of bronchoactive agents in guinea pig tracheal strips, and on arachidonic acid metabolism in isolated perfused guinea pig lung lobes. OKY-046 and OKY-1580 attenuated histamine-, serotonin-, acetylcholine-, bradykinin- and prostaglandin F2- induced contractile responses in guinea pig tracheal strips dose-dependently and potentiated isoproterenol-, salbutamol- and prostaglandin E2-induced relaxation in guinea pig tracheal strips dose-dependently. In addition, OKY-046 and OKY-1580 inhibited the biosynthesis of thromboxane A2 and accelerated the production of 6-keto prostaglandin F1 from arachidonic acid in isolated perfused guinea pig lung lobes. The above results suggest that OKY-046 and OKY-1580 might be useful therapeutic agents for the treatment of pulmonary thromboembolism and chronic obstructive lung diseases.

The trend of acquired immunity with live poliovirus vaccine and the effect of revaccination: follow-up of vaccinees for ten years.

The persistence of neutralizing antibody (NA) against three types of poliovirus acquired after two doses of trivalent live attenuated poliovirus vaccine (LPV) has been followed up for ten years in individual vaccinees. Sixty-seven children were bled once a year over a five year period following the primary vaccination. More than 80% of them retained NA against all three types of poliovirus. Thirty-two individuals whose NA titres were 1:16 or over for types 1 and 2 and 1:4 or over for type 3 at the fifth year were further followed up for a further five years and it was shown that during this period some of them had a naturally-acquired antibody rise, mostly against type 3 virus. At the sixth to eighth year after the primary vaccination, one further dose of the trivalent vaccine was administered to the children whose NA titres were down to 1:8 or less and the effect of booster vaccination on NA was followed. Other subjects were revaccinated with LPV and their fecal excretion of the vaccine virus was investigated. The results showed that a decrease in serum antibody level could be a good indicator of the local resistance of the alimentary tract and that reinfection could occur if serum NA had decreased to 1:8 or less, which allowed a virus excretion in the stools.

Serum lipid and apolipoprotein profiles after onset of acute myocardial infarction.

The concentrations of lipids and apolipoproteins in serum following onset of acute myocardial infarction were measured periodically in eleven patients for 28 days. Significant and sustained decrease in the concentration of HDL-cholesterol was observed by the seventh days post-infarction and remained low throughout the remainder of the study. The concentration of apo A-I and apo A-II assayed by electroimmunoassay was significantly low by 4th and 5th days post-infarction, remained low until day 10 and then began to increase but were not back to the initial values by 28 days post-infarction. Apo B, C-I, C-II, C-III, and E levels remained unchanged throughout the course. The concentrations of total cholesterol, triglycerides tended to fall reaching the lowest levels on day 7 and 10, but were not significant. The concentration of serum phospholipids level tended to decrease and fall significantly by day 7 and 10. The concentration of the free fatty acids in the serum was elevated during the first 2 days post-infarction and was within the normal range throughout the remainder of the study.
