Dissociation of K+-induced tension and cellular Ca2+ retention in vascular and intestinal smooth muscle in normoxia and hypoxia.

In experiments on smooth muscle preparations of rabbit aorta and guinea pig taenia coli, replacement of the external Na+ with K+ produced sustained contraction. When external K+ concentration was increased, cellular Ca2+ retention as measured by a modified lanthanum technique increased. However, when K+ concentration was above 80 mM, the tension decreased despite an increase in Ca2+ retention. Maximum amount of Ca2+ retained was 1280 nmol/g in aorta and 980 nmol/g in taenia coli while the control values for both tissues were approximately 430 nmol/g when the external Ca2+ concentration was 2.5 mM. Under hypoxia (N2 aeration), sustained contraction was induced by 80 mM K+ in aorta and by 45.4 mM K+ (and 55 mM glucose) in taenia coli. However, no increase in the cellular Ca2+ retention was observed under these conditions. During the K+-induced sustained contraction in aorta, introduction of N2 transiently increased, while readmission of O2 transiently decreased the muscle tension. In taenia coli, the introduction of N2 decreased the sustained contractile tension probably because of an ATP deficiency, while the readmission of O2 further decreased the tension transiently. From these results, it is concluded that, in the presence of a high concentration of K+, external Ca2+ enters the cell and activates the contractile machinery. A part of the cellular Ca2+ is taken up by mitochondria under normoxic but not under hypoxic conditions.

Relaxing and metabolic inhibitory action of X537A (Lasalocid) on the taenia of the guinea-pig caecum.

1. In the isolated guinea-pig taenia, application of X537A to the muscle caused a relaxation, while A23187 caused a sustained contraction. 2. Treatment with adrenergic blocking agents and tetrodotoxin had no effect on the relaxing response to X537A. 3. In the presence of a low concentration (10(-6) M) of X537A or under hypoxic conditions (bubbled with 95% N2: 5% CO2), the taenia lost the ability to respond to a high concentration (40 mM) of potassium with a sustained tonic contraction, although the rapid phasic contraction was still present. 4. When a low concentration of X537A was applied the shape of the contractile response to A23187 was changed from a sustained development of tension to an oscillatory tension which was also observed under hypoxia. 5. In tissues treated with glycerol for 3 weeks, neither X537A nor A23187 had any effect on the contractile response induced by elevation of calcium concentration (pCa = 4) in the presence of magnesium and ATP. 6. Exposure to a low concentration (10(-6) M) of X537A, or hypoxia, slightly yet significantly decreased the ATP content of the muscle. A high concentration (10(-5) M) of X537A markedly decreased the ATP content to about half normal. A23187 did not alter the ATP content. 7. Application of X537A did not alter the calcium content of the muscle and inhibited the A23187-induced increase in content. Under hypoxia, A23187 failed to increase the calcium content of the muscle. 8. The results indicate that, in contrast to A23187, X537A has a relaxing and metabolic inhibitory action on the guinea-pig taenia. The action of low concentrations of X537A resembled that of the hypoxia, indicating that X537A might exert its relaxing action, at least in part, by inhibition of aerobic energy metabolism of the muscle.

Role of glycolysis in the tension development under anoxia in guinea pig taenia coli.

The role of glycolysis in the tension development under anoxic conditions in a high-K medium was studied in the intestinal smooth muscle of guinea pig taenia coli. After exposure to the high-K medium (isotonic, 60 mM) under normal oxygen for 30 min, the muscles were exposed to a high-K medium bubbled with N2 gas. The tonic contraction decreased gradually to about 10% of the original level. Glucose was then cumulatively added to the high-K medium under anoxia. The maximum tension was observed following the addition of the higher concentrations of glucose. The muscle tension which developed in the high-K medium with a high concentration of glucose under anoxia was dependent on the external Ca2+ and was inhibited by iodoacetic acid (IAA). The addition of glucose to a high-K medium under anoxia also increased lactate release from the muscle. Pretreatment with 1 mM IAA decreased the lactate release from the muscle. In a Ca2+-free medium under anoxia, the addition of glucose did not increase the muscle tension although there was a significant increase in the lactate release. In summary, it is considered that the smooth muscle of taenia coli develops tension utilizing energy produced by the glycolytic pathway under anoxia in a high-K medium.

High K+,Na+-deficient solution inhibits tension, O2 consumption, and ATP synthesis in smooth muscle.

In guinea pig taenia coli, added 45.4 mM K+ induced a sustained contraction, increased the rate of oxygen consumption, and slightly decreased the ATP content. In substituted 154.2 mM K+, Na+-deficient solution, only a transient contraction was induced. Oxygen consumption also showed only a transient increase and ATP content of the muscle rapidly decreased. Such an inhibition of sustained contraction and the decrease in both oxygen consumption and ATP content were recovered when 5.5 mM pyruvate or 50 mM NaCl was added during the 154.2 mM K+-induced contraction. In rabbit aorta, substituted 80 mM K+, 74.2 mM Na+ solution induced a sustained contraction, increase in oxygen consumption, and no change in ATP content. The 80 mM K+ solution without added glucose also induced a sustained contraction followed by a slight increase in oxygen consumption and a slight decrease in ATP content. The 154.2 mM K+, Na+-deficient solution produced similar changes in both oxygen consumption and ATP content as the 80 mM K+, glucose depleted solution. However, the 154.2 mM K+ solution induced only a transient contraction in the vascular smooth muscle. When 100 mM sucrose was hyperosmotically added to the 154.2 mM K+ solution, the suppressed muscle tension increased again, although the ATP content did not increase. From these and the previous results, it is concluded that glucose utilization by the taenia coli is inhibited in the 154.2 mM K+, Na+-deficient solution, and the decreased energy production of the muscle cell does not compensate the increased energy consumption induced by the high concentration of K+. In the aorta, although the Na+-deficient solution also decreases ATP production, it is the cell swelling induced by a high KCI concentration in the medium, not the decrease in energy metabolism, that has a direct inhibitory effect on muscle tension.

Mode of the ciguatoxin-induced supersensitivity in the guinea-pig vas deferens.

Ciguatoxin (CTX; 10(-7) X 10(-7) g/ml), the most potent marine toxin isolated from a number of tropical and subtropical fishes, shifted the dose-contractile response curves for norepinephrine (NE) and K+ to the left in a parallel manner in the guinea-pig isolated vas deferens, indicating that CTX caused supersensitivity. The CTX-induced potentiation was inhibited or abolished in the presence of tetrodotoxin (5 X 10(-7) M) or saxitoxin (5 X 10(-7) M) and in Na+-deficient medium, but was not affected by phentolamine (10(-6) M) and verapamil (10(-6) M). Treatment with reserpine (2 mg/kg/day, twice) almost completely prevented the release of NE by CTS, such pretreatment had no affect on the ability of CTX to potentiate responses to NE and K+. Furthermore, after cold storage (4 degrees C for 7 days) of tissues, the contractile response to NE (3 X 10(-6) M) and K+ (20 mM) was still profoundly potentiated after treatment with CTX (5 X 10(-7) g/ml). CTX (10(-7)-10(-5) g/ml) by itself had no apparent effect on either Na+, K+-adenosine triphosphatase activity or Na+ content of the vas deferens. However, in the presence of ouabain, CTX elevated the Na content of the vas deferens treated with ouabain alone by 27%. This effect of CTX was abolished by tetrodotoxin. These data suggest that CTX causes an increasing Na+ permeability across the TTX sensitive Na+ channels of smooth muscle cell, and this may play an important role in its mechanism of potentiation.

[Assessment to fixed and variable ROI (region-of-interest) settings in the determination of left ventricular ejection fraction by multiple gated cardiac pool imaging: the benefit of variable ROI setting].

The accuracy of left ventricular ejection fractions (EF) determined by multiple gated cardiac pool imaging (RI-EF) and by M-mode echocardiography (Echo-EF) (Teichholz's method) was evaluated by comparing with those derived from angiocardiography (LVG-EF) in 19 patients including 9 with myocardial infarction, 3 with rest angina, 2 with effort angina, 2 with valvular disease, 1 with hypertrophic cardiomyopathy, 1 with congestive cardiomyopathy and 1 with hypertensive heart. Measurement of RI-EF was carried out by the fixed ROI method using a single end-diastolic ROI, and the variable ROI method where end-diastolic and end-systolic ROI were used.

[Analysis of the murine autologous mixed lymphocyte reaction. I. Characterization of the stimulator cells and their recognition sites].

A stimulating activity in the SMLR was found in the mice of a week after birth at low but significant level and it became to be an adult level within only two to three weeks. A responding activity however remained very low until three to four weeks after birth. But this discrepant phenomenon could not be due to immaturation of the responder cells, since some T cell population was detected in the splenocytes of young mice which suppressed SMLR of adult mice. The responder cells of SMLR were T cells. On the other hand, stimulator populations of this reaction consisted of macrophages and B lymphocytes. Spleen adherent cells and peritoneal exudate cells showed a strong stimulating activity. But in the SMLR, highly purified B cells did not have a stimulating capacity, since removal of adherent populations from surface Ig(+) spleen cells entailed a loss of stimulating activity for responder T lymphocytes. With a help of macrophages, B lymphocytes acquired a stimulating activity which was easily blocked with anti-Ia antisera. Responder T lymphocytes recognized B lymphocytes across the MHC barrier in the secondary responses, whereas they were able to respond only to the histocompatible macrophages. Specific stimulation in the SMLR therefore was determined not by B cells, but by macrophages. Recognition sites of stimulators in the SMLR were presumed to be Ia antigens, especially products of I-A(B) subregions from the results of blocking assay with anti-Ia antisera and of secondary responses of the SMLR primed cells.

[Analysis of the murine autologous mixed lymphocyte reaction. II. Role of thymus in maturation of the responder T cells].

Bone marrow chimera mice sharing an identical H-2 K end as the donor were able to respond in an autologous mixed lymphocyte reaction (AMLR). On the other hand, bone marrow chimera made from histoincompatible partners or partners having identical D end of the H-2 were unable to respond. In the secondary responses of the AMLR primed cells from semiallogeneic bone marrow chimera, T cells responded specifically only to the cells which shared identical H-2 haplotypes with the recipient. AMLR primed cells from F1 T cell deprived mice (F1 At X BM mice) transplanted with the thymus grafts were able to respond only to the cells bearing identical H-2 haplotypes with the thymus grafts. It therefore becomes clear that responder cell activity of the AMLR is genetically restricted by the MHC along the pathway of their maturation through the thymus microenvironment. However, an experiment using athymic nude mice suggests the presence of another pathway for being restricted to the stimulator cells, because nude mice which were unable to respond in the AMLR could generate the reaction after thymic grafting from allogeneic donor.

New detection and separation method for amino acids by high-performance liquid chromatography.

A simple system for simultaneous fluorescence detection of primary and secondary amines has been developed. The optimum conditions for fluorescing secondary amines in the two-stage reaction technique using sodium hypochlorite and o-phthalaldehyde have been studied, and then the switching flow method (sodium hypochlorite is added only to secondary amines) and the non-switching flow method (sodium hypochlorite is added to all the amino acids) have been compared. It was concluded that the non-switching flow method ensures high efficiency and reliability. This newly-developed system was applied to the detection of amino acids and gave satisfactory results.

Thirteen cases of pyruvate kinase deficiency found in Japan.

Thirteen cases of pyruvate kinase (PK) deficiency, considered to be heterozygous for different PK mutants because of no consanguinities in their parents, were characterized by the International Committee for Standardization in Haematology (ICSH) recommended methods. These deficiency cases are named PK "Kagoshima," PK "Kyoto," PK "Takamatsu," PK "Abeno," PK "Kobe," PK "Marugame," PK "Hoenzaka," PK "Osaka," PK "Motomachi," PK "Gifu," PK "Hiroshima" PK "Matsumoto," and PK "Tama." The characteristics of mutant PK enzymes suggest that the cause of chronic hemolysis depends mainly on decreased affinity for phosphoenolpyruvate, thermolability, increased inhibition by adenosine triphosphate, and low activation by fructose-1, 6-diphosphate.

A case of lead intoxication: clinical and biochemical studies.

A 23-year-old Japanese male with severe lead intoxication accompanied by hemolytic anemia was studied. The patient had taken 12 g to lead in about a month. He had moderate hemolytic anemia (Hb 8.9 g/100 ml) with reticulocytosis ranging from 2.5 to 11.7%. Peripheral blood smear showed nucleated red cells (42/200 white blood cells) and marked basophilic stippling in the red cells. Activities of erythrocyte enzymes were either normal or increased except for pyrimidine 5'-nucleotidase (P5N) and delta-aminolevulinic acid dehydratase (ALA-D) both of which were found to be decreased, being 48.8% and 4.1% of the normal controls respectively. Erythrocyte reduced glutathione (GSH) was high (145.6 mg/100 ml RBC). Erythrocyte pyrimidine nucleotides were accumulated up to 10.2% of total nucleotides. The level of lead in peripheral blood was 112 microgram per 100 ml blood. Ca2+-Na2-EDTA was given to the patient as treatment. The level of lead in the blood decreased gradually and hemolytic anemia improved. Basophilic stippling in the red cells disappeared. These results confirmed the findings of Valentine et al [1] that lead-induced deficiency of P5N resulted in basophilic stippling and hemolytic anemia just like hereditary hemolytic anemia due to P5N deficiency.

The localization of glial fibrillary acidic protein in brain tumors induced in hamsters with avian sarcoma virus.

The immunohistochemical localization of glial fibrillary acidic protein (GFAP) was studied by the peroxidase-antiperoxidase (PAP) method in avian sarcoma virus (ASV)-induced brain tumors in hamsters. One hundred twenty-four tumors including 54 astrocytomas, 64 pleomorphic gliomas, five sarcomas, and one unclassified tumor were stained with anti-GFAP serum. A positive immunostaining was seen in astrocytomas and in pleomorphic gliomas. Sarcomas and an unclassified tumor were negative. These results confirmed that a majority of ASV-induced brain tumors contained neoplastic glial cells, and further suggested that the astrocyte is a main target cell of ASV in the experimental neuro-oncogenesis.
