Modified lipoprotein lipase catalyzes ester synthesis in benzene. Substrate specificity.

Modified lipoprotein lipase catalyzed the synthesis of trilaurin from mono- or diacylglycerol and fatty acid and also the synthesis of ester from fatty acid and alcohol in benzene. In the ester synthesis reaction, the longer the chain length of fatty acid or alcohol, the higher the ester synthesis activity. The ester synthesis was competitively inhibited by fatty acids with branch of carbon chain at the position neighboring the carboxyl group. Other substrates including secondary and tertiary alcohols and carboxylic acids having benzene ring were tested and discussed in relation to the ester synthesis.

Effects of drug-induced reduction in oxyhemoglobin affinity on survival time of mice in severe hypoxic conditions.

We studied the effect of ortho-iodo sodium benzoate (OISB) given intraperitoneally to mice. 300 and 600 mg/kg of OISB increased P50 from the control value of 41.2 mmHg to 42.7 mmHg and 45.7 mmHg, respectively. The mice were then exposed to two experimental hypoxias, namely, hypobaric hypoxia and carbon monoxide hypoxia. Mean survival time was prolonged from 67.8 (control saline) to 94.8 sec (OISB 300 mg/kg) and 112.2 sec (OISB 600 mg/kg) in the hypobaric hypoxia, and from 201.3 sec (control saline) to 329.7 sec (OISB 300 mg/kg) and 407.7 sec (OISB 600 mg/kg) in the carbon monoxide hypoxia. We concluded that OISB had prolonged survival time by reducing oxyhemoglobin affinity, thus facilitating oxygen release in severe hypoxic conditions.

Gated cardiac imaging using a continuously rotating CT scanner: clinical evaluation of 91 patients.

To produce electrocardiographically (ECG)-gated computed tomographic (CT) images of the heart, a post-data-acquisition ECG correlation technique was used in which data for missing angular projections are derived from the original scan data to complete 360 angular projections. Improved image quality and clinical usefulness were demonstrated compared with routine nongated CT and two-dimensional echocardiography. Gated CT was better than nongated CT in 26 of 41 positive and three of five negative cases of suspected myocardial infarction, four of 10 positive and one of 12 negative cases of suspected left atrial mass, three of 10 cases with pericardial fluid collection, and three other cases. Compared with echocardiography, CT was of additional value in eight of 10 cases of myocardial infarction, five of nine positive and one of 10 negative cases of suspected left atrial mass, four of 10 positive and one of three negative cases of suspected pericardial fluid collection, and two other cases. The equipment required for CT gating is of low cost, but the examination time is lengthy and less conveniently performed than echocardiography. However, when echocardiography is indecisive or suspected to be falsely negative, gated CT imaging of the heart is recommended.

Selective inhibition of adenosine triphosphatase from plasma membranes of cultured endothelial cells by calcium antagonists.

A study has been made of the inhibitory effect of various kinds of hypotensive agents on ATPase of cultured endothelial cells from bovine carotid artery stimulated with Ca2+. The ATPase activity was selectively inhibited by calcium antagonists including a new compound, 6-ethoxycarbonyl-5,7-dimethyl-1-[3-(4-phenethyl-1-piperazinyl)propyl]-3- (alpha, alpha, alpha-trifluoro-o-tolyl)-2,4(1H,3H)-quinazolinedione dihydrochloride, while not by other hypotensive agents.

[The effect of high frequency jet ventilation on intracranial pressure in the patients with severe head injury].

The effect of high frequency jet ventilation (HFJV) on intracranial pressure (ICP) in the series of seven patients with severe head injury was studied. These patients received mechanical ventilation for respiratory failure accompanied with neural damage or circulatory shock and for ICP control by decreasing PaCO2. ICP was measured continuously by subarachnoid catheter method connected to Gould P-50 strain-gage transducer and controlled by hyperventilation and/or osmotic diuretics during acute phase. Arterial pressure, central venous pressure and intratracheal pressure were measured continuously in the same way. Arterial blood gases were analysed by BG-I of Technicon Co. Ltd. At the weaning from respirator, two methods were applied. One was conventional IMV (with PEEP) and/or CPAP by using IMV mode of Servo 900 respirator or T-piece together with PEEP valve and another was new-fashioned HFJV. Mean intratracheal pressure (mPit) was fixed at 0 mmHg (control value), 5, 10 and 15mmHg in both methods by adjusting PEEP valve or driving pressure of HFJV. The result is that in the same patients ICP was significantly lower during HFJV than IMV/CPAP when mPit was 5 and 10mmHg. Only three cases having been studied at 15mmHg of mPit because circulatory condition would not permit it, significant difference of ICP between HFJV and IMV/CPAP was not detected. PaCO2, being suspected to be the cause of lower ICP of HFJV group, was studied too. But blood gas analysis showed no significant difference neither PaCO2 nor PaO2. The fluctuation of CVP reflecting the change of intrathracic pressure was smaller in HFJV group than in IMV/CPAP group.(ABSTRACT TRUNCATED AT 250 WORDS)

Rapid modulation of platelet aggregation in plasma by oxygenated sterols.

The effect of oxygenated sterols on platelet aggregation induced by thrombin and ADP has been studied. All oxygenated sterols tested with a hydroxyl group on the side chain enhanced thrombin-induced aggregation at 25 microM. In the case of ADP-induced aggregation, however, only 22S-hydroxycholesterol[(22S)-5-cholestene-3 beta,22-diol] enhanced the aggregation, and 22R-hydroxycholesterol[(22R)-5-cholestene-3 beta,22-diol], 24S-hydroxycholesterol[(24S)-5-cholestene-3 beta,24-diol], and 25-hydroxycholesterol[5-cholestene-3 beta,25-diol] inhibited it. These effects were observed within 10 min after the addition of oxygenated sterols to platelet suspensions in plasma.

[Preparation of repeatedly and effectively usable L-asparaginase by a chemical modification].

The anti-tumor activity of L-asparaginase (EC. 3.5.1.1.) has been conclusively demonstrated. Its therapeutic application, however, has been hampered by its short clearance time in the circulation and high immunogenicity. In order to solve these problems, polyethylene glycol, a linear synthetic and non-immunogenic polymer, was introduced covalently into the amino groups in the enzyme molecule. Monomethoxypolyethylene glycol with molecular weight of 5000 was activated with cyanuric chloride to obtain activated PEGs [2, 4-bis (O-methoxypolyethyleneglycol)-6-chloro-s-triazine]. L-Asparaginase lost its immunoreactivity against its antibodies after the modification of 52 out of 92 amino groups in the molecule. This modified asparaginase retained 11% of its enzymic activity under the physiological condition. When the modified asparaginase was administered in rodents, it diminished the serum asparagine level and its enzymic activity persisted in the circulation 10 to 20 times longer than that of native enzyme. Furthermore, repeated injections of modified asparaginase did not induce any significant anti-asparaginase antibody production. Modified asparaginase showed a superior anti-tumor activity to the native counterpart irrespective of the presence of anti-asparaginase antibodies, when it was tested with murine Gardner lymphoma. This chemical modification should make it possible for the first time to repeatedly and effectively use L-asparaginase obtained from a bacterium.
