[The behavior of 7, 12-dimethylbenz(a)anthracene (DMBA)-induced mammary tumors in rats during pregnancy and lactation and hormone levels in the pituitary and plasma (author's transl)].

In order to keep a long-term lactation in primiparous SD strain rats with DMBA-induced mammary tumors, 5 pups aged 5 to 10 days were exchanged every 10 days. FSH, LH and prolactin (PRL) in pituitary tissues and FSH, LH, PRL, estradiol (E2) and progesterone (PRG) in plasma were determined during pregnancy (1,2 and 3-week), at paturition (0 to 12 hours after parturition) and during lactation (1,2,3,4,5 and 6-week). Experimental rats were divided into 6 groups according to the lactating period (non-lactating 3,6,8,12 and 17-week). Blood samples were collected by cardiac puncture 1 hour after an intraperitoneal injection of urethane (150 mg/100 g body weight), and the pituitary gland was removed simultaneously. During pregnancy, plasma PRL was low in the first and second week. PRL, E2 and PRG were high in the third week, during which DMBA-induced mammary tumors began to proliferate. PRG in plasma in the third week was lower than that of the second week. After paturition, the tumors regressed and did not develop for three weeks of lactation, during which plasma PRL was high whereas E2 and PRG were low. The tumors began to proliferate again from the fifth week of lactation, during which E2 was high and the estrous cycle reappeared. The behavior of DMBA-induced mammary tumors in rats during pregnancy and lactation seems to be closely related to plasma estrogen levels.

Effect of dietary mineral composition on nutritional equivalency of amino acid mixtures and casein in rats.

The effect of dietary mineral composition on the quality of amino acid mixtures and intact casein was examined in growing rats fed for 28 days under "conventional" conditions. When 25% demineralized casein (DC) was the sole dietary protein source, the widely-used mineral mixture by Harper (MMH) supported growth poorly (3.2 g/day), but a new formula mineral mixture (MM2) containing sufficient amounts of zinc gave a growth rate of over 8 g/day for 21 days. With a crude casein (CC) diet, rats grew at the same rate regardless of mineral mixtures. The growth-supporting power of amino acid mixtures, one of which simulated casein (AA-A) and another patterned after Rogers and Harper to give maximal growth of rats (AA-B), was compared with those of DC and CC with addition of MMH or MM2. When the protein source was DC, AA-A or AA-B, MM2 significantly improved the growth over that of rats fed MMH. The growth-stimulating effect of MM2 was greater when added to DC diet than when added to AA-A diet. When MM2 was added to AA-B diet, the growth rate greatly increased and reached that of rats fed DC diet containing MM2 (over 8 g/day), showing a nutritional equivalency between the amino acid mixture and intact protein. Drawbacks inherent in MMH did not appear with CC diet, because CC contained considerable amounts of zinc. When a highly purified amino acid-sucrose diet is used, dietary minerals become most limiting for growth even under conventional conditions. Thus, the previous conflicting results concerning nutritional equivalency of amino acid mixtures and protein can be explained by inadequate mineral sources.

Relationships between the haemolytic activities of the human complement system and complement components.

The relationships between the haemolytic activities of complement and its components were studied. The activities studied included CH50 (classical pathway), AP50 (alternative pathway), CV50 (early part of alternative pathway) and C(3--9)H50 ((the late part of both pathways). The components included C3, C4, C5, C9, B and D. There was a good correlation between CH50 and AP50. AP50 had a good correlation with B and CV50. There was no correlation between AP50 and C(3--9)H50, and none between C(3--9)H50 and C5 or C9. AP50 may primarily represent changes in the early part of the alternative pathway. C(3--9)H50 is not influenced by respective changes in the amounts of C5 or C9. Since cell lesion is now considered to be caused by a unit of C5b to C9, a change in each component of C5 to C9 may not influence haemolytic activity.

Glomerular lesions in multiple myeloma.

An autopsy case of multiple myeloma (IgG, lambda type), clinically characterized by decreased glomerular filtration rate, is reported with particular emphasis on changes in the glomeruli of kidneys. Histologically, the glomeruli revealed slight increase in mesangial matrix and focal thickening of tuft capillary wall. Electron-microscopically, deposits were observed in a subendothelial location in the glomerular capillary walls, and inclusions were noted in the cytoplasm of the visceral epithelial cells. Histoimmunofluorescent study of the kidney demonstrated intense focal and slight diffuse positivity against labelled antisera of anti-IgG and anti-lambda type of light chain on the capillary wall of the glomerular tufts. Other immunoglobulins were not demonstrable in capillary walls. These findings represent the intraglomerular deposition of paraprotein of multiple myeloma without amyloidosis.

Waldenström's macroglobulinaemia with Ig E M-component.

A case of Waldenström's macroglobulinaemia with Ig E M-component was presented. A 76-year-old Japanese woman had a history of the operation for mammary carcinoma. Although neither lymphadenopathy nor hepatosplenomegaly was present, the immunohaematological findings typical for Waldenström's macroglobulinaemia were observed in this patient. Coexistence of the Ig M and Ig E M-components was recognized in the patient's serum protein. Positive reaction for FITC-conjugated anti-epsilon or rhodamine-conjugated anti-mu was noted in proliferating lymphoplasmacytic cells of the patient's bone marrow, but these reactions were not simultaneously observed in a single cell. The finding suggests that two separate populations of M-component-producing cells were present.
