Nucleotide sequence of the transcriptional initiation region of Dictyostelium discoideum rRNA gene and comparison of the initiation regions of three lower eukaryotes' genes.

The 5' end of the rRNA precursor of D. discoideum was mapped on a cloned rDNA by S1 nuclease protection mapping, and the sequence of about 1240 nucleotides surrounding the transcriptional initiation site of the rRNA gene has been determined. Repeated sequences consisting of 16 nucleotides appeared in the region upstream from the initiation point. Comparison of the nucleotide sequences around the initiation site of rRNA genes in three lower eukaryotes, D. discoideum, Saccharomyces cerevisiae and Tetrahymena pyriformis, indicated that there was little similarity in the nontranscribed spacer regions, but in the transcribed spacer regions near the initiation point, very similar sequences consisting of 9 nucleotides were found.

Recognition patterns for exon-intron junctions in higher organisms as revealed by a computer search.

Most genes of higher eukaryotes are discontinuous. The DNA which codes for a protein is interrupted by introns. In the nucleus, certain machinery is supposed to recognize the exon-intron and intron-exon junctions. Applying computer searching to eighteen genes from various biological species, we examined what kinds of patterns or nucleotide sequences are necessary and sufficient to recognize the splice junctions. We propose that four common patterns of AG/GTA, /GTAAGT, RG/GTGAG and AG/GTXXGT, where R = A or G and X = A, T, G, or C, are often used as signals for exon-intron junctions. This proposal is based on the facts that they are not found in the exons of the genes examined and on the assumption that the recognition machinery scans the mRNA precursor from the 5'-end to the 3'-end.

Thyroid CT number and its relationship to iodine concentration.

Sixty-seven patients with thyroid disease and 24 normal controls were examined with computed tomography (CT). The mean CT number (Hounsfield units +/- SD) in the normal controls (118.1 +/- 12.2) was significantly higher (p less than 0.001) than the mean CT number in patients with diseased thyroids, except for 2 cases of simple goiter (CT numbers 113, 132). The mean CT numbers in the patients with Graves disease (69.5 +/- 17.6) and Hashimoto thyroiditis (61.4 +/- 9.1) were significantly higher than those in patients with adenoma (41.7 +/- 10.6, p less than 0.001), cyst (33.1 +/- 14.8, p less than 0.001), or cancer (48.7 +/- 13.0, p less than 0.01). In 14 patients studied, a significant correlation was observed between thyroid CT numbers and the iodine concentration of the tissue (r = 0.889; p less than 0.001).

Localization of intrarenal cross-linked fibrin in children with various renal diseases.

The localization of intrarenal cross-linked fibrin was examined by the effect of monochloroacetic acid treatment on the kidney sections. In acute glomerulonephritis or in mild diffuse or focal proliferative type of nephritis, cross-linked fibrin was observed mainly within glomerular capillary walls. Extension of cross-linked fibrin deposit over the mesangium or sclerotic area was seen in moderate to severe proliferative type of nephritis or in membranoproliferative glomerulonephritis. In hemolytic uremic syndrome or disseminated intravascular coagulation syndrome, cross-linked fibrin was detected within glomeruli and vessels.

Subjective and objective ocular disturbances in reattached retina after surgery for retinal detachment, with special reference to visual acuity and metamorphopsia.

A questionnaire about postoperative complaints was given to patients after surgery for retinal detachment. Their visual acuity and visual field problems were analyzed. Only one third of all the patients felt postoperative improvement. Blurred vision and metamorphopsia after operation were complained of in one third of all the patients. Blurred vision was apt to be complained of in patients with macular detachment before operation. About 77% of patients with blurred vision had a visual acuity of less than 0.5. Metamorphopsia was frequently complained of in patients treated with the segmental buckling procedure.

Inhibition of thyrotropin-induced adenosine 3'5'-monophosphate increase by immunoglobulins from patients with primary myxedema.

Immunoglobulin G (IgG) fractions prepared from the serum of 18 patients with primary myxedema, 9 patients with goitrous Hashimoto's thyroiditis, and 14 normal controls were tested for their ability to alter TSH stimulation of cAMP production in cultured human thyroid cells and the binding of TSH to its receptor. When compared with the cAMP increase induced by 0.1 mU/ml bovine TSH in the presence of normal IgG, cAMP accumulation was significantly inhibited (P less than 0.005) by the addition of IgG from patients with primary myxedema. TSH-induced cAMP accumulation was not affected by IgG from patients with goitrous thyroiditis. IgG from patients with primary myxedema also inhibited the cAMP increase induced by thyroid-stimulating immunoglobulins, but not against the increase induced by prostaglandin E1. None of the IgG tested affected the basal level of cAMP. Two potent inhibitory IgG were strongly positive for TSH-binding inhibitor immunoglobulins. Excluding these, no significant correlation was found between the thyroid stimulation-blocking activity and the TSH-binding inhibitory activity. These data suggest the presence of at least two different types of antibodies in primary myxedema which block adenylate cyclase stimulation by TSH and might be responsible for thyroid dysfunction and atrophy.

Partial purification and properties of the TSH receptors from human thyroid plasma membranes.

Human thyroid plasma membranes were solubilized with 0.5% Triton X-100 and TSH receptors were purified by using DEAE-Sephadex, Con A and TSH affinity chromatography. A TSH binding activity was bound to DEAE-Sephadex equilibrated with 0.05 M sodium acetate, pH 6.3, 0.2% Triton X-100 and was eluted by a linear gradient of 0.1 M to 1.0 M ammonium acetate, pH 6.3. Eighty-five percent of the activity was absorbed to Con A Sepharose and was eluted with 0.5 M alpha-methyl-D-mannoside, 0.05 M sodium acetate, pH 6.0. Seventy-five per cent of the TSH binding capacity could be absorbed to TSH-affinity column and was eluted with 0.1 M glycine-HCl, pH 3.0. By sequential application of the above procedures, more than 100-fold purification of the receptor activity was attainable. [125I]TSH binding of this fraction was inhibited by addition of unlabelled TSH in a dose-dependent manner. Scatchard analysis gave a curvilinear plot with a high affinity association constant of 0.72 X 10(9)M-1. By using Ultrogel AcA 34 gel filtration, the molecular size of the hormonereceptor complex was estimated to be 180 000.
