Dephosphorylation of "abp38" protein during the differentiation of mouse myeloid leukemia cells.

We have characterized the phosphorylation of "abp38" before and after the differentiation of mouse myeloid leukemic cells (M1 cells). The abp38 of both undifferentiated and differentiated M1 cells contained phosphoserine and phosphotyrosine. The extent of phosphorylation of abp38 decreased to about 30% of the value of undifferentiated cells during cell differentiation; phosphoserine and phosphotyrosine decreased to 41 and 11% of the values before differentiation, respectively.

Thermoluminescence dosimetry of gamma rays from the Hiroshima atomic bomb at distances of 1.27 to 1.46 kilometers from the hypocenter.

Sixteen ornamental tile samples were collected from 1982 to 1983 from the rooftops of two buildings at Hiroshima University, Hiroshima, Japan. Quartz grains 50-150 microns in size extracted from the samples were analyzed for their thermoluminescence (TL) intensities. Conversion of TL intensity to 60Co gamma exposure resulted in the following estimates: 40.5 to 27.6 mC kg-1 (157 to 107 R) for five samples (one each) collected from five sites at distances of 1.27 to 1.34 km from the hypocenter of the atomic bomb detonated in 1945; 23.7 +/- 1.4 mC kg-1 (92 +/- 5 R) for three samples from one site at a distance of 1.39 km; 21.4 to 17.0 mC kg-1 (83 to 66 R) for three samples (one sample per site) from three sites at distances of 1.40 to 1.43 km; 19.8 +/- 1.3 mC kg-1 (77 +/- 5 R) for four samples from one site at a distance of 1.45 km; and 13.2 mC kg-1 (51 R) for one sample at a distance of 1.46 km. At face value, these estimates are greater by a factor of about 2.5 than previous estimates based on the tentative 1965 radiation dose estimates for atomic bomb survivors (a tentative dosimetry model proposed in 1965), but agree within +32% to -13% (+15% on the average) with recent estimates using modern computational techniques using an improved model of the atomic bomb explosion.

A case of virilizing adrenocortical carcinoma.

A 36-year-old woman who had experienced two pregnancies consulted our hospital, because of scant menses and virilization. A 24-hour excretion of 17-ketosteroids and 17-hydroxy-corticosteroids demonstrated a decrease in 11-hydroxylase. A computed tomogram showed a huge inhomogenous tumefaction in the left adrenal. Left selective renal angiography revealed a large adrenal tumefaction. Selective adrenal venous samplings revealed that testosterone and dehydroepiandrosterone (DHA) were produced in response to stimulation by 0.25 mg exogenous adrenocorticotropic hormone (ACTH). After left adrenalectomy was performed, a diagnosis of adrenocortical carcinoma was made by pathological examination. This is the first report of a patient with a virilizing adrenocortical carcinoma, which produced testosterone and DHA in response to exogenous ACTH stimulation.

[A case of renal angiomyolipoma treated with partial nephrectomy. Diagnostic value of ultrasonography and computed tomography].

A 62-year-old woman with renal angiomyolipoma is reported. Ultrasonogram revealed a hyperechoic renal mass and computed tomography showed a fatty density area in the tumor. Preoperative diagnosis of renal angiomyolipoma was made and was confirmed by frozen section during the operation. Partial nephrectomy was performed subsequently. Renal angiomyolipoma is known by its fat content causing high echogenicity in the ultrasonogram and fatty density area in computed tomography. Renal angiomyolipoma, therefore, could be suspected by this combination of findings and conservative surgery should be planned preoperatively.

[Clinical experience of enoxacin in the treatment of nongonococcal urethritis].

Enoxacin (ENX) is a new derivative of pyridonecarboxylic acid and has a broad spectrum antibacterial activity. In this study, the effects of ENX on nongonococcal urethritis (NGU) were examined in 20 patients. ENX was administered orally 3 times a day at a daily dose of 600 mg for 2-23 days. Before treatment, Chlamydia trachomatis was isolated from urethral epithelium in 5 of the 20 patients. Of the 18 patients evaluated in this clinical trial, 15 (83%) responded well, and ENX was effective in 3 of the 5 Chlamydia positive cases and in 12 of the 13 Chlamydia-negative patients. As adverse reactions, headache was observed in one case and numbness of limbs in another case. No abnormality induced by ENX was recognized in laboratory findings. It is concluded that ENX is a useful drug in the treatment of patients with NGU, particularly in whom Chlamydia trachomatis is not isolated.

Report of a case with chronic myelomonocytic leukemia: demonstration of leukemic monocytes lacking nonspecific esterase by flow cytometry using monoclonal antibodies.

Chronic myelomonocytic leukemia (CMML) is a rare leukemia, which is now included in myelodysplastic syndromes. In a small number of patients with CMML, problems in the diagnosis have been reported, especially when atypical morphological features in both monocytic and granulocytic cells due to dysmyelopoiesis are prominent, or when cytochemical characteristics are lost in the leukemic cells. The case history of a sixty-seven year-old male patient with CMML is described. The diagnosis of CMML in the patient was supported by the following evidence: chronic course of his disease; increased monocyte-like cells without other cause; normocytic anemia; immature granulocytic cells with hypogranular feature and giant platelets were observed in the peripheral blood. The bone marrow showed myeloid hyperplasia. Serum muramidase and vitamin B12 levels were increased, while neutrophil alkaline phosphatase score was low in the peripheral blood. Ph' chromosome was negative. The monocyte-like cells completely lacked nonspecific esterase. However the cells were confirmed as monocytic cells by flow cytometry using monoclonal antibodies to monocytes (OKM5).

A multicenter double blind controlled study of lipo-PGE1, PGE1 incorporated in lipid microspheres, in peripheral vascular disease secondary to connective tissue disorders.

Lipo-PGE1 is a drug preparation of PGE1 (prostaglandin E1) incorporated in lipid microspheres similar in properties to liposomes. A multicenter, placebo controlled, double blind test of lipo-PGE1 was carried out in patients with severe peripheral vascular disease secondary to diffuse connective tissue disorders. A total of 135 patients received a 90 min intravenous infusion of either 10 micrograms lipo-PGE1 or placebo every day for 4 weeks. A significant improvement was noted in the lipo-PGE1 group compared with the placebo group in the final overall improvement of peripheral vascular disease (p less than 0.001) and in the healing of ulcers (p less than 0.01), whereas, there was no difference in Raynaud's syndrome between the 2 groups. Side effects in the lipo-PGE1 group were few and were no different from controls. These results indicate that lipo-PGE1 at a low dose is beneficial in the treatment of peripheral vascular disease, and that lipid microspheres are useful as a drug delivery system for such purposes.

[Parathyroid carcinoma: report of a case].

A case of parathyroid carcinoma is presented. A 46-year-old female was admitted to our hospital for fractures of both femurs on July 29, 1983. Laboratory data revealed a serum calcium level of 15.2 mg/dl, serum phosphate level of 1.2 mg/dl, serum immunoreactive parathyroid hormone 9.35 ng/ml (less than 0.5), and % tubular reabsorption of phosphate of 57%. X-ray examination showed marked osteitis fibrosa cystica. The diagnosis of primary hyperparathyroidism was made. A hard tumor was palpable on the left anterior side of her neck. Neck exploration was carried out on August 10. The tumor was found to be localized in contact with the left lower lobe of the thyroid gland. Parathyroid carcinoma was strongly suspected, because the tumor severely adhered to surrounding tissues, thus the tumor was resected en bloc. The histopathological diagnosis was typical parathyroid carcinoma. Postoperative course and the treatment of the fractures were uneventful, and she was discharged able to walk five months after the operation. No evidence of recurrence or metastasis has been seen during the eighteen months since the operation. This is the 80th case in the Japanese literature to our knowledge and the clinical features of these 80 cases revealed an average age of 41.5 years old; male/female ratio of 32/48; average weight of tumor of 8.65 g, palpable neck mass in 72%, bone disease in 64%, and renal disease in 34%.

[Percutaneous nephrolithotripsy: a report of 50 cases].

Fifty patients with renal (33 patients), renal and ureteral (4 patients) and ureteral (13 patients) stones were treated by percutaneous nephrolithotripsy (PNL) between November 15, 1984 and December 14, 1985. Complete stone removal was achieved in 39 cases (78%). Four of the 11 cases of incomplete stone removal were among the first 5 cases we treated. We have not experienced any severe complications in our 50 cases. The cause of the unsuccessful attempts, complications and other factors of PNL are briefly discussed.

Heterogeneity of adrenocortical ferredoxin.

Bovine adrenocortical ferredoxin (adreno-ferredoxin) was purified from adrenocortical mitochondria by an improved method that included hydrophobic chromatography on Toyopearl gels. The purified ferredoxin was electrophoretically homogeneous. It was further separated into five fractions by hydrophobic chromatography on a TSK-gel phenyl-5PW column with a high-pressure liquid chromatography system. The properties of the three main fractions were examined. The fractions had identical absorption spectra and almost the same activity in an NADPH-cytochrome c reducing system. Their amino-terminal sequences all corresponded to the reported sequence, but the carboxyl-terminal residues were glycine or serine, not alanine as reported. These results indicate that these adreno-ferredoxins had additional amino acid residues at the carboxyl end. It seems that adreno-ferredoxin extracted from mitochondria undergoes proteolytic attack during purification to become heterogeneous.
