[Determination of malignant disease-associated DNA-binding protein 2 in patients with liver cancer].

The aim of this study was to assess the value of malignant disease-associated DNA-binding protein 2(MAD2) in the diagnosis of liver cancer. The concentration of plasma MAD2 was determined in 27 patients with primary liver cancer, 14 patients with metastatic liver cancer and 12 healthy subjects by ELISA assay. The primary tumors of all the patients with metastatic liver cancer were located in the gastrointestinal tract, which had been radically resected and had no signs of local recurrence. The concentrations of plasma MAD2 in the patients with primary liver cancer, with metastatic liver cancer, and the healthy subjects were 30.56 +/- 11.38 micrograms/ml, 9.27 +/- 5.58 micrograms/ml and 8.43 +/- 5.62 micrograms/ml, respectively. The concentration of MAD2 in patients with primary liver cancer was significantly higher than that in the patients with metastatic liver cancer and healthy subjects (P < 0.001). There was no significant difference between the healthy subjects and the patients with metastatic liver cancer(P > 0.05). These results suggest that MAD2 may be an useful marker for the diagnosis of primary liver cancer.

[Establishment of the animal model of triple semicircular canal occlusion].

OBJECTIVE: To establish the animal model of triple semicircular canal occlusion(TCO) in guinea pigs. METHODS: TCO was performed on 20 guinea pigs. Electronystagmography, auditory brainstem responses (ABR) and distortion product otoacoustic emissions(DPOAE) were recorded pre- and post-operatively. The non-operated ear was used as control. RESULTS: There was spontaneous nystagmus towards non-operated side on the 1st postoperative day and no nystagmus to stimuli of sinusoidal angular acceleration in operated side. On the 30th postoperative day, left and right nystagmus returned symmetrical. Pre-operation, ice-water stimulation induced normal symmetric nystagmus in both sides. On the 30th postoperative day, ice-water stimulation only induced normal nystagmus in non-operated ear and failed in operated one. ABR thresholds increased approximately 10 dB post-operation. There were no significant changes of the amplitudes of DPOAE before and after surgery. Histologic examination revealed complete occlusion in the three semicircular canals with newly formed bone and connective tissue. CONCLUSION: This study showed that the animal model of TCO was successfully established. These findings suggests that TCO might be a safe and simple procedure for the treatment of intractable peripheral vertigo.

[Areas of brain involved in immunoregulation].

OBJECTIVE: To study the location of brain areas involved in immunoregulation. METHODS: Immunohistochemistry and immunofluorescence were used to detect the different distribution of cytokines immunopositive cells in the brain of rats immunized via both intraperitoneal and subcutaneous injections. RESULTS: The cytokines immunopositive cells were distributed in the supraoptic (SO) and paraventricular (PV) nuclei of the hypothalamus, the anterior hypothalamic (AH) nucleus, arcuate and median eminence, the lateral hypothalamic nucleus (LH) and the amygdaloid nuclear complex while only the appearance of the cytokines immunopositive cells in LH and amygdaloid nuclear complex in hypothalamus was related with immunization status of the animals. Double-labelling results showed that the cytokines immunopositive cells were neurons. CONCLUSIONS: We have observed that neurons of the LH and amygdaloid nuclear complex in hypothalamus as a main source of the neuroimmunoregulation played a key role in neuroimmunoregulation and they participated in the neuroimmunoregulation at an early stage of the immune response.

[Principle of evidence-based medicine and its application in clinical trial practice for glutamine and gut permeability].

Evidence-based medicine (EBM) was developed from the progress of traditional clinical model trial patterns. EBM was based on well-designed, randomized, double blind, controlled, multiple center trails in order to assess end pont parameters, prognosis parameters (as: mortality, morbidity of complications and hospital stay etc.), quality of life and cost/effect ratio. It has been accepted in developed countries and could be the guidelines for Chinese clinical trials in the future. This article summarized the principle of EBM and attached with a clinical trial sample about effects of glutamine dipeptide on gut barrier function, post-operative complications and the hospital stay.

[The effects of sodium nitroprusside-induced hypotension at different levels on cerebral blood flow and metabolism: a clinical study].

OBJECTIVE: The clinical experiment was designed for observing the effects of deliberate hypotension at different level on cerebral blood flow and metabolism. METHODS: 30 ASA Grade I-II patients were randomly allocated to one of three groups (each group n = 10). Hypotension was induced by sodium nitroprusside in group 1[mean arterial pressure (MAP) = 55 mmHg] and group 2 (MAP = 65 mmHg). Group 3 (control) was on normotension. The changes of cerebral blood flow during hypotension were estimated according to the variability of cerebral blood flow velocity (CBFV) in the middle cerebral artery(MCA), which was obtained through transcranial Doppler ultrasonography (TCD). RESULTS: Hypotension caused a moderate decrease in CBFV, but no significant difference, in CBFV and oxygen saturation in internal jugular vein(SjO2), either among three groups or during the procedure. CONCLUSIONS: It is safe for cerebral perfusion and metabolism with deliberate hypotension at MAP = 55 mmHg.

[Effect of propofol and etomidate for anesthesia induction on plasma total cortisol concentration].

OBJECTIVE: To investigate the effect of propofol and etomidate for anesthesia induction on adrenocortical function. METHODS: The plasma total cortisol concentration was determined in 20 patients who were treated with propofol or etomidate. RESULTS: After propofol induction, the plasma total cortisol level decreased from (308.7 +/- 27.4) nmol/L to (267.7 +/- 31.2) nmol/L at post-medication 2 hours, increased to (400.2 +/- 26.9) nmol/L at postmedication 5 hours and restored to premedication level at postmedication 24 hours [(306.4 +/- 35.4) nmol/L]. After etomidate induction, the plasma total cortisol level decreased from (309.1 +/- 36.6) nmol/L to (115.9 +/- 29.7) nmol/L at postmedication 2 hours, maintained (171.1 +/- 34.7) nmol/L at postmedication 5 hours and restored to premedication level at postmedication 24 hours [(311.8 +/- 46.2) nmol/L]. CONCLUSIONS: Propofol induction has little effect on adrenocortical function whereas etomidate induction inhibits adrenocortical function.

[Stage-dependent expression of androgen binding protein mRNA in sertoli cell of rat testis].

OBJECTIVE: To investigate the variation of rat androgen binding protein (ABP) mRNA in sertoli cell of rat testis during spermatogenesis. METHODS: In situ hybridization was performed with digoxin-labeled rat ABP cDNA probe on frozen testis tissue sections from SD rats. The expression of ABP mRNA was quantified by using laser density scanning system. RESULTS: It was shown that ABP mRNA levels were low from stage I to stage VI, increased to the peak level at the stages VII-VIII, and then declined rapidly at stages IX-XI. Thereafter, ABP mRNA levels increased again at stages XII-XIV, and then decreased to the lowest level at stages I-VI. CONCLUSIONS: ABP mRNA level was stage-dependent during the spermatogenic cycle.

[Changes of trace elements in cells of lung cancer and cervix cancer before apoptosis and after apoptosis].

The sensitivity of total reflection X-ray fluorescence analysis (TXRF) is higher than normal X-ray fluorescence analysis (XRF). The cells of lung cancer and cervix cancer were researched before apoptosis and after apoptosis employed the method of TXRF on new transformed synchrotron radiation XRF experimental station of new 3W1A beam line which built on Beijing Synchrotron Radiation Facility (BSRF) of BEPC (new detector and spectrometer systems). We acquired their fluorescence spectrum of various elements before apoptosis and after apoptosis. We discovered there were interesting results that changes even remarkable changes of some elemental contents had occurred in these cells before apoptosis and after apoptosis. These alterations of elemental contents have served an inspiration for us: if their regularities of change are found, we will hope that some contributions would be done at the treatments of cancer.

[Discussions on some problems related to fluid inclusion analysis by SXRF].

X-ray fluorescence experiments were carried out at Beijing Synchrotron Radiation Facilities (BSRF) using synchrotron radiation as excitation source. The samples were made by mixing national reference standard GBW07106 and certain amount of reagent NaCl and KCl, and pressed into pellets. The purpose of the experiment is to verify the detection ability of the XRF setup at BSRF for elements in geological samples, especially for light elements. The beam size was 20 x 20 microns2, and the current in the storage ring was about 40 mA. The gap between the sample and the Si(Li) detector was 2 cm. With measuring time of 200 sec., The relative and absolute detection limits for 10 elements have been calculated. The sampling depth and sampling weight, which are critical in individual fluid inclusion analysis, have also been estimated. Some problems concerning the individual fluid inclusion analysis by SXRF, such as inclusion depth determination, effect of the inclusion depth on XRF intensity etc., are discussed based on the experiment results.

[Preparation and spectroscopic characterization of hydrophobic metallic nanoparticles and their LB films].

Hydrophilic silver and gold nano-colloids were synthesized using sodium oleate as a stabilizer, and their hydrophobic nano-colloids were further obtained using tetradecanethiol as a co-stabilizer and using concentrated hydrogen chloride acid as an initiator. UV-visible spectra of the gold and silver nanoparticles at different states were recorded and compared. The spectra show that surfactant stabilized colloids are stable and monodispersed both in aqueous solution and in organic solvents. Surface pressure (pi)-area (A) curves of the nanoparticulate monolayers were measured and their LB films were constructed. IR spectra show that polar head group of the oleate was adsorbed on the surface of the nanoparticles.

Hepatic arterial infusion of chemotherapy after resection of hepatic metastases from colorectal cancer.

BACKGROUND: Two years after undergoing resection of liver metastases from colorectal cancer, about 65 percent of patients are alive and 25 percent are free of detectable disease. We tried to improve these outcomes by treating patients with hepatic arterial infusion of floxuridine plus systemic fluorouracil after liver resection. METHODS: We randomly assigned 156 patients at the time of resection of hepatic metastases from colorectal cancer to receive six cycles of hepatic arterial infusion with floxuridine and dexamethasone plus intravenous fluorouracil, with or without leucovorin, or six weeks of similar systemic therapy alone. Patients were stratified according to previous treatment and the number of liver metastases identified at operation. The study end points were overall survival, survival without recurrence of hepatic metastases, and survival without any metastases at two years. RESULTS: The actuarial rate of overall survival at two years was 86 percent in the group treated with local plus systemic chemotherapy and 72 percent in the group given systemic therapy alone (P=0.03). The median survival was 72.2 months in the combined-therapy group and 59.3 months in the monotherapy group, with a median follow-up of 62.7 months. After two years, the rates of survival free of hepatic recurrence were 90 percent in the monotherapy group and 60 percent in the monotherapy group (P<0.001), and the respective rates of progression-free survival were 57 percent and 42 percent (P=0.07). At two years, the risk ratio for death was 2.34 among patients treated with systemic therapy alone, as compared with patients who received combined therapy (95 percent confidence interval, 1.10 to 4.98; P=0.027), after adjustment for important variables. The rates of adverse effects of at least moderate severity were similar in the two groups, except for a higher frequency of diarrhea and hepatic effects in the combined-therapy group. CONCLUSIONS: For patients who undergo resection of liver metastases from colorectal cancer, postoperative treatment with a combination of hepatic arterial infusion of floxuridine and intravenous fluorouracil improves the outcome at two years.

A single base substitution in the variable pocket of yeast tRNA(Arg) eliminates species-specific aminoacylation.

Early biochemical data showed that aminoacyl-tRNA synthetases often displayed species-specific recognition of tRNA. We compared the ability of purified Saccharomyces cerevisiae and Escherichia coli arginyl-tRNA synthetases to aminoacylate native and transcribed yeast tRNA(Arg) as well as E. coli tRNA(Arg). The kinetic data revealed that yeast ArgRS could charge E. coli tRNA(Arg), but at a lower efficiency than it charged either the transcribed or native yeast tRNA(Arg). E. coli ArgRS can acylate only its cognate E. coli tRNA. Strikingly, a single base change from C to A at position 20 in yeast tRNA(3)(Arg) altered the species specificity. The transcript of yeast tRNA(3)(Arg)CA20 mutant was aminoacylated by E. coli ArgRS with a 10(6) increase in k(cat)/K(m) over that for aminoacylation of yeast tRNA(3)(Arg) transcript. This indicates that A20 is not only an important identity of E. coli tRNA(Arg), but is also the key to altering species-specific aminoacylation of yeast tRNA(Arg).

The presence of an endogenous endo-D-arabinase in Mycobacterium smegmatis and characterization of its oligoarabinoside product.

Endogenous mycobacterial endo-D-arabinase activity, which degrades cell wall polysaccharide arabinogalactan, was found in Mycobacterium smegmatis. The arabinan product contains 20-30 arabinosyl residues but no galactofuranosyl residues. Recognition of this endogenous activity results in the possibility of developing antituberculosis drugs that do not require bacterial growth for activity.

Functional analysis of R651 mutations in the putative helix 6 of rat glucocorticoid receptors.

Trypsin digestion of steroid-free, but not steroid-bound, rat glucocorticoid receptor (GR) has recently been reported to occur at arginine-651 (R651). This residue is close to the affinity labeled Cys-656 and thus could be a sensitive probe of steroid binding. This hypothesis is supported by the current model of the GR ligand binding domain (LBD), which is based on the X-ray structures of several related receptor LBDs and places R651 in the middle of the putative alpha-helix 6 (649-EQRMS-653 of rat GR), close to the bound steroid. To test this model, R651, which could be involved in hydrophilic and/or hydrogen bonding, was mutated to alanine (A), which favors alpha-helices, the helix breakers proline (P) and glycine (G), or tryptophan (W). All receptors were expressed at about the same level, as determined by Western blots, but the cell-free binding activity of R651P was reduced twofold. The cell-free binding affinities were all within a factor of 10 of wild type receptors. Whole cell biological activity with transiently transfected receptors was determined with a variety of GR agonists (dexamethasone and deacylcortivazol) or antagonists (dexamethasone mesylate, RU486, and progesterone). Reporters containing both simple (GRE) and complex (MMTV) enhancers were used to test for alterations in GR interactions with enhancer/promoter complexes. Surprisingly, no correlation was observed between biological activity and ability to preserve alpha-helical structures for each point mutation. Finally, similar trypsin digestion patterns indicated no major differences in the tertiary structure of the mutant receptors. Collectively, these results argue that the polar/ionizable residue R651 is not required for GR activity and is not part of an alpha-helix in the steroid-free or bound GR. The effect of these mutations on GR structure and activity may result from a cascade of initially smaller perturbations. These LBD alterations were the most varied for interactions with deacylcortivazol and RU 486, which have recently been predicted to be sub-optimal binders due to their large size. However, further analyses of ligand size versus affinity suggest that there is no narrowly defined optimal size for ligand binding, although larger ligands may be more sensitive to modifications of LBD structure. Finally, the changes in GR activity with the various mutations seem to result from altered LBD interactions with common, as opposed to enhancer specific, transcription factors.

Case study of the effects of atmospheric aerosols and regional haze on agriculture: an opportunity to enhance crop yields in China through emission controls?

The effect of atmospheric aerosols and regional haze from air pollution on the yields of rice and winter wheat grown in China is assessed. The assessment is based on estimates of aerosol optical depths over China, the effect of these optical depths on the solar irradiance reaching the earth's surface, and the response of rice and winter wheat grown in Nanjing to the change in solar irradiance. Two sets of aerosol optical depths are presented: one based on a coupled, regional climate/air quality model simulation and the other inferred from solar radiation measurements made over a 12-year period at meteorological stations in China. The model-estimated optical depths are significantly smaller than those derived from observations, perhaps because of errors in one or both sets of optical depths or because the data from the meteorological stations has been affected by local pollution. Radiative transfer calculations using the smaller, model-estimated aerosol optical depths indicate that the so-called "direct effect" of regional haze results in an approximately 5-30% reduction in the solar irradiance reaching some of China's most productive agricultural regions. Crop-response model simulations suggest an approximately 1:1 relationship between a percentage increase (decrease) in total surface solar irradiance and a percentage increase (decrease) in the yields of rice and wheat. Collectively, these calculations suggest that regional haze in China is currently depressing optimal yields of approximately 70% of the crops grown in China by at least 5-30%. Reducing the severity of regional haze in China through air pollution control could potentially result in a significant increase in crop yields and help the nation meet its growing food demands in the coming decades.

PET imaging of serotonin 1A receptor binding in depression.

BACKGROUND: The serotonin-1A (5HT1A) receptor system has been implicated in the pathophysiology of major depression by postmortem studies of suicide victims and depressed subjects dying of natural causes. This literature is in disagreement, however, regarding the brain regions where 5HT1A receptor binding differs between depressives and controls and the direction of such differences relative to the normal baseline, possibly reflecting the diagnostic heterogeneity inherent within suicide samples. PET imaging using the 5HT1A receptor radioligand, [11C]WAY-100635, may clarify the clinical conditions under which 5HT1A receptor binding potential (BP) is abnormal in depression. METHODS: Regional 5HT1A receptor BP values were compared between 12 unmedicated depressives with primary, recurrent, familial mood disorders and 8 healthy controls using PET and [carbonyl-11C]WAY-100635. Regions-of-interest (ROI) assessed were the mesiotemporal cortex (hippocampus-amygdala) and midbrain raphe, where previous postmortem studies suggested 5HT1A receptor binding is abnormal in depression. RESULTS: The mean 5HT1A receptor BP was reduced 41.5% in the raphe (p < .02) and 26.8% in the mesiotemporal cortex (p < .025) in the depressives relative to the controls. Post hoc comparisons showed the abnormal reduction in 5HT1A receptor BP was not limited to these regions, but extended to control ROI in the occipital cortex and postcentral gyrus as well. The magnitude of these abnormalities was most prominent in bipolar depressives (n = 4) and unipolar depressives with bipolar relatives (n = 4). CONCLUSIONS: Serotonin-1A receptor BP is abnormally decreased in the depressed phase of familial mood disorders in multiple brain regions. Of the regions tested, the magnitude of this reduction was most prominent in the midbrain raphe. Converging evidence from postmortem studies of mood disorders suggests these reductions of 5HT1A receptor BP may be associated with histopathological changes involving the raphe.

A genetic map and recombination parameters of the human malaria parasite Plasmodium falciparum.

Genetic investigations of malaria require a genome-wide, high-resolution linkage map of Plasmodium falciparum. A genetic cross was used to construct such a map from 901 markers that fall into 14 inferred linkage groups corresponding to the 14 nuclear chromosomes. Meiotic crossover activity in the genome proved high (17 kilobases per centimorgan) and notably uniform over chromosome length. Gene conversion events and spontaneous microsatellite length changes were evident in the inheritance data. The markers, map, and recombination parameters are facilitating genome sequence assembly, localization of determinants for such traits as virulence and drug resistance, and genetic studies of parasite field populations.
