[The study on effects of human fetal liver cells on immunity of BALB/c mice].

This study sought to better understand the effects of human fetal liver cells(FLC) on immunity. FLC were injected into BALB/c mice intraperitoneally. The design included four groups: negative control and three dose (125 x 10(5), 25 x 10(5), 5 x 10(5) FLC/day) groups. The results showed that FLC increased the thymus visceral coefficient, numbers of T-lymphocytes(TLC) and lymphocytes(LC) in blood, total number of spleen cells, the number of plaque forming cells(PFC) of spleen, the rate(%) of transformation and stimulus index of TLC of spleen, and the function of macrophages. FLC also effected the function of natural killer cells(NK cells) of BALB/c mice. The findings suggest that human FLC can strengthen the nonspecific immunity, humoral immunity and cellular immunity in mice. In this study, however, no effect of FLC on spleen visceral coefficient was noticed, and no sign of delayed type allergy was observed.

[A histopathologic and immunohistochemical study of 68 cases of hemangioblastoma].

The aim of this study was to evaluate the histologic characteristics of hemangioblastoma (HB) and investigate the histogenesis of its stromal cells. Sixty-eight cases of hemangioblastoma were studied by light microscopy and thirty-three cases were examined by immunohistochemical stainning. Forty-eight of the 68 patients were males and 20 were females. The average age was 37. Fifty-two cases (76.6%) had been present for two months to one year before the diagnosis was made. Sixty cases (88.2%) arose in the cerebellum. Fifty cases (73.5%) appeared as cystic nodules. Histologically, the 68 cases of HB were further categorized into three subtypes: typical (30 cases), cellular (21 cases) and reticular (17 cases). In 33 cases, the stromal cell were positive for NSE, but negative for F-VIII, UEA-1, GFAP and EMA. In summary, there are three essential histologic categories of hemangioblastoma, and its stromal cells may well have the characters of neuroendocrine differentiation.

[Pharmacognostical identification of one Rhizoma Drynariae habitually used species from Yunnan].

The species recorded on the "Rhizoma Drynariae" item of Chinese Pharmacopeia is the dry rhizoma of Drynaria fortunei from Polypodiacae. But in Yunnan province, many plants (including D. delavayi) are habitually used as Rhizoma Drynariae. In this article the authors identified D. delavayi on Chinese medinal materials discriptions, microscopic characters and TLC. This proved evidences for the utilization and identification of D. delavayi in some extent.

[Clinical investigation of 0.3% levofloxacin eyedrops on the treatment of cases with acute bacterial conjunctivitis and bacterial keratitis].

PURPOSE: To investigate the effects of 0.3% levofloxacin eyedrops on the treatment of acute bacterial conjunctivitis and bacterial keratitis. METHODS: The patients were randomly divided into treated group and control group. They were given 0.3% levofloxacin and 0.3% ofloxacin eyedrops respectively. RESULTS: From April to December in 1999, 132 cases were enrolled in the study. It showed that there was no significant difference between two drops in the cure and effective rates, but the mean cure day of keratitis in levofloxacin group was significantly shorter than that in ofloxacin group. No serious side effects occurred. CONCLUSION: Levofloxacin eyedrops was effective and safe antibiotics in treating bacterial conjunctivitis and keratitis.

[Application of extended high frequency audiometry in the early diagnosis of noise--induced hearing loss].

OBJECTIVE: To assess the application of 10-20 kHz audiometry in early diagnosis of noise-induced hearing loss and to explore the relationship between hearing loss and subjects' age, the exposure time as well as the noise level. METHODS: One thousand workers with noise exposure history were examined by pure tone audiometry(0.5-20 kHz). One hundred and twenty normal subjects served as the control group. RESULTS: The noise levels at working sites ranged from 95 to 115dB(A). Compared with the control group, noise-exposed subjects showed minor changes in the thresholds from 0.5 to 6 KHz, whereas the thresholds were elevated in the high frequency range of 10-16 kHz (P < 0.001). The incidence of ears without any response to the maximum output at the frequency range of 14 to 20 kHz was much higher (P < 0.01) than that of the control group. In the animals exposed to noise, the thresholds of 10-18 kHz increased with the aging and with increase of working time (6-10 years), but there were no significant differences among subjects exposed to the noises at different levels(P > 0.05). CONCLUSION: In noise-exposed ears, the occurrence of threshold changes in the high frequency range (10 to 20 kHz) is earlier than that in the low frequency range (0.5 to 6 kHz). Aging and working time also affect the 10-20 kHz hearing thresholds. The lack of responses to maximum output and the changes in thresholds in the high frequency range (10-20 kHz) can be used as indices for the diagnosis of early noise-induced hearing loss and for the assessment of the susceptibility of the subject to noise damage.

[Effects of 1 alpha(OH)D3 on trabecular and cortical bone in ovariectomized rats].

OBJECTIVE: To study the pharmaceutical effects of 1 alpha(OH)D3 on trabecular and cortical bone in ovariectomized (OVX) rats. METHODS: 41 female Wistar rats of six months old were randomly divided into 5 groups: (1) Baseline control; (2) Sham control; (3) 6 weeks after OVX (OVXb); (4) 14 weeks after OVX (OVXe); (5) OVX + 0.1 microgram/(kg.d) 1 alpha(OH)D3 (O + VD), treatment started 6 weeks after OVX and lasted 8 weeks. Histomorphometry analysis of tibia, peripheral quantitative computed tomography (pQCT) scanning of femur, bone biomechanical test and serum and urinary biochemical parameters were determined. RESULTS: The levels of bone turnover indexes increased in OVX rats, OVX also resulted in reducing of trabecular, bone mass and biomechanical properties. The ratio of urinary deoxypyridinoline crosslink/creatinine was decreased by 67.0% in O + VD group compared with OVXe group 67.0% [(43.50 +/- 11.20) nmol.L-1/mmol.L-1 vs(131.80 +/- 14.90) nmol.L-1/mmol.L-1, P < 0.01]. Percent trabecular area (Cn-BV/TV) was increased by 89.8% in O + VD group compared with OVXe group (11.03 +/- 0.73 vs 5.81 +/- 1.29, P < 0.05). Trabecular bone mineral content and density were increased by 77.3% and 91.3% compared with OVXe respectively (P < 0.05). Although cancellous maximal load and stiffness increased in O + VD group, but no statistical significance. 1 alpha(OH)D3 also enhanced polar moment of inertia (PMI) and maximal load of cortical bone in femur compared with OVXe (23.70 +/- 1.63 vs 18.23 +/- 1.41, P < 0.01 and 171.69 +/- 9.92 vs 147.58 +/- 11.29, P < 0.05 respectively). CONCLUSION: 1 alpha(OH)D3 inhibited the higher bone turnover induced by OVX, increased trabecular bone in proximal tibia and bone mass in distal femur. 1 alpha(OH)D3 also improved the mechanical properties of cortical bone in femur.

[Determination of eight impurity elements in organic dye oil red by ICP-AES].

Simultaneous determination of metal impurities of Fe, Al, Cu, Pb, Zn, Cr, Mn and Ti in organic dye oil red by inductively coupled plasma atomic emission spectrometry (ICP-AES) is presented in this paper. Different sample digestion procedures were compared, and methods for the elimination of spectrum interference were investigated. The proposed sample digestion procedure includes charring with concentrated sulfuric acid and nitric acid, cineration at 500 degrees C, and step-wisely extracting the residue with hydrochloric acid plus peroxide of hydrogen, and hydrochloric acid plus nitric acid (VHCl:VHNO3 = 1:1). The detection limits are 0.15 mg.L-1 for Fe, 1.7 x 10(-2) mg.L-1 for Al, 5.0 x 10(-3) mg.L-1 for Cu, 0.20 mg.L-1 for Pb, 2.7 x 10(-2) mg.L-1 for Zn, 5.7 x 10(-3) mg.L-1 for Cr, 1.3 x 10(-3) mg.L-1 for Mn, 2.1 x 10(-3) mg.L-1 for Ti, respectively. The precision of the method is in the range of 0.8%-21.5% (n = 5), and recovery in the range of 84.6%-118.6%.

Ascorbate and glutathione regulation in hibernating ground squirrels.

Ground squirrels withstand up to 90% reductions in cerebral blood flow during hibernation as well as rapid reperfusion upon periodic arousals from torpor. Metabolic suppression likely plays a primary adaptive role which allows hibernating species to tolerate such phenomena. However, several other aspects of hibernation physiology are also consistent with tolerance to dramatic fluctuations in cerebral blood flow, suggesting that multiple neuroprotective adaptations may work in concert during hibernation. The purpose of the present work was to study the dynamics of the low molecular weight antioxidants, ascorbate and glutathione (GSH), during hibernation. Alterations in concentrations of ascorbate during hibernation and arousal in two species of hibernating ground squirrels suggest that it could play a protective role during hibernation or arousal. Samples were collected during the hibernation season from arctic ground squirrels (AGS; Spermophilus parryii) and 13-lined ground squirrels (TLS; S. tridecemlineatus) during prolonged torpor and in squirrels that did not hibernate or had not been hibernating for several weeks. We determined antioxidant levels in plasma, cerebrospinal fluid (CSF), and in frontal cortex, hippocampus and cerebellum using high-performance liquid chromatography (HPLC). Plasma ascorbate concentrations increased dramatically (3-4-fold) in both species during hibernation and rapidly returned to prehibernation levels upon arousal. By contrast, plasma GSH concentrations fell slightly or remained stable during hibernation. Ascorbate levels in the CSF doubled in hibernating AGS (not determined in TLS), while brain ascorbate content fell slightly (10-15%) in both species. Substantial increases in plasma and CSF ascorbate concentrations suggest that this antioxidant could play a protective role during hibernation and reperfusion upon arousal from hibernation.

The Arabidopsis SKP1-LIKE1 gene is essential for male meiosis and may control homologue separation.

The yeast and human SKP1 genes regulate the mitotic cell cycle but are not yet known to be required for meiosis. Nine Arabidopsis SKP1 homologues have been uncovered and are named ASK1 through ASK9. Here, we report the isolation and characterization of a male sterile Arabidopsis mutant and show that the mutant defect was caused by a Ds transposon insertion into the ASK1 gene. In the ask1-1 mutant, abnormal microspores exhibit a range of sizes. Furthermore, during mutant male meiosis, although homologous chromosome pairing appeared normal at metaphase I, chromosome segregation at anaphase I is unequal, and some chromosomes are abnormally extended. Therefore, in ask1-1, at least some homologues remain associated after metaphase I. In addition, immunofluorescence microscopy indicates that the mutant spindle morphology at both metaphase I and early anaphase I is normal; thus, the abnormal chromosome segregation is not likely caused by a spindle defect. Because the yeast Skp1p is required for targeting specific proteins for ubiquitin-mediated proteolysis, we propose that ASK1 controls homologue separation by degrading or otherwise removing a protein that is required directly or indirectly for homologue association before anaphase I.

Structural and functional characterization of the 5'-flanking region of the rat and human cytochrome P450 2E1 genes: identification of a polymorphic repeat in the human gene.

Cytochrome P450 2E1 (CYP2E1) is a toxicologically very important enzyme with a high extent of interindividual variability in expression. We sequenced and characterized the 5'-flanking region of the human and rat CYP2E1 genes. The identity between the human and rat sequences (-3.8 kb to +1 kb) was generally between 35 and 60%, and the most similar regions were found in the proximal part of the sequence. Two more distant regions at -1.6 to -2.0 kb and -2.5 to -2. 8 kb in the human sequence were also found to have high identity to the rat sequence. A polymorphic repeat sequence in the human gene was found between -2178 to -1945 bp. The common allele (CYP2E1*1C) contained 6 repeats (each 42-60 bp long) and the rare allele (CYP2E1*1D) had 8 repeats with an allele frequency of 1% among Caucasians and 23% among Chinese. The CYP2E1 5'-flanking regions of the human (-3712 bp to +10 bp) and rat (-3685 bp to +28 bp) genes were ligated in front of a luciferase reporter gene and transfected into rat hepatoma Fao and human hepatoma B16A2 cells. Important species specificity was noted in the control of gene expression and regions of negative and positive cis-acting elements were localized. No difference was seen in the constitutive expression between the two polymorphic forms. The importance of this repeat polymorphism for high and low inducible CYP2E1 phenotypes is discussed.

Cyclic strain stress-induced mitogen-activated protein kinase (MAPK) phosphatase 1 expression in vascular smooth muscle cells is regulated by Ras/Rac-MAPK pathways.

Recently, we demonstrated that mechanical stress results in rapid phosphorylation or activation of platelet-derived growth factor receptors in vascular smooth muscle cells (VSMCs) followed by activation of mitogen-activated protein kinases (MAPKs) and AP-1 transcription factors (Hu, Y., Bock, G., Wick, G., and Xu, Q. (1998) FASEB J. 12, 1135-1142). Herein, we provide evidence that VSMC responses to mechanical stress also include induction of MAPK phosphatase-1 (MKP-1), which may serve as a negative regulator of MAPK signaling pathways. When rat VSMCs cultivated on a flexible membrane were subjected to cyclic strain stress (60 cycles/min, 5-30% elongation), induction of MKP-1 proteins and mRNA was observed in time- and strength-dependent manners. Concomitantly, mechanical forces evoked rapid and transient activation of all three members of MAPKs, i.e. extracellular signal-regulated kinases (ERKs), c-Jun NH(2)-terminal protein kinases (JNKs), or stress-activated protein kinases (SAPKs), and p38 MAPKs. Suramin, a growth factor receptor antagonist, completely abolished ERK activation, significantly blocked MKP-1 expression, but not JNK/SAPK and p38 MAPK activation, in response to mechanical stress. Interestingly, VSMC lines stably expressing dominant negative Ras (Ras N17) or Rac (Rac N17) exhibited a marked decrease in MKP-1 expression; the inhibition of ERK kinases (MEK1/2) by PD 98059 or of p38 MAPKs by SB 202190 resulted in a down-regulation of MKP-1 induction. Furthermore, overexpressing MKP-1 in VSMCs led to the dephosphorylation and inactivation of ERKs, JNKs/SAPKs, and p38 MAPKs and inhibition of DNA synthesis. Taken together, our findings demonstrate that mechanical stress induces MKP-1 expression regulated by two signal pathways, including growth factor receptor-Ras-ERK and Rac-JNK/SAPK or p38 MAPK, and that MKP-1 inhibits VSMC proliferation via MAPK inactivation. These results suggest that MKP-1 plays a crucial role in mechanical stress-stimulated signaling leading to VSMC growth and differentiation.

Variability of somatosensory-evoked potentials in different stages of scoliosis surgery.

STUDY DESIGN: A comparison of the latencies and amplitudes of the somatosensory-evoked potential in different stages of scoliosis surgery. OBJECTIVES: To investigate the normal variability of the somatosensory-evoked potential at different stages of scoliosis surgery and to improve the reliability of spinal cord monitoring. SUMMARY OF BACKGROUND DATA: False alarms in intra-operative spinal cord monitoring have been reported when the somatosensory-evoked potential measured before skin incision was used as the reference baseline value. However, the normal variability during different stages of surgery and the specificity of somatosensory-evoked potential is not well documented. METHODS: The somatosensory-evoked potential of 90 patients who underwent surgical correction for idiopathic scoliosis was monitored intraoperatively, with stimuli applied to the posterior tibial nerve. The amplitudes and latencies of the somatosensory-evoked potential were recorded over Cz'-Fz and Cv-Fz. The values of latencies and amplitudes and their percentage variabilities were analyzed. RESULTS: The values of latencies and their percentage variabilities did not differ significantly from each other in different stages of surgery. The values of amplitudes between different stages showed statistically significant differences. In particular, the amplitude of the somatosensory-evoked potential was found to be significantly decreased after exposure of the spine when compared with the amplitude before skin incision. CONCLUSION: Some variability in latencies and amplitudes between different stages of scoliosis surgery is normal. These results suggest that the somatosensory-evoked potential when the spine is exposed may be used as the reference baseline to determine whether somatosensory-evoked potentials are subnormal at the subsequent stages of surgery.

Lepidopteran DALP, and its mammalian ortholog HIC-5, function as negative regulators of muscle differentiation.

During myogenesis, reductions in trophic factor availability signal most myoblasts to fuse, up-regulate the expression of muscle-specific genes, and form myotubes. Those cells failing to differentiate into myotubes initiate apoptosis and rapidly die. At present, the signal-transduction molecules that determine whether myoblasts should differentiate or die are largely unknown. In this report, we describe the cloning and characterization of DALP, a small LIM-only type zinc-finger protein that is induced when the intersegmental muscles (ISMs) of the moth Manduca sexta become committed to die at the end of metamorphosis. Forced expression of death-associated LIM-only protein (DALP) in Drosophila results in skeletal muscle atrophy. Ectopic expression of DALP, or its mammalian ortholog Hic-5, blocks differentiation and induces apoptosis in mouse C(2)C(12) myoblasts. Both of these effects can be overcome by contact with normal myoblasts or by ectopic expression of the muscle-specific transcription factor MyoD. Hic-5 expression is specifically and dramatically induced in normal myoblasts that die after removal of trophic support. Taken together, these data suggest that DALP and Hic-5 act upstream of MyoD and function as phylogenetically conserved "switches" to block muscle differentiation and induce death.

Inhibition of neointima hyperplasia of mouse vein grafts by locally applied suramin.

BACKGROUND: Saphenous vein grafts are widely used for aortocoronary bypass surgery as treatment for severe atherosclerosis and often are complicated by subsequent occlusion of the graft vessel. METHODS AND RESULTS: We described a mouse model of venous bypass graft arteriosclerosis that can be effectively retarded by locally applied suramin, a growth factor receptor antagonist. Mouse isogeneic vessels of the vena cava veins pretreated with suramin were grafted end to end into the carotid arteries and enveloped with a mixture of suramin (1 mmol/L) and pluronic-127 gel. In the untreated group, vessel wall thickening was observed as early as 1 week after surgery and progressed to 4-fold and 10-fold the original thickness in grafted veins at 4 and 8 weeks, respectively. Pluronic-127 gel alone did not influence neointima formation. Suramin treatment reduced the neointima hyperplasia 50% to 70% compared with untreated controls. Immunohistochemical studies demonstrated that a significant proliferation of vascular smooth muscle cells (SMCs) constituted neointimal lesions between 4 and 8 weeks. The majority of SMCs expressed platelet-derived growth factor (PDGF) receptors-alpha and -beta, which were significantly reduced by suramin treatment. In vitro studies indicated that suramin completely blocked PDGF receptor activation or phosphorylation stimulated by PDGF-AB, inhibited activation of mitogen-activated protein kinase (ERK) kinases (MEK1/2) and ERK1/2, and abrogated transcription factor AP-1 DNA-binding activity. CONCLUSIONS: Suramin inhibited SMC migration and proliferation in vivo and in vitro by blocking PDGF-initiated PDGF receptor and MAPK-AP-1 signaling. These findings indicate that locally applied suramin is effective in a mouse model of venous bypass graft arteriosclerosis.

Determination of striatal extracellular gamma-aminobutyric acid in non-hibernating and hibernating arctic ground squirrels using quantitative microdialysis.

This study determined extracellular concentrations of gamma-aminobutyric acid ([GABA](ecf)) in striatum of non-hibernating and hibernating arctic ground squirrels to test the hypothesis that an increase in [GABA](ecf) was associated with profound CNS depression during hibernation. Quantitative microdialysis procedures were employed to circumvent the effects of low temperature on the relative recovery of the analyte across the dialysis membrane and yielded for the first time quantitative in vivo estimates of [GABA](ecf) in any brain region or any species. Laboratory housed, wild caught Arctic ground squirrels (Spermophilus parryii) were implanted intraperitoneally with radio transmitters that enabled the telemetric monitoring of activity and core body temperature (T(b)) and bilaterally implanted with cranial guide tubes that enabled the implantation of microdialysis probes into the striatum. Striatal [GABA](ecf) was determined in unrestrained, non-hibernating ground squirrels (T(b) range 34.7-38.9 degrees C) and hibernating ground squirrels (T(b) range 2.9-3.9 degrees C) using extrapolation to zero flow and very slow flow microdialysis techniques. The results show that [GABA](ecf) in non-hibernating squirrels was 73 nM and this level was decreased by approximately 50% during hibernation thereby suggesting that an increase in [GABA](ecf) does not play a major role in CNS depression during hibernation. The reduction of [GABA](ecf) parallels a decrease in plasma and CSF [glucose] and may be related to a decrease in GABA synthesis or reduced voltage dependent release. This paper demonstrates that measurement of extracellular concentrations of neurotransmitters in animals with vastly different body temperatures is possible using microdialysis techniques of extrapolation to zero flow or very slow flow rates that enable 100% recovery. Such quantitative techniques may prove valuable in the study of the neurochemistry of the cerebral mechanisms of hibernation and tolerance to cerebral ischemia exhibited by hibernating animals.

An ultrastructural analysis of tissue surrounding a microdialysis probe.

Microdialysis is a widely used in vivo sampling technique commonly used to monitor extracellular levels of a variety of molecules including neurotransmitters and metabolites. To facilitate interpretation of microdialysis results, this study critically examines changes in synaptic morphology induced by microdialysis. Tissue surrounding microdialysis probes was examined using light and electron microscopy at three distances from the probe tract. Microdialysis probes were implanted into rat striatum, and after 40 h of post-operative recovery were perfused with a modified Ringer's solution. Light microscope analysis revealed tissue disruption up to 1.4 mm from the probe site. Axonal damage indicative of non-excitotoxic insult was also seen as far away from the probe as was examined. The presence of dark-degenerating neurons was also noted and estimates of neuronal densities revealed loss up to 400 microm from the probe tract. This study, the first qualitative ultrastructural investigation of neuropil surrounding the probe site, indicated swollen processes up to 1.4 mm from the probe tract. Swollen mitochondria and bloated endoplasmic reticulum suggest intracellular chemical disruption. Tissue damage resulting in synaptic and neuronal disruption may affect neurotransmitter efflux or extracellular concentrations of metabolites.

Randomization of the receptor alpha chain recruitment epitope reveals a functional interleukin-5 with charge depletion in the CD loop.

We report the functional phage display of single chain human interleukin-5 (scIL-5) and its use for receptor-binding epitope randomization. Enzyme-linked immunosorbent assays and optical biosensor analyses verified expression of scIL-5 on the phage surface and binding of scIL-5 phage to interleukin-5 receptor alpha chain. Furthermore, an asymmetrically disabled but functional scIL-5 mutant, (wt/A5)scIL-5, was displayed on phage. (wt/A5)scIL-5 was constructed from an N-terminal half containing the original five charged residues (88EERRR92) in the CD loop, including the Glu89 and Arg91 believed key in the alpha chain recognition site, combined with a C-terminal half containing a disabled CD loop sequence (88AAAAA92) missing the key recognition residues. This asymmetric variant was used as a starting point to generate an scIL-5 library in which the intact 88-92 N-terminal CD loop was randomized. From this epitope library, a receptor-binding variant of IL-5 was detected, (SLRGG/A5)scIL-5, in which the only charged residue in the CD loop is an Arg at position 90. Characterization of this variant expressed as a soluble protein in E. coli shows that the IL-5 pharmacophore for receptor alpha chain binding can function with a single positive charge in the CD loop. Charge-depleted CD loop mimetics of IL-5 suggest the importance of charge distribution in functional IL-5 receptor recruitment.
