Detection of IgG rheumatoid factor by concanavalin A treatment and complement fixation with IgG rheumatoid factor.

Concanavalin A (Con A) froms precipitates with carbohydrate-rich protein such as IgM, IgD, IgE, and IgA. Since IgG contains little carbohydrate and does not react with Con A, the activity of IgG-rheumatoid factor (RF) can be measured in the supernate of the Con A-treated serum. When the latex fixation test (LFT) and the sensitized sheep cell agglutination test (SSCA) were perfromed in the supernate for the detection of IgG-RF, LFT was positive in 32-1% of sera, out of 137 sera originally positive for LFT, and SSCA was positive in 18-5% of sera, out of 119 sera originally positive for SSCA. IgG-RF exhibited lower complement fixing ability than IgM-RF and correlated with agglutination titres of IgG-RF, while the CH50 of the original serum did not correlate with haemolytic activities of either IgM-RF or IgG-RF.

Elevated levels of IgE antibodies to ascaris and mite antigens in Papua New Guinea.

Levels of IgE as well as specific IgE antibodies to ascaris and mite have been studied in two groups in Papua New Guinea (PNG), patients with primary liver carcinoma (PLC) and normal blood donors, and in two groups in Japan, normal subjects and asthmatic patients. A radioallergosorbent test was used to measure IgE antibodies to ascaris and mite antigens. Much higher levels of IgE were found in the Papua New Guinean subjects than the normal Japanese subjects. Also levels of IgE antibodies to ascaris were much higher in Papua New Guineans than in the Japanese subjects. Though similar results were observed in IgE antibodies to mite, difference between normal Papua New Guineans and normal Japanese was small, whereas difference between the patients with PLC in PNG and the normal Japanese subjects was very large. Highest levels of IgE antibodies to mite were detected in Japanese with asthma. No correlations were observed among IgE levels, levels of IgE antibodies to ascaris and levels of IgE antibodies to mite. It is concluded that both ascaris and mite IgE antibodies contribute the very high levels of IgE seen in Papua New Guineans, but many other factors may be operative.

Delayed-type hypersensitivity reaction in the lungs of guinea pigs due to potassium dichromate.

Potassium dichromate was inhaled by guinea pigs previously immunized by potassium dichromate until strong positive patch tests were obtained. No obvious respiratory changes were noted during and after inhalation. Histologically, however, mononuclear cells infiltrated the interstitial spaces in large areas of the lung, producing considerable thickening of the alveolar spaces in 24 to 48 hr after inhalation. Polymorphonuclear cells were predominant initially. These changes were similar to the delayed-type hypersensitivity reaction in the lung elicited by the inhalation of purified protein derivative (PPD) in the guinea pigs immunized by an injection of dry-killed tubercle bacilli. A less marked reaction was observed in guinea pigs passively sensitized with peritoneal exudate cells and lymph node cells. Consequently, the pulmonary changes were thought to be elicited by delayed-type hypersensitivity reaction due to a simple chemical. The clinical implications of delayed-type hypersensitivity reaction in the lung due to simple chemicals are discussed.

The distribution of polymorphic traits in atopic asthmatic patients.

The phenotypic distribution and gene frequencies of haptoglobin (Hp), transferrin (Tf), group specific component (Gc), cholinesterase (Cho E2), and alpha1-antitrypsin (Pi) in plasma proteins, and phosphoglucomutase (PGM), 6-phosphogluconate dehydrogenase ((6-PGD), esterase D (Es D), phosphohexose isomerase (PHI), adenosine deaminase (ADA) and acid phosphatase (AcP) in red cells were studied in 127 atopic, asthmatic patients. The gene frequencies were compared with normal groups. The phenotypic distribution of the Pi system in atopic patients was somewhat different from the normal. No significant differences were found between the two groups in protein systems or in enzyme systems, except Pi systems. In conclusion, except for the Pi system, no definite association between polymorphic traits and atopic asthma was found in this study.

Fc-rosette inhibition by hypocomplementaemic systemic lupus erythematosus sera.

Human red cells sensitized with one of the Rh antisera (Ripley) form rosettes (Fc-rosette) with human B lymphocytes and the rosettes are well inhibited by aggregated human IgG. Since sera of hypocomplementaemic patients with systemic lupus erythematosus (SLE) have frequently been reported to contain immune complexes, they were used for the inhibition of Fc-rosette formation in this study. The results of Fc-rosette inhibition rates of the sera were inversely correlated with the serum CH50 levels. When the sera were separated into top, middle, and bottom fractions by ultracentrifugation, the bottom fractions showed more effective inhititions than the others. Similarly, the strongest inhibition was found in the void volume of the serum separated by Sephadex G200 gel filtration. Reduction and alkylation of IgG resulted in the loss of reactivity with Fc receptor of B lymphocytes, and the rosette inhibiting activities of the SLE sera were markedly reduced after reduction and alkylation. Some of anti-HLA sera were inhibitory for the Fc-rosette formation, while the tested sera did not contain anti-HLA activity assessed by the microcytotoxicity test. These results indicated that circulating immune complexes contained in the sera inhibit the rosette formation, and that the Fc-rosette inhibition test is a simple and relatively sensitive method for the detection of circulating immune complexes. Antinuclear antibody activities of the sera were tested by the indirect immunofluorescent method; however, clear correlations were not obtained between Fc-rosette inhibition rates and staining patterns of antinuclear antibodies. On the other hand, the positive groups of LE-test exhibited slightly greater inhibition rates of the rosette than the negative groups.

Further studies in allergenic identity between house dust and the house dust mite, Dermatophagoides farinae Hughes, 1961.

The sera from 99 Japanese asthmatic subjects were studied for the allergenic similarity between house dust and the House-dust Mite, Dermatophagoides farinae Hughes, 1961. A close correlation (correlation coefficient 0.92, P less than 0.01) was obtained between house dust and the mite with the radioallergosorbent test (RAST). High correlation was not noted between house dust and other allergens such as smoky brown cockroach, Periplaneta fuliginosa, Aspergillus, book lice, Liposcelis bostricophilus, Cheyletus malaccensis and Tribolium confusum. IgE antibody to the mite reduced significantly after the incubation of the sera with house dust and IgE antibody to house dust reduced markedly after the incubation of the sera with the mite. These results further support the view that the mite is the most important allergenic component in house dust.
