The use of Fourier transform infrared spectrometry for monitoring the retention of polymers in the vitreous humour.

A frequent cause of the failure of various materials tested as artificial vitreous substitutes is their short retention in the vitreous humour of the eye. Previous techniques to monitor the residence time of the vitreous substitutes have been traditionally complicated and usually involved the chemical modification of the test fluids. Fourier transform infrared (FTIR) spectrometry is used in this study to determine the rate of disappearance of a crosslinked poly(1-vinyl-2-pyrrolidinone) (PVP) hydrogel after injection into the vitreous humour of rabbits. The fluid removed from the eye at various postoperative intervals of time (1 day, 1 week, and 1 month) was placed on a horizontal attenuated reflectance (ATR) unit, and the infrared spectrum from 700 to 3000 cm-1 was obtained directly. The absorption band at 1295 cm-1 due to C-N stretching vibration does not overlap with bands of any other species (natural vitreous, blood) present in the test fluid. By estimating the intensity of this band, it was found that about 80% of the injected polymer was removed from the vitreous cavity within 1 month. This study shows that (1) FTIR spectrometry is a convenient method to assess the residence time of foreign materials in the vitreous humour, and (2) the PVP gel is unsuitable in this current form as a permanent vitreous substitute, regardless of the pathways of its removal, such as phagocytosis of fragments produced by the injection process, or enzymatic biodegradation.

[Studies on the deltamethrin-medicated bath of domestic dogs for interrupting visceral leishmaniasis transmission].

In a previous paper, it was reported that deltamethrin-medicated bath of domestic dogs had a strong killing effect against Ph. chinensis, and could also inhibit its blood sucking ability or cause its death shortly after blood sucking. The effect could last for 30 days to 114 days, and mostly for 2 months in the local area. Based on our previous study, three villages (Yongle, Yongfeng, Anle) and a town (Chengguan) were selected from the mountainous region with high incidence of visceral leishmaniasis in Nanping County, northern Sichuan for carrying out an extensive experiment of medicated bathing of domestic dogs, where there were 13 new infected cases in the year of 1991 before the experiment. During the sandfly season in 1992, two rounds of medicated bathing of dogs were conducted, the drug concentration used was 50 ppm. From September 1992 to August 1993, the number of new infected cases was 11 in the above mentioned three villages and a town. Another two rounds of medicated bathing of dogs had been conducted during the sandfly season in 1993. The results show that there was no new infection from September 1993 to October 1994 in the three villages and a town, indicating that through two years' consecutive extensive medicated bathing of dogs in the endemic areas, canine visceral leishmaniasis transmission had been significantly interrupted. The authors consider that in the mountainous region of southern Gansu and northern Sichuan where it has been difficult to treat dogs, it might be more practical to use deltamethrin bathing of dogs for interrupting the transmission of canine visceral leishmaniasis.

[Study on LDL receptors of liver non-parenchymal cells in hypertriglyceridemic rats induced by high carbohydrate diet].

In the present study, hypertriglyceridemia was induced by feeding S. D. rats with high carbohydrate diet (carbohydrate accounting for 80% of total calorie) for 5 days. Rat liver parenchymal cells (PC) and non-parenchymal cells (NPC) were prepared by collagenase method. Human plasma LDL, VLDL and HDL were isolated by density gradient ultracentrifugation method. 125I-LDL was labelled by ICI method. 125I-LDL binding to rat liver PC and NPC were measured by Goldstein and Brown's method. The results showed that both rat liver PC and NPC had LDL receptors to bind, uptake and degradate 125I labelled human plasma LDL. The Bmax of 125I-LDL binding to NPC was 7-fold higher than that of PC (P < 0.01), but the Kd values remained unchanged. The Bmax of 125I-LDL binding to liver NPC from hypertriglyceridemic rats was significantly higher than that of NPC from the normal diet (carbohydrate accounting for 60% of total calorie) rats (435.4 + 100.0 vs 307.1 +/- 76.8 ng/mg cell protein, n = 6, P > 0.05), but there was no difference in Kd between the HTG rats and normal rats (18.1 +/- 4.1 vs 17.2 +/- 4.0 micrograms/ml n = 6, P > 0.05). These results suggest that the increase in fractional catabolic rate of LDL in patients with hypertriglyceridemia reported by Vega and Grundy might be induced by the increase in number of LDL receptor on liver non-parenchymal cells.

[Studies on the bionomics of Phlebotomus chinensis in mountain cave and its relation to control in northern Sichuan].

After a survey of the gonotrophic cycle of 5147 female Phlebotomus chinensis collected from the caves in the mountain area 1500 m above sea level in Nanping County, Sichuan Province where this sandfly was proved to be the typical predominant wild species living mostly in the wild cave at a very high density, a large cave was sprayed with alphamethrin at a dosage of 50 mg/m2 in July, 1993. As a result, the sandfly density dropped to zero spanning over a period of two peak seasons. The authors consider that the residual effect of alphamethrin as long enough to suppress sandfly density in the cave for at least two years, and stress that the spray is a favourable measure in all the year round in reducing the sandfly population density and preventing visceral leishmaniasis.

Human T cells express specific binding sites for C1q. Role in T cell activation and proliferation.

Although the receptor that binds to the collagen-like domain of human C1q (C1qR) is expressed on a wide variety of cell types, the presence or absence of this receptor on human T lymphocytes has been debatable. The current studies were undertaken to re-examine whether human T cells possess specific binding sites for C1q by using a combination of techniques, including radioligand binding studies, flow cytometric analysis, and epifluorescence imaging techniques. Radioligand binding studies indicate that both peripheral T cells and the cultured T cell line, MOLT4, bind 125I-labeled C1q in a specific and apparently saturable manner, reaching equilibrium within 30 min at 37 degrees C under conditions of subphysiologic (90 mM NaCl) ionic strength. Western blot analysis with anti-C1qR of membrane proteins derived from Raji and MOLT4 cells showed an apparent single band of approximately 60 kDa under nonreducing conditions. Furthermore, when peripheral blood T cells were stimulated with 12,-o-tetradecanoyl phorbol-13-ester acetate for 5 days at 37 degrees C and assessed by FACS for their ability to bind anti-C1qR, the mitogen-induced cells were found to bind 40 to 50% more than their unstimulated counterparts. In addition, both CD4+ and CD8+ T cells were found to bind anti-C1qR. When the cells were mitogen induced with either 12,-o-tetradecanoyl phorbol-13-ester acetate, Con A, or PWM for 48 h in the presence or absence of 50 micrograms/ml C1q then pulsed with 1 microCi [3H]thymidine for 16 h at 37 degrees C, proliferation was significantly inhibited (40 to 80%, n = 7) as assessed by reduced [3H]thymidine incorporation. Taken together, the data suggest that: 1) Human T cells express C1qR in which immunoblots reveal a 60-kDa single chain protein. 2) C1qR expression is up-regulated by mitogens that induce T cell proliferation. 3) The primary ligand, C1q, induces an antiproliferative signal, which suggests that the C1qR plays a role in T cell activation and proliferation. In addition, the data contribute to the characterization of C1qRs on cells in peripheral blood and indicate that all cells, with the exception of erythrocytes, bear functional C1q receptors.

Molecular analysis of the glycoprotein C-negative phenotype of attenuated Marek's disease virus.

Serial passage of oncogenic serotype 1 Marek's Disease virus (MDV) in cultured avian cells results in attenuation of viral oncogenicity and pathogenicity. Coincident with attenuation, expression of MDV glycoprotein C (gC) is significantly reduced. Regulation of MDV gC may have important implications for virus-host interactions and have important consequences in the process of viral attenuation. To investigate the mechanism by which MDV gC expression is reduced during attenuation of the very virulent serotype 1 MDV strain Md11, protein levels, gene structure, steady-state RNA levels, and transcription rates of MDV gC from oncogenic and attenuated isolates of Md11 were determined. Comparison of these data with similar studies on MDV proteins whose expression is not altered during attenuation indicates that reduced expression of MDV gC is directly related to reduction in transcription rate of the MDV gC gene in attenuated Md11. Reduced transcription rates and lack of any gross structural alterations in the attenuated MDV gC gene suggest that MDV regulatory protein(s) which interact with the MDV gC promoter are altered during attenuation of MDV strain Md11. This conclusion is supported by DNA sequence data which indicate that the promoters of oncogenic and attenuated Md11 gC genes are identical.

Behavioural effects of intraplantar injection of inflammatory mediators in the rat.

The behavioural response to intraplantar injection of inflammatory mediators was examined using the rating scale developed to assess formalin-induced pain. Serotonin, bradykinin, prostaglandin E2, substance P and histamine induced dose-dependent favouring of the injected paw. Serotonin, bradykinin and prostaglandin E2 also induced transient dose-dependent paw elevation (lifting) and licking. Noradrenaline produced only a weak favouring response. Serotonin produced a synergistic increase in lifting and licking when combined with any of the other mediators, while all other combinations of agents taken two at a time showed additivity. There was apparent antagonism between some combinations in the favouring response; this may reflect overestimation of the baseline. The data indicate that (i) the overt spontaneous behaviour of rats can be used to evaluate spontaneous pain, (ii) the favouring response and the lifting and licking responses are qualitatively different, the former being similar to hyperalgesia and the latter possibility representing overt pain, (iii) hyperalgesia and overt pain are related, but the generation of overt pain involves specific mechanisms in addition to those required to induce hyperalgesia, and (iv) serotonin may function to enhance the pain-producing effects of inflammatory mediators, even when they lack intrinsic activity. The data show that some inflammatory mediators produce transient overt pain when high doses are injected into normal tissue in rats. Combination of inflammatory mediators with low doses of serotonin produced a synergistic increase in the pain response. The data suggest that serotonin released from platelets in injured tissue plays a central role in the pain associated with injury, and that serotonin antagonists may have promise as peripherally acting analgesics or analgesic adjuncts by blocking a synergistic process involved in algogenesis.

Tritium isotope effect in high-performance liquid chromatography of eicosanoids.

A significant difference in retention time between unlabeled and the corresponding multi-tritium-labeled eicosanoid has been observed in the high performance liquid chromatography (HPLC) analysis of 11 eicosanoids. Variations in retention time range from 3-7%, depending on the separation conditions as well as the number and position of the tritium substitution. Multi-tritium-labeled eicosanoids were eluted earlier than the corresponding unlabeled eicosanoid in reversed phase HPLC, whereas no isotope effect was seen with 14C- and 3H2-eicosanoids. Considerations must be given to this tritium isotope effect whenever both multi-tritium-labeled and unlabeled eicosanoids are used for HPLC cochromatography or recovery studies.

Physical activity and cardiovascular risk factors in rural Shanghai, China.

BACKGROUND: Data on the association between physical activity and cardiovascular risk factors among populations with a relatively high level of physical activity such as the Chinese, are sparse. METHODS: In 1991, as part of the Sino-Shanghai Cardiovascular Disease Registry Project, a cross-sectional survey was performed in a random sample of 1206 residents, aged 35-64 years, living in rural Shanghai, China. Information on physical activity was obtained by questionnaire. The level of physical activity was categorized into low, moderate and high, based on the presence of a weekly frequency (< 1, 1-2, > or = 3) of periods of 20 minutes that cause shortness of breath, increase in pulse rate and perspiration. Body mass index (BMI), systolic (SBP) and diastolic blood pressure (DBP) level, serum lipids levels, smoking status and heart rate were assessed. RESULTS: Across groups with low, moderate and high levels of physical activity, significant inverse trends were found for hypertension, total cholesterol, BMI and heart rate in men, and for hypertension, SBP and DBP, BMI and heart rate in women. High density lipoprotein cholesterol and current smoking were not related to physical activity. When differences in BMI were allowed for, the association between physical activity and hypertension and serum cholesterol in men, and with hypertension, SBP and DBP in women, were attenuated. CONCLUSIONS: Our findings from a population-based survey in China suggest that a high level of physical activity is associated with favourable levels of some of the established cardiovascular risk factors in men and women.

Effect of halothane on large-conductance calcium-dependent potassium channels in cerebrovascular smooth muscle cells of the rat.

BACKGROUND: Patch-clamp methods were used to examine the effects of the volatile general anesthetic halothane on large-conductance calcium-dependent potassium channels (BK channels) in dispersed cerebrovascular smooth muscle cells of adult rats. METHODS: Inside-out membrane patches were used for recordings at 21-23 degrees C. Halothane was administered at aqueous concentrations of 0.5-2.8 mM in conjunction with free cytoplasmic calcium concentrations of 1 or 100 microM and at a membrane potential of -60 mV or +60 mV. RESULTS: At a free cytoplasmic calcium concentration of 1 microM, the clinically relevant dose of 0.5 mM (2 MAC) halothane reduced the open probability of large-conductance calcium-dependent potassium channels without altering the single-channel conductance. This effect was blocked by increasing the concentration of cytoplasmic free calcium to 100 microM, but was not intrinsically voltage dependent. CONCLUSION: The marked dilation of cerebral vessels seen during surgical anesthesia with halothane cannot be attributed to direct effects of the drug on large-conductance calcium-dependent potassium channels. The protective effect of calcium suggests that halothane exerts its effects at channel sites located within the cell membrane.

Identification of an immediate-early gene in the Marek's disease virus long internal repeat region which encodes a unique 14-kilodalton polypeptide.

Marek's disease virus (MDV) is an oncogenic avian herpesvirus whose genomic structure is similar to those of herpes simplex virus and varicella-zoster virus. Repeat regions of the MDV genome have been intensively investigated because of a potential relationship to MDV oncogenicity and abundant expression of immediate-early transcripts. In this study, a 1.6-kb immediate-early transcript was localized to the BamHI-I2 region by Northern (RNA) hybridization analysis. With cDNA cloning and sequencing, two cDNAs of 1.4 kb (C1) and 1.35 kb (C2) were identified. Both cDNAs are derived from spliced mRNAs spanning the BamHI-H and -I2 fragments. C1 and C2 use the same splice acceptors and 3' ends, but they differ at their 5' ends and utilize different splice donors. The upstream promoter-enhancer region of C1 cDNA has been defined as a bidirectional regulatory region shared by the MDV pp38 gene. Sequencing analysis shows two small open reading frames (ORFs) within each cDNA (ORF1a and ORF2 in C1, ORF1b and ORF2 in C2). Potential ORFs of the sequence have no significant homology with any known protein in the Swiss-Protein data base. DNA fragments encoding ORF1a and ORF1b were cloned into pGEX-3X vectors to produce glutathione S-transferase fusion proteins and induce antisera. In Western blot (immunoblot) analysis of MDV-infected-cell lysates, a 14-kDa polypeptide was identified by antisera against both ORF1a and ORF1b. This 14-kDa protein is expressed in cells which are lytically infected with MDV strains GA, Md11 passage 14 (oncogenic), and Md11 passage 83 (attenuated), as well as in the latently MDV-infected and transformed MSB-1 cell line.

Spinal cord regulation of sympathetic activity in intact and spinal rats.

Excitatory amino acid (EAA) and cholinergic neurotransmission in the spinal cord of urethan-anesthetized rats was investigated to assess mechanisms regulating sympathetic activity after spinal cord injury. Blockade of EAA transmission by intrathecal injection of kynurenic acid decreased arterial blood pressure by 24 +/- 4 mmHg, heart rate by 15 +/- 10 beats/min, and renal sympathetic nerve activity (RSNA) by 85 +/- 4% in intact rats. In rats with cervical spinal transections, this blockade decreased RSNA by 51 +/- 5% and had no effect on arterial pressure and heart rate. Muscarinic blockade by intrathecal atropine decreased RSNA by 12 +/- 3 and 32 +/- 6% in intact and spinal rats, respectively, and caused no cardiovascular responses in either group. Combined blockade of EAA and muscarinic receptors in spinal rats decreased RSNA by 77 +/- 1%. Intrathecal injections of the EAA agonist D,L-homocysteic acid in spinal rats caused initial increases (335 +/- 28%) in RSNA lasting approximately 3 min and later sustained increases (157 +/- 19%) lasting 36 +/- 8 min. Only the early excitation increased arterial pressure by 17 +/- 3 mmHg, and then pressure returned to baseline values. The EAA agonist kainic acid increased RSNA by 402 +/- 90% in spinal rats, an effect lasting 70 +/- 5 min, and increased arterial pressure by only 8 +/- 2 mmHg for 12 +/- 5 min. These findings suggest that tonic activity of spinal neurons with EAA and cholinergic receptors maintains tonic RSNA after spinal cord transection. However, this activity does not play a major role in maintaining arterial pressure, even if it is increased substantially by EAA receptor stimulation.

Genetic and environmental influences on blood pressure in elderly twins.

We used 289 pairs of Swedish twins reared apart or together to evaluate the importance of genetic and environmental influences on blood pressure. Unlike other twin and family studies, the adoption/twin design allows a distinction between estimates of the importance of shared rearing environments and genetic effects. Genetic factors were observed to play an important role for individual differences in blood pressure. Model-fitting analyses suggested upper limits of heritability for systolic and diastolic blood pressures in the entire sample of 0.44 and 0.34, respectively. More interestingly, substantial influences of shared family effects accounting for up to 27% of the variation were also revealed. Effects of correlated environment, which might reflect, for example, the intrauterine environment, existed to some extent later in life. The influence of genetic factors tended to decrease across age groups for systolic blood pressure (0.62 in individuals less than 65 years old; 0.12 in those 65 years and older) but not for diastolic blood pressure (0.22 for the middle-aged group; 0.26 for the older group). However, this declining trend for systolic blood pressure did not reach significance (chi 2 = 8.07, df = 4, P = .09).

Stroke incidence and mortality in rural and urban Shanghai from 1984 through 1991. Findings from a community-based registry.

BACKGROUND: Mortality from stroke has declined in many countries. This decline may result from a fall in incidence of stroke or a decrease in case fatality from stroke. The present study was conducted to investigate the temporal trends of incidence rate, mortality rate, and case fatality of stroke in an urban and rural Chinese population. METHODS: A community-based registry study was established in 1983 in defined rural and urban areas of Shanghai with a population aged 35 to 74 years of approximately 86,000 subjects, adhering to the methods and definitions of the World Health Organization MONICA protocol. All stroke events occurring in the population aged 35 to 74 years in these areas were registered by a special three-level case-registration system. RESULTS: From 1984 through 1991, 1391 stroke cases were identified. No significant change in the incidence rate of stroke was observed from 1984 through 1991 in men and women living in rural and urban areas. Age-standardized mortality rates (per 100,000 person years) of stroke declined significantly during this period, except for rural women. The annual changes and 95% confidence intervals of age-standardized mortality rates were: for rural men -4.6% (-5.4, -3.9), for rural women -0.6% (-1.6, 0.5), for urban men -2.5% (-3.5, -1.6), and for urban women -4.7% (-5.2, -4.2). A significant decrease in case fatality from stroke from 1984 through 1991 was found among men living in rural areas (from 62.4% to 46.0%) and among women living in urban areas (from 48.4% to 33.3%). Overall case fatality, however, showed a nonsignificant decline over time. CONCLUSIONS: We observed a decline in stroke mortality rate in a rural and urban Chinese population. No significant changes in stroke incidence were found in this study, whereas case fatality appeared to have decreased, in particular among men living in rural areas and women living in urban areas.

Synthetic polymers as materials for artificial vitreous body: review and recent advances.

In spite of a century of recorded attempts to replace the vitreous body of the eye with a foreign material, there is no permanent substitute currently available, and the success of some materials as temporary substitutes is still very limited. Among the large number of materials proposed or tested as vitreous substitutes, synthetic polymers have been episodically considered during the past four decades. This article will review these attempts, their outcomes, and recent progress in this field. There were only seven synthetic polymers and a few semisynthetic polymers (modified biopolymers) so far proposed or tested as vitreous substitutes. It appears that probably a synthetic hydrogel with very high water content would eventually be the material of choice for a permanent vitreous substitute, but the ideal material has not yet been found.
