[Mutation characteristic of STK].

OBJECTIVE: To identify the mutation characteristic of STK(11) gene in Chinese with Peutz-Jeghers syndrome(PJS) and establish the base of the gene diagnosis of PJS. METHODS: STK(11) germline mutation was analysed by DNA sequencing in 18 unrelation patients with PJS. RESULTS: Six novel mutations of STK (11) gene were detected in six unrelation patients. These mutations will lead to production of truncated protein. CONCLUSION: STK (11) gene mutation accounts for one third of the Chinese with PJS. The content of mutation includes single base substitution or deletion and one or two bases insertion. The mutations were widely found in different regions of the whole coding sequence, and 2/3 of those concentr ate in exon 1. Mutation frequency is 66.7% in the family suffering PJS in two or more generations, and 16.7% in the disseminated cases.

Differential regulation of interleukin-1 beta-induced cyclooxygenase-2 gene expression by nimesulide in human synovial fibroblasts.

Osteoarthritic (OA) human synovial fibroblasts (HSF) in culture were treated with the preferential COX-2 inhibitors nimesulide or NS-398, the non-specific COX-1/COX-2 inhibitor naproxen, or dexamethasone, in the presence or absence of IL-1 beta or LPS. Nimesulide or NS-398 inhibited IL-1 beta-induced PGE2 production at all concentrations tested, and in addition they suppressed IL-1 beta-induced COX-2 mRNA expression and protein synthesis. These suppressive effects were most evident at therapeutic levels of the drugs. Mechanistic studies revealed that the drug-induced inhibition of COX-2 expression and synthesis was not promoter-based, but may be associated with the blockade of IL-1 beta-dependent calcium flux and increased cellular calcium levels.

Modulation of TIMP-1 synthesis by antiinflammatory cytokines and prostaglandin E2 in interleukin 17 stimulated human monocytes/macrophages.

OBJECTIVE: To examine the regulation of tissue inhibitor of metalloproteinase 1 (TIMP-1) synthesis by interleukin 17 (IL-17) stimulated human monocytes/macrophages in primary culture in the presence of prostaglandin E2 (PGE2) and antiinflammatory cytokines, and to compare this with the regulation of matrix metalloproteinase (MMP-9) production. METHODS: IL-17 stimulated human monocytes isolated from the peripheral blood of healthy donors were cultured in the presence of PGE2, cyclic adenosine monophosphate (cAMP) mimetics (IBMX, cAMP, forskolin, cholera toxin), or antiinflammatory cytokines (IL-4, IL-10, IL-13), or with protein kinase inhibitors of diverse specificity. MMP-9 and TIMP-1 were measured using specific ELISA, while expression of specific messenger RNA was determined by Northern blotting. RESULTS: IL-17 stimulated an increased level of MMP-9 production relative to TIMP-1 production in monocytes/macrophages. Stimulation was accompanied by upregulation of specific MMP-9 mRNA expression relative to TIMP-1 mRNA. Exogenous PGE2, cAMP, and cAMP-mimetics completely inhibited both basal and IL-17 induced MMP-9 synthesis, while only IL-17 induced TIMP-1 synthesis was abrogated. The same effect was found for the antiinflammatory cytokines. Both basal and IL-17 induced production of TIMP-1 involved p42/44 and p38 kinases and nuclear factor kappaB signaling pathways. CONCLUSION: The excess of MMP-9 over TIMP-1 production, and decreased inhibition of MMP-9 activity in chronic rheumatoid diseases, may result in cartilage degradation and joint destruction.

[Diagnostic value of five serum markers for liver fibrosis].

OBJECTIVE: To research the diagnostic value of serum hyaluronic acid (HA), type III procollagen (PCIII), type IV collagen (CIV), laminin (LN), and transforming growth factor-beta(1) (TGF-beta(1)) for liver fibrosis in patients with chronic hepatitis. METHODS: Serum levels of HA, PCIII, CIV, LN and TGF-beta(1) in 116 patients with chronic hepatitis and cirrhosis were investigated and compared with hepatic histological findings of 87 patients. RESULTS: The correlation between serum HA and histologically assessed grade of inflammatory activity was weak (r=0.393, P<0.05). The correlation between serum HA, PCIII, LN, TGF-beta(1) and histologically assessed stage of liver fibrosis were all moderate (r=0.584, 0.454, 0.441 and 0.612, respectively, P<0.05), while that between serum CIV and histologically assessed stage of liver fibrosis was weak (r=0.319, P<0.05). As shown by the ROC curves in cases of chronic hepatitis, the ability to differentiate patients with cirrhosis from those without cirrhosis was greater for serum HA than that for serum PCIII, CIV, LN, and TGF-beta(1) (the areas under the curves=0.904 vs 0.784, 0.815, 0.805, 0.828, P<0.05). The ability of serum HA, LN and TGF-beta(1) to differentiate patients with extensive liver fibrosis from those with no or mild liver fibrosis exceeded that of serum PCIII and CIV (the areas under the curves=0.849, 0.819, 0.836 vs 0.702, 0.721, P<0.05). To discriminate the stage of liver fibrosis, serum HA and TGF-beta(1) were selected from the five markers by Bayes discriminate analysis. If S(1), S(2) and S(3) were not required to be discriminated, discrimination between the three stages showed significant difference (P<0.05). The predictive accurate percentage was 72.90%. CONCLUSIONS: The five markers all have the ability not only to judge liver cirrhosis, in which the ability of serum HA is the best, but also to differentiate chronic hepatitis with extensive liver fibrosis from that with no or mild liver fibrosis, in which the ability of serum HA, LN, TGF-beta(1) is stronger than the other two. It is useful for detection of serum HA and TGF-beta(1) at the same time to discriminate the histologically assessed stage of fibrosis. Serum HA and TGF-beta(1) can be helpful in discriminating patients of chronic hepatitis with "no liver fibrosis", "liver fibrosis but no cirrhosis" and "liver cirrhosis", but cannot discriminate them accurately as the histologically assessed stage of fibrosis. They cannot displace liver biopsy for the judgement of liver fibrosis.

Telomerase activity and homozygous deletions of the p16 gene in liver metastases of colorectal carcinoma.

OBJECTIVE: To study the interaction between telomerase activity and abnormalities of the p16 gene in liver metastases of colorectal carcinoma. METHODS: Telomerase activity was detected by a non-isotopic PCR-based telomeric repeat amplification protocol (TRAP) assay, and homozygous deletions of the p16 gene were detected by a semiquantitative multiplex polymerase chain reaction in tissue samples from 24 liver metastases of colorectal carcinoma and 5 primary colorectal carcinomas. RESULTS: Telomerase activity was observed in 19 (79.2%) of 24 liver metastases of colorectal carcinoma. Telomerase activity was also observed in all 5 primary colorectal carcinomas and in 3 of their liver metastatic samples. The incidence of telomerase activity in liver metastases of colorectal carcinoma was not significantly correlated to tumor diameter, number of tumors, cirrhosis, and HBsAg. Homozygous deletions of the p16 gene were found in 9 of 24 (37.5%) liver metastases of colorectal carcinoma. Homozygous deletions of the p16 gene were observed in 2 of the 5 primary colorectal carcinomas and in 1 of the matching liver metastatic cancers. There was a correlation between telomerase activity and homozygous deletions of the p16 gene. CONCLUSIONS: There is a correlation between telomerase activity and homozygous deletions of the p16 gene in liver metastases of colorectal carcinoma, suggesting its crucial role in liver metastases. However, telomerase activation and homozygous deletions of the p16 gene might not be the initiating event in liver metastases of colorectal carcinoma.

[Association of human leukocyte antigen-DQA1 with endometriosis of women in southern China].

OBJECTIVE: To study the genetic susceptibility in patients with endometriosis by genotyping their human leukocyte antigen-DQA1 (HLA-DQA1) alleles. METHODS: The allelic types of HLA-DQA1 were detected by polymerase chain reaction single specific primers (PCR-SSP) technique in 51 patients with endometriosis proven by laparoscopy or histological examination, and 44 control women who had laparoscopic sterilization and without endometriosis. RESULTS: The frequency of HLA-DQA * 0401 allele (12%) in patients with endometriosis group was markedly higher than that in the control group (0%) (P = 0.019). Contrarily, the frequency of HLA-DQA1 * 0301 allele (39%) in control group was higher than that in endometriosis group (14%) (P = 0.005, odds ratio = 0.253). CONCLUSION: Our result indicated that HLA-DQA1 * 0401 may be a susceptible gene in correlation with endometriosis, whereas HLA-DQA1 * 0301 may be a protective gene against endometriosis.

[Effects of Fas ligand expression on pancreatic islet allografts].

OBJECTIVE: To investigate the immune privilege of islet allgrafts induced by the Fas ligand (FasL) expressed by co-transplanted testicular Sertoli cells and the effects of FasL gene transfected into islet cells on pancreatic islet allografts. METHODS: The allogeneic islets and testicular cells from rats were co-transplanted into diabetic recipients. Pancreatic islet cells were firstly infected with the recombinant adenovirus AdV-FasL and then transplanted into the diabetic recipients. Allograft survival, islet function and apoptosis of infiltrative lymphocytes in allografts and gene-transfected islet allografts were observed. RESULTS: All the animals receiving islet allografts alone returned to a diabetic status in several days (mean survival time = 6.3 +/- 0.56 days). When the number of testicular cells co-transplanted with islets increased to 1 x 10(7), all the animals remained normoglycemic throughout the follow-up period (P < 0.05). Sertoli cells expressing FasL induced apoptosis of infiltrative lymphocytes in the allografts. In the group of FasL gene transfection, the rejection of allografts was accelerated and the allograft survival time shortened to 3.4 +/- 0.24 days (P < 0.05). Pancreatic islets infected with AdV-FasL demonstrated positive staining for FasL at the 24th hour and increased intensity at the 48th hour after transplantation. After the transplantation, apoptosis of FasL-transfected islet cells occurred. CONCLUSIONS: Co-transplantation of testicular Sertoli cells expressing FasL and islets can induce apoptosis of activated lymphocytes and allows long-term survival of allogeneic islets because of immune privilege. However, direct expression of FasL by islet allografts infected with AdV-FasL accelerates the islet rejection by islet cell apoptosis and granulocytic infiltration.

Will the polio niche remain vacant?

C-Cluster enteroviruses (C-CEVs), consisting of Coxsackie A viruses (C-CAV1, 11, 13, 15, 17, 18, 19, 20, 21, 22, 24, 24v) and polioviruses (PV1, 2, 3), have been grouped together in relation to their genomic sequences. On the basis of disease syndromes caused in humans, however, C-CAVs and PVs are vastly different: the former cause respiratory disease, just like the major receptor group rhinoviruses (magHRV), whereas PVs, on invasion of the CNS, can cause poliomyelitis. It is assumed that the difference in pathogenesis of C-CEVs is governed predominantly by cellular receptor specificity. C-CAVs use ICAM-1, just like magHRV, whereas PVs uniquely use CD155. Both ICAM-1 and CD155 are Ig-like molecules. Remarkably, based on a phylogenetic analysis of non-structural proteins, CAV 11, 13, 17 and 18 are interleaved with, rather than separated from, the three PV serotypes, e.g. PV1 is more closely related to CAV18 that to PV2. This observation suggests that PVs may have emerged from a pool of C-CAVs by evolving a unique receptor specificity. We have been studying virion structure, virion/receptor interactions, genetics, and the molecular biology of C-CEVs with the objective of identifying the molecular basis of phenotypic diversity of these viruses. Of particular interest is the prospect that C-CEVs can be genetically manipulated to switch their receptor affinity: from CD155 to ICAM-1 for PVs, or from ICAM-1 to CD155 for C-CAVs. We propose a hypothesis that in a world free of poliovirus and anti-poliovirus neutralizing antibodies C-CAVs would be given a greater chance to switch receptor specificity from ICAM-1 to CD155 and thus, to evolve gradually into a new polio-like virus.

[Ultrasonic removal of organic pollutants in tannery waste water].

The effect of ultrasonic treatment on flocculating settling of tannery effluent was studied, removal of COD in particular was did. The influences of irradiation method and duration of ultrasonic as well prescription of coagulants on the efficiency of minimizing organic chemicals in waste water were investigated. The results show that: 1. when waste water was treated only by ultrasonic for 60s, the removal rate of COD was 40.6%; 2. when total concentration of coagulants increased to 100 mg/L, the highest removal rate of COD was achieved; 3. when waste water was firstly treated with ultrasonic for 60s, then flocculated with coagulants, the removal rate of COD came to 73.2%, 10% higher than the control without ultrasonic. Thus the treatment of tannery waste water with flocculating settling technology can be effectively strengthened by ultrasonic.

A novel mis-sense mutation (G1381A) in the G6PD gene identified in a Chinese man.

OBJECTIVE: To detect new mutations among 29 glucose-6-phosphate dehydrogenase (G6PD) deficient individuals from Yunnan province. METHODS: The nitroblue tetrazolium (NBT) method was used to screen G6PD deficient individuals. Mutation was identified by single strand conformation polymorphism (SSCP), amplification created restriction site (ACRS), amplification refractory mutation system (ARMS) and DNA sequencing. RESULTS: Among 29 cases, 18 cases of G1388A, 1 case of C1004A, and 1 case of G1381A were identified. Nine cases remained to be defined. The G1381A mutation is a novel mis-sense mutation, with a substitution of threonine for alanine (A461T). The resultant G6PD had reduced enzymatic activity. In addition, G1381A caused a restriction site of Stu I to disappear, providing a rapid method for the detection of this mutation. CONCLUSION: A novel mis-sense mutation G1381A was found. This mutation results in a substitution of threonine for alanine, producing enzyme with reduced activity. The loss of the Stu I restriction site offers a rapid method for the detection of this mutation.

[Influence of transfection of sense- and antisense-cyclin A gene on human squamous cell carcinoma of tongue].

OBJECTIVE: To investigate the effect of human sense- and antisense-cyclin A gene on human squamous cell carcinoma of tongue cell line (Tca 8113). METHODS: Tca8113 cells were transfected with an expression vector containing a full length human sense-cyclin A or antisense-cyclin A gene by gene transfer with DNA-liposome. A series of methods, such as the changes of cell proliferation, DNA-flow cytometric analysis, silver binding nucleolar organizer regions (AgNORs) and expressions of cyclin A and Ki-67 were studied on transfected cell and untransfected cell. RESULTS: The cells transfected by antisense-cyclin A gene showed growth inhibition in vitro and decreased proliferation. The proliferation index(PI) was reduced (0.231 vs 0.421) as compared with untransfected cell group, and FCM analysis showed that Tca8113 cells transfected with antisense-cyclin A were arrested at G0/1 phase. The mechanism of such changes described might be associated with the lower expression of cyclin A mRNA in antisense-cyclin A gene transfected cells. However, opposing changes were observed in the sense-cyclin A gene transfected cells. CONCLUSIONS: Antisense cyclin A could inhibit target gene expression and partly reverse malignant phenotype of squamous cell carcinoma of tongue.

[Effects of soil water condition and microclimate on transpiration rate of rice].

The transpiration rate and water use efficiency (WUE) of rice were studied under high, moderate and low soil water conditions (treatments A, B and C) in the red soil area of southern China. Diurnal variation of transpiration showed that rice transpiration rate was influenced by several microclimatic factors in the field. Correlation analysis and stepping multi-regression analysis suggested that the difference of relative humidity between leaf and air, and the leaf temperature were the two main factors affecting the transpiration rate of rice. The results also showed that WUE was and yield were increased significantly in treatment B. While in treatment C, the WUE increased, but the yield of late rice decreased. The transpiration rate of early rice was decreased under moderate and low soil water treatments, but this did not happen to late rice.

Clinical features of diffuse axonal injury.

OBJECTIVE: To analyze the mechanism of diffuse axonal injury (DAI) and study the relationship between DAI and brain concussion, brain contusion, and primary brain stem injury. METHODS: The clinical data and iconographic characteristics of 56 patients with DAI were analyzed retrospectively. RESULTS: Traffic accidents were the main cause of DAI. Among t he 56 cases, 34 were injured for at least twice, and 71.43% of the patients were complicated with contusion. CONCLUSIONS: It is considered that DAI is a common pattern of primary brain injury, which is often underestimated. And DAI includes cerebral concussion and primary brain injury, and is often complicated by cerebral cortex contusion. Therefore, it is very simple and practical to divide primary brain injuries into local and diffuse injuries.

[Experimental study of the effect of Hpudiod cream on random flap survival in rats].

OBJECTIVE: To explore the effect of Hpudiod cream on blood circulation of flap. METHODS: 80 rats were casually divided into two groups: the experiment and the control group. A random flap was created on the back of the animal. Flap survival and necrosis were observed in the two groups. The flaps were examined histologically and the capillaries were counted. RESULTS: On the 5th and 7th day after operation, the distant ends of the flaps necrosed in both groups. On the 5th day there was no significant difference between the two groups in the number of the capillary in the middle of the flap (P > 0.05). On the 7th day, the capillary in the middle of the flap increased in the treatment group(P < 0.05). CONCLUSION: Hpudiod cream can improve the blood circulation of the random flap evidently.

[Studies on epidemic feature of enteric adenoviruses infection in feces of infants with diarrhea in Shenzhen].

OBJECTIVE: To estimate the enteric adenoviruses infection in feces of infants with diarrhea in Shenzhen. METHODS: All 195 fecal samples of infants were tested by PCR. Amplimers were analysed by Rsa I endonucleases and the type of enteric adenovirus identified. RESULTS: The positive rate of adenovirus DNA in fecal samples was 13.33% (26/195) while the prevalence rates of enteric adenovirus type 40, adenovirus type 41 and the other type of adenovirus were 2.56% (5/195), 8.21% (16/195) and 2.56% (5/195) respectively. Fever was the characteristic symptom of adenovirus infection. CONCLUSION: Enteric adenovirus type 40 and 41 were important pathogens which caused infant diarrhea in Shenzhen, with latter the majorty.

[The levels of angiotensin and aldosterone in gases of hyperthyroid Graves' disease with and without hypokalemic periodic paralysis].

OBJECTIVE: To investigate the relationships between the changes of plasma angiotensin I (AT I). Angiotensin II (AT II). serum aldosterone (Ald) levels and hypokalemic periodic paralysis (THPP) complicating hyperthyroid Graves' disease. METHODS: The levels of serum potassium, TT3, TT4, FT3, FT4, Ald and the plasma AT I, AT II and 24 hours urine potassium output were determined in 73 cases of hyperthroid Graves' disease, of which 43 were complicated by THPP and the other 30 were grouped as NTHPP cases. RESULTS: The mean level of serum potassium in THPP group was significantly lower than that in NTHPP group (P = 0.000), and the mean level of 24 hours urine potassium output in THPP group was significantly higher than that in NTHPP group(P = 0.000); The mean levels of plasma AT II and serum Ald in THPP group in upright position were significantly higher than those in NTHPP group (P = 0.008 and 0.013, respectively). The mean levels of 24 hours urine potassium output, plasma AT II and serum Ald in upright position were negatively correlated to serum potassium in both groups. The level of serum Ald in upright position was positively correlated to the level of 24 hours urine potassium out-put. The mean levels of plasma AT I and AT II in supine position were positively correlated to serum T3, and the level of plasma AT II in upright position was positively correlated to serum FT3. CONCLUSION: The level of 24 hours urine potassium output rises in THPP patients; the high levels of AT and Ald may contribute to the pathogenesis of the disease.

[Relationship between the expression of Bax and apoptosis induced by hyperthermia in BcaCD885 cells].

OBJECTIVE: This study aimed to investigate the role of Bax gene in apoptosis induced by hyperthermia in BcaCD885 cells. METHODS: After inducing apoptosis through hyperthermia, the expression of Bax protein and mRNA were examined with FCM antibody analysis and RT-PCR technique respectively. RESULTS: Up regulation of Bax protein was noted after hyperthermia. The correlation coefficient r between the apoptotic rate and the expression Bax protein was 0.9240. The expression of Bax mRNA was also raised after hyperthermia. CONCLUSION: Hyperthermia acting as an external signal may up-regulates expression of Bax gene and then induces apoptosis in BcaCD885 cells.

[Preliminary study of closing cleft of soft palate at an early age for complete cleft palate].

OBJECTIVE: The aim is to introduce the method closing the soft palate at an early age in order to produce better speech and lessen interruption to the development of maxilla. METHODS: Clinical data of 37 cases with treated cleft were preliminarily analyzed. Some dental casts were measured. RESULTS: 1. No complications of anaesthesia and surgery. Except 2 cases whose soft palates were split after surgery were observed as they were coughing. 2. A reduction in the width of the hard palate cleft was noticed. CONCLUSION: Closing the soft palate at an early age is possible and safe. It shows that the Langebeck method could be used to close the hard palate of patients at an early age.
