Effects of carotenoids on aflatoxin B1-induced mutagenesis in S. typhimurium TA 100 and TA 98.

The effects of beta-carotene, canthaxanthin, and extracts of tomato paste (containing lycopene) and orange juice (containing cryptoxanthin) on aflatoxin B1 (AFB1)-induced mutagenesis in S. typhimurium TA 100 and TA 98 were investigated. Inhibition of mutagenesis was studied during and following completion of AFB1 metabolism (i.e., after the addition of menadione), thereby permitting separate examination of the metabolic activation and phenotypic expression phases. Each experimental carotenoid, except lycopene, inhibited AFB1-induced mutagenesis in both tester strains. Cryptoxanthin was the most potent inhibitor, being at least an order of magnitude more potent than the other carotenoids. Inhibition by beta-carotene and canthaxanthin was more prominent during the activation phase, whereas cryptoxanthin was more effective during the subsequent phenotypic expression phase. These inhibitory effects were not dependent on conversion to retinol.

[Parasitological and pathological studies on rhesus monkeys infected with Chinese mainland strain of Schistosoma japonicum].

The present study was undertaken to obtain fundamental data from rhesus monkey in the course of infection with Chinese mainland strain of Schistosoma japonicum under laboratory condition. 15 normal monkeys were infected by cutaneous exposure to 200 cercariae for each animal. The following items were observed: the dynamic distribution of schistosomulum in the skin; the average of worm recovery rate; the size of mature worms of both sexes; the size of mature eggs from the feces; the mean prepatent period; the duration of egg passage in the feces and the histopathological changes of the skin, liver and gastro-intestinal tract at different time intervals after infection. Our results showed that rhesus monkey is a permissive host for Chinese mainland strain of S. japonicum and a spontaneous recovery happened due to gradual disappearance of the worms at later stage.

[Serum apolipoprotein AI, B levels in patients with coronary diseases].

The levels of total cholesterol, triglyceride, LDL-C, HDL-C, apolipoprotein A1 and apolipoprotein B in the serum were measured in a selected series of 100 CAD patients (77 men and 23 women) who underwent coronary angiography and 141 non-CAD controls. Mean values of those variables differed significantly between the CAD and non-CAD groups matched in age, body weight, hypertension and smoking. There are significant difference in apolipoproteins A1, B and the ratio of apolipoprotein B to A1 between angina and myocardial infarction groups. Using stratified and multivariate stepwise regression analysis, it was shown that the apo A1, apoB/apoA1 are more sensitive and specific than the ordinary indices (e.g. total cholesterol, triglycerides, LDL-C and HDL-C) in estimating the degree of coronary artery stenosis and the differentiation of CAD from other diseases.

[Effects of clonidine on experimental stomach ulcer in rats].

Clonidine 2 mg/kg ig inhibited the rat gastric ulcers induced by pyloric ligation, stress and indomethacin by 71%, 77% and 82%, respectively. Clonidine 2 mg/kg ig tended to accelerate the healing of gastric ulcer induced by acetic acid, and the healing rate was 61%. Clonidine decreased the secretion of gastric acid and pepsin, and increased the release of gastric barrier mucus. These actions may contribute to its protective effect against ulceration.

[Function of large plasmids of the plague pathogen].

Function of the largest plasmid of Yersinia pestis was studied. Assay of four strains of highly virulent Y. pestis isolated from different natural sources demonstrated that the characters of endotoxin production, sugar and alcohol fermentation, nitrate reduction as well as requirement in nutrients are mediated by no Y. pestis plasmids. On the contrary, FI and FII antigens are determined by the largest plasmid in some strains and are chromosome-mediated in others. It may well be that the genes for these antigens are comprised within the transposon-like DNA fragment.

[Effect of high carbohydrate and high fat diets on plasma glucose, insulin and lipids in normal and hypertriglyceridemic subjects].

The fasting plasma glucose, insulin and lipids response to high-carbohydrate-low-fat diets and low-carbohydrate-high-fat diets in normal subjects and patients with endogenous hypertriglyceridemia was observed. The normal, high carbohydrate and high fat diets consisted of 60%, 80% and 40% carbohydrate, 25%, 5% and 45% fat and 15% protein of total calorie respectively. The fasting plasma glucose levels increased (P less than 0.01) on day 1, insulin increased on day 3 (P less than 0.01 and P less than 0.05) and TG levels increased on days 3 and 5 after taking high carbohydrate diets both in the normal and hypertriglyceridemic groups. The fasting plasma TC and HDL-C decreased on high carbohydrate diets mainly in the hypertriglyceridemic group. The fasting plasma glucose slightly decreased on day 5, TG decreased on day 1 (P less than 0.05) and TC increased on day 1 and 3 (P less than 0.01) after taking high fat diets, but no differences were observed in insulin levels. Fasting plasma HDL-C levels in the normal group remained unchanged, but significantly decreased 3 days after taking high fat diets in the hypertriglyceridemic group (P less than 0.001). Our results indicate that high carbohydrate intake may be an important cause of hypertriglyceridemia in our country. This diet may cause a slight increase in the blood glucose level, which in turn stimulates an increase in plasma lipids and decrease in HDL-C, which is associated with an increase in the incident of coronary heart disease.(ABSTRACT TRUNCATED AT 250 WORDS)

[Detection of sorbitol content in crystalline lens of normal rats and rats with diabetic cataract by 1H-NMR].

The pulse Fourier NMR was employed to measure the artificial diabetic cataract lens at various stages of its formation, and the lenses of the normal rats. Data obtained by using this method show that all the peaks that of water concentrate in the range of delta less than 4 ppm. The peak value at delta = 3.20 ppm is on a marked increase during the formation of cataract which is caused by the phosphate metabolites, such as GPC, ATP, ... etc, in cataract lens. With the development of the disease, the peak width at delta = 3.73 ppm becomes greater and greater, which shows that the activity of sorbitol dehydrogenase has decreased. This leads to a high concentration of the sorbitol in the cataract lens. Consequently, the osmosis pressure in the cataract lens is increased, and excessive water might dip into the crystalline lens to keep the balance of the osmosis pressure. And this might result in the hydration of the fiber cell of the crystalline lens, which might cause a swelling or blisters. These results are in favour of the prolongation of the relaxation time of cataractous lens reported in our other papers, and also support those gained by biochemical studies issued in the medical literature.

Ticlopidine in China: comparative study on the effect of two dose levels on bleeding time and platelet function in healthy volunteers.

The compared inhibitory effects of two dose levels of ticlopidine (250 and 500 mg/day) on bleeding time and platelet function in Chinese volunteers have been studied in a placebo-controlled study. Ticlopidine exerted several significant effects: namely prolongation of bleeding time, inhibition of ADP-induced platelet aggregation, reduction of the thromboxane B2 synthesis induced by collagen as well as by ADP. The daily dose of 250 mg did not show significantly different effects as compared to the western, usually recommended daily dose of 500 mg. At the daily dose of 500 mg, adverse reactions occurred more frequently than at the daily dose of 250 mg, but no serious side effects necessitating discontinuation of treatment, or abnormalities in hematological and liver function parameters were observed at these two dose levels. In Chinese people, ticlopidine therapy should be administered at a daily dose of 250 mg.

The effect of transposition of pedicled omentum to spinal cord ischemia segments. An animal experiment.

This animal experiment was in succession to the research on paraplegia caused by spinal cord local ischemia in an animal model. Immediately after the operative production of local spinal cord ischemia, pedicled omentum spinal transposition onto the ischemic segments of spinal cord was performed. The animals recovered from paraplegia around two months afterwards. Spinal cord specimens showed adhesion of omentum to the spinal cord without compression. Omental vessels grew into the cord. Gross sections of specimen stained with Nissl method revealed that normal structure of nerve cells were preserved but number of nerve cell decreased.

[Hypertension and angiographically defined coronary heart disease].

The associations of CHD, as defined by coronary arteriography, with hypertension history and causal blood pressure in 103 CHD cases and 103 non-CHD controls matching on sex, race, age(within 3 years)was studied. The odds ratio (i.e.OR) of CHD for patients with hypertension history is 4.64. For patients with higher causal blood pressure, it is 3.53 with both 99% confidence intervals not include 1. There are significant dose-response relationships between level of systolic hypertension and duration of hypertension with regards to odds ratio of CHD or the degree of coronary atherosclerosis. The relative risk of CHD may reduce 70-80% for hypertension patients by taking drugs to lower blood pressure. There are significant correlation between hypertension and myocardial infarction in clinical types of CHD. The CHD average incubation period caused by hypertension is 10 years. It is concluded that the relationship between hypertension and CHD is seemingly causality with part of patients suffering from other consequences.

[Leprous peripheral neuropathy: clinical observation of 11 cases and ultrastructural study of nerves in 2 cases].

Two cases of tubercular form of leprosy associated with peripheral neuropathy and having biopsy of sural nerve performed, were reported. Pathologically, there was diffuse and massive infiltration of lymphocytes in the endoneurium and perineurium. The pathogenic organisms were found in the Schwann cells and endoneurium. The clinical features of leprous neuropathy were studied in 11 cases and the clinical diagnosis, pathological features, pathogenesis and treatment of this disease were discussed.

[Women's passive smoking and coronary heart disease].

Thirty-four women CHD cases (22 cases diagnosed by coronary arteriography and 12 myocardial infarction) and 68 of non-CHD controls (34 hospital based and 34 population based), matched on age (within five years), race, residence, occupation (and case is to control as 1:2), were interviewed regarding the smoking habits of themselves and their husbands. The odds ratio (i.e. OR) of non-smoking women CHD associated with having a smoking husband are 3.00-3.52, OR 95% CI do not include 1. Significant dose-response relationships between OR of women's CHD and their husband's cigarette consumption, duration of passive smoking and cumulative quantity of passive smoking were found in the study. The logistic regression model analysis with other CHD risk factors showed that the relationship with CHD and passive smoking still existed. It was found that the metabolism of HDL-cholesterol and apolipoproteins with passive smokers was abnormal.
