P-element insertion alleles of essential genes on the third chromosome of Drosophila melanogaster: mutations affecting embryonic PNS development.

To identify novel genes and to isolate tagged mutations in known genes that are required for the development of the peripheral nervous system (PNS), we have screened a novel collection of 2460 strains carrying lethal or semilethal P element insertions on the third chromosome. Monoclonal antibody 22C10 was used as a marker to visualize the embryonic PNS. We identified 109 mutant strains that exhibited reproducible phenotypes in the PNS. Cytological and genetic analyses of these strains indicated that 87 mutations affect previously identified genes: tramtrack (n = 18 alleles), string (n = 15), cyclin A (n = 13), single-minded (n = 13), Delta (n = 9), neuralized (n = 4), pointed (n = 4), extra macrochaetae (n = 4), prospero (n = 3), tartan (n = 2), and pebble (n = 2). In addition, 13 mutations affect genes that we identified recently in a chemical mutagenesis screen designed to isolate similar mutants: hearty (n = 3), dorsotonals (n = 2), pavarotti (n = 2), sanpodo (n = 2), dalmatian (n = 1), missensed (n = 1), senseless (n = 1), and sticky ch1 (n = 1). The remaining nine mutations define seven novel complementation groups. The data presented here demonstrate that this collection of P elements will be useful for the identification and cloning of novel genes on the third chromosome, since >70% of mutations identified in the screen are caused by the insertion of a P element. A comparison between this screen and a chemical mutagenesis screen undertaken earlier highlights the complementarity of the two types of genetic screens.

Cloning, characterization, and mapping of a murine promiscuous chemokine receptor gene: homolog of the human Duffy gene.

We report here the isolation and genomic organization of the orthologous mouse Duffy gene, named Dfy. It is a single copy gene located in chromosome 1 in a region homologous to the human Duffy gene (FY). Sequence analyses indicate that Dfy consists of two exons: exon 1 of 55 nucleotides, which encodes 7 amino acid residues; and exon 2 of 1038 nucleotides, which encodes 327 residues. The single intron consists of 462 nucleotides. The 5'-end promoter region contains motifs involved in vertebrate development in addition to potential binding sites of factors for globin transcription. The open reading frame (ORF) shows 60% homology with the human Duffy protein. However, mouse erythrocytes are serologically Duffy-negative and mouse erythrocyte membrane proteins do not cross-react with two Duffy-specific rabbit polyclonal antibodies. The deduced protein predicts a M(r) of 36,692 and carries three potential N-glycosylation sites to asparagine residues. Hydropathy analysis predicts an exocellular amino-terminal domain of 57 residues, seven transmembrane alpha-helices, and an endocellular carboxy-terminal domain of 29 residues. In bone marrow and spleen, Dfy expresses a major 1.4-kb and a minor 1.8-kb mRNA. Contrary to humans, Dfy is expressed in liver, synthesizing a 1.4-kb mRNA, and is repressed in kidney. Dfy is highly expressed in mouse brain and produces a major 8.5-kb and a minor 10.2-kb mRNA. The human erythroleukemia K562 cells, transfected with cDNA encoding the mouse Duffy-like protein and mouse erythrocytes, have the same chemokine binding profiles indicating that they contain the same protein.

Passive smoking and coronary heart disease: a brief review.

1. Whether passive smoking or environmental tobacco smoke (ETS) can cause coronary heart disease (CHD) is controversial. We have undertaken a comprehensive review of the epidemiological studies regarding the relationship between ETS and CHD. 2. We searched for all original papers and reviews and calculated pooled odds ratio using the Mantel-Haenszel method. 3. Ten prospective studies, nine case-control studies and one cross-sectional study were found. Almost all studies showed positive associations between ETS and CHD and a few were statistically significant, with dose-response relationships being evident. Six review papers calculated pooled estimates of the relative risks, which ranged from 1.23 to 1.51. Other studies also showed that ETS could have other cardiovascular effects. 4. The association observed is likely to be real. The criteria for causation of time sequence, consistency, coherence and biological plausibility are satisfied. We conclude that ETS may cause CHD, particularly in women who have never smoked.

Production and characterization of monoclonal antibodies specific for the lipopolysaccharide of Escherichia coli O157.

Identification of the O157 antigen is an essential part of the detection of Escherichia coli O157:H7, which is recognized as a major etiologic agent of hemorrhagic colitis. However, polyclonal antibodies produced against E. coli O157:H7 lipopolysaccharide (LPS) may react with several other bacteria including Brucella abortus, Brucella melitensis, Yersinia enterocolitica O9, Escherichia hermannii, and Stenotrophomonas maltophilia. We produced eight monoclonal antibodies (MAbs) specific for the LPS of E. coli O157. Western blots (immunoblots) of both the phenol phase (smooth) and the aqueous phase (rough) of hot phenol-water-purified LPS indicated that three of the MAbs were specific for the O antigen and five were reactive with the LPS core. The eight MAbs could be further differentiated by their reactivities to Salmonella O30 LPS (group N), which is reported to be identical to the E. coli O157 antigen. All eight MAbs reacted strongly to all of the 64 strains of E. coli O157 tested, which included 47 isolates of O157:H7 and 17 other O157 strains. None of the eight MAbs cross-reacted with any of the 38 other E. coli serotypes tested, which consisted of 29 different O-antigen serotypes, or with 38 strains (22 genera) of non-E. coli gram-negative enteric bacteria.

Increased expression of nitric oxide synthase in the myometrium of the pregnant rat uterus.

Nitric oxide (NO) relaxes uterine smooth muscle and is produced by the pregnant uterus. Our previous studies revealed an increase in rat uterine NO synthase (NOS) activity in pregnancy and a decline at term. In the present study, we have examined the distribution of NOS isoform expression to determine whether their regulation is consistent with a role in the inhibition of uterine contractions before term. At day 17-18 of pregnancy, NOS immunohistochemistry revealed expression of two isoforms: endothelial constitutive form of NOS (ecNOS) in vascular endothelium and inducible form of NOS (iNOS) in myometrial and vascular smooth muscle and in decidual epithelium. Immunoblotting revealed that expression of iNOS declined nearly fivefold, whereas ecNOS declined twofold in laboring rats at term. We conclude that iNOS is expressed in myometrium of pregnant rat uterus but not the virgin rat and that iNOS expression declines at term when labor is present. The pattern of changes in myometrial iNOS expression with advancing gestation suggests that NO could act in an autocrine and/or paracrine manner to inhibit uterine contractions before term.

[An analysis of the cause of misdiagnosis with ocular opportunistic infection].

OBJECTIVE: To evaluate the causes of misdiagnosis with ocular opportunistic infection. METHOD: The causes of misdiagnosis on 4 cases (6 eyes) with ocular opportunistic infection were analyzed. RESULTS: Acanthoamoeba keratitis was misdiagnosed as herpes simplex keratitis and fungal keratitis in 1 case (1 eye). Propionibacterium acne endophthalmitis was misdiagnosed as exogenous fungal keratitis in 1 case (1 eye). Endogenous fungal endophthalmitis was misdiagnosed as metastatic choroid cancer from lung in 1 case (2 eyes). Cytomegalovirus retinitis was misdiagnosed as endogenous fungal endophthalmitis in 1 case (2 eyes). CONCLUSION: If common therapies are of no effect with some ocular infections and the causative organisms responsible for the infection could not be found out after repeated tests, the rare ocular opportunistic infection should be considered. Special examinations (such as acanthoamoeba culture, anaerobic culture, fungus blood culture, cytomegalovirus serological assays, etc.) should be made for a definite diagnosis.

[Diagnosis and treatment of blowout fracture of the orbit].

OBJECTIVE: To analyze the diagnosis and treatment of blowout fracture of the orbit. METHODS: 78 cases with blowout fracture of the orbit seen in 20 years were retrospectively analyzed. In different years, different methods of examinations were used. There were 16 cases who had undertaken X-ray examinations, 31 cases, ultrasonographic examinations and 72 cases, CT. RESULTS: The positive rate of the CT examinations with blowout fracture of the orbit was 100%, CT having the tendency to eventually replace the X-ray. Ultrasonography shows only changes of intraorbital tissue, providing an auxiliary method for diagnosis. CONCLUSION: With the progress of modern photographic methods, blowout fracture of the orbit is known more deeply and the advance helps in the improvement of the treatment of the disease.

[Chemical studies on peptidepolysaccharides of Ganoderma lucidum (W. Curt. Fr.) Karst].

Seven homogeneous peptidepolysaccharides have been obtained from the fruit-body of Ganoderma lucidum. The main constituents are TGLP-2, TGLP-3, TGLP-6 and TGLP-7, whose molecular weights are 20.9 x 10(4), 4.5 x 10(4), 3.2 x 10(4) and 10 x 10(4) respectively. TGLP-2 is a heteroglycan peptide, TGLP-3 and TGLP-6 are glycan peptides and TGLP-7 is a galactan peptide.

[The clinical analysis of eosinophil infiltration in human squamous cell carcinoma of buccal mucosa].

Tumor-associated tissue eosinophils (TATE) is a host's defensive reaction to tumor. In this study, authors examined the eosinophil of 40 squamous cell carcinomas of buccal mucosa. Infiltration of the primary tumor by eosinophil was found to have marked prognostic significance. High-grade TATE was associated with the absence of head and neck lymphonode's metastasis (P < 0.05). There was a statistical significance (P < 0.05) that high-grade TATE was a favorable prognostic indicator for squamous cell carcinoma of the buccal mucosa. Authors concluded that quantitative assessment of eosinophils was one of the most important aspects of the microscopic evaluation of this human carcinoma.

[Study on effects of combined hearing protector on communication quality and noise protection].

To study the communication quality and the protection effects of single protector and the combined ones, sound attenuation experiments and the communication experiments were conducted in 14 male subjects. The results showed that the sound attenuation of combined protectors were 13 dB or 6dB more than that of single earplug or earmuff respectively, and combined protectors could avoid sound attenuation caused by the harmonics on some frequency bands in single one. After combined usage, the single protector reduced the distortion which came from the overload of hearing organs and maintained the communication quality. The combined protector is an effective hearing protection device in high noise environment in aerospace field.

[Pharmacological study of the rhizome powder of Pinellia pedatisecta processed by different procedures].

The potentiation of hypnotic activity of a barbituate and acute tocicity by the rhizome powder of Pinellia pedatisecta Schott processed by pigbile with two different traditional medicinal procedures (fermentation mixing and heating alternatively) were studied in mice. The results showed that aquatic extracts of two processed drugs could potentiate the action of hypnotics; and ethanol extracts of the drug processed with mixing and heating alterntively could significantly enhance this action, compared with that of the drug pricessed with fermentation; while no acute toxicity was measured by the two processed drugs.

[An analysis of the infection state of hepatitis C virus in liver diseases].

UNLABELLED: The local state of hepatitis C virus infection was probed. METHOD: The markers of hepatitis viruses were tested by nested RT-PCR and ELISA. RESULTS: In the patients with transfusion history, the incidence of acute hepatitis C (60.53%) was higher than that of acute hepatitis B (10.53%, P<0.01). In the patients with acute hepatitis without transfusion, however, the percentage of type A (41.67%) and type B (30.56%) hepatitis were higher than that of type C hepatitis (4.17%, P<0.01). The incidences of acute and chronic posttransfusion hepatitis C and asymptomatic HCV-infected cases, respectively 60.53%, 20.34% and 29.17%, were higher than that of the cases without transfusion, which were 4.2%, 8.74% and 3.33% respectively (P<0.01-0.05). No matter whether the patients with chronic hepatitis, severe hepatitis, cirrhosis, hepatocellular carcinoma or asymptomatic had transfusion history or not the incidences of HBV infection in them were higher than that of HCV infection (P<0.01). CONCLUSION: Most of the source of HCV infection were from transfusion, but infective rate of HCV by way of non-transfusion was lower. Severe hepatitis, chronic hepatitis, cirrhosis and hepatocellular carcinoma in local area was mostly induced by HBV.

[Isolation of coltivirus from mosquitoes in Beijing and it's biological characteristics].

Two strains (Beijing 95-70 and 95-75) of coltivirus were isolated from mosquitoes collected in Beijing during summer-autumn in 1994. The viruses caused cytopathogenic effect on C6/36 cells but not on BHK-21 and Vero cells, and were not lethal for new born mice and three week old mice. The isolates were resistant to 5'-IdU and ether, but sensitive to acid pH 3.0. There were 12 segments RNA shown by polyacrylamide gel electrophoresis (PAGE) and PAGE profile of the two isolates were similar (6-6). The PAGE profile was also similar to that of the TRT2 strain isolated in 1991, but there were at least 8 segments slightly different between the new isolates and TRT2 in band migration distance. Tissue culture cross-neutralization test showed that the new isolates were antigenically related to TRT2 strain identified as coltivirus. Growth curve of the two isolates on C6/36 cells showed that virus titer began to rise at the first day after infection and continued to increase up to 7.0 Log TCID50 per 0.1 ml and maintained a high level until the 21st day after infection. It seems that the virus may cause persistent infection on C6/36 cell.

[Serological survey of human T lymphotropic virus type I (HTLV-1) IgG antibody in south region of Xinjiang].

Human T lymphotropic virus type-1 (HTLV-1) is endemic in southwestern Japan, Seychelles Islands, Caribbean basin, Brazil, and Sub Saharan Africa. Recently, the prevalence of HTLV-1 of domestic source has been reported from Beijing, Fujian and so forth. The object of our study is to know whether the prevalence of HTLV-1 is present in Xinjiang. We collected 2642 serm samples of various ages and different nationalities (Uigur, Han and Khalkhas) from south region of Xinjiang and tested for determination of HTLV-1 IgG antibody by IFA. The results showed that the total positive rate of HTLV-1 IgG was 0.34% (9/2642), of which Uigur nationality was 0.74% (8/1082), Khalkhas nationality was 0.21 (1/471), Han nationality was zero (0/1089). This data indicated that there are HTLV-1 infection among the population of Xinjiang and especially in Uigur and Khalkhas. Why did the minority nationalities have relatively high frequency of HTLV-1 infection? This needs to be studied further.

[Generation of human monoclonal antibodies against hemorrhagic fever with renal sydrome (HFRS) virus].

Peripheral blood lymphocytes (PBL) were obtained from healthy donors and treated with L-leucy-leucin methy (LeuLeu-OMe). The LeuLeu-OMe treated PBL were in vitro immunized with HFRS virus proteins (55kD, 67kD) for 6 days, and then fused with K6H6/B5 cells in medium containing 45% (PEG) and 7.5% DMSO. The fused cells were seeded in 96-well plates at cell density 0.5-1 x 10(5) cells/well in HAT supplemented RPMI-1640 medium. After 14-20 days, 3 positive wells were deteced by ELISA and two (1B7, 2D5) of the 3 hybridoma were stable after 4 time cloning. The hybridoma 2D5 appears to be with high ability of production of the antibodies at a concentration of 20-30 microg/ml/10(6) cells/24h. It was proved by IFAT that 2D5 McAb was specific for HFRSV.

[Biological characteristics of residual leukemic cells and their drug-resistant regrowth].

OBJECTIVE: To elucidate the mechanisms of residual leukemia and its drug-resistant regrowth. METHODS: Bone marrow cells from transplantable mouse leukemia models L615 and L7811 were cultivated in long-term culture system respectively. Daunorubicin (DNR) was added to the systems to mimick in vivo chemotherapy. RESULTS: Residual leukemic cells on stromal layers escaped DNR killing. They derived from the adherent leukemic cell population, which was heterogeneous with many undifferentiated and dormant cells. They were drug-resistant and expressed bcl-2 gene. CONCLUSION: Bone marrow stromal layer was the vital microenvironment for the residual leukemic cells to survive and regrow, and might increase the drug-resistance by up-regulating bcl-2 gene expression.
