[Methodology for designing pathological models of acute renal failure].

This paper reports the methodological study on the following five pathological models of acute renal failure (ARF): 1. glycerine-induced ARF in rats; 2. noradrenaline-induced ARF in dogs; 3. ARF model caused by shutting left renal artery in dogs; 4. hydrargyrism-induced ARF in rabbits; and 5. gentamycin-induced ARF in rats.

Effects of proglumide, a gastrin receptor antagonist, on human large intestine carcinoma SW480 cell line.

OBJECTIVE: To explore the effects of proglumide, a gastrin receptor antagonist, on the amount of viable cells, synthesis of DNA and protein, and cell proliferation cycle in human large intestine carcinoma SW480 cell line in order to provide experimental basis for treatment of large intestine carcinoma using proglumide. METHODS: Large intestine carcinoma SW480 cells at logarithmic growth stage were cultivated with different concentrations of proglumide for different periods of time, then the amount of viable cells was determined by MTT colorimetric analysis. The SW480 cells were cultivated with proglumide, pentagastrin, proglumide + pentagastrin for the same period of time, then the contents of DNA and protein and the cell proliferation cycle were determined by flow-cytometry. RESULTS: The amount of viable cells, synthesis of DNA and protein, distribution of cell cycle, and proliferation index (PI) in the proglumide group did not differ significantly from those in the pentagastrin group (P > 0.05). The amount of viable cells in the pentagastrin group was significantly higher than that in the pentagastrin group (P < 0.01). In the proglumide + pentagastrin group the amount of viable cells, synthesis of DNA and protein, amount of S and G2M phase cells, and PI were all significantly lower than those in the pentagastrin group (all P < 0.01), and the amount of G0/G1 phase cells was significantly higher than that in the pentagastrin group (P < 0.01), but none of the above differed from those in the control group (all P > 0.05). CONCLUSIONS: Proglumide has no obvious effect on the growth of human large intestine carcinoma SW480 cell line, but can inhibit the growth-promoting effect of pentagastrin on large intestine carcinoma cells. The mechanism may be that proglumide inhibits the promoting effect of pentagastrin on the synthesis of DNA and protein of carcinoma cells, and then inhibits carcinoma cell growth from G0/G1 phase to S and G2M phase.

[The effects of transcutaneous electrical stimulation during sleep on obstructive sleep apnea].

OBJECTIVE: To study the effects of transcutaneous electrical stimulation during sleep on obstructive sleep apnea syndrom (OSAS). METHOD: Sixteen patients with OSAS were studied by polysomnography during all-night sleep with and without submental transcutaneous electrical stimulation (TES) of the genioglossus. RESULT: Fourteen of the 16 patients (87%) who accepted the TES were treated successfully (reduction of Al > 50%). The average apnea index showed a decrease of 29 events per hour when the TES were performed (P < 0.001). The average apnea time decreased from 22 to 7 sec (P < 0.001). The apnea time/TST decreased from 27% +/- 11% to 7% +/- 3%. The lowest SaO2 increased from 71% to 87% (P < 0.001). TES did not cause arousal. The sleep stages of SWS, and REM sleep effect (SE) increased significantly. CONCLUSION: TES is a conservative but effective treatment in patients with obstructive sleep apnea syndrome, although it failed to improve central sleep apnea.

[Experimental study on inhibitory effect of ganlu xiaodu Dan on coxackie virus in vitro].

OBJECTIVE: To observe the Coxackie virus inhibitory effect of Ganlu Xiaodu Dan (GLXDD), including complete recipe, incomplete recipe and supplemented recipe of it. METHODS: Using tissue culture technique, the maximal non-toxic concentration of drug on cultured cells, the influence of drug on proliferation of virus in cultured cells and the virus inhibitory effect in different time of action were observed. RESULTS: The maximal non-toxic concentration of GLXDD was 1:40 (25 g/L), no matter whether the recipe was complete, incomplete or supplementary. CLXDD in that concentration could inhibit significantly the proliferation of Coxackie virus B2, B3 and B4 strains, the inhibition index being over 2 and the output of these strains treated with complete recipe of GLXDD was lower significantly than that of the control group (P < 0.05). CONCLUSION: GLXDD, including its complete recipe, incomplete recipe and supplementary recipe, could inhibit the replication of Coxackie virus in cultured cells.

[Separation and determination of eight plant hormones by reversed-phase high performance liquid chromatography].

In this paper, reversed-phase high performance liquid chromatographic technique was used for the separation and determination of eight plant hormones. Methanol-water-acetic acid system was chosen as the mobile phase. The effects of different separation conditions, such as the methanol and acetic acid concentrations in mobile phase, on the retention behaviours of eight plant hormones in this system were studied. The general trends in retention behaviours could be correlated to the methanol concentration in mobile phase. The experimental results showed that the optimum separation was achieved with following gradient elution condition: 0-3 minutes, 70% (water percentage in mobile phase), 3-13 minutes, 70%-20%, 13-48 minutes, 20%. Benzene was added to be as the internal standard. Under this experimental condition, the eight plant hormones could be separated completely and detected quantitatively at 260 nm within 16 minutes. The calibration curves for the eight compounds gave linearity over a wide range. The correlation coefficients of each components were r(ZT) = 0.9971, r(GAs) = 0.9999, r(K) = 0.9997, r(BA) = 0.9995, r(IAA) = 0.9998, r(IPA) = 0.9982, r(IBA) = 0.9995 and r(NAA) = 0.9995. The method is rapid, simple and efficient. It is a suitable method for the accurate determination of gibberellic acid (GA) and alpha-naphthaleneacetic acid (alpha-NAA) in products for agricultural use.

[Observation on sleep improvement under noise environment by wearing earplugs].

Sleep experiments were done in 6 subjects under 3 conditions. (1) Quiet, background noise level lower than 45dB (A); (2) Noisy with earplugs, ambient noise level 90dB (A); (3) Noisy, ambient noise level 90dB (A), and subjects wearing earplugs. EEG, REM and EMG during sleep were recorded. Sleep latency, times and duration of waking up, sleep rate of each stage and total sleep rate were analyzed. It shows that sleep under noise condition can be improved by wearing earplugs.

[The responses of the microvasculature of the periodontal ligament to horizontal force].

OBJECTIVE: The purpose of this study is to examine the responses of the microvasculature in periodontal ligament(PDL) to horizontal force. METHODS: Using ink injections and celloidin embed, serial sections were cut, then observed by light micrascope, quantitative analysis was conducted by the picture-analysis instrument. Vascular corrosion casts were prepared and examined by scanning electron microscopy. The forth premolars of dogs were pulled by 200 g force in a mesial direction for two hours, and the pressure effects in different positions were demonstrated. RESULTS: The section area of the microvasculature per unit PDL area is obvious larger on the tension sides than on the pressure sides. There is not an obvious distinction on the pressure sides between the experiment groups and control groups. However, there are clear distinctions in the one-third of cervical, and apical segment on the tension sides between the experiment groups and control groups. CONCLUSION: The microvascular changes in PDL had importance to the cushioning effect of PDL when a teeth received a kind of force in a short-time.

[A long-term evaluation for opticfull-thickness lamellar keratoplasty].

OBJECTIVE: To investigate the long-term optic effects of optic full thickness lamellar keratoplasty (OFLK) and penetrating keratoplasty (PKP) and compare their effects. METHODS: 44 cases (48 eyes) with stromal disorder with normal endothelial function were treated by OFLK (25 eyes) or PKP (23 eyes). The optic effect of OFLK group was compared with that of the PKP group, and the endothelial density in partial cases was observed. RESULTS: The rate of transparent graft of OFLK group was 84.0%, and that of PKP group was 69.6% at postoperative 2 - 3 years, the rate of escaping blindness being 80% and 78.3% respectively. The rate of obtaining more than 0.3 vision in OFLK group was 40.0%, and in PKP group was 21.7%. Postoperatively, the endothelial cell density tended to decrease with time in PKP group, but was relatively stable in OFLK group. CONCLUSION: The results show that the optic effect of OFLK group is similar to that of PKP group, and the safety, less complications of OFLK group are the same as that of lamellar keratoplasty. It is considered that OFLK might have a bright future and the operative technique should be further studied.

[Peripheral corneal edema after cataract extraction].

OBJECTIVE: To report a rare type of corneal edema, i.e. peripheral corneal edema after cataract extraction---Brown-Mclean syndrome. METHODS: Clinical charts of 8 affected eyes in 5 patients were reviewed. Ultrasound pachymetry was used to determine central corneal thickness. Cell densities were measured by specular microscopy. The clinical characteristics and pathogenesis were discussed. RESULTS: The average time from operation to initial diagnosis was 10.5 years. The typical edema started inferiorly and progressed circumferentially, but central portion of the cornea was clear. The corneal thickness at edematous area was increased. The cell counts of endothelium was decreased in our patients. Additionally, the edema was associated with a punctate orange-brown pigmentation on the endothelium. CONCLUSIONS: So far this new syndrome of corneal edema has not been reported in china. Its clinical characteristics are not similar to that of conventional edema after intracapsular cataract extraction. The pathogenesis of this curious condition is still unknown. It may be associated with an aphakic eye, chronic uveitis and hereditary factors.

[Clinical analysis of primary orbital varix].

OBJECTIVE: To analyze and summarize the clinical and imaging signs, diagnosis and treatment of primary orbital varix. METHOD: 218 cases of primary orbital varix in 20 years were analyzed. RESULTS: Positional proptosis was the primary sign. Ultrasound examination showed that a characteristic well-out-lined lesion with low or without reflection of sound appeared in the orbital fat when the internal carotid vein was compressed. CONCLUSION: To recognize the characteristic imaging signs of orbital varix in ultrasound examination is valuable for the choice of treatment.

[Construction and screening of type 1 human immunodeficiency virus specific phage antibodies combinatorial library].

UNLABELLED: Human monoclonal antibodies to type 1 immunodeficiency virus (HIV-1) gp120 were generated from phage antibody combinatorial library. METHODS: The human immunoglobulin heavy chain Fd and light chain k genes were amplified by half-nested PCR from PBMC of patient infected with HIV. Phage antibody combinatorial library was constructed with the Fd and k chain genes using Pcomb3 as vector. The affinity selection and ELISA were adopted for generating specific phage antibodies. Partial DNA of a positive clone was sequenced and its soluble Fab was expressed in E coli. HIV-1 specific phage antibodies combinatorial library were constructed using the Fd and k genes and Pcomb3 vector. The library capacity was about 1.95 x 10(7). The specific phage antibodies were highly enriched after three rounds of biopanning selection against HIV-1 gp120 and 32% positive clones were detected by ELISA screening. DNA fragment coding for CH1 and CL derived from a positive clone was sequenced and its product was successfully expressed as soluble Fab which was specific for HIV-1 gp120. The HIV-1 specific phage antibody combinatorial library, and human monoclonal antibodies to HIV-1 gp120 have been used as tools for screening of neutralizing antibody to HIV-1, and the methods seem to be very crucial and applicable.

[Studies on target localization of Parkinson's disease treated with gamma knife].

Ninety cases of Parkinson's disease were treated in our center from Dec. 1993 to Aug. 1997. Based on our work, we have developed a method for the MR localization in the treatment of Parkinson's disease with Gamma knife, including the fixing of stereotactic frame, the selection of target, and the correction of the target's coordinates. In this paper, the effects of fixing of coordinate frame on the localization of target have been analyzed. Also presented are the errors of the coordinates of the selected targets, compared with those corrected. The method reflects a new idea for target localization.

An analysis of 1,001 blinding patients with corneal disease in 1960-1989.

PURPOSE: To investigate the condition and change of corneal blindness (CB) in the past 30 years. METHODS: 1,001 blinding patients of corneal disease were clinically analysed from 1960 to 1989, including etiology, sex, age, occupation, difference between the urban and rural areas etc. RESULTS: The male predominated. Infection stood the first cause of blinding corneal diseases, followed by trauma, malnutrition and the others. HSV-1 keratitis was the most frequent infection in 1980s. The highest incidence of CB was 20-40 year-old. The number of CB caused by occupational trauma was more than that by ordinary trauma in 1960s, but from 1970s, the latter was more than the former. The number of CB in rural area was more than that in the urban in 1960s, but from 1970s, there was no significant difference between the two areas. The percentage of bilateral CB was gradually decreased. CONCLUSION: We should try to find more effective strategies to prevent and treat CB caused by HSV-1 keratitis, pay special attention to preschool blindness caused by perforation injuries and avoid infection after corneal injuries of foreign bodies.

[The genetic analysis of amplitude of progesterone secretion in serial blood collection from tail in min sows].

28 adult Min sows (3-7 parities), represented 9 sire families were selected to conduct the continual blood collection from the tail for two hours at the interval of ten minutes between 9:00-11:00 in the morning on the sixth day after the last mating. The content of progesterone was assayed by radio-immunological assay. The amplitudes of the secretion of progesterone were generated by the procedure of HORM fft. exe complied by the author. It is difficult to get a reasonable conclusion from the content analysis, because the content varied widely between different points of blood collection in single sow. After Fourier conversion, the heritability of amplitude for the fifth partial wave was high (h2 = 0.932); middle for the basal, 2nd and 3rd partial waves; lower for others. This supported the idea that prolific sows had more active secretion of progesterone in luteal cells. The coexist of low and high heritability components in amplitude of progesterone secretion showed that the activities of progesterone secretion were influenced by both the genetics and environments. The genetic correlation of basal, 1st, 5th partial waves were negative, the others were positive, with the litter size and litter size alive. It showed that the reproductive performance of Min pigs were not expressed perfectly, as the balance of secretion of progesterone did not reach the best. The genetic correlation of all partial waves with duration of estrus was negative which supported that the progesterone suppresses the estrus. The genetic correlation of 3rd, 5th partial waves were stronger than others, but selection for 5th partial wave would make high responses than the 3rd wave, because the genetic correlation of 3rd partial wave with litter size and litter size alive was positive, therefore, the more appropriats method of selection for progesterone was adopted, the better results would be achieved in the improvement of indirect selection response for litter size.

Impact of oral immunization with Acanthamoeba antigens on parasite adhesion and corneal infection.

PURPOSE: To determine whether oral immunization mitigates ongoing Acanthamoeba castellanii corneal infections in pigs. METHODS: Pigs were orally immunized with aqueous Acanthamoeba antigen mixed with cholera toxin (Ac-CT) or with saline, before or after ocular infection with A. castellanii. Mucosal secretions (i.e., tears and enteric wash) were tested for Acanthamoeba-specific IgA by enzyme-linked immunosorbent assay. Enteric washes were used as a source of IgA in assays measuring the binding of trophozoites to Chinese hamster corneal epithelial (CHCE) cells. RESULTS: Pigs immunized with Ac-CT before ocular challenge with A. castellanii had significant anti-Acanthamoeba IgA antibody titers in their tears and enteric washes and were protected against ocular infection. Enteric washes from orally immunized pigs inhibited trophozoite binding to CHCE cells in vitro by more than 75%. By contrast, pigs immunized after corneal infections had been established displayed keratitis of the same severity and duration as that in control pigs. However, 80% of the orally immunized animals were resistant to rechallenge with Acanthamoeba-laden contact lenses, whereas none of the control animals was resistant. CONCLUSIONS: Oral immunization with Ac-CT protects against Acanthamoeba keratitis when administered before corneal challenge. However, delaying oral immunization until after corneal disease is established fails to mitigate keratitis. The appearance of parasite-specific tear IgA correlates with protection and may act by preventing the parasite's binding to the corneal epithelium.

Mortality attributable to cigarette smoking in China.

CONTEXT: The few published prospective studies of smoking and mortality in China have reported low relative risks, but the durations of follow-up were short. OBJECTIVE: To assess the mortality of ever- and never-smokers in a cohort after 20 years of follow-up. DESIGN, SETTING, AND SUBJECTS: A cohort analytic study in a machinery factory in Xi'an, China, involving 1696 people aged 35 years or older (1124 men and 572 women) examined in May 1976. MAIN OUTCOME MEASURES: All-cause and tobacco-associated mortality. RESULTS: A total of 56% of the men and 12% of the women were ever-smokers at baseline. Through August 31, 1996, 218 persons (173 men and 45 women) had died. The relative risks (95% confidence intervals [CIs]) for ever smoking (after adjusting for age, marital status, occupation, education, diastolic blood pressure, and triglyceride and cholesterol levels) for deaths resulting from all causes, all cancer, and coronary heart disease were, respectively, 2.42 (95% CI, 1.72-3.42), 2.50 (95% CI, 1.41-4.43), and 3.61 (95% CI, 1.35-9.67) in men and 2.32 (95% CI, 1.18-4.56), 1.98 (95% CI, 0.50-7.92), and 4.67 (95% CI, 0.78-27.8) in women. CONCLUSIONS: Previous prospective studies of smoking-related mortality in China tended to underestimate the risks, probably because of short durations of follow-up. We have demonstrated that smoking is a major cause of death in China, and the risks are similar to those seen in the United States and the United Kingdom. Thus, about half of the 300 million smokers in China will eventually die of smoking-related diseases if urgent tobacco-control measures are not instituted to prevent this growing epidemic.

CooA, a CO-sensing transcription factor from Rhodospirillum rubrum, is a CO-binding heme protein.

Biological sensing of small molecules such as NO, O2, and CO is an important area of research; however, little is know about how CO is sensed biologically. The photosynthetic bacterium Rhodospirillum rubrum responds to CO by activating transcription of two operons that encode a CO-oxidizing system. A protein, CooA, has been identified as necessary for this response. CooA is a member of a family of transcriptional regulators similar to the cAMP receptor protein and fumavate nitrate reduction from Escherichia coli. In this study we report the purification of wild-type CooA from its native organism, R. rubrum, to greater than 95% purity. The purified protein is active in sequence-specific DNA binding in the presence of CO, but not in the absence of CO. Gel filtration experiments reveal the protein to be a dimer in the absence of CO. Purified CooA contains 1.6 mol heme per mol of dimer. Upon interacting with CO, the electronic spectrum of CooA is perturbed, indicating the direct binding of CO to the heme of CooA. A hypothesis for the mechanism of the protein's response to CO is proposed.
