Alternative splicing of vascular endothelial growth factor (VEGF)-R1 (FLT-1) pre-mRNA is important for the regulation of VEGF activity.

Angiogenesis is essential for normal mammalian development and is controlled by the local balance of pro- and antiangiogenic factors. Here we describe a novel mouse cDNA sequence encoding sFLT-1 that is a potent antagonist to vascular endothelial growth factor (VEGF) and show for the first time its in vivo production. In situ hybridization and Northern blot analysis with probes specific for sFLT-1 or FLT-1 showed that the relative abundance of their mRNAs changed markedly in spongiotrophoblast cells in the placenta as gestation progressed. On day 11 of pregnancy, sFLT-1 mRNA was undetectable but FLT-1 readily apparent, and by day 17 sFLT-1 mRNA was abundant but FLT-1 barely detectable. sFLT-1 was identified in conditioned medium of cultured placenta from day 17 pregnant mice and likely to be present in the circulation, as there is a substantial increase of VEGF-binding activity in the serum from day 13 of pregnancy, which coincides with the abundant sFLT-1 expression in placenta. Expression of sFLT-1 was also observed in adult lung, kidney, liver, and uterus. These data suggest a novel mechanism of regulation of angiogenesis by alternative splicing of FLT-1 pre-mRNA. Treatment of pregnant mice with exogenous VEGF from day 9 to 17 of pregnancy, which alters the ratio of VEGF to sFLT-1, resulted in an increase in the number of resorption sites and fibrin deposition in the placenta of ongoing pregnancies. These findings have important implications for understanding placental function and may be relevant in a range of disease states.

Identification of selective mechanism-based inactivators of cytochromes P-450 2B4 and 2B5, and determination of the molecular basis for differential susceptibility.

Rabbit cytochromes P-450 (P-450) 2B4 and 2B5 differ by only 12 amino acid residues yet they exhibit unique steroid hydroxylation profiles. Previous studies have led to the identification of active site residues that are determinants of these specificities. In this study, mechanism-based inactivators were identified that discriminate between the closely related 2B4 and 2B5 enzymes. A previously characterized inhibitor, 2-ethynylnaphthalene (2EN), was found to be selective for 2B4 inactivation. As inhibitor metabolism and the partition ratio affect susceptibility, molecular dynamics simulations were performed to assess the stability of the productive binding orientation of 2EN within 2B4 and 2B5 three-dimensional models. Although 2EN was stable within the 2B4 model, it exhibited substantial movement away from the heme moiety in the 2B5 model. However, heterologously expressed 2B5 was found to catalyze the oxidation of 2EN to the stable product 2-naphthylacetic acid. Thus, the increased mobility of 2EN may result in reduced susceptibility of 2B5 by increasing the probability that the reactive ketene intermediate hydrolyzes with water instead of reacting with active site residues. Another compound, 1-adamantyl propargyl ether (1APE), selectively inactivated 2B5. The structural basis for 2EN and 1APE susceptibility was assessed using active site mutants. Interconversion of 2EN susceptibility was observed for 2B4 or 2B5 mutants containing a single alteration at residue 363. Single substitutions in 2B4 also conferred susceptibility to 1APE; however, multiple alterations were required to reduce the susceptibility of 2B5. These alterations may influence inhibitor susceptibility by affecting the stability of the productive binding orientation.

Reduced Nurr1 expression increases the vulnerability of mesencephalic dopamine neurons to MPTP-induced injury.

Mutation in the Nurr1 gene, a member of the nuclear receptor superfamily, causes selective agenesis of dopaminergic neurons in the midbrain of null mice. Homozygous Nurr1 knockout mice (Nurr1-/-) die 1 day after birth, but heterozygous mice (Nurr1 +/-) survive postnatally without obvious locomotor deficits. Although adult Nurr1 +/- mice show significantly reduced Nurr1 protein levels in the substantia nigra (SN), they display a normal range of tyrosine hydroxylase-positive neuron numbers in the SN and normal levels of dopamine in the striatum. The reduction in Nurr1 expression in Nurr1 +/- mice, however, confers increased vulnerability to the selective dopaminergic neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) compared with wild-type (Nurr1 +/+) mice. This study suggests that Nurr1 may play an important role in maintaining mature mesencephalic dopaminergic neuron function and that a defect in Nurr1 may increase susceptibility to SN injury.

An animal and clinical study on the change of neuropeptide Y release evoked by electrical stimulation and myocardial ischemia.

OBJECTIVE: To investigate the change characteristics of neuropeptide Y (NPY) release during acute myocardial ischemia period. METHODS: The animal test was carried out in in situ perfused guinea pig hearts with intact sympathetic innervation. Electrical stimulation-evoked exocytotic release of NPY during ischemia and reperfusion was tested by radioimmunoassay (RIA). The plasma NPY concentrations were measured in patients with acute myocardial infarction (AMI) and angina pectoris (AP) in different times. RESULTS: Electric stimulation of the left ganglion in guinea pig heart evoked an exocytic release of neuropeptide Y. Stimulation after 20 minutes of global ischemia (S2), compared with control period stimulation (S1) produced the inhibition of NPY to a certain extent (S2/S1: 0.72, P < 0.05), whereas the inhibition of NPY release disappeared after 5 minutes reperfusion (with S2/S1 of 1.01, P > 0.05). Ischemia alone, without the electric stimulation, did not apparently induce NPY release. The clinical test found that the plasma NPY level was increased significantly during the acute ischemia attack period of coronary heart disease (CHD). The plasma NPY level reached peak (136.3 +/- 66.5 pg/ml) in patients during the first day after AMI. It began to decrease from the third day and came to normal level in the end of the first week. The plasma NPY level was 159.3 +/- 98.5 pg/ml in AP patients during angina attack. After two weeks treatment, the plasma NPY level was decreased to 118.9 +/- 54.3 pg/ml (P < 0.05). CONCLUSIONS: The NPY release of global ischemia have some relation with sympathetic nerve activity. At the early stage of ischemia, NPY release is inhibited to some degree and the inhibition factors will fade away on reperfusion. NPY interferes with the pathogenesis and the pathophysiolgy.

A review on studies of smoking and coronary heart disease in China and Hong Kong.

PURPOSE: To document and review all the published reports on smoking and coronary heart disease (CHD) in China and Hong Kong, and to estimate the risks of CHD due to smoking. DATA SOURCES: Papers cited in Medline, Chinese Biomedical Literature Disc and Chinese Medical Current Contents, references in review papers, conference papers and abstracts. STUDY SELECTION: All published reports which had findings on smoking and CHD were included. DATA EXTRACTION: Reports on 13 cross-sectional, 16 case-control and 13 prospective studies were included. Available data on risk estimates from 23 studies were extracted and pooled odds ratios (OR) and relative risks (RR) were calculated using Mantel-Haenszel's methods. RESULTS: In China in 1970s, most results, which were from cross-sectional studies, did not show an association between smoking and CHD. In the 1980s, most case control studies found an association but the prospective studies did not. In the 1990s, there was stronger evidence from prospective studies mainly in men. The relative risk was about 1.90 in men and about 2.50-3.50 in women although the 95% confidence interval for the latter was wide. Dose response relationships were observed in 12 studies. In Hong Kong, there were one cross-sectional and two case control studies. CONCLUSIONS: The existing evidence supports that smoking is a cause of CHD in Chinese, particularly in men. More data in women are needed to quantify the risk more precisely. The risk estimates probably indicate only the early stage of a growing tobacco epidemic.

[Identification of three common G6PD gene mutations in Yi minority in Yunnan province].

OBJECTIVE: To identify three common G6PD gene mutations in Yi minority in Yunnan province. METHODS: G6PD gene mutation of 28 samples from G6PD deficient patients was detected by using amplification refractory mutation system(ARMS). Samples were collected from Yi minority in Yunnan province. RESULTS: In 28 cases, 22 were identified to be three known mutations, including 12 of 1388(G-->A), 9 of 1376(G-->T) and 1 of 95(A-->G). The remaining 6 cases were undefined. CONCLUSIONS: This is the first report of G6PD gene G1376T and A95G mutations in Yi minority. The three common mutations account for 78.6% of the 28 cases.

Immunological characteristics of natural resistance in Microtus fortis to infection with Schistosoma japonicum.

OBJECTIVE: To explore the immunological characteristics of natural resistance to Schistosoma japonicum infection in Microtus fortis (MF) living in the Dongting Lake area. METHODS: Passive transfer of sera from uninfected laboratory bred MF (BMF) to albinao mice (AM) was performed to observe the acquired protection. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and enzyme-linked immunoblotting (ELIB) methods were used to recognize 4 different life-cycle stage antigens of S. japonicum by sera from wild MF (WMF), BMF, BMF13 and BMF19. Tests were also performed on in vitro killing effect of sera and/or lymphocytes from BMF and WMF to schistosomulae; quantitative determination of C3 and C4 by immunoturbidometry, and interleukin-4 (IL-4) and antibodies to the 4 stage antigens in sera from WMF, BMF and infected BMF by ELISA. RESULTS: Compared with the control group, stool eggs per gram (EPG) of AM in the test group was significantly reduced by 81.54%, miracidium hatching rate, by 50.67%, liver egg counts, by 72.07%, the diameter of hepatic egg granuloma, by 70.39 microns. Western blotting probed with the 4 MF sera all revealed 7 specific bands for SSA, 3 for AWA and SEA, but none for CA antigens. The sera and/or lymphocytes from WMF and BMF gave obvious killing effects on schistosomulae with an adjusted death rate of 64.12%-78.83%. The levels of "natural antibodies" produced by MF to S. japonicum were in the following order: anti-SSA > anti-AWA > anti-SEA > anti-CA, all of which increased significantly after the infection. Serum levels of C3, C4 and IL-4 in uninfected BMF were significantly higher than those in AM. After infection, levels of C3 and C4 were further increased respectively by 72.83% and 295.49% in the 4th week and IL-4 by 303.83% in the 9th day. CONCLUSIONS: Immunological characteristics of innate resistance in M. fortis to S. japonicum infection were existed with no significant difference between WMF and BMF.

[The relationship of Apo E2 and renal insufficiency lipid levels in NIDDM].

OBJECTIVE: To study the relationship of ApoE2 and renal insufficiency, lipid levels in NIDDM patients. METHODS: PCR/ASO probe technique was used to determine ApoE genotypes. 46 NIDDM patients with renal insufficiency and 84 NIDDM patients without renal insufficiency as well as 50 healthy individuals were selected. Of the patients with renal insufficiency, 10 belonged to ApoE2 and 38 to ApoE3/3. In addition, 57 NIDDM patients with normalbuminuria (6 with ApoE2, 51 with ApoE3/3) were selected from the 84 NIDDM patients without renal insufficiency. RESULTS: Allele frequency was significantly higher in diabetic patients with renal insufficiency than in diabetic patients without renal insufficiency and in healthy individuals (P < 0.05). Tc, TG levels were significantly higher in ApoE2 group with renal insufficiency than in E3/3 group with renal insufficiency, ApoE3/3 and ApoE2 groups with normoal buminuria. HDL-c level was significantly lower in ApoE2 group with renal insufficiency than in other 3 groups. CONCLUSION: ApoE2 is associated with renal insufficiency in NIDDM patients and that ApoE2 may be a factor that aggravates lipid abnormalities in NIDDM patients with renal insufficiency.

[Protective effects of bactericidal/permeability-increasing protein on the vital organ function after endotoxic shock in rats].

OBJECTIVE: To investigate the protective effects of BPI on the vital organ function after endotoxic shock in rats, and to provide experimental evidence for further clinical studies. METHODS: 12.5 mg/kg of E. coli 026:B6 LPS were bolus injected via i.a. to reproduce endotoxic shock. Immediately after injection of LPS, 5 mg/kg of BPI or equal volume of physiological saline were bolus injected via i.a. RESULTS: The survival time of shocked animals was prolonged in the BPI group, 24 h survival rate was higher than that in the saline group. The values of mean arterial pressure, left intraventricular systolic pressure, isovolemic ventricular pressure, and dp/dtmax in the BPI group were markedly higher than those in the saline group. The plasma glutamic-pyruvic transaminase and urea nitrogen levels were obviously lower than those in the saline group. The plasma endotoxin and TNF levels in the BPI group were significantly lower than those in the saline group. CONCLUSIONS: BPI has good protective effects on cardiac, hepatic, and renal function in endotoxic shock in rats, indicating that BPI might be useful in clinic treatment of sepsis/septic shock.

[A study on chemical constituents in seeds of Crataegus pinnatifida Bge. var. major N. E. Br].

OBJECTIVE: To study the chemical constituents in the seed of Crataegus pinnatifida var. major. METHOD: Four compounds were isolated and identified by column chromatography and 1H, 13C-NMR data. RESULTS: The compounds obtained were identified as protocatechuic acid, gallic acid, catechol and p-hydroxybenzoic acid. CONCLUSION: All the compounds were isolated from this plant for the first time.

[Chemical studies on polysaccharides from mycelium of Hericium erinaceum (Bull. Ex Fr.) Pers].

OBJECTIVE: To study the chemical composition and structure of polysaccharides from the mycelium of Hericium erinaceum. METHOD: DEAE-cellulose and DEAE-Sephadex column chromatography were employed for isolation and purification. Sephadex column chromatography and agarose gel electrophoresis were used to measure the purity. Gel filtration was employed to measure MW. Acid hydrolysis, enzymolysis, GC and spectral analysis etc. were employed for determination of the composition and construction of the unique homogeneous polysaccharides. RESULT: The five homogeneous polysaccharides were obtained from the mycelium. The main compounds(HEPA1, HEPA4 and HEPB2) were identified. CONCLUSION: HEPA1 (MW 62,000) is a heteropolysaccharide consisting mainly of glucose. HEPA4 (MW 26,000) is a heteropolysaccharide peptide. HEPB2 (MW 12,000) is a heteropolysaccharide consisting mainly of glucose. The main linkage of all the afore-said homogeneous monosaccharides is alpha-linkage.

[Analysis of chiral compounds by micelle electrokinetic capillary chromatography].

An overview on applications of micelle electrokinetic capillary chromatography (MECC) in chiral compounds analysis is presented. Ionic chiral surfactants such as bile salts, long-alkyl-chain surfactants and high-molecular-weight surfactants, which can migrate with electroosmosis and its electrophoretic mobility, and interact with the solutes, are employed as chiral selectors. Addition of electrically neutral chiral selectors such as cyclodextrins is also successful in MECC separation. Theory of MECC enantiomer separation is briefly described.

[The effect of age on the regional blood flow of neck of femur in normal Chinese].

This study aimed to assess the effect of age on the regional blood flow of neck of femur in normal Chinese. 133Xe washout method was adopted. The result showed that the femur regional blood flow of normal Chinese decreased slowly with the increase of their age. The relation between age and femur blood flow might be expressed as F = -0.042Y + 9.01(F refers to blood flow, Y refers to age). The average femur regional blood flow of normal Chinese was 7.37 +/- 0.62 ml.(100 g.min)-1 for 21-40 years old. The authors suggest this result can be considered to stand for regional femur blood flow of young healthy adults.

[SDS-PAGE and EITB analysis of the protein components of different isolates of Schistosoma japonicum in China and Japan].

AIM: To make a supplementary observation on the protein components prepared from adult S. japonicum of 4 different isolates from Zhejiang, Jiangxi, Taiwan Provinces of China and Japan in origin and to observe antigen reactivity of the above-mentioned isolates together with adult worms from Anhwi, Hubei, Sichuan and Yunnan isolates against heterologous anti-Oncomelania h. hupensis (collected from Guangxi, China and Japan) sera by EITB. METHODS: SDS-PAGE and EITB. RESULTS AND CONCLUSION: SDS-PAGE showed that by Coomassie blue staining, male S. japonicum from Jiangxi, Zhejiang, Taiwan Provinces and Japan isolales revealed 7-17 bands while female worms revealed 1-6 bands. The protein patterns of Taiwan and Zhejiang male worm were similar, but slight difference could be seen above 81 kDa. By silver staining, male worms of the 4 isolates revealed 10-23 bands while female worms revealed 1-19 bands. Male worms from Japan not only showed less bands but also differed in their pattern as compared to those of other Chinese isolates. Results of EITB revealed that each of the 4 isolates had slightly different pattern but all of the tested isolates had common antigens with their heterologous snail hosts i.e., Oncomelania h. hupensis from Guangxi, China and Japan.

[Preliminary study on the levels of natural antibodies against Schistosoma japonicum in Microtus fortis in Dongting lake area].

AIM: To explore the detection rate and the levels of natural antibodies to cercarial antigen(CA), juvenile antigen(JA), adult worm antigen (AWA) and soluble egg antigen (SEA), respectively, derived from Schistosoma japoncum in Microtus fortis distributed in Dongting Lake area. METHODS: The antibodies in wild Microtus fortis(WMF), laboratory-bred Microtus fortis (BMF) and laboratory-bred Microtus fortis 15 days after S. japonicum infection (BMFi15) were detected by indirect ELISA and a comparison of the antibody levels between Microtus fortis including WMF, BMF, BMFi15 and albino mice without infection (AM), 15 days after infection(AMi15) and 42 days after infection (AMi42) was made. RESULTS: The positive rates of detection for anti-JA-Ab in WMF and BMF were 94.6% and 94.4%, respectively, followed by 85.5% and 83.3% for anti-AWA-Ab, 58.1% and 59.4% for anti-SEA-Ab, and 14.3% and 13.9% for anti-CA-Ab. The levels of antibodies to the above four antigens in BMFi15 increased significantly, the detection rates being 100% for anti-JA-Ab and anti-AWA-Ab, 84.4% for anti-SEA-Ab and 65.6% for anti-CA-Ab. The levels of antibodies in WME, BMF and BMFi15 had the similar trend(JA > AWA > SEA > CA), and those in BMFi15 to CA, JA, AWA and SEA increased by 1.9, 2.2, 2.1 and 1.5 folds, respectively, when compared with BMF. CONCLUSION: Natural antibodies to Schistosoma japonicum were found in wild and labortorybred Microtus fortis. The levels of antibodies in BMFi15 to four above antigens were apparently higher than those in AMi15.

[Study on cultivation of Dendrobium flexicaule by habitat imitation].

Dendrobium drug, a rare medicinal herb, reproduces differcut for growth environment to lead rare resources. By studying of cultivation for many many years in Fuliu mountain area, authors had found out the cultivated method simulating the habitat of wild Dendrobium flexicaule. The results could supply a basis for its development and utilization.
