Inhibition of prostaglandin synthesizing enzymes by haptoglobin and plasma of rats with inflammation.

Administration of carrageenin or adjuvant to rats to induce inflammation produced increases in the plasma haptoglobin level. Simultaneously, there was an increase in the activity of the plasma inhibitor of prostaglandin E2 synthesis. Partially purified haptoglobin inhibited the microsomal over-all conversion of arachidonic acid to prostaglandin E2. The addition of the haptoglobin inhibited two heme-dependent reactions catalyzed by a purified enzyme of seminal vesicle microsome, i.e., the prostaglandin G2 synthesis from arachidonic acid and the conversion of prostaglandin G2 to H2. However, the plasma inhibitory activity was accounted for only partially by the haptoglobin contained in the plasma from treated rats.

Inotropic effects of non-depolarizing muscle relaxants in isolated canine heart muscle.

The inotropic effects of five non-depolarizing muscle relaxants were examined using an isolated canine heart muscle preparation. Except for fazadinium, all drugs were studied in their commercially available forms. d-Tubocurarine chloride (dTc) and metocurine iodide (MTC) produced dose-dependent decreases in isometric force (F) and the maximum velocity of force development (dF/dt) at concentrations greater than 22.5 x 10(-3) g/L for dTc and greater than 15.0 x 10(-3) g/L for MTC, concentrations which are 3 and 6 times higher than estimated clinical serum concentrations, respectively. Myocardial depression was about 3 times less with MTC than with dTc at equipotent concentrations. The degree of depression in F and dF/dt produced by MTC was almost identical with that produced by phenol, a preservative of MTC, indicating that MTC-induced myocardial depression may be due to the effect of the preservative. Pancuronium bromide (PC) produced a dose-dependent increase in F and dF/dt and decrease in the time to peak force. PC-induced changes in F, dF/dt, and time to peak force were inhibited by administration of propranolol 10(-6) M. The results indicate that PC possesses a positive inotropic effect mediated by beta-adrenergic stimulation. Alcuronium chloride did not change F or dF/dt at concentrations from 5.0 x 10(-3) to 60.0 x 10(-3) g/L. Frazadinium bromide increased F and dF/ dt slightly at a low concentration (1.875 x 10(-2) g/L), but further increases in its concentration returned the values of F and dF/dt to control levels. F and dF/dt were not altered in vitro by concentrations of relaxants that would be anticipated in plasma in vivo in patients given clinically effective doses of 0.3 mg/kg of dTc, 0.1 mg/kg of MTC or PC, 0.2 mg/kg of alcuronium chloride, or 0.75 mg/kg of fazadinium bromide.

Vitamin A receptor in cancers and in vitro-transformed epithelial cells of the rat urinary bladder: its implication in the preventive effect of vitamin A on in vitro keratinization.

Cellular retinol-binding protein (CRBP) in cancers and in vitro-transformed epithelial cells of the ACI/N rat urinary bladder was assayed with radiolabeled retinol by sucrose density gradient centrifugation. CRBP was detected in abundance in tumor tissues of all 5 transplantation bladder cancer lines. However, this protein was below the limits of detection in bladder cancer cells of culture lines that had originated from the same primary cancers as the corresponding transplantation lines. CRBP was detected in small quantity in in vitro-transformed epithelial cells of a culture line but it was absent in another line of similar origin. The addition of retinol to the culture medium prevented the in vitro keratinization of CRBP-positive cells but it had no effect on the keratinization of CRBP-negative cells.

Quantitative histological studies on the spleen of ducks after neonatal thymectomy and bursectomy.

The duck spleen was examined histoplanimetrically after neonatal surgical thymectomy, cyclophosphamide treatment, and/or X-ray irradiation. The development of the periarterial lymphoid tissues, which consisted mainly of small lymphocytes, was inhibited in the spleen of the Tx and Tx-X ducks. The peri-ellipsoidal lymphoid tissues, consisting of pyroninophilic medium or large sized lymphocytes, was greatly depleted in the Bx and/or CY ducks. The germinal centres also showed probable bursa dependency. On the other hand, the development of the perivenous lymphoid tissue was not inhibited after Tx and Bx. A number of plasma cells was distributed in the red pulp of the Bx and/or CY duck spleen.

Reactions of potent mutagens, 3-amino-1-methyl-5H-pyrido[4,3-b]indole (Trp-P-2) and 2-amino-6-methyl-dipyrido[1,2-a:3',2'-d]imidazole (Glu-P-1) with nucleic acid.

Two potent mutagens, 3-amino-1-methyl-5H-pyrido[4,3-b]indole (Trp-P-2), isolated from a tryptophan pyrolysate, and 2-amino-6-methyldipyrido[1,2-a:3',2'-d]imidazole (Glu-P-1), isolated from a glutamic acid pyrolysate, modified calf thymus DNA in the presence of rat liver microsomes. The major base modified by Trp-P-2 was identified wih 3-(3-guanyl)amino-1-methyl-5H-pyrido[4,3-b]indole. The major base modified by Glu-P-1 was identified with 2-(8-guanyl)amino-6-methyldipyrido[1,2-a:3',2'-d]imidazole. N-acetoxy-Glu-P-1 efficiently modified DNA without microsomes.

Changes in type of collagen during the development of human post-burn hypertrophic scars.

Neutral-salt soluble collagen was relatively abundant in young post-burn hypertrophic scars. The content of insoluble collagen was fairly low in granulation tissues, whereas it gradually increased over 2 years and reached a significantly higher value than that in normal skins. Interrupted gel electrophoresis was carried out to estimate the relative proportions of type III and type I collagens in the pepsin digests of human normal skins and post-burn scars. Granulation tissues showed an abnormally higher ratio of type III to type I (0.37--0.72) than the mean value of normal skins (0.17 +/- 0.04 (S.D.)). The ratio gradually decreased to the normal range after 2 years or so.

Simultaneous determination of endogenous norepinephrine and dopamine-beta-hydroxylase activity in biological materials by chemical ionization mass fragmentography.

A new clean-up procedure is described for the quantitation of biogenic amines and their metabolites by gas chromatography-chemical ionization mass fragmentography. The procedure is carried out in an acidic, non-aqueous medium. The metabolites as well as the amines are extracted with greater than 92% recovery. Using a CPG-10 column, the procedure is most suitable for the purification of these amines and their metabolites from the extract. It has been applied to the simultaneous determination of endogenous norepinephrine and dopamine-beta-hydroxylase (DBH) activity in rat adrenal by use of [2H2]dopamine as a substrate and [2H6]norepinephrine as an internal standard. The quantitation limit of the DBH activity is 0.1 pmole/mg of rat adrenal per hour.
