[Partial deletion of mitochondrial DNA in mitochondrial encephalomyopathies].

OBJECTIVE: To characterize the deletions of mitochondrial DNA (mtDNA) in Chinese patients with Kearns-Sayre syndrome (KSS) and chronic progressive external ophthalmolegia (CPEO) and identify deletion mutations of mtDNA be the etiology of these diseases. METHODS: Patients and preparation of total DNA: Two patients with KSS and two with CPEO and ten with other mitochondrial myopathies or encephalomyopathies were determined by histological and biochemical assays. Total DNA was isolated from 100 mg to 150 mg of frozen muscle obtained by biopsy using the methods described by Zeviani et al. (1988). Preparation of mtDNA probes: MtDNA were isolated and purified according to Palva's method (1985). The mtDNA was linearized by digestion with the restriction endonuclease Pvu II and separated by electrophoresis through low melting agarose gel. The 16.5 kb DNA band was recovered from the gel and labeled with alpha-p32 dCTP by random primer labeling. Southern blot analysis: Portions of five micrograms total DNA obtained from the patients and controls were digested with Pvu II, EcoR I, Hind III And Xba I respectively. The digested DNA was separated by agarose gel electrophoresis and transferred to nitrocellulose membrane and then hybridized with P32 labeled mtDNA as previously described (Zhang, 1991). RESULTS: A large-scale deletions of mtDNA were identified in two patients with KSS and two patients with CEPO. The deletions ranged in size from 2.4 kb to 5.5 kb. The proportion of mutated mtDNA in each patients ranged from 54.6% to 84.6% of total mtDNA. No detectable deletions were found in mtDNA of ten patients with other mitochondrial myopathies or encepholomyopathies and normal controls. CONCLUSIONS: Deletions of mtDNA were found in all KSS and CEPO patients detected. These results supported The view point of the deletions are important causes of physical defect in KSS and CEPO.

[Scanning electron microscopic changes of maxillary sinus mucosa before and after functional endoscopic sinus surgery].

We attempted to evaluate postoperative maxillary sinus mucosal changes in 20 maxillary sinuses of 16 patients who underwent functional endoscopic sinus surgery (FESS). Ciliated area of the maxillary sinus mucosa and ostium of maxillary sinus were quantitatively observed by scanning electron microscope and image analysis before and after FESS. In the present study, the number of preoperative ciliated cells of maxillary sinus mucosa and ostium was significantly reduced, goblet cells and microvillous cells were increased, and squamous cell metaplasia observed in part of the maxillary sinuses. Many damaged ciliated cells tend to recover and ciliated area significantly increased at 6-12 months after the surgery. The results showed that the damaged ciliated epithelium could return to normal by the improvement of ventilation and drainage of the maxillary sinus following FESS.

[Modulation of potassium channel by arachidonic acid].

Arachidonic acid is an important modulator for the inflammatory process. It can directly or indirectly regulate potassium channels which exist widely in the membrane of various types of cells. The direct modulation changes the conformation of potassium channel protein by the interaction between arachidonic acid and the protein, or through the interference with cell membranes around channels. The indirect influence of arachidonic acid upon channels results from its metabolites caused by cylcooxygenase and lipooxygenase, protein kinase C, and [Ca2+]i. All these aspects provide new potential targets for pharmacological investigations.

[DNA polymorphism in the genomes of different Escherichia coli strains].

Bacteria have been traditionally classified on the basis of their morphology, biochemical reaction, serology and etc. However, in some case these methods could not authenticate the closely related bacteria strains. In this study, we cloned two repetitive DNA sequences with 0.9 and 0.6 kb in length from Escherichia coli K12 strain JM109 and designated as ECR-1 and ECR-6 respectively. Using ECR-1 and ECR-6 sequences or their combination as the probes for DNA polymorphism analysis, we were be able to develop a molecular method of biotyping for the identification of very closely related strains of Escherichia coli. Both of ECR-1 and ECR-6 probes could be applied for the taxonomy, epidemiological and microecological studies, and clinical diagnosis for pathogenic Escherichia coli strains.

Mutation effect of high altitude balloon flight on rice and green pepper seeds.

Dry seeds of rice and green pepper were carried in the basket of a high altitude balloon to 30-40 km for 8 h. The treated seeds were planted on the ground and characters of these seeds and their progenies, such as development, fruiting, disease resistance were observed. The results showed that the SP2 generation of the treated rice showed strong mutation. A Japonica male fertility restorer rice line was found out. It could be hybridized with sterile line of Indica rice, and their progeny had strong hybrid vigor, high setting percentage and good plumpness of grain. Disease resistance, high-yield, big-fruit and high vitamin C content strains were selected from the progenies of treated green pepper seeds. The above results indicated that the special conditions in high altitude could produce a variety of mutations and most of them are hereditable. Therefore, high altitude balloon flight can be used as a new method of mutation breeding.

[Discovery of a novel reassortant H1N2 influenza virus].

Three strains of influenza A virus were isolated from patients suffering from influenza like disease in Railway Hygiene and Anti-epidemic Station of Taiyuan in January 1996. The identification results by serological method indicated that the isolates were different from H1N2 subtype of influenza A virus isolated from men in 1989 and 1992 and that their HA antigenicity was similar to that of A/RP/8/34 (H1N1) virus, but distinguishable from that of influenza A (H1N1) virus circulating in mans recently. The comparison of migration patterns of the RNA among the isolates, the A/PR//34 (H1N1) and the A/Wuhan/359/95 (H3N2) viruses revealed that the migration patterns of RNA 1-4 segments of the isolates were similar to those of A/PR/8/34 (H1N1) virus, but those of RNA 5-6 segments were indistinguishable from those of A/Wuhan/359/95 (H3N2) strain. However, the migration patterns of RNA 7-8 segments of the isolates were not only different from those of A/PR/8/34 (H1N1) virus, but also distingrasgable from those of A/Wuhan/359/95 (H3N2) strain. Therefore, it could be considered that the isolates were a novel reassortant H1N2 influenza A virus.

[Aetiologic study on the outbreaks of influenza-like disease in Shenzhen].

During March to May in 1994, the outbreaks of influenza like disease occurred in Shenzhen city. The outbreaks were caused by influenza B virus demonstrated by aetiologic and serological studies. The antigenic analysis indicated that the isolates were antigenically similar to B/Shanghai/1/93 and B/Guangdong/8/93 strains (both are B/Victoria/2/87. like virus). The genetic analysis showed that the HA1 genes of the isolates were 1038 nucleotides in length coding for HA1 protein of 346 amino acids which were same as those of B/Shanghai/1/93 virus and that the homogeneity of amino acid sequence on HA1 protein between isolates and B/Shanghai/1/93 virus was as high as 95%. These results further demonstrated that the isolates were B/Victoria/2/87 like strains. Meanwhile, this paper also revealed that the two antigenically distinct genetic lineages of influenza B virus were cocirculating in human population.

[HBsAg autologous red cell agglutination assay].

Three lot of HBsAg autologous red cell agglutination kits (ARCA kit) have been qualified in lab, clinical trial and the stability confirmation have been performed. The sensitivity of the kits is 1ng/ml HBsAg. A, B, O, AB type of peripheral blood or antiagglutinated specimens can be used for assay. No Holk effect has been found. The repeated rate of 10 samples assayed for five times reached 100%. The specimens collected from HAV, HCV, Schistosoma Japonicum infected patients or RF positive patients do not interfere the test. The specificity of the kits is very high. 1712 clinical specimens were inspected comparing with the EIA kits in market. The relativity between the two methods is 0.985. 18 of the 26 specimens which two methods did not accord with each other have been confirmed. The false positive and missing detected for ARCA kits is 1, 3 and for EIA kits are 6, 8 respectively. ARCA kits are stable in 4 degrees C and 22 degrees C for more than one year. The kits can stand in 35 degrees C, 45 degrees C for 45 and 10 days without losing activity respectively.

[The prognostic significance of regular bone marrow aspirations during induction chemotherapy in acute myeloid leukemia].

OBJECTIVE: To evaluate the prognostic significance of regular aspirations during induction chemotherapy in acute myeloid leukemia. METHODS: Regular bone marrow aspirations were performed during the first course of induction chemotherapy in 96 cases of de novo AML. RESULTS: Complete remission (CR) rate after the first course in patients with < or = 0.10 bone marrow blasts at the 7th day on chemotherapy was higher than that in patients with > 0.10 blasts (64.6% vs 40.0%, P < 0.05). Prolonged the chemotherapy course for a few more days in the latter group of patients increased CR rate from 25.0% to 50.0%. CR rate in patients with < or = 0.10 bone marrow blasts at the nadir of white blood cell count after chemotherapy was higher than that in patients with > 0.10 blasts (66.0% vs 25.8%, P < 0.005), and the refractory cases were significantly fewer in the former group of patients (7.5% vs 45.2%, P < 0.005). CONCLUSION: Regular bone marrow aspirations during induction chemotherapy were helpful in predicting the prognosis and guiding the treatment in AML.

[Tetraploid clones characterized by double 8;21 translocations in two patients with acute myeloid leukemia].

OBJECTIVE: To report two cases of leukemia with t(8;21) and tetraploid clones. METHODS: R-banding technique was used. RESULTS: Cytogenetic studies of BM cells at diagnosis revealed that each of the two cases had three related abnormal clones besides a normal one: 46, XY, t(8;21) (2%), 46, idem, add((7)(q31)(6.8%) and 92, idem x 2 (80.6%) in case 1 and 46, XX, t(8;21) (13.4%), 47, idem, + 4 (46.3%) and 94, idem x 2 (39.1%) in case 2. They did not obtain complete remission on chemotherapy and survived 4 and 6 months, respectively. CONCLUSION: Tetraploid or near tetraploid clones are secondary events which are specifically associated with t(8;21) leukemia and the prognosis of patients with this kind of abnormal karyotype is poor.

[The effect of antisense human Fas RNA on activation induced apoptosis of T cell].

OBJECTIVE: To explore an approach to the reduction of activation induced apoptosis of T cell so as to maximize the number of CTL and enhance the tumor cytotoxicity. METHODS: CD3-induced Jurkat cell line was used as the activated T cell apoptosis model, and the antisense Fas cDNA was introduced into Jurkat cells with retroviral vector. RESULTS: The expression level of Fas protein in Jurkat cells transducted with the retroviral vector was decreased. Apoptosis was reduced in antisense Fas-transfected Jurkat cells after anti-CD3 or anti-Fas treatment. CONCLUSION: Antisense blocking of Fas expression can partially inhibit Jurkat cell apoptosis induced by anti-CD3 or anti Fas.

[Rapid determination of low molecular weight aldehydes in air by gas chromatography].

This paper reports a method for determination of low molecular weight aldehydes in polluted air by GC-FID using a GDX-401 packed column (3 mm x 3 m). The advantages of this method are simple, rapid, accurate and sensitive. The method requires only 8 minutes for the whole analysis. In the concentration range of 1.7-442.4 mg/m3 for formaldehyde and 0.5-680 mg/m3 for acetaldehyde and propionaldehyde the calibration curves are linear. The minimun detectable amounts are 3.5 ng, 1 ng and 1 ng, and the average recoveries (n=5) are 89.4%, 89.6%, 88.1% for formaldehyde, acetaldehyde and propionaldehyde respectively. The quantitative determination is performed by external standard calibration method. The results of sample analysis are satisfactory.

[Nondectructive analysis of liquid samples with near-infrared spectroscopy].

This paper studied the near-infrared spectra of liquid samples. The plots of weights at each wavelength showed the spectral characteristics of pure materials. The samples could be classified by using plots of the scores of their first principal components. The principal component regression of samples was made. The obtained results were identical with the reference analytical values. The advantages of near-infrared spectroscopy, combined with principal component analysis, lie in its nondectructivity for samples and suitability for on-line measurements.
