[Effects of estradiol and medroxyprogesterone on the growth and doxorubin-resistance of drug-resistant human epithelial ovarian cancer cell line OVCAR-3].

OBJECTIVE: To study the effects of estradiol (E2) and medroxyprogesterone acetate (MPA) on the growth of drug resistant human epithelial ovarian cancer cell line. METHODS: OVCAR-3 cells, treated by different concentrations of E2, MPA and verapamil, were examined for their growth inhibitions with methyl thiazolyl tetrazolium (MTT, a tetrazolium dye) rapid photocolorimetric assay. RESULTS: OVCAR-3 cell growth was inhibited by E2 at high concentrations (10.00-100.00 mumol/L) and by MPA at 100.00 mumol/L, while lower concentrations of E2 (0.01-1.00 mumol/L) and MPA (0.01-10.00 mumol/L) had no effect on OVCAR-3 cell growth, neither stimulation nor inhibition. The effect of E2 combined with MPA was more obvious than either one used separately. E2 and MPA in combination with doxorubin showed a synergistic inhibition effect on OVCAR-3 cell growth, which suggested that both of them could reverse the doxorubin resistance of OVCAR-3 cell line. The inhibition effect of Verapamil combined with doxorubin was obviously weaker than that of E2 and MPA. CONCLUSIONS: E2 and MPA could reverse the doxorubin resistance of OVCAR-3 cell line and their inhibition were more obvious than that of verapamil, which suggested that E2 and MPA may be used clinically for adjuvant chemotherapy.

Intercellular adhesion molecule-1 and accumulation of eosinophils in nasal polyp tissue.

OBJECTIVE: To investigate the relationship between intercellular adhesion molecule-1 (ICAM-1) and the accumulation of eosinophils in nasal polyp tissue to better understand the mechanism of airway eosinophilic inflammation. METHODS: The expression of ICAM-1 and its natural ligand, lymphocyte function-associated antigen-1 (LFA-1), in normal nasal mucosa from 6 controls and in nasal polyp tissue from 19 patients with nasal polyposis were determined with immunohistochemistry. With dual immunohistochemistry and May-Griünwald Giemsa stain (MGG), the expression of LFA-1 and infiltrating eosinophils in nasal polyp tissue was observed. RESULTS: The expression of ICAM-1 and LFA-1 was stronger in the nasal polyp tissue than in normal nasal mucosa. There was a positive relationship between the infiltration of eosinophils and the expression of LFA-1 on eosinophils. CONCLUSION: Accumulation of eosinophils in nasal polyp tissue is associated with the counter-effect between adhesion molecules and its ligand on eosinophils.

Aberrant expression of growth factor Wnt-5A in six urinary malignant cell lines.

OBJECTIVE: To investigate the expression of growth factor Wnt-5A in urinary tumor cell lines. METHODS: By semi-quantitative RT-PCR (reverse transcriptase polymerase chain reaction), the expression of Wnt-5A in six urinary malignant cell lines, one primary cultured renal fibroblast and one case of normal human renal tissue was detected using Gel Doc 1000 computer controlled molecular analysis system. RESULTS: The expression of Wnt-5A in-urinary tumor cell lines was much higher than that in primary cultured renal fibroblasts (PRF) and normal human renal tissue (NRT). The levels of Wnt-5A mRNA were different between six malignant cell lines. CONCLUSION: The overexpression of growth factor Wnt-5A has a potential effect on the oncogenesis of urinary malignancies.

Interstitial brachytherapy using iridium-192 for malignant brain tumors: clinical results.

OBJECTIVE: To determine the effects and toxicity of interstitial brachytherapy using iridium-192 on brain malignant gliomas. METHODS: Between January 1992 and January 1995, 56 patients with anaplastic astrocytoma and glioblastoma multiforme were treated with stereotactic brachytherapy using temporary high-activity iridium-192 sources. RESULTS: The median survival for patients receiving brachytherapy was 28 months. The survival rates at 1, 2, 3 years were 92.8%, 83.9% and 71.4% respectively. CONCLUSIONS: Brachytherapy may improve the control of local tumor and prolong the survival, when used in deep malignant brain gliomas, by temporary implanted high doses of iridium-192 sources.

[Study of thrombopoietin and its receptor C-mpl in acute leukemia].

OBJECTIVE: To study the expression of C-mpl gene in acute leukemia (AL) and its implication and investigate the effect of recombinant human thrombopoietin (rhTpo) on acute leukemic cells as well as its relation to C-mpl expression. METHODS: C-mpl expression was detected in 43 AL patients by using semi-quantitative reverse transcription polymerase chain reaction and the effect of rhTpo on acute leukemia cells by MTT assay. RESULTS: C-mpl was expressed in 22 of 35 patients with acute myeloblastic leukemia (AML), but did not in 8 patients with ALL. The percentage of C-mpl expression (16/22, 72.7%) was significantly higher in CD34 positive AML patients than in CD34 negative group (4/12, 33.3%, P = 0.031), and was higher in patients with M2b, M3 and M4EO than in those with other AML subtypes (40.0% vs 80.0%, P = 0.019). The complete remission rate in C-mpl positive AML patients was lower than that in negative patients (70.0% vs 81.8%, P = 0.394), although the difference was not significant. In 9 of 28 cases of AML, the in vitro treatment with rhTpo induced proliferation of leukemia cells. Among these 28 patients, leukemic cells from 8 of 17 (47.0%) patients expressing C-mpl responded to rhTpo, but only the cells from one of 11 (9.1%) non-expressing patients did. The rhTpo induced proliferation of AML cells was enhanced when combined with IL-3, GM-CSF or SCF. CONCLUSIONS: C-mpl was expressed in some AML, but did not in ALL. Tpo could induce AML cells to proliferate and the effect was augmented when combined with other hematopoietic growth factors.

[Dose distribution in human body in external irradiation accidents].

OBJECTIVE: To study the dose distribution in human body and dose parameters under various exposure conditions. METHODS: In consideration of various factors, we calculated the dose distributions in human body by tissue-air ratio. Then dosimetric parameters were obtained. The calculation was verified by experiments using tissue equivalent anthropomophic phantom. RESULTS: The more homogeneous the dose distribution was, the stem cell dose was closer to the average dose. Stem cell dose could be larger than average dose. The difference between stem cell dose calculated and that by experiments was less than 5%. CONCLUSION: The calculation method and the program were reliable. The computer program has been applied in radiation accidents.

[Efficacy of transcatheter arterial infusion chemotherapy and transcatheter arterial embolization in 132 patients with primary hepatocellular carcinoma].

OBJECTIVE: To find out factors influencing efficacy of transcatheter arterial infusion chemotherapy (TAI) and transcatheter arterial embolization (TAE) for unresectable primary hepatocellular carcinoma. METHODS: From January 1993 to October 1997, 132 patients with unresectable primary hepatocellular carcinoma (HCC) received 597 intervention treatments. Of the 132 patients, 122 patients were given TAI plus TAE (92.4%), while the remaining 10 patients were given TAI alone (7.6%). RESULTS: The 1-, 2-, and 3-year overall survival rate was 81.8%, 36.4% and 18.2%, respectively. The clinical stage of the disease, the embolizing agents used and their dosage, formation of collateral circulation to the tumor and proper application of superselective hepatic arterial catheterization are factors affecting treatment efficacy. CONCLUSION: Adequately performed TAI and TAE can be routinely used for the treatment of unresectable liver cancer.

[The effect of combined endocrine therapy on prostate and testis].

OBJECTIVE: To study the pathological changes of the effect of combined endocrine therapy on normal prostate, benign prostatic hyperplasia, prostatic adenocarcinoma and testis. METHODS: 11 radical prostatectomy specimens, 3 core-needle biopsies of prostate and 3 testes obtained after at least 3 months of enatone-flutamide inhibition therapy were studied. Step-section was performed on radical specimens and average 16 sections per case were reviewed. PSA, PSAP and AE1/AE3 were immunostained on 14 cases of prostate cancer and a comparative study pre- and post-treatment was made. RESULTS: No residual tumor was recognized in 2 cases. Such characteristic changes were found in 9 cases as prominent acinar atrophy, decreased ratio of acini to stroma, stromal fibrosis, squamous metaplasia of carcinoma, cytoplasmic vaculation, nuclear shrinkage, and nucleolar shrinkage. No apparent change was discovered in 3 cases. Secretory epithelial atrophy and basal cell hyperplasia were the popular change in BPH and normal prostate. The expressions of both PSA and PSAP were markedly reduced in prostate carcinoma and nonneoplastic glands. Pathological downstaging of the tumor was not found statistically. Epithelial atrophy also existed in seminal vesicles and Leydig cells of the testes after hormone therapy. CONCLUSIONS: Combined endocrine therapy results in histologically distinctive changes that can be found in both nonneoplastic and neoplastic prostate tissue. However, the drugs can not eradicate prostate cancer completely. Testis atrophy is the direct action of the therapy.

[Early use of Chinese drug rhodiola compound for patients with post-trauma and inflammation in prevention of ALI/ARDS].

OBJECTIVE: To evaluate the effects of the Chinese drug rhodiola on the preventive treatment of acute lung injury (ALI) caused by post-traumatic/inflammatory and thoracic-cardiovascular operations. METHODS: 104 patients who coincide with their diagnosis and standard scale were divided randomly in to single number groups A(1), B(1), C(1) and D(1) (early preventive treatment group EG) and double number groups A(2), B(2), C(2) and D(2) (control group CG). The treatment of primary disease both in EG and CG was similar, but in EG four grams of rhodiola compound (RCO) were used every day for a period of 3-15 days since admission and 5-7 days after operation. The indicators related to ALI in each groups were measured. RESULTS: PaO(2), AaDO(2) and hemorheology were significant in EG than in CG (P < 0.05) or (P < 0.01). The complication rate of ARDS was decreased about of 50.0% in EG than in CG. CONCLUSIONS: Early use of RCO may protect against major risk factors of ALI/ARDS caused by massive trauma/infection and thoracic-cardio-operations.

[Clinicopathologic study of 11 patients with Paget's disease of scrotum].

OBJECTIVE: To probe the relationship of clinicopathologic presentations and prognosis of Paget's disease primarily involving the scrotum. METHODS: 11 cases of scrotal Paget's disease were studied retrospectively from 1984 to 1997. PAS was stained and EMA, S-100, HMB45, CEA and CA15-3 were immunostained in all cases for the differential diagnosis from Bowen's disease and malignant melanoma. RESULTS: Six of 11 cases involved hair follicle and/or sweat glands, and 3 cases involved dermis (one had metastasis of left superficial and deep inguinal lymph nodes, and left internal iliac lymph nodes). Paget's cells were positive for PAS (11/11), EMA (11/11), CEA (6/11), CA15-3 (11/11), and negative for S-100, HMB45. Wide local excision of the lesions was performed in 10 of 11 patients both for primary disease and recurrent disease. 3 of the 10 patients underwent lymph nodes resection and metastasis was found in 1 case pathologically. One patient accepted chemotherapy and died, from respiratory insufficiency after one month. The other 10 patients were followed up for 6 to 127 months and no patient died of Paget's disease. CONCLUSIONS: Involvement of hair follicles and sweat glands by Paget's cell does not influence the prognosis.However, invasion of dermis by Paget's cells may suggest possible metastasis.

[The experimental research of purification and character of cultured retinal ganglion cell].

OBJECTIVE: To establish a cell line and purification model of retinal ganglion cells (RGCs) in vitro. METHOD: RGCs from Sprague Dawley neonatal rats (postnatal 1 - 3 days) were cultured in basal medium eagle (BME) basal medium. The growth regularity of RGCs in vitro was observed under phase-contrast microscope. RGCs were purified by Thy 1.1 with FITC antibody and detected under fluorescent microscope and phase-contrast microscope. RESULTS: Higher density of retinal cells and tectal extract facilitate cultured RGCs to survive. The purification rate of retinal ganglion cells in the experiment arrived at 95 percent. CONCLUSION: Cytokine and trophic factors from other cells in the retina and tectal extract can promote RGCs to survive, and they can be purified by Thy 1.1 antibody.

[Studies on immunoassay for detecting fumonisin B1].

Direct competitive inhibition enzyme-linked immunosorbent assay for determination of fumonisin B1 (FB1) in corns were developed by using anti-FB1 monoclonal antibody. The minimum detectable concentration of FB1 was 10 ng/ml, and the standard curve was linear between 10 ng/ml and 5 micrograms/ml. The FB1 concentration of some corn samples from Xinjiang, Anhui, Hilongjiang provinces and Haerbin city were detected. The result imply that 34 out of 156 samples were positive for FB1. The concentrations were in the range of 0.18-31.32 micrograms/g. The average concentration was 12.04 micrograms/g.

[Analysis of measured results of transient-evoked otoacoustic emissions].

OBJECTIVE: In order to provide grounds for clinical diagnosis, the normal sensitive parameters of the transient evoked otoacoustic emissions (TEOAE) in normal ears were studied. METHOD: 60 ears with normal hearing subjects using ILO-88 otodynamic analyzer system were measured, the click stimuli level is 80 dB SPL and the stimuli pattern used non-linear model. RESULT: Analysis of the normal values of the stability, A-B difference, response amplitude, wave reproducibility, band reproducibility and band SNR, dominant frequencies were obtained. CONCLUSION: The findings from comprehensive analysis of spectrum, response amplitude and reproducibility can be used as main sensitive parameters to evaluated whether or not of normal-hearing.

[Study of the relation between MMP2, MMP9 and nasopharyngeal carcinoma].

OBJECTIVE: To evaluate the role of matrix metalloproteinase (MMP) in the invasion and metastasis of nasopharyngeal carcinoma (NPC). METHOD: Twenty-six cases of NPC were studied by immunohistochmical method using monocolonal antibody to MMP2 and MMP9. RESULT: It was showed that there were significantly correlation between MMP2 or MMP9 and the invasion and metastasis of NPC. The positive rate of MMP2 and MMP9 were higher in NPC with cervical lymph node involvement than that of no cervical lymph node involvement. And they were lower in well differentiated NPC than poorly differentiated NPC. CONCLUSION: The results indicate that MMP2 and MMP9 may play a critical role in assisting carcinoma cell to degrade basement membrane to invade surrounding tissues and to form metastatic colonies in lymph nodes.

[Fusion expression of porcine IFN-gamma and Cysticercus cellulosae antigen cC1 in Escherichia coli cells].

AIM: To construct an expression vector, including a chimeric cDNA of porcine IFN-gamma and Cysticercus cellulosae antigen cC1. METHODS: DNA fragments of porcine IFN-gamma and cC1 including linkers were generated by polymerase chain reaction (PCR). The recombinant vector pJLA-PRcC1 was constructed by inserting a chimeric cDNA of porcine IFN-gamma and Cysticercus cellulosae antigen cC1, and transformed to E. coli XL-Blue. RESULTS: The recombinant vector was identified by restriction analysis. An inserted fragment about 1.5 kb could be found. After induction, a 52 kDa new protein band appeared in SDS-PAGE. CONCLUSION: The fusion expression of porcine IFN-gamma and cC1 antigen in Escherichia coli cells is successful.
