[Experimental studies on the cancer chemoprevention of tea pigments].

A batch of short-term tests were used to examine the effects of tea pigments on three stages of carcinogenesis, i.e. initiation, promotion and progression. Forward gene mutation test and micronuclei test were used to study the initiation stage of carcinogenesis; metabolic cooperation test and mice ear test to study the promotion stage. Viability and growth ability of Hela cells in soft agar and S180 solid tumor test in mice were used to examine the effect of tea pigments on the third stage of carcinogenesis. The results showed that both tea pigments and tea polyphenols had significantly protective effects on initiation, promotion and progression stages in carcinogenesis. In vitro study showed that tea pigments and tea polyphenols could induce QR and GST activity in Hep G2 cells. Oral administration of 0.1% tea polyphenols and 0.1% tea pigments could increase GST activity in rat liver by 25% and 18% respectively, and this increase was accompanied by the significant increase of GST 1-1, 1-2 and 3-3 protein expression level in rat liver. Our results suggested that the anticancer effect of tea pigments was the same as that of tea polyphenols, and the anticancer properties of tea pigments might be mediated by activating the enzymes such as QR and GST, which play important roles in the detoxification and exclusion of carcinogen.

[Effect of long-term experimental stress on blood glucose level in mice with diabetes-prone: the age difference and its mechanism].

OBJECTIVE: To compare the effects of long-term experimental stressors on blood glucose level and insulin secretion in the aged and young mice with diabetes-prone and to explore the mechanism. METHODS: Diabetes-prone was induced by streptozotocin (STZ) in 40 1.5-month-old mice and 40 15-month-old mice. The young and the aged STZ mice were divided randomly into the stress group and the control group, respectively. Each group contained 20 mice. Both the aged and the young stress mice were exposed to multiple stressors (restrain, rotation, crowding) for 6 weeks. Fasting blood glucose (FBG), fasting plasma insulin (FINS), pancreas lipid peroxidase (LPO) and the metabolic products of nitric oxide (NO2-, NO3-) levels and superoxide dismutase (SOD) activity were measured. The grade of insulitis in STZ mice was also evaluated. RESULTS: After 6-week stress, FBG level, pancreatic tissue LPO and NO2-, NO3- contents were significantly higher while FINS level and SOD activity lower in the aged stress mice than those in the youngs. The insulitis malignization in the aged stress group was severer than that in the young stress group. There were correlativities among the above variables. CONCLUSION: The effects of long-term stress in promoting hyperglycemia exacerbating insulitis, and inhibiting insulin secretion are more significant in the aged STZ mice than in the young ones. This difference may result from the excessive NO and the peroxide injury to the pancreas.

cDNA sequence analysis and expression of four long neurotoxin homologues from Naja naja atra.

The novel cDNAs encoding four long neurotoxin homologues were firstly cloned from the venom gland of Naja naja atra by reverse transcriptase-polymerase chain reaction. Amino acid sequence comparison showed that they may come from two different evolutionary origins. The mature proteins were expressed as soluble fusion proteins in Escherichia coli and purified for immunoblotting. The results revealed that they displayed different immunochemical properties.

NMR studies of internal dynamics of serine proteinase protein inhibitors: Binding region mobilities of intact and reactive-site hydrolyzed Cucurbita maxima trypsin inhibitor (CMTI)-III of the squash family and comparison with those of counterparts of CMTI-V of the potato I family.

Serine proteinase protein inhibitors follow the standard mechanism of inhibition (Laskowski M Jr, Kato I, 1980, Annu Rev Biochem 49:593-626), whereby an enzyme-catalyzed equilibrium between intact (I) and reactive-site hydrolyzed inhibitor (I*) is reached. The hydrolysis constant, Khyd, is defined as [I*]/[I]. Here, we explore the role of internal dynamics in the resynthesis of the scissile bond by comparing the internal mobility data of intact and cleaved inhibitors belonging to two different families. The inhibitors studied are recombinant Cucurbita maxima trypsin inhibitor III (rCMTI-III; Mr 3 kDa) of the squash family and rCMTI-V (Mr approximately 7 kDa) of the potato I family. These two inhibitors have different binding loop-scaffold interactions and different Khyd values--2.4 (CMTI-III) and 9 (CMTI-V)--at 25 degrees C. The reactive-site peptide bond (P1-P1') is that between Arg5 and Ile6 in CMTI-III, and that between Lys44 and Asp45 in CMTI-V. The order parameters (S2) of backbone NHs of uniformly 15N-labeled rCMTI-III and rCMTI-III* were determined from measurements of 15N spin-lattice and spin-spin relaxation rates, and [1H]-15N steady-state heteronuclear Overhauser effects, using the model-free formalism, and compared with the data reported previously for rCMTI-V and rCMTI-V*. The backbones of rCMTI-III [(S2) = 0.71] and rCMTI-III* [(S2) = 0.63] are more flexible than those of rCMTI-V [(S2) = 0.83] and rCMTI-V* [(S2) = 0.85]. The binding loop residues, P4-P1, in the two proteins show the following average order parameters: 0.57 (rCMTI-III) and 0.44 (rCMTI-III*); 0.70 (rCMTI-V) and 0.40 (rCMTI-V*). The P1'-P4' residues, on the other hand, are associated with (S2) values of 0.56 (rCMTI-III) and 0.47 (rCMTI-III*); and 0.73 (rCMTI-V) and 0.83 (rCMTI-V*). The newly formed C-terminal (Pn residues) gains a smaller magnitude of flexibility in rCMTI-III* due to the Cys3-Cys20 crosslink. In contrast, the newly formed N-terminal (Pn' residues) becomes more flexible only in rCMTI-III*, most likely due to lack of an interaction between the P1' residue and the scaffold in rCMTI-III. Thus, diminished flexibility gain of the Pn residues and, surprisingly, increased flexibility of the Pn' residues seem to facilitate the resynthesis of the P1-P1' bond, leading to a lower Khyd value.

Studies on the mechanism of the selenite-induced decrease in cell attachment: effect of selenite on the levels of fibronectin receptor (alpha5beta1 integrin) mRNAs.

We previously reported that exposure of HeLa cells to selenite for 2 h results in a decrease in their ability to attach to fibronectin (Yan and Frenkel, Cancer Res. 52, 5803-5807 [1992]), as well as a decrease in the level of fibronectin receptor (alpha5beta1 integrin) at the cell surface (Yan and Frenkel, Biol. Trace Element Res. 46, 79-89 [1994]). We have now found that after exposure to selenite, there was a decrease in the total cellular content of the receptor protein, as well as in the level of the mRNAs for both of the subunits. Exposure of cells to actinomycin D (an inhibitor of RNA synthesis) also resulted in a decrease in the level of these mRNAs, suggesting that the effect of selenite is the result of its known inhibitory effect on RNA synthesis (Frenkel, Toxicol. Lett. 25, 219-223 [1985]). Exposure of cells to actinomycin D for 2 h also resulted in a decrease in the ability of cells to attach to fibronectin. Furthermore, both selenite and actinomycin D caused a decrease in integrin mRNA levels and in cell attachment to fibronectin only when high-density cells were exposed to the agents. In contrast, when low-density cells were exposed, neither agent had any detectable effect on mRNA levels or on cell attachment. These results have suggested the following scheme for the mechanism of the inhibition of cell attachment by selenite: After exposure to selenite for 2 h, there is a significant inhibition of cellular RNA synthesis, which results in a general decrease in the cellular level of those mRNAs with relatively short half-lives, including in particular those of the fibronectin receptor. This leads to a decrease in the intracellular level of the receptor protein and, consequently, in its level at the cell surface, which in turn causes a decrease in the rate of cell attachment to fibronectin.

Determining the degree of chaos from analysis of ISI time series in the nervous system: a comparison between correlation dimension and nonlinear forecasting methods.

Two different chaotic time series analysis methods--the correlation dimension and nonlinear forecasting--are introduced and then used to process the interspike intervals (ISI) of the action potential trains propagated along a single nerve fiber of the anesthetized rat. From the results, the conclusion is drawn that compared with the correlation dimension, nonlinear forecasting is more efficient and robust for chaotic ISI time series analysis in a noisy environment. Moreover, the evolution of the correlation coefficient curves calculated from nonlinear forecasting can qualitatively give a better reflection of the unpredictability of the system's future behavior and is in good agreement with the values of the largest Lyapunov exponent that quantitatively measures the degree of chaos.

The replicative intermediate molecule of bovine viral diarrhoea virus contains multiple nascent strands.

The replication of bovine viral diarrhoea virus (BVDV) RNA is considered to involve replicative intermediates (RI) and replicative forms (RF) of virus RNA. These RNA species were radiolabelled metabolically by 3H-uridine in BVDV-infected cells and separated by sucrose gradient centrifugation. RNA in the fractions was then digested with RNase A in high salt (2 x SSC) and the RNase-resistance was determined. Using the Baltimore formula, this led to the calculation that the RI of BVDV contained 6-7 nascent strands on the template. This suggests that the structure of the BVDV RI is similar to that of poliovirus and dengue 2 virus.

Use of proliferating cell nuclear antigen as a marker of liver regeneration after partial hepatectomy in rats.

In order to document and compare proliferating cell nuclear antigen (PCNA) mRNA and protein levels with more traditional parameters of hepatic regenerative activity in a rat model, adult male Sprague-Dawley rats (4 to 6 per group) were killed at various times up to 96 hours after 70% partial hepatectomy. At each time interval, tissue PCNA mRNA abundance and protein levels were documented (by Northern and Western blot analysis, respectively) and compared with the results of PCNA immunostaining and 3H-thymidine incorporation into hepatic DNA. Tissue PCNA protein levels were also documented in additional groups of rats 12, 24, 36, and 48 hours after sham or 30% partial hepatectomy. PCNA mRNA expression after partial hepatectomy was variable: almost undetectable at 24 hours, levels returned to baseline at 36 hours, then fell again to low levels at 96 hours. PCNA protein levels remained stable for 36 hours, increased to fourfold above baseline (p < 0.01) at 48 hours, then remained elevated for the duration of the 96-hour study. Changes in PCNA by immunostaining were similar but tended to occur somewhat earlier (significant increases being detectable at 24 hours), whereas 3H-thymidine incorporation detected the earliest increases in DNA synthesis at 12 hours and peaked at 36 hours. Peak PCNA protein levels correlated with the extent (0%, 30%, or 70%) of hepatic resection. The results indicate that PCNA protein level as determined by Western blot analysis, but not PCNA mRNA expression, correlates with PCNA immunostaining and 3H-thymidine incorporation in the regenerating liver. These findings support the use of PCNA protein determinations as an additional quantitative measure of hepatic regenerative activity after partial hepatectomy in rats.

A stable cell line with a proportion of cells persistently infected with bovine viral diarrhoea virus.

Bovine turbinate (BTu) and lamb testis (LT) cell lines persistently infected with bovine viral diarrhoea virus (BVDV) arose as a result of a single change of medium containing commercial foetal calf serum. Infected cells comprise 30% and 50% respectively, of the total cell population, determined by immunohistochemical staining. The ratio of positive cells has remained unchanged during successive passages. Characterization of the persistently-infected BTu cells (BTuI) showed that only full length viral RNA was detected by northern blot hybridisation, indicating that DI particles were not involved. Secreted and intracellular virus from these cells was fully infectious for fresh BTu and LT cells. The BTuI cell line was fully permissive for a cytopathic BVDV isolate and a bovine herpesvirus, but non-permissive for two non-cytopathic BVDV isolates. Attempts to induce the permissive state in the BVDV-negative cells of the BTuI culture by treatment with actinomycin D and 5'-aza-cytidine failed. These cells provide a convenient model to study aspects of BVDV pathogenesis and replication.

cDNA cloning and sequence analysis of six neurotoxin-like proteins from Chinese continental banded krait.

The cDNAs encoding six neurotoxin-like proteins were cloned from the total RNA isolated from one venom gland of Bungarus multicinctus multicinctus by reverse transcription-polymerase chain reaction. Although they shared a high degree of nucleotide sequence homology, the deduced proteins displayed a rather low extent of identical residues with the exception that the 21 residue signal peptides were highly conserved. Each mature protein consisted of 65, 66 or 68 amino acids with four or five disulfide bonds respectively. Comparative sequence analyses showed that they were structurally related to neurotoxin homologues. Phylogenetic analyses implied that they might represent two different evolutionary branches.

The duck hepatitis B virus polymerase is activated by its RNA packaging signal, epsilon.

The epsilon stem-loop at the 5' end of the pregenomic RNA of the hepatitis B viruses is both the primary element of the RNA packaging signal and the origin of reverse transcription. We have previously presented evidence for a third essential role for epsilon, that of an essential cofactor in the maturation of the viral polymerase (J. E. Tavis and D. Ganem, J. Virol. 70:5741-5750, 1996). In this case, binding of epsilon to the polymerase is proposed to induce a physical alteration to the polymerase that is needed for it to develop enzymatic activity. Three lines of evidence employing duck hepatitis B virus supporting this hypothesis are presented here. First, an unusual DNA polymerase activity employing exogenous RNAs (the trans reaction) that was originally discovered with recombinant duck hepatitis B virus polymerase expressed in Saccharomyces cerevisiae yeasts was shown to be an authentic property of the viral polymerase. The trans reaction was found to be template-dependent reverse transcription of the exogenous RNA. The trans reaction occurred independently of the hepadnavirus protein-priming mechanism, yet it was still strongly stimulated by epsilon. This directly demonstrates a role for epsilon in activation of the polymerase. Second, the reverse transcriptase domain of the polymerase was shown to be physically altered following binding to epsilon, as would be expected if the alteration was required for maturation of the polymerase to an enzymatically active form. Finally, analysis of 15 mutations throughout the duck hepatitis B virus polymerase demonstrated that the epsilon-dependent alteration to the polymerase was a prerequisite for DNA priming, reverse transcription, and the trans reaction.

[Protective effect of tea on immune function in mice].

OBJECTIVE: To study the mechanism of preventive effect of tea on cancer by immune regulation. METHODS: A tumor model was induced in mice using carcinogen, 4-methyl-nitrosoamino-1-(3-pyridyl)-1-butanone (NNK), to examine their changes in immune function and the effects of green tea, mixed tea and polyphenol on protection from tumor. RESULTS: During the four weeks of observation after injection of NNK into mice, their immunological indicators, such as cytophagocytosis of macrophage in the abdominal cavity, chemiluminescence of peripheral leukocyte, delayed allergic reaction, count of antibody-forming spleen cells and activity of spleen nature killer cells, etc. increased or decreased to various extent, as compared with those in normal controls. It was found that whether green tea, mixed tea or polyphenol all showed significant protection from adverse changes in immune functions. CONCLUSION: Tea and its components had significant protection from early adverse changes in immune function in tumorgenesis induced by NNK.

Construction of engineering host by non-random method in Saccharomyces cerevisiae.

A yeast engineering host, GJ30, was constructed by a non-random method. GJ30 derivated from wild type yeast have leu2- and ura3- deficients which can be used as selection markers. The PCR analysis showed that the homologous recombinations occurred in the leu2 and ura3 locus of GJ30, respectively. The biological properties (cellular growth density, stability and efficiency for gene expression) of GJ30 were compared with other several yeast stains. The results showed that GJ30 is a suitable host strain for expression of foreign genes.

[Ultrastructure changes of liver autolysis].

Ultra-structure changes of autolysis of hepatocyte were studied using transmission electron microscope(TEM). It was found that autolysis started 0.5 hour after death, with the heterochromatin of nuclei shrinking abnormally and smooth endoplasmic reticulum expanding. The changes developed to distinction after 1 h, and progressed continuously after 2 h, 3 h, 4 h and 5 h. It was found that (1) the mitochondria were swelling, its matrix density decreasing and flocculent dense bodies (FDB) emerging; (2) the rough endoplasmic reticulum and perinuclear cistern were expanding. It was suggested that ultra-structure changes of the autolysis be correlated to the postmortem time.
