Fusimotor-free spindles in reinnervated muscles of neonatal rats treated with nerve growth factor.

Crushing the nerve to the medial gastrocnemius muscle in newborn rats and administering nerve growth factor afterwards results in a reinnervated muscle containing supernumerary muscle spindles. The structure and innervation of 88 spindles in the reinnervated muscles were reconstructed from serial thick and thin transverse sections at 30-35 days after the nerve crush, and compared to those of five control spindles. The spindles consisted of one to four small-diameter encapsulated fibers with features of nuclear chain intrafusal fibers, or infrequently a nuclear bag intrafusal fiber. Some of the spindles were located within a capsule that also contained an extrafusal fiber. Each spindle was innervated by an afferent with features of the primary afferent. The density of secondary afferents was lower in reinnervated muscles than in controls. Endplates were observed on extrafusal fibers in the experimental muscles, attesting to restoration of skeletomotor (alpha) innervation after the nerve crush. However, 78% of the experimental spindles were entirely devoid of efferent innervation. The remainder received either one or two fusimotor (gamma) axons or a skeletofusimotor (beta) axon, compared to the six to eight motor axons that innervated control spindles. The presence of supernumerary spindles composed of fibers that resemble normal intrafusal fibers in the absence of motor innervation suggests that afferents alone can induce the formation and subsequent differentiation of intrafusal fibers in nerve-crushed muscles of neonatal rats. In addition, the paucity of gamma innervation in nerve-crushed muscles suggests that immature gamma neurons are more susceptible than spindle afferents or alpha efferents to cell death after axotomy at birth.

Influence of muscle cell substrates on differentiation of intrafusal fiber types in neonatal rats.

Rat muscle spindles contain one nuclear bag2, one nuclear bag1, and two nuclear chain fibers. The three different types of intrafusal fiber in spindles may be a reflection of concomitant changes in proportions of slow primary, slow/fast secondary, and fast secondary myotubes during the period of spindle development. We examined whether experimentally altering the available muscle substrates would impact the intrafusal fiber type composition of spindles. De novo formation of spindles in muscles devoid of primary myotubes was induced by crushing the nerve to the medial gastrocnemius muscle in newborn rats and administering nerve growth factor for ten days afterwards. Encapsulated fibers of the reinnervated muscles examined one month after nerve crush had myofibrillar adenosine triphosphatase and myosin heavy chain profiles similar to normal bag2, bag1, or chain intrafusal fibers. However, spindles in reinnervated muscles contained fewer fibers than controls. Most experimental spindles contained chain and/or bag1 fibers, the two fiber types which ordinarily arise during secondary myogenesis. In contrast, bag2 fibers, fibers that normally form concomitant with primary myogenesis, were absent from nearly 90% of spindles in reinnervated muscles. The paucity of bag2 fibers may reflect the absence of primary myotubes, whereas the prevalence of chain and/or bag1 fibers may reflect that secondary myotubes or myofibers that descended from the secondary myotubes were the principal muscle substrates available for spindle formation in the nerve-crushed muscles.(ABSTRACT TRUNCATED AT 250 WORDS)

[11C] beta-CIT, a cocaine analogue. Preparation, autoradiography and preliminary PET investigations.

beta-CIT (2 beta-carbomethoxy-3 beta-(4-iodophenyl)tropane) is a cocaine analogue with a high affinity for the dopamine transporter. [11C] beta-CIT was prepared by N-methylation of nor-beta-CIT with [11C]methyl iodide. The total radiochemical yield of [11C] beta-CIT was 40-50% with an overall synthesis time of 35-40 min. The radiochemical purity was > 99% and the specific radioactivity at the time of injection was about 1000 Ci/mmol (37 GBq/mumol). Autoradiographic examination of [11C] beta-CIT binding in human brains post-mortem demonstrated a high level of specific binding in the striatum. PET examination of [11C] beta-CIT in a Cynomolgus monkey showed a marked accumulation of radioactivity in the striatum. The ratio of radioactivity in the striatum-to-cerebellum approached 5 after 87 min. In a displacement experiment, radioactivity in the striatum but not in the cerebellum, was markedly reduced after injection of unlabelled cocaine. [11C] beta-CIT has a potential as ligand for PET examination of cocaine effects in man.

Immunohistochemical analysis of E-cadherin expression in human colorectal tumours.

The homotypic homophilic cell-cell adhesion molecule E-cadherin is crucial in the organization and maintenance of most epithelia. The expression of E-cadherin was studied immunohistochemically in various human colorectal tumours. Therefore we stained 1 tubular adenoma with low grade dysplasia, 18 adenocarcinomas with different histologic degrees of differentiation and invasion, and 1 metastasis using a modified peroxidase-anti-peroxidase technique. In the adenoma as well as in all well differentiated adenocarcinomas we found E-cadherin immunopositivity at the cell membrane of almost all cancer cells. The immunopositivity of E-cadherin was clearly weaker and sometimes even absent in isolated neoplastic cells and glands of less differentiated adenocarcinomas. The moderately differentiated adenocarcinomas showed an intermediate staining pattern. These findings are in line with experimental evidence that downregulation of E-cadherin favours invasion, eventually leading to metastasis.

Attenuation of contractions to acetylcholine in canine bronchi by an endogenous nitric oxide-like substance.

1. The involvement was assessed of an endogenous nitric oxide-like substance in contractions of canine bronchi to acetylcholine. 2. Canine third order bronchial rings, in some of which the epithelium was removed mechanically, were suspended in organ chambers and isometric tension was recorded. In some experiments, the content of guanosine 3',5'-cyclic monophosphate (cyclic GMP) of the bronchi was also measured. 3. Acetylcholine induced concentration-dependent contractions. The contractions were potentiated by nitro-L-arginine (an inhibitor of the synthesis of nitric oxide), oxyhaemoglobin (a scavenger of nitric oxide), and methylene blue (an inhibitor of soluble guanylate cyclase). The magnitude of the potentiation to acetylcholine-induced contractions by these inhibitors were not significantly different between tissues with and without epithelium. 4. Acetylcholine induced a concentration-dependent increase in intracellular content of cyclic GMP, which was similar in bronchi with and without epithelium. These increases were abolished by nitro-L-arginine and methylene blue. 5. During contractions to acetylcholine, exogenous nitric oxide relaxed the canine bronchi. The relaxations were not affected by nitro-L-arginine, but were augmented by superoxide dismutase plus catalase, and were abolished by methylene blue. 6. These observations suggest that, during contraction evoked by acetylcholine, the production of an endogenous nitric oxide-like substance increases and in turn attenuates the response of the airways to the muscarinic agonist. However, the endogenous nitric oxide-like substance does not play a major role in the epithelium-dependent attenuation of the contraction to acetylcholine in canine bronchi.

Complex nature of the major viral polyadenylated transcripts in Epstein-Barr virus-associated tumors.

The most abundant polyadenylated viral transcripts in the Epstein-Barr virus (EBV)-associated tumor nasopharyngeal carcinoma are a family (apparent sizes, 4.8, 5.2, 6.2, and 7.0 kb) of highly spliced cytoplasmic RNAs expressed from the BamHI-I and -A regions of the viral genome in an antisense direction with respect to several viral lytic functions encoded within the same region and concerned with the lytic cycle of the virus. We have called these complementary-strand transcripts. They are also expressed in B cells, including Burkitt's lymphoma and EBV-immortalized marmoset cell lines, and tumors generated in cottontop tamarins in response to EBV infection, but at a lower level. The complete structure of the major 4.8-kb RNAs (seven or eight exons) was determined in this study; the larger, but related, transcripts appear to be produced by differential splicing. The transcriptional promoter for the major complementary-strand transcripts, located in BamHI-I, contains several well-characterized transcriptional control elements (E2A, SP1, and AP1) and is functionally active in both B lymphocytes and epithelial cells. It appears to be a bifunctional viral promoter, as it also contains the initiation codon for a gene (BILF2) that encodes a glycoprotein that is expressed off the other strand. Splicing events create a number of small AUG-initiated open reading frames, one of which has homology to functionally significant regions of the EBV-encoded nuclear antigen 2 and to E2 (in papillomavirus). The complex nature of these transcripts and their potential role in the virus association with malignancy are considered.

Two cofactors and cytoplasmic chaperonin are required for the folding of alpha- and beta-tubulin.

Though the chaperonins that mediate folding in prokaryotes, mitochondria, and chloroplasts have been relatively well characterized, the folding of proteins in the eukaryotic cytosol is much less well understood. We recently identified a cytoplasmic chaperonin as an 800-kDa multisubunit toroid which forms a binary complex with unfolded actin; the correctly folded polypeptide is released upon incubation with Mg-ATP (Y. Gao, J. O. Thomas, R. L. Chow, G.-H. Lee, and N. J. Cowan, Cell 69:1043-1050, 1992). Here we show that the same chaperonin also forms a binary complex with unfolded alpha- or beta-tubulin; however, there is no detectable release of the correctly folded product, irrespective of the concentration of added Mg-ATP and Mg-GTP or the presence of added carrier tubulin heterodimers with which newly folded alpha- or beta-tubulin polypeptides might exchange. Rather, two additional protein cofactors are required for the generation of properly folded alpha- or beta-tubulin, which is then competent for exchange into preexisting alpha/beta-tubulin heterodimers. We show that actin and tubulins compete efficiently with one another for association with cytoplasmic chaperonin complexes. These data imply that actin and alpha- and beta-tubulin interact with the same site(s) on chaperonin complexes.

Vasopressin depolymerizes apical F-actin in rat inner medullary collecting duct.

In amphibian bladder, arginine vasopressin (AVP) depolymerizes F-actin in the apical region of the granular cell, promoting fusion of water channel-carrying vesicles with the apical membrane. We now report the effect of AVP on F-actin in the mid- and terminal segments of rat inner medullary collecting duct (IMCD2 and IMCD3). In IMCD3, 5 min of stimulation by 2.5-250 nM AVP significantly depolymerized F-actin by 13-24% in whole cell assays employing the rhodamine-phalloidin binding technique. The IMCD2 was more sensitive, responding to subnanomolar (0.25 nM) AVP with 6 +/- 2% depolymerization. Depolymerization occurred as early as 2 min after 2.5 and 25 nM but not 250 nM AVP. 8-Bromoadenosine 3',5'-cyclic monophosphate depolymerized F-actin in IMCD3 at both 2 and 5 min. Immunogold labeling of the apical actin pool in IMCD3 principal cells was reduced by 26 +/- 5% (P < 0.05) by 2.5 nM AVP; the lateral and basal pools showed no significant changes. Capillary endothelial, thin limb of Henle, and intercalated cells showed no changes in immunogold labeling after AVP. Thus reorganization of the apical actin network by AVP is a consistent finding in both mammalian and amphibian target cells.

Products of cyclooxygenase mediate the responses of the guinea pig trachea to hydrogen peroxide.

The role of products of cyclooxygenase was investigated in the responses of isolated airways to H2O2. Strips of guinea pig trachea, in some of which the epithelium had been removed mechanically, were suspended in organ chambers, and isometric tension was recorded. Under basal conditions, H2O2 induced indomethacin-sensitive contractions, which were larger in preparations without than in those with epithelium; the difference was abolished by inhibitors of thromboxane synthase or thromboxane A2 receptors. During contractions to acetylcholine, low concentrations of H2O2 induced relaxation in preparations with but had no significant effect in those without epithelium. At higher concentrations of H2O2, the epithelium-dependent relaxation was attenuated but an epithelium-independent relaxation appeared. The epithelium-dependent but not the epithelium-independent responses to H2O2 were blocked by indomethacin. Under basal conditions, prostaglandin E2 (PGE2; < or = 10(-7) M), U-46619, prostaglandin PGF2 alpha (PGF2 alpha), prostaglandin PGD2 (PGD2), and prostacyclin (PGI2) caused contractions. During contractions to acetylcholine, PGE2 induced larger relaxations in preparations with than in those without epithelium. Radioimmunoassay revealed that lower concentrations of H2O2 predominantly increased the release of PGE2 and 6-ketoprostaglandin F1 alpha (6-keto-PGF1 alpha); in preparations without epithelium, the release of thromboxane B2 was augmented also. At higher concentrations of H2O2, the release of PGE2, PGF2 alpha, PGD2, 6-keto-PGF1 alpha, and thromboxane B2 increased in preparations with and without epithelium. These findings demonstrate that the responses of the guinea pig trachea to H2O2 are mediated mainly by products of cyclooxygenase and that the effects of H2O2 are modulated by the epithelium.

Respiratory epithelium modulates the responses of canine bronchi to cooling.

The present study was designed to investigate the effect of cooling on the modulatory role of the respiratory epithelium on the underlying smooth muscle. Canine bronchial rings and segments (with or without epithelium) were suspended in organ chambers and perfused with modified Krebs-Ringer bicarbonate solution, respectively. Isometric force was recorded. Cooling did not affect the resting tension of the bronchi. During contractions to carbachol, cooling evoked contractions in bronchi with epithelium but relaxations in those without epithelium. In the presence of indomethacin, cooling induced contractions in both preparations with and without epithelium. The contractions in bronchi with epithelium were significantly larger than those in bronchi without epithelium. After treatment with indomethacin, exogenous arachidonic acid potentiated the cooling-induced contractions in preparations with epithelium but not in those without epithelium. This potentiation was not affected by nordihydroguaiaretic acid. SKF 525-A and metyrapone, inhibitors of cytochrome P-450 monooxygenases, converted the cooling-induced contractions of preparations with epithelium to relaxations and had no significant effects on the responses of preparations without epithelium. These observations suggest that cooling induces from the epithelium the release of a cytochrome P-450-derived eicosanoid that potentiates contractions of the underlying airway smooth muscle to carbachol.

Responsiveness of the guinea pig trachea to stretch: role of the epithelium and cyclooxygenase products.

The role of the epithelium and cyclooxygenase products was investigated in the responses of isolated airways to sudden stretch. Strips of guinea pig trachea, in some of which the epithelium had been removed mechanically, were suspended in organ chambers; isometric tension was recorded. After rapid stretching to their optimal tension, the preparations (with and without epithelium) relaxed initially and then contracted to a level close to the imposed tension. Afterward, tissues with epithelium maintained this level of tension, but those without epithelium relaxed. After treatment with papaverine or isoproterenol (at concentrations causing maximal relaxation), stretch was followed only by a decrease in tension; a similar response to stretch was also obtained in tissues treated with indomethacin or acetylsalicylic acid (inhibitors of cyclooxygenase). Dazmegrel (an inhibitor of thromboxane synthase) and SQ-29548 (an antagonist of prostaglandin H2 or thromboxane A2 receptors) did not affect the response of tissues with epithelium but abolished the stretch-induced contraction in those without epithelium. Tranylcypromine, which inhibits prostacyclin synthase, and tetrodotoxin, which blocks local reflexes, did not significantly affect the responses of the tissues to stretch. These observations suggest that thromboxane may mediate the epithelium-independent contraction and that another product of cyclooxygenase contributes to the maintenance of tension on stretching observed in tissues with epithelium.

The effects of chemotherapy including cisplatin on vitamin D metabolism.

Plasma concentrations of three major vitamin D3 metabolites, 25-hydroxyvitamin D3 (25-OHD3), 24R, 25-dihydroxyvitamin D3 (24,25-(OH)2D3), and 1 alpha, 25-dihydroxyvitamin D3 (1,25-(OH)2D3), were determined serially in patients with gynecological malignancies treated by chemotherapy which included cisplatin. While 25-OHD3 and 24,25-(OH)2D3 levels did not change regularly, 1,25-(OH)2D3 decreased significantly (50%) after only one or two courses of the treatment in all cases. In concert with declining 1,25-(OH)2D3, the PTH levels increased two or three times. The results were explained by the impairment by cisplatin of mitochondrial function in proximal tubules in the kidney.

Use of an oestrogen, androgen or gestagen as a potential chemosterilant for control of rat and mouse populations.

Three synthetic steroids were evaluated as potential chemosterilants for rodent control. Ethinyl oestradiol, methyl testosterone or Org 5933, a synthetic gestagen, were incorporated into paraffin blocks containing cereal grains and offered to laboratory rats and mice in addition to their standard laboratory diet. Ethinyl oestradiol (50 mg kg-1 paraffin block) was highly unpalatable to female rats, and the amount of steroid ingested was not sufficient to interfere with their oestrous cycles or inhibit ovulation. Methyl testosterone (5000 mg kg-1 paraffin block), although not as palatable as untreated blocks, was effective in inducing almost immediate infertility in female rats and mice at an ingested dose of about 35 micrograms g-1 body weight day-1. This infertility persisted throughout the duration of treatment, and lasted for several weeks after the cessation of treatment. Male rats became infertile after 3 months of treatment owing to suppression of spermatogenesis. Female rats developed a specific aversion to methyl testosterone when they were pregnant or lactating; it was therefore not possible to masculinize the brains of their female offspring. In mice, the androgen treatment induced high levels of aggression in the females so that they fought with males and with one another. One female died of her wounds. Org 5933 (4 mg kg-1 paraffin block) was highly palatable to female rats and mice, and at doses of about 420 ng g-1 body weight day-1 was effective in inhibiting ovulation in rats within 3 to 4 days after the start of treatment. This infertility persisted throughout the duration of treatment, and the animals conceived within 5 days of cessation of treatment. A dose of about 930 ng g-1 body weight day-1 was not completely effective in inhibiting ovulation in mice, but females that became pregnant during treatment gave birth to dead young. When the gestagen was given to female rats and mice in the last few days of pregnancy, the duration of gestation was significantly prolonged, and most young were born dead; some of the females also died in labour. The gestagen did not appear to inhibit lactogenesis, since the few animals that gave birth to live young reared them normally for the first 5 days of life. These results show that either methyl testosterone or Org 5933 in paraffin blocks could perhaps be used as a chemosterilant for the control of rat and mouse populations.(ABSTRACT TRUNCATED AT 400 WORDS)

The sydnonimine C87-3754 evokes endothelium-independent relaxations and prevents endothelium-dependent contractions in blood vessels of the dog.

Experiments were designed to compare the relaxing activities of the new sydnonimine C87-3754 with SIN-1 in arteries and veins of the dog, and to determine whether C87-3754 can prevent endothelium-dependent contractions. Rings of coronary and femoral arteries, and saphenous veins were suspended in organ chambers for the measurement of changes in isometric tension. SIN-1 and C87-3754 evoked concentration-dependent relaxations in all rings of blood vessels contracted with a submaximal concentration of either prostaglandin F2 alpha, endothelin-1, phenylephrine, or norepinephrine. In both arteries and veins, the concentration-relaxation curves to C87-3754 were shifted significantly to the right (by two to three logarithmic units) of that to SIN-1. The presence of endothelium significantly inhibited the relaxations to SIN-1 but did not affect those to C87-3754. The treatment of coronary arteries with methylene blue or oxyhemoglobin significantly impaired the relaxation to SIN-1 and C87-3754. Neither C87-3754 nor its prodrug pirsidomine (CAS 936) affected the membrane potential in coronary arteries. The endothelium-dependent contractions evoked by nitro L-arginine, arachidonic acid, and the calcium ionophore A23187 in basilar arteries of the dog were inhibited by C87-3754. These results indicate that the sydnonimine C87-3754 is a dilator of both arterial and venous smooth muscle, and can prevent endothelium-mediated contractions in cerebral arteries of the dog. The inhibition of vascular tone is likely to involve the activation of soluble guanylate cyclase, causing enhanced production of cyclic guanosine monophosphate in the smooth muscle without a change in membrane potential.

Amplification and analysis of a male-specific SRY homologous sequence of cattle.

Based on the highly conserved region of human, rabbit and mouse SRY (sex-determining region Y) sequences, primers were designed for the amplification of the unknown homologous sequence of cattle by polymerase chain reaction (PCR). A 200-bp male-specific amplifying fragment was obtained. The fragment shares high homology with the known SRY gene. By using the cloned fragment as a probe to hybridize to the Southern blot of HindIII-digested cattle genomic DNA, a 1.7-kb male-specific fragment was detected, which is considered to contain the 200-bp fragment. When the same pair of primers was used in amplification of human and goat DNA samples, the 200-bp male-specific fragment could be detected.

[Cell proliferation in oral candidal leukoplakia].

Epithelial thickness, mitotic activity and DNA synthesis in oral candidal leukoplakia (chronic hyperplastic candidosis) were studied. The results showed that the epithelium was thicker, the number of mitosis and DNA synthesis cells were more in candidal leukoplakia than in non-candidal leukoplakia. These findings indicate an active cell proliferation exists in candidal leukoplakia which may be a precancerous lesion.
