[beta-Endorphin enhances IL-2 and IFN-gamma gene expression in human blood mononuclear cells].

The effect of beta-endorphin (beta-END) stimulation of cytokines production by PHA-induced human peripheral blood mononuclear cells (PBMC) was studied by reverse transcription in combination with polymerase chain reaction (RT/PCR), slot blotting and hybridization to detect and measure messenger RNA (mRNA). beta-END significantly and dose dependently enhanced IFN-gamma and IL-2 mRNA expression of T cells and naloxone (Nal) reversed the effect of beta-END on IL-2 mRNA expression. The results further demonstrate the important link between the neural system and the immune system.

[IL-2, IFN-gamma, and TNF-alpha mRNA expression in peripheral blood mononuclear cells in patients with multiple sclerosis].

We determined the cytokine messenger RNA (mRNA) expression patterns of peripheral blood mononuclear cells from 17 patients with multiple sclerosis (MS) and 11 normal controls, by using a quantitative reverse transcription in combination with polymerase reaction (RT/PCR) and Southern blot hybridization method. A significantly higher levels of IL-2, IFN-gamma mRNA in patient with MS compared to normal controls and significantly higher level of expressions of IFN-gamma, TNF-alpha mRNA were detected in active MS patients compared to those with stable desease (P < 0.05). These data further support the central role of some cytokines in the pathophysiology of this autoimmune disease.

[Clinical observations on syringomyelia treated by syrinx-peritoneal shunting with "T" tube].

We 22 cases of syringomyelia were verified by MR and treated by syrinx-peritoneal shunting with "T" tube. After the operation, the patients were reexamined by MR and B ultrasound and confirmed that their shunting was functional. Symptoms of the patients disappeared. But four patients got slight numbness in one leg or at the saddle area. One patient suffered from a difficult movement in one hand. With the exception of 6 patients, 16 showed that both of the newly and original symptoms improved significantly. Follow up for 2 years with regular MR and B ultrasound reexaminations was made. The patients resumed their work and study. MR and B ultrasound reexaminations showed that all of syrinxes collapsed significantly as the same as that after the operation immediately. These mean that this kind of operation is useful and effective for most syringomeylias.

[Expression of transforming growth factor beta 1 in keratinocytes of oral submucous fibrosis tissue].

To explore significance of transforming growth factor beta 1 (TGF beta 1) in the pathogenesis of oral submucous fibrosis (OSF), TGF beta 1 mRNAs in keratinocytes of the paraffin embeded tissues of 25 OSF cases, 5 normals (NOR) and 10 oral lichen planus (OLP) were determined by the in situ hybridization technique. The result showed that there was an expression of TGF beta 1 mRNA in keratinocytes of 15 OSFs (60%). The positive expression of TGF beta 1 mRNA was mainly in kerationcytes of early and middle stage OSF. There was no expression in that of 5 NORs and 10 OLPs. The study suggests that keratinocytes of OSF tissue may synthesize and release TGF beta 1 which may play an important role in the pathogenesis of OSF and participate as a mediator in the pathogenetic process of OSF.

[Clear cell odontogenic carcinoma].

Histopathologic and immunohistochemical features of 3 cases of clear cell odontogenic carcinoma were studied. The tumors were composed of sheets or islands of clear cells separated by mature collageneous tissue. Basaloid cells were also seen in the tumors. Tumor cells showed positive reactions for epithelial membrane antigen and cytokeratin. More PCNA positive cells were seen in basaloid cells than in clear cells. Metastasis to regional lymph nodes was found in all 3 cases.

[An analysis of causes of death in 329 emergency cancer patients].

OBJECTIVE: To analyze the causes of death in cancer patients. METHODS: In collection of 2,167 cancer cases coming to the emergency clinic, the causes of death were analyzed in 329 cases. RESULTS: Of the 329 cases, 25.8% (85/329) was due to the cancer growth itself while 59.9% (197/329) was resulted from complications. Thirty-seven cases (11.2%) died within 12 h in the emergency clinic. Most of the deaths were seen in patients with cancer of the lung, stomach and liver. Infection, respiratory failure, shock, hemorrhage and cardiac attack were the causes of death. The first two causes accounted for 36.2% and 32.5% of the total deaths in this series, respectively. CONCLUSION: Complications are the major causees of death. The importance of their proper prevention needs to be emphysized.

[Metallothionein expression and its significance in salivary gland tumors].

Expression of metallothionein (MT) in 75 cases of salivary gland tumors was studied by immunohistochemistry. All tumors containing myoepithelial cells were positive for MT immunostaining. In the tumors composed of both adenoepithelial cells and myoepithelial cells, only myoepithelial cells were MT positive. A rather large portion of cells was noted MT positive in myoepithelial carcinoma and myoepithelioma. Compared with S-100 and smooth muscle actin (SMA) antibodies, the effect of staining by MT antibody for myoepithelial cells was more satisfactory. The results indicate that MT may be a better marker for neoplastic myoepithelial cells and useful in the differential diagnosis of salivary gland tumors.

[Study on diagnosis of oral mucosal melanoma with histology, electron microscopy (EM) and immunohistochemistry].

Fifteen cases of primary oral mucosal melanoma were studied with the methods of Fontana Staining, EM, and immunohistochemistry. The results showed that 4 of 15 cases were non-pigmented lesions. In these cases, two were positive with Fontana staining. Pigment body could not be found in three cases observed under EM, but all the 15 cases were positive with immunohistochemical staining. The results suggested that the immunohistochemical staining should be done for those cases suspected as non-pigment melanoma.

[Comparision on the effects of ALI aeute liver injury of three species of concha haliotidis in mice].

The liver injury mice model with Haliotis ruber (Leach), Haliotis disus hannai Ino and Haliotis lacvigata (Donovan) was studied. The results indicated three species Haliotidis could versus carbon tetrachloride-induced ALI aeute liver injury in mice. Comparision of reduced serum glutamic-pyruvic transaminass (SGPT) activity showed Halitis disacus hannai Ino > Haliotis Lacvigata (Donovan) > Haliotis ruber (Leach). The Liver starch was determined. It is suggested protection liver effect of Haliotis discus hannai Ino and Haliotis lacvigata (Donovan) are most powerful.

Antibodies to glycosphingolipid antigens in patients with immune-mediated cochleovestibular disorders.

HYPOTHESIS: To determine the presence of antibodies against the glycosphingolipid antigen sulfated glucuronic lactosominyl paragloboside (SGLPG) in the sera of patients suspected of immune-mediated cochleovestibular disorders (IMCVD). BACKGROUND: Glycospingolipids are molecules present on the surface of normal nerve cells and are considered antigenic. Previous studies have isolated these antigens in vestibular neuroepithelia, cochleovestibular nerves and endolymphatic sacs. METHODS: The sera of 22 patients suspected of IMCVD were tested for antibodies against the antigen SGLPG. Thin-layer chromatography-immunostaining method was used. RESULTS: Antibody titers were elevated in 63.6% of patients tested. Statistical significance (p < 0.0001) was achieved since reactivity was seen in only 7% of 43 age-matched healthy controls. CONCLUSIONS: Antibodies to SGLPG antigens are present in some patients with IMCVD. Because SGLPD antigens have been previously isolated in the inner ear and the cochleovestibular nerve, these structures can potentially become targets for anti-SGLPG antibodies.

The effect of ethanol on muscarinic receptor-G protein coupling in the rat cortex.

To understand the mechanism of ethanol action on G protein-mediated signal transduction pathway, the effect of ethanol on muscarinic receptor-G protein coupling in the rat cerebral cortex was examined. Acetylcholine (ACh)-stimulated G protein GTPase activity was used as an index of receptor-G protein coupling. ACh stimulation of G protein GTPase activity was time- and concentration-dependent, and atropine-sensitive. Rats injected with ethanol (3 g/kg body weight) were sacrificed after 4 hr, and the cerebral cortices removed. The ability of ACh to stimulate GTPase activity was similar in cortical cell membranes obtained from control and ethanol-treated rats; ACh maximally stimulated the enzymatic activity by 22% in membranes from both groups of rats. Next, in cortical cell membranes obtained from control rats (i.e., not injected with ethanol) the ability of ACh to stimulate GTPase activity in the presence of ethanol was examined. ACh stimulated GTPase activity in a concentration-dependent manner; the activity was 12.3 +/- 0.1, 14.5 +/- 0.64, 15.7 +/- 0.54, and 16.1 +/- 0.33 Pi pmol/min/mg protein, at 0, 0.01, 0.1, and 1 mM ACh, respectively (P < 0.05). In the presence of 100 mM ethanol ACh-stimulated GTPase activity was significantly inhibited. The IC50 value of ethanol inhibition of ACh-stimulated GTPase activity was approximately 50 mM. These results suggest that: 1) in vitro, ethanol impairs ACh-stimulated G protein GTPase activity in the rat cortical cell membranes, and 2) in vivo, the acute effects of alcohol on G protein function may be transient and reversible.

Detection of neuroblastoma cells in blood by reverse transcriptase-polymerase chain reaction.

OBJECTIVE: To determine whether circulating tumor cells are present in patients with localized or disseminated neuroblastoma (NB). METHODS: Sixteen pediatric malignant tumor cell lines, including 10 NB, 4 primitive neuroectodermal tumor (PENT), 2 Wilms' tumor and 39 samples (tumor tissue, bone marrow and peripheral blood) from 8 patients with NB, were evaluated by reverse transcriptase-polymerase chain reaction (RT-PCR) using two markers: neuroendocrine protein gene products 9.5 (PGP 9.5) and tyrosine hydroxylase (TH). RESULTS: Analysis of RT-PCR products by agarose electrophoresis demonstrated that in culture cell lines the specificity for NB was highest in TH (9/10 with NB, 3/4 with PNET, 0/2 with Wilms' tumor) and the sensitivity was highest in PGP 9.5 10(-7). PGP 9.5 and TH mRNA were undetectable in the peripheral blood mononuclear cells (PBMNC) from normal children. Expression of these markers in 8 patients with NB at diagnosis and during treatment was found positive in 5 out of 8, all had been proved paralleling with their disease status. Two of the 5 became negative after further treatment. CONCLUSIONS: These systems allow the detection of circulating NB cells with higher sensitivity and specificity. When RT-PCR is clinically used for the detection of NB cells in blood, that both PGP 9.5 and TH should be selected is recommended. The assessment of bone marrow alone might not be used as a single index of tumor dissemination.

Sex hormone receptors of hemangiomas in children.

OBJECTIVE: To investigate whether or not estrogen receptor and other sex hormone receptors are present in hemangioma tissues of children. METHODS: Fifty-two specimens of hemangiomas were taken from 52 children. The specimens were doubly stained with hematoxylin-eosin and enzyme-linking affinity immunohistochemistry. The prepared tissue slides were observed under the microscope to search for estrogen receptors (ER), progestogen receptors (PR) and androgen receptors (AR) in all the cells. RESULTS: ER, PR and AR were detected in various kinds of hemangiomas. The mean positive cell rate of each sex hormone receptor in different hemangioma tissues revealed no significant distinction in statistics (P > 0.05). CONCLUSIONS: The results suggest that hemangiomas are one of the target tissues of estrogen. Estrogen, ER, PR, and AR may play an important role in the growth and development of hemangiomas.

[Causes of active phase arrest].

OBJECTIVE: To evaluate factors affecting active phase arrest. METHOD: Retrospective study from 1992 to 1994 was performed. A total of 166 pregnant women were included, 83 in to the active phase arrest group and 83 in normal group. RESULTS: There were significant differences in the average birth weight and the prevalence of abnormal fetal presentation between the 2 groups. The time length of the latent phase was another factor influencing active phase arrest. CONCLUSION: With normal pelvis, the high birth weight, abnormal presentation and prolonged latent phase were active phase arrest.

[The human SRY gene for prenatal diagnosis].

OBJECTIVE: To establish the method for prenatal sex diagnosis of the fetus carrying sex-linked genetic disorder. METHOD: Human SRY gene was amplified by polymerase chain reaction. A 422-bp male specific fragment was obtained. RESULTS: The fragment was identified in 10 men, but unidentified in 10 women. The diagnostic accordance rate of 20 amniotic fluid samples was 100%, 22 of 47 chorionic villi samples were positive. The rate of positive/negative (22/25) was nearly the sex rate of newborn babies. In the meantime, direct-PCR amplification of blood and amniotic fluid was completed. The fragment was shown from 4 microliters to 0.5 microliters of blood and from 2 ml to 0.5 ml of amniotic fluid. CONCLUSION: The results show that fetal sex determination by PCR will be suitable for clinical prenatal diagnosis of sex-linked genetic disorders.
