The suppression of long-term potentiation in rat hippocampus by inhibitors of nitric oxide synthase is temperature and age dependent.

At room temperature (23 degrees C-25 degrees C), the induction of long-term potentiation (LTP) in area CA1 of slices from young male Sprague-Dawley rats was depressed by preincubation with the nitric oxide synthase inhibitors NG-nitro-L-arginine (L-NA, 100 microM) and NG-nitro-L-arginine methyl ester (L-NAME, 100 microM). The D isomers were ineffective under the same conditions. Hemoglobin (20 microM) reduced but did not completely block LTP. Neither L-NA (at concentrations up to 1 mM) nor hemoglobin (20 microM) had any significant effect on LTP in slices from adult rats at room temperature, or in young rats at 29 degrees C-30 degrees C. These results suggest that nitric oxide is unlikely to play a role in the induction of LTP under physiological conditions.

Relationship between immunohistochemically detectable p53 protein and prognostic factors in head and neck tumors.

This study examined the relationship between immunohistochemically detectable p53 protein and prognostic factors in squamous cell carcinoma of the head and neck (SCCHN). Twenty-seven tumor specimens were evaluated utilizing a panel of three monoclonal antibodies (MAbs) directed against different epitopes of the p53 protein (PAb 421, PAb 1801, and PAb 240). The overall incidence of p53 protein detection with a panel of MAbs was 78%, which was significantly higher than with any one of the tested antibodies. Comparison of the tumors that were negative for p53 with tumors that stained positive with one or multiple antibodies, however, revealed no statistically significant differences with respect to the stage of disease, metastatic node involvement, size of the primary tumor, or degree of tumor differentiation. The results of our study suggest that levels of p53 protein, although commonly immunohistochemically detected in head and neck tumors, do not correlate with known prognostic factors for SCCHN.

Ser-His-Glu triad forms the catalytic site of the lipase from Geotrichum candidum.

The Ser-His-Asp triad is a well known structural feature of the serine proteases. It has also been directly observed in the catalytic sites of two lipases, whose high-resolution three-dimensional structures have been determined 1,2. Lipases show a wide variety of sizes, substrate and positional specificities, and catalytic rates 3. They achieve maximal catalytic rates at oil-water interfaces. The fungus Geotrichum candidum produces several different forms of lipases, two of which have been purified to homogeneity 4,5. Two lipase genes have been identified, cloned and sequenced 6,7. Both code for proteins of 544 amino acids with a total relative molecular mass of about 60,000 (Mr 60K). The two forms are 86% identical. Their isoelectric points differ slightly, being between 4.3 and 4.6. About 7% of the total Mr is carbohydrate. Until now, only a low resolution structure of GCL has been reported 8, but no high resolution structure has followed. We now report the three-dimensional structure of a lipase from G. candidum (GCL) at 2.2 A resolution. Unlike the other lipases and serine proteases, the catalytic triad of GCL is Ser-His-Glu, with glutamic acid replacing the usual aspartate. Although the sequence similarity with the other two lipases is limited to the region near the active-site serine, there is some similarity in their three-dimensional structures. The GCL is also an alpha/beta protein with a central mixed beta sheet whose topology is similar to that of the N-terminal domain of human pancreatic lipase. As in the other lipases 1,2, the catalytic site is buried under surface loops. Sequence comparisons with proteins from the cholinesterase family suggest that they also contain the Ser-His-Glu triad.

[Determination of ionization constants of primaquine and study of its coordination ratio with vitamin C].

Primaquine (P) has long been used as an antimalarial drug. The following formulas are derived for determination of the ionization constants of PH3+3 by pH-titration method: Ka1 = [aH + (3 - a)Cp]/[(a - 2)Cp/aH - 1] Ka3 = [(1 - a)Cp aH - Kw]/(a Cp + Kw/aH) = (1/Ka2)[(2 - a)Cp a2H - Kw aH]/(Kw/aH + a Cp) + [(1 - a) Cp aH - Kw]/(Kw/aH + a Cp) in which Kw is the ionic product of water, a is the mole ratio of HClO4 to primaquine and Cp is the total concentration of primaquine. The ionization constants of primaquine in 50% (v/v) ethanol in water determined at 25 degrees C in the ionic strength range of 5 x 10(-3)-5 x 10(-2) mol/L are: Ka1 = (3.84 +/- 2.35) x 10(-2) (attributed to the secondary ammonium group of primaquine); Ka2 = (1.50 +/- 1.17) x 10(-8) (attributed to the tertiary ammonium group); Ka3 = (2.07 +/- 0.27) x 10(-10) (attributed to the primary ammonium group). The coordination ratio of primaquine to vitamin C in the above solvent is determined by continuous variation and mole ratio methods based on pH and conductance measurements to be 1:1, indicating that the coordination compound formed in the solution is mainly a 1:1 compound.

[Surveillance of case findings of pulmonary tuberculosis].

As a result of the changing epidemic situation of tuberculosis, collective chest fluoroscopy as a method to detect pulmonary tuberculosis is becoming increasingly inadequate, and "symptom-dependent case finding" should be taken as the chief means for detection of the disease in our district. However, in collective units with high detected rates and in populations of high prevalence or in combination with general survey of lung cancer, regular fluoroscopic chest examination is an important route to discover patients with pulmonary tuberculosis.

Critical phenomena and thermodynamics of dilute aqueous sodium chloride to 823 K.

Semiempirical equations are developed that represent the behavior of dilute solutions of NaCl in water (steam) in the range 723-823 K where ion pairing is extensive. This supplements the equations given earlier for more concentrated solutions. In this temperature range the system NaCl/H(2)O shows critical behavior with two phases below the critical pressure. The equations for the dilute solutions yield critical behavior. Though the equations for concentrated solutions do not yield critical behavior at the critical pressure, only a very small interpolation function is required to connect smoothly the two equations. The ion-pairing equilibrium constants are reported as well as the Gibbs energies of hydration for both ions and ion pairs.

Thermodynamics of aqueous sodium chloride to 823 K and 1 kilobar (100 MPa).

It is shown that a very simple semiempirical equation represents quite accurately the thermodynamic properties of aqueous sodium chloride from 373 to 823 K. The equation comprises one Margules term and a Debye-Hückel term. Just the one Margules parameter is freely adjustable because the Debye-Hückel parameter is determined by the properties of water. The equation is valid from the saturation composition down to infinite dilution for solvent density above 0.75 g.cm(-3) but at lower density only above a solute mole fraction of about 0.1 on an ionized basis. Both solute and solvent activity coefficients are fitted from the lowest pressure of solution existence up to 1 kilobar (1 bar = 100 kPA). Derivation of enthalpy and other related functions is discussed.
