[Mechanisms of the pulmonary congestion in ligature strangulation (IX)].

The authors summarized 10 our papers on the mechanisms of pulmonary congestion in ligature strangulation. Since old times, a hypothesis based on hypersecretion of adrenaline at agonal stage in asphyxia is well-known. From our investigations, this hypothesis seemed to be applicable to ligature strangulated guinea-pig's lungs at least.

[Mechanisms of the pulmonary congestion in ligature strangulation (IV)].

The authors determined by radioimmunoassay, spectrophotometry or fibrin plate method the changes of levels of plasma or serum vasomotor (renin, bradykinin, plasmin, lactic acid and ferritin) of guinea-pigs sacrificed by ligature strangulation. This experiment was a part of the studies on the mechanisms of the pulmonary congestion in ligature strangulation. 1) In the ligature strangulation group, significant high levels of plasma renin was observed when compared with the presacrificed group and the beating on occiput group. 2) In both the ligature strangulation group and the beating on occiput group, significant high levels of plasma lactic acid were observed when compared with the presacrificed group. However, between the above-mentioned two groups, there were no significant differences in the levels of this substance. 3) Between the ligature strangulation group and the presacrificed group, there were no significant differences in the levels of plasma bradykinin and plasmin and serum ferritin.

Acute and chronic effects of carbon monoxide on mitochondrial function.

The purpose of the present study was to examine whether there were some differences between metabolic responses induced by carbon monoxide (CO) poisoning and those by hypoxic hypoxia. Significant decrease in cytochrome oxidase (COX) activity in heart and liver was observed during acute exposure, while no change was observed in brain. When the control of respiration was investigated, cerebral energy homeostasis was maintained even after severe exposure. In order to document this phenomenon we measured blood flow in liver and brain, but increase in cerebral blood flow was not observed and the experiments failed to show a relationship between cerebral homeostasis and blood flow. The prolonged exposure to CO and hypoxia resulted in an increase in COX and these responses were obviously considered to be adaptive to chronic exposure. From the results of acute and chronic exposure it is considered to be difficult to determine if CO has the direct effect on the respiratory chain. Further studies of the mechanism of the CO toxicity may be necessary.

[Transmission electron microscopy of the consumed hairs in an Australian lobster's stomach].

The present report is concerned with the transmission electron microscopic observations of the consumed hairs in an Australian lobster's stomach. The lamellar cuticles of these hairs were observed and their minimum diameters of melanin granules of cortices were measured by an imaging analyzer. The lamellar cuticles of these hairs consisted of two or three layers and there was no significant difference between them and Aberdeen Angus's hairs in mean the minimum diameters of melanin granules of cortices. Consequently, these hairs in an Australian lobster's stomach were identical to Aberdeen Angus's hairs.

[Mechanisms of the pulmonary congestion in ligature strangulation (V)].

The authors measured the iron contents and 3H-water contents of the lung et al.'s tissues of the guinea-pigs sacrificed by ligature strangulation. Congestion of the heart, lung, liver and kidney and anemia of the spleen were clearly recognized. And, the increase of hematocrit value of the heart blood was observed.

[Mechanisms of the pulmonary congestion in ligature strangulation (VI)].

The authors measured the minimum diameters of the hepatic, splenic and renal blood vessels of the guinea-pigs sacrificed by ligature strangulation. Constrictions of the splenic central arteries and the splenic trabecular veins, and dilatations of the renal interlobular veins and the renal cortical and medullary capillaries were clearly recognized.

[Mechanisms of the pulmonary congestion in ligature strangulation (VII)].

The authors observed the endothelial cells of the pulmonary veins and the immunoreactions of histamine in the pulmonary blood vessels and measured the 3H-histamine contents of the pulmonary tissues of the guinea-pigs sacrificed by ligature strangulation. Endothelial cells of the pulmonary veins possessed abundant pores and intracytoplasmic vacuoles. Heavy reactions of histamine exclusively demonstrated on the endothelial surface of the pulmonary veins and in the Weibel-Palade body of the pulmonary artery.

[Mechanisms of the pulmonary congestion in ligature strangulation (VIII)].

The authors observed the localization of adrenaline, noradrenaline and histamine in the walls of splenic and renal blood vessels of ligature strangulated guinea-pigs by autoradiography and immunocytochemistry, and measured the 3H-adrenaline and 3H-noradrenaline contents of the spleen et al.'s tissues of the guinea-pigs sacrificed by ligature strangulation. In both the experimental and control groups, some silver grains were observed in the endothelial cells of splenic central arteries, splenic trabecular veins, splenic venous sinuses and renal interlobular arteries after 3H-adrenaline administrations. In the ligature strangulation, the reactions of histamine exclusively demonstrated in the Weibel-Palade bodies of the splenic central arterial endothelial cells. By radioassay, the 3H-adrenaline contents of splenic and renal tissues were more than the control groups. From the above-mentioned facts, adrenaline-induced constrictions of splenic central arteries, splenic trabecular veins and splenic venous sinuses were recognized.

[Mammary fibroadenoma showing osseous metaplasia: a case report].

Discussed is a 33-year-old premenopausal woman who noted a mass in her right breast. On palpation, the tumor was determined as being 3.5 x 2.5 cm in size, well circumscribed, of a firm consistency, and freely movable. Mammography showed a well-defined oval lesion which contained a coarse calcification in the upper external quadrant. An ultrasound study revealed a well-defined oval low echoic lesion with a high echoic portion in the internal echo. The tumor was extirpated and a gross inspection found it to be an ordinary fibroadenoma, 3.2 x 2.5 x 1.5 cm in size. Histologically the lesion was a hyalinized fibroadenoma showing osseous metaplasia. A review of the literature has not revealed cases of a benign breast tumors showing an osseous and/or cartilagenous metaplasia. Notable however is that many reports show mammary osteosarcomas as originating from a fibroadenoma. Thus, this tumor also might have possibly developed into a osteosarcoma.

Anti-ischemic effect of the novel anti-ischemic agent 2-(4-(p-fluorobenzoyl)-piperidin-1-yl)-2'-acetonaphthone hydrochloride and possible mechanism of its action.

The protective effect of E-2001 (2-(4-(p-fluorobenzoyl)-piperidin-1-yl)-2'-acetonaphthone hydrochloride) was examined in various ischemic models, and the mechanisms of its action were investigated in vitro and in vivo. 1. Pretreatment with E-2001 ameliorated the degeneration of pyramidal neurons in the hippocampal CA1 sector following transient ischemia in Mongolian gerbils. 2. E-2001 improved stroke symptoms induced by permanent unilateral carotid artery ligation in gerbils. 3. E-2001 prolonged the survival time following permanent bilateral carotid artery ligation in gerbils and mice. E-2001 also prolonged the survival time following intravenous injection of KCN into mice. 4. E-2001 suppressed the high potassium-evoked release of glutamate from rat hippocampal slices. Furthermore, E-2001 prevented the excessive accumulation of extracellular glutamate induced by a brief ischemia in gerbils. 5. E-2001 exerted calcium antagonistic action, i.e., a relaxing effect on high potassium-induced contraction of rat aorta. 6. E-2001 exerted inhibitory action on the lipoperoxide production in vitro and in vivo, and exhibited a radical scavenging effect against O- and N-radicals. These results suggest that E-2001 might be effective as a novel anti-ischemic agent.

Biological and pharmaceutical factors affecting the absorption and lymphatic delivery of ciclosporin A from gastrointestinal tract.

Absorption site and some biological and pharmaceutical factors affecting the absorption and transport of ciclosporin A (CiA) from the gastrointestinal (GI) tract into both the thoracic lymphatics and the systemic circulation have been studied in rats. In a group of rats whose gastric emptying of orally administered CiA in HCO-60 formulation, 7.0 mg/kg, was physically prevented, both the lymphatic and the systemic availabilities of CiA were negligibly low. CiA was orally administered to another group of rats whose major intestinal lymphatics as well as thoracic lymph ducts were cannulated, and it was revealed that the amount of CiA delivered to the major intestinal lymphatics was about six times greater than that of CiA transferred into the thoracic lymphatics. In bile fistulous rats, the systemic availability of CiA was predominantly decreased but the lymphatic availability was not so decreased. In contrast, solvents such as propylene glycol and polyethylene glycol affected both the systemic and the lymphatic availabilities of CiA, which were also dependent on the absorption site. In particular, the lower small intestine does not contribute to the lymphatic availability of CiA.

A new method for studying working memory by using the three-panel runway apparatus in rats.

We designed a new method for studying working memory, by using a repeated acquisition procedure in the three-panel runway apparatus. This apparatus is composed of a start box, a goal box and four consecutive choice points; each choice point consists of three panel gates. Male Wistar rats were trained with 6 consecutive trials (one session) per day. Each trial was performed every two minutes. In this apparatus, rats could pass through only one gate (correct gate) among three panel gates in the direction of the goal box and were given 100 mg food pellets as the positive reinforcement. The sequence of correct gate position in each rat was changed everyday, but not in each session. Error responses (pushing the incorrect gate) were gradually reduced as training was repeated, and the learning was established within 16 training sessions to achieve criterion performance. Intraperitoneal scopolamine and intrahippocampal ethylcholine aziridinium ion (AF64A) produced increases in both the number of errors and the latency in a dose-dependent manner. The increase in errors induced by AF64A did not return to the control level, though the prolonged latency returned to normal. As a conclusion, this experimental procedure using the three-panel runway apparatus would be a useful method for studying working memory, and its memory deficit is involved at least in the dysfunction of the cholinergic system in the hippocampus.

Suppression of hepatic HMG-CoA reductase activity by beta-muricholic acid in mice fed a diet containing cholesterol and cholic acid.

A diet containing cholesterol and cholic acid (SID) is known to induce the formation of cholesterol fatty liver as well as cholesterol gallstones. The activity of HMG-CoA reductase, one of the key enzymes for cholesterol synthesis in the liver, is significantly lowered by addition of beta-muricholic acid to SID. The prevention of fatty liver formation by beta-muricholic acid was accompanied by the suppression of HMG-CoA reductase activity.

Electroencephalographic effects of the new antidepressant paroxetine in the rabbit.

The electroencephalographic (EEG) effect of (-)-trans-4-(4'-fluorophenyl)-3-(3',4'-methylene-dioxyphenoxy-met hyl)piperidine hydrochloride (paroxetine, BRL 29060A) a new antidepressant, was investigated in conscious rabbits with chronic electrode implants and was compared with those of imipramine and amitriptyline. Paroxetine induced an arousal pattern of the spontaneous EEG consisting of low voltage fast waves in the cortex and synchronization of hippocampal theta waves with decreased amplitude, while imipramine and amitriptyline elicited drowsy patterns of the spontaneous EEG. Paroxetine failed to suppress the EEG arousal responses induced not only by auditory stimulation but also by electrical stimulation of the mesencephalic reticular formation, centromedian thalamus and posterior hypothalamus, whereas imipramine and amitriptyline markedly inhibited these responses. The EEG arousal response induced by i.v. injection of physostigmine 0.2 mg/kg was slightly enhanced by paroxetine, while the response was significantly suppressed by imipramine and amitriptyline. Paroxetine, imipramine and amitriptyline showed no significant effect on the photic driving response and recruiting response. Paroxetine did not show any effects on the limbic afterdischarges elicited by either hippocampal or amygdaloid stimulation, while imipramine and amitriptyline caused an initial suppression followed by slight enhancement of these afterdischarges. These results indicate paroxetine to be an antidepressant of a new type which induces a sustained arousal pattern of the spontaneous EEG and has no central anticholinergic action.
