Springtime peaks of trace metals in Antarctic snow.

Drifting snow samples were collected at Asuka Station (71 degrees 32'S, 24 degrees 08'E, 930 m above sea level) over a period from July to December 1991; 36 elements (including Na, Mg, K, Ca, Fe, Al, Li, V, Cr, Mn, Co, Ni, Cu, Zn, Ga, Se, Rb, Sr, Cd, Pb, Y, La, Ce, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Yb, Lu, and Th) in snow were analyzed by inductively coupled plasma-mass spectrometry (ICP-MS) by direct sample introduction. Concentrations of Cl-, NO3-, and SO(4)2- in the snow were also determined by ion chromatography. In late September to early October, there was a pronounced peak concentration of most of the elements together with non-sea salt sulfate. Enrichment factor analyses suggest that Na, Mg, Ca, K, and Sr are of marine origin and Al, Fe, Mn, Rb, Cr, Ni, Ga, V, and all the rare earth elements are of crustal origins. Volcanic eruption of Mt. Pinatubo (June 1991) and Mt. Hudson (August 1991) could be the reason for the precipitation of Pb, Cd, Cu, Zn, and Se together with non-sea salt sulfates in the austral spring at Asuka Station.

Effects of 6-chloro-2',3'-dideoxyguanosine (6-Cl-ddG) in surface lymph nodes of rhesus monkeys (Macaca mulatta) chronically infected with simian immunodeficiency virus (SIVmac239).

We studied the effects of 6-chloro-2',3'-dideoxyguanosine (6-Cl-ddG), an antiretroviral drug, in surface lymph nodes of rhesus monkeys (Macaca mulatta) chronically infected with simian immunodeficiency virus (SIV). The rhesus monkeys were treated with 25 mg/kg of 6-Cl-ddG every 8 hr for 2 weeks. We performed sequential biopsies of the surface lymph nodes three times: before, during, and after the drug treatment. The 6-Cl-ddG dramatically decreased the number of infectious virus (measured by limiting dilution assay) in lymph node mononuclear cells. This decrease was consistent with the decrease in the number of viral RNA-positive cells in lymph nodes (analyzed by in situ hybridization). Histopathological analysis revealed that hyperplastic lymphoid follicles were reduced in size, especially, enlarged areas of centroblasts in lymphoid follicles (the so-called dark areas of germinal centers) were declined. Our results demonstrated that 6-Cl-ddG decreased the viral burden concomitantly with reduced hyper-activation of germinal centers in lymphoid follicles of SIV-infected rhesus monkeys.

Epitope mapping of murine monoclonal antibodies against human immunodeficiency virus type 1 Nef.

It has shown that the human immunodeficiency virus type 1 (HIV-1) Nef protein has the high antigenicity in HIV-1 seropostive individuals. We newly obtained seven monoclonal antibodies (mAbs). To identify the antigenic determinants of HIV-1 Nef protein against murine, epitope mapping of the mAbs was performed by enzyme-linked immunosorbent assay (ELISA) by using several recombinant truncated Nef fusion proteins, that were expressed in Esherichia coli, and synthetic peptides. The results showed that mAbs A6, A7, F2, F3, F4, F8 and E5 recognized epitopes on Nef protein located at amino acid residues 18-26, 28-45, 115-137, 128-137, 115-126, 128-137, and 170-181, respectively.

Efficacy of 6-chloro-2',3'-dideoxyguanosine (6-Cl-ddG) on an ARC/AIDS rhesus macaque (Macaca mulatta) infected with simian immunodeficiency virus.

The efficacy of 6-chloro-2',3'-dideoxyguanosine (6-Cl-ddG) was investigated in vivo by using a male ARC/AIDS rhesus macaque infected with simian immunodeficiency virus (SIVmac251/32H). He was administered subcutaneously 6-Cl-ddG (50 mg/kg B.W.) every 8 hr for 14 days when he showed clinical features of recurrent weight loss, severe diarrhea and neuropathy. The number of CD4+, CD8+ cells and total T cells increased rapidly after administration of 6-Cl-ddG and a high level was maintained for 2 months, but the B cell count decreased during the treatment. The antibody titer to SIV did not change significantly during or after the treatment, but the virus load in the plasma measured by RT-PCR dropped to one-third at the start of the 6-Cl-ddG treatment. Within 3 days after the start of 6-Cl-ddG administration, he began to show recovery in clinical signs including weight increase, and disappearance of diarrhea and neuropathy. These findings suggested that 6-Cl-ddG was effective at the stage of ARC/AIDS in a rhesus monkey infected with SIV.

Vomiting induction by ipecac syrup in dogs and ferrets.

The dog has been used as an experimental animal in emesis research. In this study, we analyzed the emetic effects of ipecac syrup using a smaller animal, the ferret, and compared its response to that of the dog. Dogs and ferrets were divided into 4 groups (n = 4, each). Each group was given either 0.1, 0.25, 0.5, or 1.0 ml/kg of ipecac syrup, and the latency and numbers of retching and vomiting were recorded. Animals given an equal volume of saline served as controls. The numbers of vomiting and retching increased dose-dependently in both dogs and ferrets, and there was no difference in latency and numbers of vomiting between them. The numbers of retching were greater in ferrets than in dogs at > or = 0.25 ml/kg. Taking these results into consideration, the ferret seems to be as useful as the dog in studies on emetic effects of ipecac syrup.

[Correction of urinary calcium levels for urine osmotic pressure].

The usefulness of the urinary calcium/creatinine (Ca/Cr) ratio, the oldest marker of bone resorption, is limited because of the influence of sodium intake and urine volume. The urinary Ca/Cr ratio was therefore compared with urinary calcium corrected for the urine osmotic pressure (Ca/OSM). Significant correlations are found between the Ca/Cr ratio and both creatinine (r = -0.386) and osmotic pressure (r = -0.473) in random urine samples from normal subjects, but similar correlations were not found for urine Ca/OSM. No significant correlation was found between Ca/OSM and Na/OSM, but the correlation coefficient between Ca/Cr ratio and Na/Cr ratio was 0.399. Thus, the Ca/Cr ratio is affected by urinary volume and sodium concentration, but the Ca/OSM ratio is not. The Ca/OSM ratio may therefore be a more accurate marker of bone resorption, because one of the main components of urinary osmotic pressure is sodium that inhibits tubular calcium reabsorption. The Ca/OSM ratios measured from 24-hour urine samples were found to be strongly correlated with the values measured from samples obtained during sleep (sleep urine, 0.823) and were slightly less strongly correlated with the values measured from second morning samples (0.641). Because bone resorption is especially active at night, the Ca/OSM ratio in sleep urine may be the most sensitive marker of bone resorption. The Ca/OSM ratio in sleep urine samples increased with age in normal women.

[Effects of calcium supplementation using AAACa or milk on nocturnal bone resorption in young women].

In order to study the effect of calcium supplementation on bone resorption, a randomized controlled crossover study was carried out on eight healthy 18-19 year old female volunteers using either AAACa heated oyster shell with vacuum-heated seaweed or milk. Regimen A consisted of an oral dose of 200 mg calcium in the form of AAACa. B 200 ml milk after breakfast and supper and at bedtime, and C control with no calcium supplement. Early morning fasting blood and urine sampling was carried out after 7 days of calcium supplementation. Serum calcium was higher in groups A and B than in C, and serum intact parathyroid hormone (PTH) was significantly lower in group A than in groups B and C, according to a paired t-test. Urinary excretion of crosslinked collagen degradation product, pyridinoline and deoxypyridinoline showed a similar decrease in groups A and B but not in C. The more effective suppression of PTH by AAACa than by milk may be due to its higher bioavailability and the absence of phosphate stimulating PTH secretion.

Serial changes of hemostasis in aneurysmal subarachnoid hemorrhage with special reference to delayed ischemic neurological deficits.

This study was undertaken to elucidate comprehensively the serial changes occurring in hemostatic systems after aneurysmal subarachnoid hemorrhage (SAH) and thereby to ascertain whether the examination of the integrity of these systems is helpful in predicting delayed ischemic neurological deficits (DINDs). The authors examined 117 patients admitted to the hospital within 24 hours after onset of SAH. Blood samples were collected from each patient on Days 0 (at admission), 3, 6, 14, and 30. A number of hemostatic parameters were examined in these samples, and the relationships between their changes and DINDs were assessed. Eighteen (15.4%) of the patients exhibited DINDs, and their frequency increased as the severity of subarachnoid clotting increased. Also, the frequency of DINDs was significantly higher in the patients with hydrocephalus on initial computerized tomography (CT) scans than in those without hydrocephalus. Regarding the hemostatic parameters at admission, there was no significant difference between the patients with and without DINDs. On Day 3, however, the fibrinogen and D-dimer levels were higher in the patients with than in those without DINDs. The fibrinogen and thrombin-antithrombin complex levels on Day 6 and the D-dimer level on Day 14 in the patients with DINDs were higher than the corresponding levels in those without DINDs. Multivariate analyses revealed that the following variables (in order of importance) were independent predictors of DINDs: the levels of D-dimer on Day 3, fibrinogen on Day 6, and the presence of hydrocephalus on admission. These data indicate that the levels of hemostatic parameters in concert with the CT findings may enable us to predict the appearance of DINDs.

[A case of lung adenocarcinoma associated with remarkably high levels of CA 19-9 and lymphangitis carcinomatosa].

A 69-year-old male was admitted to our hospital because of dry cough. Chest X-P and CT scans showed a mass shadow in the right lung, thickening of pulmonary vessels and pleural effusion. Cytological examination of transbronchial brushing specimen revealed lung adenocarcinoma. Cancer cells were also detected in pleural effusion. High levels of CA 19.9 were noticed: 48,400 U/ml in serum and 395,000 U/ml in pleural effusion, respectively. Two courses of combined chemotherapy (CDDP + VDS) were done. Concurrent chest radiation therapy (40 Gy) to primary tumor was also performed. After treatment the primary tumor decreased in size on CT scan analysis, but the patient suffered from respiratory failure due to the increase of sputa and pleural effusion and died 104 days after admission.

[Characterization of spontaneous liver metastatic model of BALB/c retroperitoneal sarcoma (LMFS) injected in footpad and antitumor effects of HCFU].

A new spontaneous liver metastasis model from LMFS tumor, a retroperitoneal sarcoma of BALB/c mouse origin, was reported. After a subcutaneous injection of LMFS cells in the footpad, amputation of the leg was performed every 5 days from day 5 to day 20, and mice were killed at day 25 under anesthesia. The LMFS cells proliferated at the inoculation site (100% take), and all mice operated after day 15 had metastatic nodules spontaneously in the liver. Local recurrence was not observed in any cases. Liver weight was correlated with tumor growth in the liver. The combination therapy of operation and HCFU was examined in comparison with the footpad injection model. The foot on the injection side was amputated on day 12 with or without HCFU administration, and mice were sacrificed on day 33 under anesthesia. Histological liver metastases were as follows: 1. no treatments: 100%; 2. early phase chemotherapy without operation: 80%; 3. operation without chemotherapy: 80%; 4. postoperative chemotherapy: 20%; 5. preoperative chemotherapy: 40%; and 6. late phase chemotherapy without operation: 100%. These results indicate that operation with chemotherapy is the most useful treatment for liver metastases. Using this liver metastatic model, we are investigating a new combination therapy of operation and anti-cancer drugs for liver metastases.

[A novel diagnostic method for allergy "LUCICA HRT"].

We developed a novel glass microfiber-based histamine release test (HRT) which is characteristic of response of basophil leukocyte to allergens in vitro. It allows the determination of 20 allergens (10 for inhalation allergy, and the other for food allergy) at the same time using a small amount of whole blood. For 158 patients of inhalation allergy and 135 patients of food allergy, they were evaluated by HRT, CAP-RAST, skin test, and provocation test. The concordance of HRT, CAP-RAST, and skin test were 78%, 72%, and 62% in inhalation allergy, and 84%, 71%, and 81% in food allergy, respectively. The specificity of HRT, CAP-RAST, and skin test were 63%, 37%, and 19% in inhalation allergy, and 92%, 63%, and 83% in food allergy, respectively. The positive predictive value of HRT, CAP-RAST, and skin test were 79%, 71%, and 63%, respectively. The false positive ratio of HRT (14%) was lower than that of CAP-RAST (42%). The sensitivity of CAP-RAST was higher than that of HRT. In addition, we experienced a case of patient who showed significant improvement by treatment. At initial stage, not only the symptoms were erythema and scratching but also HRT, CAP-RAST, and skin test resulted in positive. Four months later, he had no allergic symptoms, HRT resulted in negative, but CAP-RAST and skin test resulted in still positive. From these results, we concluded that CAP-RAST is good for screening of etiological allergens and that HRT is a useful diagnostic method for the confirmation of a clinical allergy.

[Ultrastructural study of lens in rat hereditary cataract by quick-freezing and deep-etching].

The cytoskeleton of lens fiber cells in rats with hereditary cataract (ICR/f) was studied by quick-freezing and deep-etching. Two kinds of filamentous structure with diameters of 5 and 10 approximately 15 nm were observed in the cortical fiber cells. They showed meshwork structures which were buried in globular particles. These filamentous structures were mostly absent in the nucleus fiber cells, but aggregated and fused globular particles with diameters of 10 approximately 20 nm were observed. Small cavities were sometimes observed in their cytoplasm. Between lens fiber cells, many globular structures were also seen. These changes might represent the degeneration of the lens fiber cells in cataract lenses.

Membrane skeletons in avian erythrocytes as revealed by the quick-freezing and deep-etching method.

Ultrastructure of chicken erythrocytes were examined by the quick-freezing and deep-etching (QF-DE) method. Some erythrocytes were fixed with paraformaldehyde and prepared with erythrocyte-splitting method or saponin treatment to remove soluble proteins before quick-freezing. Others were prepared in the cytosol buffer with the erythrocyte-splitting method to obtain natural state of cytoskeletons. Non-expanding membrane skeletons were highly condensed on the cytoplasmic side of lipid membrane in the paraformaldehyde-fixed specimens. Under unilateral extension of the specimens, long stretched filaments were connected alternately with condensed filamentous or granular structures under erythrocyte membranes. As the membrane skeletons got closer to the marginal bands, they become more dense network structures. Moreover, in the fresh unfixed specimens, dense networks of filaments were localized underlying erythrocyte membranes in a relatively intact state. Fine filaments connected the marginal microtubule bands to the cytoplasmic sides of erythrocyte membranes. The different distribution of each cytoskeletal component and the association of these structures may support the elliptocytic shape of chicken erythrocytes and resist the dynamic circumstance.

Immunocytochemical study of dystrophin localization in cone cells of mouse retinas.

PURPOSE: Previously, the authors reported that dystrophin was observed under the rod cell membranes in rat retinas. However, it was not determined whether dystrophin is located in cone cells. In the current study, the authors clarify dystrophin localization in cone cells of mouse retinas. METHODS: Immunoblotting, confocal laser scanning microscopy, and immunoelectron microscopy were used to investigate retinal dystrophin with a monoclonal antibody raised against the human dystrophin C-terminus. RESULTS: Immunoblotting analysis showed some immunoreactive bands from retinal extracts. Confocal images indicated two different immunostaining patterns: One was a tiny dot, and the other was a larger, aggregated dot. Immunoelectron microscopy revealed that retinal dystrophin was localized in cone cells as well as in rod cells. CONCLUSIONS: Retinal dystrophin is a common component of cone and rod cells and probably is related to the physiological function of photoreceptor cells.

Concomitant cardiac and pulmonary operation. Pulmonary mechanics and outcome of phrenic nerve injury.

OBJECTIVE: We describe the postoperative respiratory failure due to the phrenic nerve injury in the setting of concomitant cardiac and pulmonary operation. EXPERIMENTAL STUDY: Prospective study. SETTING: Department of Cardiac and Thoracic Surgery Osaka University Medical School. PATIENTS AND INTERVENTIONS: From January 1984 to December 1993, 5 patients (1.4%) underwent the concomitant cardiac and pulmonary operation out of 359 patients who received surgical treatment for lung cancer at our institution. MEASURES AND RESULTS: Three (60%) out of 5 patients required prolonged mechanical ventilation despite the absence of cardiac complication, lung edema or pneumonia. Diaphragm function and work of breathing were measured in two patients before and after weaning from mechanical ventilation. Phrenic nerve dysfunction was consistent with the result that trans-diaphragmatic pressure (delta Pdi) was low, a ratio of gastric to esophageal pressure swing (delta Pga/delta Pes) was abnormally negative, and work of breathing (WOB) was high. Phrenic nerve function restored associated with clinical improvement. CONCLUSIONS: Diaphragm dysfunction and an increase in work of breathing may be potential causes of respiratory failure in patients after concomitant cardiac and pulmonary operation. This compromise in respiratory mechanics should not be overlooked in the postoperative care, which may lead to the best management in postoperative respiratory care.
