Diaminobenzidine peroxidase in rat brain.

Peroxidase has several roles in each organ. In neural tissue, it is considered from pathological examination to be related to the aging process. To study biochemically the presence of peroxidase, diaminobenzidine was used as the hydrogen donor and the enzyme was partially purified from rat brain and characterized. Diaminobenzidine peroxidase was purified 670-fold from the original homogenate. The presence of p-phenylenediamine and guaiacol peroxidase was not detected in either the crude homogenate or the partially purified fraction. The possible presence of isozyme(s) in peroxidase was implied and this may be related to the physiologic differences in organs.

Various sulfatase activities in leukocytes and cultured skin fibroblasts from heterozygotes for the multiple sulfatase deficiency (mukosulfatidosis).

In heterozygotes for multiple sulfatase deficiency (MSD), several sulfatase activities including arylsulfatases A, B1, B2, and C, and cholesterol sulfatase were 40-50% of normals in cultured skin fibroblasts and 70-80% of normals in leukocytes. In MSD patients, these enzyme activities were deficient or reduced. DEAE-Sepharose column chromatographic patterns of 4-methylumbelliferyl sulfatases A, B1, and B2 in leukocytes and cultured skin fibroblasts from MSD patients and heterozygotes were also consistent with the above data. These data indicate that several sulfatase activities in heterozygotes of MSD exhibited intermediate activities as observed in the heterozygote state of other autosomal recessive inherited diseases.

[A case of alpha-fetoprotein producing embryonal carcinoma of the stomach].

A 67-year-old man with complaints of general malaise and epigastralgia was admitted to our hospital for further examination. Upper GI series and endoscopy showed advanced cancer, Borrmann type II; biopsy specimens revealed pathological pictures of malignancy. The serum alpha-fetoprotein values were remarkably high (473,000 ng/dl); the liver scintigram revealed a space-occupying lesion in the right lobe and the abdominal CT-scan showed ascites and lymphadenopathy of the abdominal para-aortic region. Therefore, double cancer of the stomach and the liver was suspected. The patient died in a state of hepatic coma with a rapid increase of jaundice, ascites and right pleural effusion at 3 weeks after admission despite the anti-cancer treatment. Autopsy revealed embryonal carcinoma with yolk sac tumor elements of stomach origin, extensive metastasis to the liver, lung, peritoneum, and portal vein. The peroxidase anti-peroxidase technique revealed that primary and secondary tumor cells produced alpha-fetoprotein.

A comparative study of brain arylsulfatases B1 and B2: the difference between the two forms in rates of uptake by multiple sulfatase deficient (MSD) disorder fibroblasts.

Arylsulfatases B1 and B2 isolated from human brain were introduced into neonatal type MSD fibroblast culture. Arylsulfatase B2 was linearly taken up into MSD fibroblasts with respect to time and the dose of enzyme, whereas arylsulfatase B1 enzyme was practically not taken up into cells. Uptake of arylsulfatase B2 was inhibited in the presence of mannose 6-phosphate. These data suggest that the biological functional difference between arylsulfatases B1 and B2 is due to the presence of a phosphorylated residue acting as a recognition marker on one of the enzymes.

[Usefulness of contrast echoangiographic diagnosis of congenital tricuspid stenosis].

Congenital tricuspid stenosis is an uncommon congenital anomaly and usually associated with other lesions such as severe pulmonic stenosis or atresia. Sometimes, it is difficult to diagnose this malformation non-invasively, but the differential diagnosis from tricuspid atresia is essential for the surgical considerations. Described here were three patients with congenital tricuspid stenosis who were successfully diagnosed by contrast echoangiography using peripheral venous injection of the contrast medium. Contrast echoes appeared in the right atrium initially and then in the right ventricle through the stenotic tricuspid valve. In conclusion, contrast echoangiography is mandatory in the diagnosis of this entity.

Characterization of glycosphingolipids from cells of various types of human leukemia: occurrence of two glycosphingolipids, one reacting with anti-asialo GM1 antibody and one with anti-Forssman antibody.

Glycosphingolipids of neutrophils, lymphocytes and leukocytes from patients with various types of human leukemia [acute lymphoblastic (ALL), acute unclassified type (AUL), acute myeloblastic (AML), acute monocytic (AMoL), chronic myeloblastic (CML)] and the hypereosinophilic syndrome (HES) were analyzed chemically and immunochemically. No distinct difference was found in the molar ratio of lipid-bound sialic acid to lipid-bound phosphorus in these cells, but a low ratio of cholesterol to lipid-bound phosphorus was found in ALL (3 of 4 cases), AML, CML and AMoL (one of 2 cases). The predominant glycosphingolipid was ceramide dihexoside (CDH) in all cells analyzed, but the amount and the molar ratio of lipid-bound phosphorus to CDH were clearly different in different cell types, indicating that the molar ratio is a useful criterion in the classification of types of leukemia. In addition, molecular diversity of minor glycosphingolipid components was observed in various leukemic cells. Two of the neutral glycosphingolipids in AMoL were tentatively identified as asialo GM1 and Forssman glycolipids by comparing their mobilities on thin-layer chromatography with those of standard glycolipids and by observing the formation of precipitin lines on a double diffusion agar plate with anti-asialo GM1 and anti-Forssman antibodies.

Prenatal diagnosis of metachromatic leukodystrophy: a diagnosis by amniotic fluid and its confirmation.

Late infantile metachromatic leukodystrophy (MLD) was successfully diagnosed in utero by demonstrating the absence of arylsulfatase-A in amniotic fluid using diethylaminoethyl-Sepharose column chromatography. Diagnosis by amniotic fluid using an ion-exchange column is more rapid and reproducible as compared with those reported previously. The diagnosis was confirmed by the absence of arylsulfatase-A in fetal brain, liver, and kidney tissues as well as by the marked accumulation of sulfatide in kidney. The kidney is the most appropriate organ for the demonstration of sulfatide accumulation in fetal tissues in MLD.

Acid mucopolysaccharide (AMPS) abnormality in multiple sulfatase deficiency: chemical compositions of AMPS in urine and liver.

Extensive chemical analyses of acid mucopolysaccharides (AMPS) were carried out in the urine and tissue (liver and brain) from a Japanese patient and two European patients with multiple sulfatase deficiency (MSD). The Japanese patient with MSD contained excessive quantities of heparan sulfate and moderately increased chondroitin sulfate A/C. Urinary excretion of AMPS in MSD heterozygotes was increased 2-fold compared to our controls. The urinary pattern of AMPS in the mother of the MSD patient showed an increase of 18% heparan sulfate and 36% dermatan sulfate whereas the urinary excretion pattern in the father was increased 21% for heparan sulfate as contrasted to controls (chondroitin sulfate A, 50-52%; chondroitin sulfate C, 38-46%; and heparan sulfate, 3-10%). Seventy-five % of the AMPS and the MSD liver was heparan sulfate rather than dermatan sulfate. The degree of accumulation of AMPS in the MSD liver was 30-50 times that of the control. Cerebral gray matter from the MSD patient contained 30-40 times that of control (relative increase of heparan and dermatan sulfate) whereas only a 5-fold increase was observed in white matter. It seems that a major site of accumulated AMPS appears to be in the gray matter. Carbohydrate analysis of the AMPS obtained from MSD urine and tissues was performed by: enzyme digestion with testicular hyaluronidase, heparitinase and chondroitinase ABC, cellulose acetate electrophoresis, Dowex-1 column chromatography and amino sugar analysis by amino acid analyzer. These findings indicate that the major accumulated AMPS in MSD urine and liver is heparan sulfate and thus, the predominant AMPS metabolic defect in MSD is heparan sulfate degradation.
