IgM anti-HBc in anti-HBe positive chronic type B hepatitis with acute exacerbations.

IgM anti-HBc was measured by radioimmunoassay in serially collected serum samples during 20 acute exacerbations which developed in 14 patients with anti-HBe positive chronic type B hepatitis. IgM anti-HBc became positive in 12 of the 14 (86%) patients and in 18 of the 20 (90%) exacerbations, and elevation of IgM anti-HBc which was believed to be significant was also observed in the remaining two patients. In most of the patients who also had determinations of serum hepatitis B virus DNA, the hepatitis B virus DNA became positive. These results suggest that ALT elevation in anti-HBe positive chronic type B hepatitis is associated with active replication of hepatitis B virus. It is considered to be useful to measure IgM anti-HBc for the purpose of identifying the causes of ALT elevation in anti-HBe positive chronic type B hepatitis.

The biochemical properties of urinary human chorionic gonadotropin from the patients with trophoblastic disease.

Human chorionic gonadotropin (hCG) was extracted and purified from urine of normal pregnant women and patients with hydatidiform mole and choriocarcinoma using the sam methods. Both hCG-hydatidiform mole and hCG-choriocarcinoma as well as hCG-normal pregnancy was separated into alpha and beta subunits by SDS disc electrophoresis upon treatment with 2-mercaptoethanol and showed the same immunoreactivities against anti-hCG, -alpha hCG, and -beta hCG as hCG in each radioimmunoassay. In vivo bioassay, bioactivities of hCG- normal pregnancy and hCG-hydatidiform mole were approximately 7,000 IU/mg (2nd IS), while that of hCG--choriocarcinoma was only 400 IU/mg. Conversely, the receptor binding activities in vitro of hCG-chorio carcinoma was about 3 times more effective than the other 2. Although the amino acid composition of these hCG preparations were practically identical, a great difference in the carbohydrate composition was observed. The significant difference was that while sialic acid was undetectable in hCG-choriocarcinoma approximately 8.5% of sialic acid was found in hCG-normal pregnancy and hCG-hydatidiform mole. A parallel finding was that iodinated hCG-choriocarcinoma was taken up in large quantities by the liver in comparison to the ovary which differed from that observed with hCG-normal pregnancy and hCG-hydatidiform mole in Parlow rats. The present findings support the thesis that neoplastic or malignant transformation of trophoblasts may result in an alteration of the glycosylation process, especially the sialylation, in the biosynthesis of hCG rather than the translation steps.

Behavior and body temperature in rats following chronic foot shock or psychological stress exposure.

In an attempt to examine stress-induced behavioral disorders, including circadian rhythm disturbances, we measured motor activity, feeding, drinking, and body temperature over a 14-day period following a long-term stress exposure in rats. Male Wistar rats were exposed to foot shock (physical) or non-foot shock stress (psychological) induced by the communication box for 1 h daily over 12 weeks. Two to three months after the termination of the stress sessions, motor activity, food intake, water intake, and body temperature were measured by means of an automatic behavioral measurement system under a 12:12-h light:dark cycle. Motor activity, feeding, and drinking patterns were not influenced by either of the previous stress exposures. Daily rhythm of body temperature was also unchanged in either stress group, however, a significant elevation in body temperature (by 0.20 degrees C, p<0.05) was observed only in non-foot-shocked rats. The present study suggests that only psychological stress induces an elevation of body temperature following the stress exposures; however, long-term stress exposures in the present experiment do not disturb behavioral activities and daily rhythms of behaviors.
