Interleukin 1-like factors can accumulate 5-hydroxytryptamine in the liver of mice and can induce hypoglycaemia.

After the injection into mice of culture medium of P388D1 cells, a murine macrophage cell line, 5-hydroxytryptamine accumulated in the liver and blood glucose declined. The factors capable of inducing these responses were purified by gel filtration and chromatofocusing. With these procedures, the activity to induce the increase in 5-hydroxytryptamine in the liver accompanied the activity to induce hypoglycaemia. Moreover, through the purification, the factors were found in the fraction of interleukin 1, a lymphocyte-activating factor. These results suggest that the factors capable of inducing the increase in 5-hydroxytryptamine and hypoglycaemia are likely to be interleukin 1 molecules or molecules closely related to interleukin 1. The present and previous findings together with those in the literature support the idea that the increase in 5-hydroxytryptamine in the liver might be a cause of hypoglycaemia. These findings may provide new and important information about the roles of macrophages in inflammation or in immune responses.

Induction of ornithine decarboxylase in the liver and spleen of mice by interleukin 1-like factors produced from a macrophage cell line.

Injection of culture medium of P388D1 cells, a murine macrophage cell line, into mice produced a rapid induction of ornithine decarboxylase in the liver and spleen. The ornithine decarboxylase-inducing factor in the medium was purified by gel-filtration and chromatofocusing. With these procedures, the factor was usually found in the fraction of interleukin 1 (a lymphocyte-activating factor), indicating that the factor is identical with interleukin 1 or a molecule closely related to interleukin 1. The ornithine decarboxylase induction by the factor was suppressed by cycloheximide, but actinomycin D did not suppress the induction, or rather enhanced it. These findings may provide valuable information on the important roles of macrophages in immune responses or in inflammatory reactions.

Fractionation of polyclonal antibody by isoelectric focusing and chromatofocusing: separation of high-affinity rabbit clonotype anti-thyroxine antibody.

Immunoglobulin G fractions prepared from conventional rabbit anti-thyroxine (T4) antisera were fractionated by agarose gel isoelectric focusing (IEF) in the range of pH 3 to 10, and by chromatofocusing using a Fast Protein Liquid Chromatography (FPLC) system. The clonotype antibodies were recovered from the fractions and subjected to Scatchard plot analysis. The highest affinity constants of the initial antibody (shown in parentheses) and those of the antibodies recovered were IEF, 1.8 X 10(9) to 8.3 X 10(9) M-1 (2.2 X 10(9) M-1); FPLC, 2.4 X 10(9) to 6.0 X 10(9) M-1 (2.5 X 10(9) M-1). A sensitive radioimmunoassay of T4 was achieved with the isolated high-affinity anti-T4 antibody. The minimum detectable concentration of T4 was 6.3 X 10(-15) to 1.5 X 10(-14) mol/tube, which was three to five times lower than detectable with the initial antibodies.

Differences in the structure of alpha-fetoprotein and its clinical use in pediatric surgery.

Techniques have been studied which distinguish two variants of human alpha-fetoprotein (AFP) on the basis of characteristics of the carbohydrate moiety of this glycoprotein. AFP in serum samples from six children with tumors of yolk sac origin showed little concanavalin-A (Con A) binding. In contrast, Con A binding of AFP was almost complete in serum samples from 14 other subjects with elevated AFP, including two with liver-cell tumors, eight with neonatal cholestasis, and four normal newborn infants. Differences were confirmed by immunoelectrophoretic studies. Thus, AFP from cells of yolk sac origin can be distinguished from AFP from liver cells or from tumors of hepatic cell origin.

A case of nonfunctioning paraganglioma arising from the retroperitoneum: angiographic and scintigraphic features.

We report a case of a nonfunctioning nonchromaffin paraganglioma arising from organ of Zuckerkandl. Angiography manifested a cartwheel-shaped image in the arterial phase and a characteristic tortoise shell-shaped lobular structure with homogeneous opacification in the venous phase. Just after administration of 99mtechnetium-E-HIDA scintigraphy disclosed a globular hot density below the left kidney. These findings may represent characteristic signs of paragangliomas.

Effective use of portions of the supernumerary digit to correct polydactyly of the thumb.

The purpose of surgery for polydactyly of the thumb is to ablate the supernumerary digit and to reconstruct a thumb with adequate function that approximates normal size and shape. A further aim is to prevent the occurrence of deformity in the future. To achieve these goals, one good thumb is reconstructed using portions from the supernumerary digit as follows: To increase the diameter of the reconstructed thumb, a skin flap is formed from the supernumerary digit. A portion of the joint from the supernumerary digit is used to correct joint deviation in the reconstructed thumb. The collateral ligament and abductor muscle are detached from the supernumerary digit with the bone fragment into which they are inserted, and transplanted together with the bone fragment into the reconstructed thumb. Osseous tissue to be used in open wedge osteotomy to correct longitudinal deviation is obtained from the phalanx of the supernumerary digit.

Evidence that the accumulation of 5-hydroxytryptamine in the liver but not in the brain may cause the hypoglycaemia induced by 5-hydroxytryptophan.

Experiments were undertaken to determine whether the site of the hypoglycaemic action of 5-hydroxytryptophan (5-HTP), a direct precursor of 5-hydroxytryptamine (5-HT), was in the central nervous system or in the liver. The fall in blood glucose followed the rapid increase in the amount of 5-HT both in the brain and liver after 5-HTP injection into pargyline-treated and non-treated mice. Carbidopa, an inhibitor of peripheral aromatic amino acid decarboxylase, prevented the elevation of 5-HT levels in the liver of both pargyline-treated and non-treated mice. In contrast, carbidopa did not prevent but rather enhanced the elevation of 5-HT levels in the brain of both groups of mice. Corresponding to the prevention of 5-HT elevation in the liver, the fall in blood glucose was prevented by carbidopa. These results support the idea that the accumulation of 5-HT in the liver but not in the brain causes the hypoglycaemia induced by 5-HTP.

A practical heparin reduction algorithm: execution and operational characteristics.

A limited pilot study has been made of a newly devised heparin reduction algorithm (HRA). This formulation is a derivative of the alternative surveillance plan known as the activated partial thromboplastin time after heparin removal (aPTT/HR) scheme. Unlike the traditional plan, the HRA is the first approach to provide information about the individual and collective pharmacological effects of heparin and coumarins when the drugs are administered simultaneously. In this feasibility study the HRA was used without incident in six patients every 24 h to calculate the trend of the evolving anticoagulant effect of coumarin. The computations provided by a laboratory based data management group permitted the clinician to titrate precisely the withdrawal of heparin in response to the daily fluctuations in coumarin effect. In this way, the activated partial thromboplastin time could always be maintained within the desired therapeutic interval. Three divergent patient experiences are presented to demonstrate the operational characteristics and responsiveness of the new HRA plan.

Antitumor activity of aqueous extracts of marine animals.

Aqueous extracts (macromolecule fractions in particular) of 25 species of well-known marine animals were tested for their antitumor activity against transplanted sarcoma 180 solid form in ICR mice. All of the macromolecule fractions from the extracts of animals in the shellfish category except that of Mytilus edulis inhibited the growth of sarcoma 180. The use of aqueous extracts from Strongylocentrotus nudus, Halocynthia hilgendorfi f. ritteri, Styela plicata, Ecteinascidia turbinata, and Megabalanus rosa also revealed relatively high inhibition ratios of over 60%.

Effects of riboflavin-2', 3', 4', 5'-tetrabutyrate on platelet function and plasma lipids.

Riboflavin-2', 3', 4', 5'-tetrabutyrate (B2-But) inhibited, in vitro, ADP (4 micrograms/ml)-induced platelet aggregation when added at the concentration of more than 1 microgram/ml and arachidonic acid (Aa) (1 mM)-induced platelet aggregation when given at the concentration of more than 0.1 microgram/ml. These inhibitions were dose-dependent. Both cyclic AMP and prostaglandin metabolites in the platelets, however, remained unchanged when the amount of B2-But added was between 0.1-10 micrograms/ml. In a volunteer study, 80 mg of B2-But taken in one bolus inhibited both ADP- and Aa-induced platelet aggregation 4 hr after oral administration. The administration of B2-But at this dose was associated with a significant decrease in malondialdehyde formation in plasma. The oral administration of 40 mg (t.i.d.) of B2-But daily did not result in significant changes in ADP- and Aa-induced platelet aggregation. B2-But showed no significant effects on either platelet adhesiveness or collagen-induced platelet aggregation, in vitro or in vivo. Our data suggest that a decrease of lipoperoxide in plasma, not in platelets, influences platelet aggregation induced by ADP and Aa.

[Hyperthermia: microwave hyperthermia in combination with intra-arterial infusion chemotherapy of cancer of the pancreas].

Fourty eight patients with cancer of head of pancreas have been treated by the intra-arterial infusion of 5-FU and Mitomycin C alone or in combination with regional hyperthermia, employing microwave machine operating at a frequency of 2450 MHz. In patients treated with intra-arterial infusion chemotherapy alone the average survival of 4.2 month was attained, whereas the average survival of 9.0 month has been seen in 39 patients treated with infusion chemotherapy and hyperthermia. The hyperthermia treatment has improved the local therapeutic effects and the effects on the survival of the patients. Thanks to this combination therapy 27 of 39 patients (69%) survived more than 6 month, and 12 patients (31%) survived more than one year, longest survival being 7 years. This study confirms that heat administered by the microwave unit potentiated the effects of the intraarterial infusion chemotherapy.

Hypomethylation of c-myc and epidermal growth factor receptor genes in human hepatocellular carcinoma and fetal liver.

The methylation state of cellular oncogenes (c-oncs) and epidermal growth factor (EGF) receptor gene from human liver tissues was examined by means of restriction endonuclease analysis. c-myc and EGF receptor gene from hepatocellular carcinoma and fetal liver were substantially hypomethylated in comparison with those genes from normal liver, while the extents of methylation of c-mos and c-Ki-ras genes were the same among these tissues. It can be speculated that the specific hypomethylation of c-myc and EGF receptor genes may be associated with the development of hepatocellular carcinoma.
