[Effect of dingxin recipe on arrhythmia and injury induced by ischemia and reperfusion in rabbits].

OBJECTIVE: To investigate the protective effect of Dingxin Recipe (DXR) on arrhythmia and injury induced by ischemia and reperfusion. METHODS: Rabbits were randomly divided into four groups and administered orally with DXR of high dose, low dose, propanolol and distilled water. Two-step coronary artery ligature was used to form ischemia and reperfusion model. Superoxide dismutase (SOD), cyclic adenosine monophosphate (cAMP), norepinephrine (NE), dopamine (DA), 5-hydroxytryptamine (5-HT), myocyte ultrastructural injury and occurrence of arrhythmia were investigated. RESULTS: DXR could obviously antagonize arrhythmia induced by myocardial infarction or ischemia and reperfusion in rabbits. Compared with the control group, DXR of high and low doses could improve the changes in ST-T. In rabbits treated with DXR, the levels of SOD were elevated while cAMP, NE, DA, 5-HT and the whole blood viscosity were lowered. CONCLUSIONS: DXR could obviously antagonize arrhythmia and injury induced by ischemia and reperfusion, DXR may clear oxygen free radical, regulate the second messenger, inhibit sympathetic nerve system, improve local circulation, protect mitochondria and prevent the activation of lysosome and safeguard the cardiac myocyte to be free from injury.

[Experimental study of influence of yiqi huoxue serial recipes on basement membrane in membranous nephritis in rabbits].

OBJECTIVE: To clucidate the mechanism of the therapeutic effect of Yiqi Huoxue (YQHX) serial recipes on membranous nephritis. METHODS: Forty-nine New Zealand male rabbits were made to menbranous nephritis model by cation bovine serum albumin and divided into 5 groups, the group A (treated by Qingre Moshen granule), B (treated by Bushen Moshen granule), C (treated by steroid), D (the control group) and E (the normal group). Twenty-four hours' urinary protein content of the animals was determined every week, and plasma albumin, blood lipid, renal function and prostaglandins were tested by the end of experiment. And pathological changes of basement membrane were observed by using light, electronic and immunofluorescent microscopy with polyethylene imine stain. RESULTS: The 24 hours urinary protein content, plasma albumin and blood lipid in the group A and B were lower than those in the control group significantly, P < 0.01 or 0.05, while those in the group C and the D were similar, P > 0.05. In comparing the group A and B with the group C, the difference was also significant, P < 0.05. Light, electronic and immunofluorescent microscopic examination all showed that the pathologic changes in the group A, B and C were lesser than that of the control, the effect was in the order A > B > C. CONCLUSION: YQHX serial recipes can reduce urinary protein content, elevate plasma albumin level, restore the charge barrier effect of and attenuate the immune complex deposition on the basement membrane of glomeruli.

[The expression of bombesin receptor in gastric cancer].

OBJECTIVE: To observe the expression of receptor mRNA and its protein in SGC 7901 cells. METHODS: (1) RT-PCR and Southern blot hybridization were performed for detection of gastrin releasing peptide GRP-R mRNA; (2) Covalent cross-linking study with biotinyl-bombesin and enhanced chemoluminescence (ECL) was performed to identify GRP-R at protein levels in SGC 7901 cells. RESULTS: (1) It is shown that the molecular weight of the cDNA was consistent with that of expected. Southern blot was then used to further prove that specific GRP-R mRNA was expressed by SGC 7901. (2) GRP-R protein was shown by covalent cross-linking in SGC 7901 and the molecular weight of bombesin/GRP-R in SGC 7901 cells was about 75000. CONCLUSION: By using RT-PCR, Southern blot and covalent cross-linking techniques, it was demonstrated that SGC 7901 cells can express bombesin receptor mRNA and protein. The results of this study provides a basis for further researches on bombesin intracellular signal transmission pathway and the role of bombesin and bombesin receptor antagonists in oncological research.

[The mediation effects of endothelin-1 on human mesangial cell proliferation stimulated by thrombin].

OBJECTIVE: To investigate the effects of endothelin-1 (ET-1) on mesangial cell proliferation stimulated by thrombin. METHODS: With cultured human mesangial cells (HMC), the following tests were performed. (1) Stimulative tests: After thrombin stimulation, HMC proliferation was determined by MTT method, concentration of ET-1 in supernatant was measured with radioimmunoassay (RIA) and preproendothelin-1 (ppET-1) mRNA expression of HMC was analysed by Northern blot hybridization. (2) Inhibitory test: The influence of ET-1 inhibitors on the HMC proliferation following thrombin stimulation at three different levels, i.e. ET-1 antiserum, selective ET receptor-type A (ET(A)) antagonist and antisense oligodeoxynucleotide for ppET-1, was measured by MTT test respectively. RESULTS: (1) HMC proliferation was stimulated by thrombin in dose dependent manner and in relation to the time of stimulation. The peak growth level was attained in thrombin concentration of 16 U/ml and incubation time of 16 hours. ET-1 secretion by HMC was increased in parallel with the dose curve and time curve of HMC proliferation; HMC ppET-1 mRNA expression was up-regulated by thrombin 16 U/ml with incubation time of 8 hours and 16 hours respectively (7.9 fold, 3.4 fold versus control group). (2) ET-1 inhibitors, ET-1 antiserum, selective ET(A) receptor antagonist and antisense oligodeoxynucleotide for ppET-1 all significantly inhibit HMC proliferation stimulated by thrombin. CONCLUSION: Thrombin stimulates HMC to produce ET-1 and ET-1 mediates the HMC proliferation.

[Reversal of multidrug resistance of leukemia cell line in vitro by antisense phosphorothioate oligonucleotide].

OBJECTIVE: To investigate the effectiveness of 14 mer antisense phosphorothioate oligonucleotide (AS-SODN) complementary to the published multidrug resistance gene (mdr(1) gene) sequence to reverse multidrug resistance of leukemia cell line. METHODS: 50% inhibitory concentration (IC(50)) of K562 cell line resistant to adriamycin (K562/ADM cell line) was determined by the tetrazolium dye, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method, the concentration of daunorubicin (DNR) in the cell by flow cytometry, the level of P170 by immunohistochemical staining technique and the level of mdr(1)-mRNA by half-quantitative RT-PCR (the ratio of mdr(1)/beta(2)-MG). RESULTS: Treatment of K562/ADM cell line with 2 micromol/L AS-SODN reduced the level of both the mdr(1)-mRNA and P170. The ratio of mdr(1)/beta(2)-MG reduced from (2.07 +/- 0.16) to (1.57 +/- 0.14), and positive rate of P170 expression decreased from (98.26 +/- 1.35)% to (18.30 +/- 6.50)%. Accordingly, the concentration of DNR in cell increased. With these findings, IC(50) to ADM reduced from 25.72 mg/L to 6.97 mg/L and relative efficiency of modulation was 73.01%. CONCLUSION: Antisense oligonucleotide targeting mdr(1) gene can reduce md(1)-mRNA, block P170 expression so as to increase the sensitivity of K562/ADM cell line to chemotherapy drugs.

[A study on the mechanism of the resistance of Shigellae to fluoroquinolones].

OBJECTIVE: To study the mechanism of the resistance of Shigellae to fluoroquinolones. METHODS: A total of 157 clinical isolates were collected. Antibiotic susceptibility testing, reserpine reverse reaction and plasmid conjugation experiment were performed. Accumulation of norfloxacin by shigellae was determined. The N-terminal coding region of gyrA gene was amplified by polymerase chain reaction (PCR), cloned and sequenced in eight fluoroquinolone-resistant isolates and one fluoroquinolone-sensitive isolate as well as in Shigella 51573 (the standard strain). RESULTS: The positive rate of norfloxacin-resistant isolates was 51.2% in the R plasmid conjugation experiment. R plasmid could not mediate resistance to fluoroquinolones. Reserpine could not decrease the minimum inhibition concentration of fluoroquinolones to Shigellae. Norfloxacin uptake by fluoroquinolone-resistant isolates was the same as that by the sensitive one. DNA sequence analysis of gyrA gene from fluoroquinolone-resistant isolates revealed five point mutations that resulted in amino acid change: Gly-81-->Ser, Ser-83-->Leu, Asp-87-->Asn, Asp-87-->Gly, Met-92-->Lys. CONCLUSION: The mutations of gyrA gene were implicated in the resistance of Shigellae to fluoroquinolones.

[K-ras mutation in pancreatic juice from patients with pancreatic carcinoma].

OBJECTIVE: Pancreatic neoplasm, which is considered as one of the most malignant tumors in humans, is now increasing steadily in frequency. The 5-year-survival rate is less than 5%. In order to detect it in early stage, we tried to analyze K-ras mutation in pancreatic juice collected during ERCP after injection of secretin. Mutation of coden 12 in exon 1 of K-ras gene was reported as high as 90% in pancreatic cancer, but much less in benign pancreatic disease. METHODS: 47 patients at Peking Union Medical College Hospital between 1994 and 1998 were enrolled. Pancreatic juice collected during ERCP after injection of 100u secretin, was briefly centrifuged and the sediment was resuspended with PBS. Pancreatic juice supernatant and pancreatic cells were stored at -80 degrees C separately. DNA of the cells and supernatant of the juice were extracted. K-ras point mutation was investigated in the DNA of these supernatant and cells by means of two-stage polymerase chain reaction/restriction fragment length polymorphism (PCR/RFLP). RESULTS: When the supernatant of the pancreatic juice was used, the PCR successful rate was 82%, K-ras mutation rate of the pancreatic carcinomas was 71% (15/21) compared with 0% (0/7) in benign pancreatic disease (3 chronic pancreatitis, 3 insulinoma and 1 pancreatic cyst). When the centrifuged pancreatic cells were used, the successful rate increased to 96%, sixty-five percent (11/17) of pancreatic carcinomas had K-ras mutation; mutation was also detected in 1/7 of the cases with benign pancreatic diseases (an insulinoma). In total, 78% (21/27) of the cases with pancreatic carcinomas has mutant alleles compared with 8% (1/13) of the benign pancreatic diseases (P < 0.001) when pancreatic juice was used to detect K-ras gene point mutation. In terms of the location of the pancreatic cancer, K-ras mutation rate was not statistically different between the head (80%) and the body + tail (75%). The mutation rate did not vary with the size of the tumor. CONCLUSION: Detection of coden 12 of K-ras mutation in pancreatic juice can be used in differentiating pancreatic carcinomas from benign pancreatic diseases. It has high sensitivity and specificity.

[A preliminary study on hepatitis E virus antibody IgG and IgM for the diagnosis of acute hepatitis E].

OBJECTIVE: To evaluate the significance of hepatitis E virus antibody (anti-HEV) IgG and IgM for the diagnosis of acute hepatitis E. METHODS: Acute phase sera from a total of 143 patients with sporadic hepatitis E in 7 cities of China were determined for anti-HEV IgM and IgG by using an enzyme-linked immunosorbent assay (EIA). 359 serial sera of 56 patients with hepatitis E and 68 serial sera of 4 rhesus macaques experimentally infected with hepatitis E virus (HEV) were also detected for anti-HEV IgG and IgM. RESULTS: In the 143 patients the positive rate of anti-HEV IgG was 100.0%, which was significantly higher than that of anti-HEV IgM (73.4%, 105/143). 97.2% of anti-HEV IgG positive patients had a titer over 1:40. The positive rate of anti-HEV IgM increased with the titer of anti-HEV IgG in sera. It was 0% (0/4), 44.4% (8/18) and 80.2% (97/121), respectively in patients with the anti-HEV IgG titer of 1:20, 1:40 and >or=1:80 (P < 0.001). All the anti-HEV IgM positive patients were also anti-HEV IgG positive. No patients were found to be anti-HEV IgM positive alone. Anti-HEV IgG was detected as early as 2 days after onset of the disease, with a cumulative positive seroconversion rate of 100% by 1 month. The negative seroconversion rate of anti-HEV IgG increased with course of the disease and 43.3% of the patients lost their anti-HEV IgG by 6 months after illness. Though anti-HEV IgM also seroconverted at the same time as anti-HEV IgG, its cumulative positive seroconversion rate was only 71.4% and declined rapidly. Up to 37.5% of anti-HEV IgM positive patients became negative by 1 month after onset. Similar antibody responses were observed in 4 rhesus macaques experimentally infected with HEV. CONCLUSION: It is suggested that as a result of the poor sensitivity of currently available anti-HEV IgM EIA kits, anti-HEV IgG will be a more reliable marker for the diagnosis of acute hepatitis E as compared with anti-HEV IgM.

[Changes of apoptosis and Fas gene expression in cardiomyocytes of rats with myocardial reperfusion and the effects of ischemic preconditioning].

OBJECTIVE: To study the changes of apoptosis and Fas gene expression in cardiomyocytes of rats with different duration of reperfusion after myocardial ischemia and the effects of ischemic preconditioning (IPC). METHODS: 108 rats were divided randomly into 6 groups, i.e. sham-operated (observing for 24h in operated control), 30min of ischemia followed by 6h of reperfusion(I(30 min) R(6h)), I(30min)R(12h), I(30min)R(24h), I(30min)R(48h) and IPC. The myocardial cell apoptosis was determined with terminal deoxynucleotidyl transferase-mediated dUTP-fluorescein nick end labeling (TUNEL) method; The protein and mRNA expression of Fas gene were studied with S-P immunohistochemical staining and RT-PCR analysis respectively. RESULTS: Cardiomyocyte apoptosis and the level of Fas gene expression varied with the duration of reperfusion. The apoptotic index (AI) and protein expression index (PEI) of Fas gene in myocytes were highest in rats with I(30min)R(48h) [AI: (38.15 +/- 13.26)%; Fas PEI: (24.77 +/- 12.92)%], but mRNA induction peaked at I(30min)R(24h) (Fas/beta-actin: 0.76 +/- 0.21); PEI of Fas gene correlated significantly with AI of myocytes (r = 0.87, P < 0.01); IPC decreased myocyte apoptosis and down-regulated the protein expression of Fas gene (all P < 0.05). CONCLUSION: The number of apoptotic myocytes and the expression level of Fas gene varied with different duration of myocardial reperfusion. The change of apoptosis and Fas gene expression may be involved in the process of myocardial ischemia reperfusion injury. IPC prevent myocardial injury partly by inhibiting myocyte apoptosis and down-regulating protein expression of Fas gene.

[The PCNA immunohistochemical study of pterygium].

OBJECTIVE: To study the proliferating status of pterygium and the drug's influence on it. METHOD: Fifty four patients with pterygium were randomly assigned into three groups: Before the surgical excision of the pterygium, group 1 (18 cases) had received no drug, group 2 (16 cases) received local dexamethasone treatment, and group 3 (20 cases), local homoharringtonine and dexamethasone treatment. The immunohistochemical study on proliferating cell nuclear antigen (PCNA) was performed on the surgical specimens of the pterygium. RESULT: The PCNA positive rate was (11.53 +/- 3.62)% in the untreated group, (10.28 +/- 4.18)% in the dexamethasone group (P > 0.05), and (5.59 +/- 3.82)% in the homoharringtonine and dexamethasone group (P < 0.01). The PCNA positive rate was higher in the younger patients (< 60 years) with progressive type of the disease than that in the elder patients (>or= 60 years) with quiet type. CONCLUSIONS: There are cell proliferation and PCNA expression in pterygium. The homoharringtonine and dexamethasone local injection before the excision of pterygium can effectively decrease the PCNA expression and recurrence rate of pterygium. Local injection of dexamethasone before surgical excision has no influence on the PCNA expression of pterygium.

[Effects of environmental lead exposure in kindergartens on children's blood lead level].

OBJECTIVE: To determine if kindergartens' environmental status can influence children's lead exposure. METHODS: Environmental specimens, such as floor dust, peeled-off paint chip, soil and drinking water, as well as children's hand dust and blood samples, were collected and measured for their lead levels in 19 kindergartens, to analyze the relationship between children's blood lead levels and their environmental lead exposure. RESULTS: Geometric means of lead concentrations in indoor floor dust, peeled-off paint chip, dust fallout, outdoor floor dust, soil and drinking water were 86.5 microg/m(2), 235.5 microg/g, 445.9 microg/g, 172.4 microg/m(2), 70.1 microg/g and 12.5 microg/L, respectively. Lead level on children's hands averaged 3.4 microg at both hands. Blood lead levels in children correlated positively with the lead concentrations of outdoor floor dust and their hand dust, with correlation coefficients of 0.5186 and 0.2206, respectively. Multiple regression analysis showed that hand dust lead level in children entered the regression model with a largest standardized partial regression coefficient of 0.3842 and a coefficient of determination of 0.673 for the full equation with F = 6.52 and P < 0.01. CONCLUSION: Status of environmental health in kindergartens plays an important role in children's lead exposure. It is necessary to offer health education for children and make them wash their hands often and overcome unhealthy behavior of sucking their fingers.

[Effects of lead exposure on their brain-stem auditory evoked potential in children].

OBJECTIVE: To determine if low lead exposure can influence auditory development in children. METHODS: Children were divided into two groups, i.e., low (< 0.483 micromol/L) and high (>or= 0.483 micromol/L) blood lead levels with atomic spectrophotometry (AAS). Latencies and latent intervals between peak waves of their brain-stem auditory evoked potentials (BAEP) in the two groups were measured and compared. RESULTS: There was positive correlation between blood lead levels and BAEP in wave III at right ear, wave V at both right and left ears and latent intervals between peak waves I to V, with correlation coefficients of 0.2991, 0.2748, 0.3248, 0.2064 and 0.2412, respectively. There was significant difference in latencies of wave III at right ear, wave V at both right and left ears and latent intervals between peak waves I to V at both right and left ears and those between waves of III to V at right ear in the low blood lead group. Latencies and latent intervals between peak waves were longer in the high blood level group (with a mean of 0.78 micromol/L and a range of 0.58 to 1.42 micromol/L) than those in the low one (with a mean of 0.30 micromol/L and a range of 0.17 to 0.45 micromol/L). CONCLUSION: Elevated blood lead level could cause prolongation of latencies and latent intervals between peak waves of BAEP and slowdown of the nerve conduction velocity, and so auditory development was influenced.

[The detection of microsatellite instability in carcinoma and adenoma of large bowel and its significance].

OBJECTIVE: To explore the role of microsatellite instability (MSI) in colorectal carcinogenesis and the relationship between MSI, proliferation activity and prognosis. METHODS: PCR-SSLP and immunohistochemistry methods were used to detect MSI and the expression of PCNA in 56 cases of carcinomas, 9 cases of adenomas and 6 cases of adenomas with malignant changes. RESULTS: The total positive rate of MSI were 25/56 cases in colorectal carcinomas. The MSI positive cases were 3/4 in HNPCCs, 22/52 in sporadic colorectal carcinomas, 2/9 in adenomas and 2/6 in adenoma with malignant changes respectively. The PCNA labelling index of MSI positive tumors were significantly lower than that of MSI negative tumors (P < 0.01). The 3 and 5 year survival rates in patients with MSI positive colorectal carcinomas were higher than those of MSI negative tumors. CONCLUSION: MSI may be an early molecular alteration and another molecular mechanism in colorectal carcinogenesis. The MSI positive tumors have low proliferating activity and a better clinical outcome. The detection of MSI might be useful in predicting prognosis of colorectal carcinomas.
