[Effects of gallbladder ruptured under laparoscopic cholecystectomy on pulmonary function].

To assess effects of gallbladder ruptured and bile running out under laparoscopic cholecystectomy on pulmonary function, we preprospectively studied 20 patients without gallbladder rupture (group A), 10 with gallbladder rupture (group B), and 10 open cholecystectomy (group C). Significant differences were found in the group B and C to all pulmonary function values compared with preoperative values until the 5th to 7th postoperative day (P < 0.05). The group A showed only a significant difference on the first day after operation (P < 0.05). There are no significant differences between groups B and C. Comparing the group A with the group B and C respectively showed a significant differences from the 1st to 7th day (P < 0.05). This study confirms that the group A is superior to the groups B and C. Postoperative peritonitis is also an important cause of pulmonary dysfunction to reduce small airway function. Performing LC, we should try to avoid rupture of gall bladder or hemorrhage to fasten the recovery of patients' pulmonary function and reduce the chance of pulmonary complications.

[Long-term effects of stapes elevation].

To assess the clinical value of stapes elevation, 38 cases had been followed-up for 1-12 years (average 6.7 years). The results revealed that hearing improvement rate (over 10 dB) was 94.7% in short term and 86.8% in long term follow-up. The long term regression rate was similar to that in stapedectomy, and no severe sensorineural hearing loss occurred. In addition, the vestibular reaction was slight, especially in reoperated cases. The stapes elevation was easy to perform as compared with stapedectomy. Therefore, this technique may be the first choice for stapes surgery.

[Relationship between human cytomegalovirus infection and soluble interleukin-2 receptor level].

In order to study the relationship between human cytomegalovirus infection and soluble interleukin-2 receptor (sIL-2R) level, 104 serum samples were selected from women of childbearing age in the city of Wuhan and examined with ELISA for anti-HCMV antibody IgG and IgM. Then we detected the sIL-2R level with double antibody ELISA in these sera with different conditions of HCMV infection. The results of different groups were compared with those women without cytomegalovirus infection and also compared with each other. The positive rate for anti-HCMV antibody IgG in the women of Wuhan was 89.4% (93/104) and for anti-HCMV antibody IgM only was 9.6% (10/104). The sIL-2R level of these women with HCMV infection was significantly higher than that in the control group (178.1 +/- 57.3 U/ml), P > 0.05. The highest level of sIL-2R were in those women whose anti-HCMV antibody IgM were positive (910 +/- 465.6 U/ml) and secondly was in those with anti-HCMV antibody IgG, IgM positive (905 +/- 347.8 U/ml). They were both higher than that in those women only with anti-HCMV antibody IgG positive (446.8 +/- 158.9 U/ml), P > 0.05. The results showed that HCMV infection can raise the sIL-2R level in serum of childbearing age women, especially in those with active HCMV infection. sIL-2R may be one of the pathogenic factors of HCMV.

[Dual wavelength multiplication subtractive spectrophotometric determination of sulfadiazinum and trimethoprimum in compound sulfadiazine tablets].

This paper described the simultaneous determination of sulfadiazine (SD) and trimethoprim (TMP) in tablets co-Trimediazini by dual wavelength spectrophotometry multriplication subtractive method. This method was based on the measurement of both absorbance of standard solution of SD (8-14) x 10(-6)g/mL, TMP (1-2) x 10(-6)g/mL and sample solution at wavelength 242.5 nm and 228.0nm. The average recovery and RSD of SD and TMP were 100.60%, 0.33% and 98.27%, 1.58%, respectively (n=20). The method was simple, quick and results satisfactory.

Linkage of a microsatellite marker to the canine copper toxicosis locus in Bedlington terriers.

OBJECTIVE: To identify a DNA marker for the copper toxicosis (CT) locus in Bedlington Terriers (BT). ANIMALS: 77 BT, of which 25 were affected. Diagnosis of affected or unaffected with CT was made in all cases by quantitative copper determinations on liver biopsy samples by use of established criteria. PROCEDURE: BT pedigrees segregating for CT were identified. Linkage studies were carried out using polymorphic microsatellite markers developed for the canine genome in these pedigrees. DNA was isolated from blood samples of pedigree members. Polymerase chain reaction was used to amplify and type alleles at 213 microsatellite loci in each dog, and findings were subjected to linkage analysis. RESULTS: One microsatellite marker was identified to be closely linked to CT with logarithm of odds score of 5.96 at a recombination fraction of zero. CONCLUSIONS: Using the linked marker, it has become possible to distinguish affected, homozygous normal, and carrier dogs in some BT pedigrees. CLINICAL RELEVANCE: In informative pedigrees where the marker is variable in the parents, it is possible to identify which dogs will require anticopper therapy and provide breeders with sound scientific advice about breeding strategies.

[Optokinetic nystagmus induced by moving compound gratings].

Optokinetic Nystagmus (OKN) induced by moving compound gratings was investigated. It was found that OKN sometimes tracks the coherent motion of the rigid plaid in a single direction, and sometimes tracks alternately the transparent motion of two component gratings in different directions, as Dual Alternate OKN. It was also found that the duration of OKN tracking coherence increases as the moving speed of the component gratings decreases, and as the angle between their moving directions decreases. The results also showed that the effect of angle plays a dominant role.

[Effect of biological electric stimulation on free muscle transfer].

The rectus femoris muscles of rabbits were used as muscle model. The electrical stimulation which resembled the normal motor-unit activity was used to observe its effects on free transferred muscle. After three months, the moist muscle weight (MW), its maximum cross-section area, its contractility and its histochemical characteristics were examined. The results showed that the function and morphology of the muscles were well preserved. These findings might encourage its clinical application.

[No point mutation of the 2.8 kb EcORI fragment of the nasopharyngeal carcinoma transforming gene TX in nasopharyngeal carcinoma].

We have previously cloned a nasopharyngeal carcinoma transforming gene from human nasopharyngeal carcinoma cell line CNE-2 using mouse JB6 cell line as the recipient cells. This transforming gene, designated Tx, exhibits moderate transforming activity upon transfection into JB6 cells. Several lines of evidence indicate that the 2.8 kb EcoRI fragment which lies in the middle of the Tx gene is responsible for the transforming activity. The sequence of the 2.8 kb EcoRI fragment was determined, which showed striking homology with the human immunoglobulin light chain C region gene. Using the modified polymerase chain reaction-single strand conformation polymorphism analysis (PCR-SSCP) silver staining technique, we studied the possible point mutations of the nasopharyngeal carcinoma gene Tx in nasopharyngeal carcinoma. No mobility shift was detected in eleven paired cases of nasopharyngeal carcinoma biopsies suggesting that in most cases of nasopharyngeal carcinoma, the Tx gene is not activated via a mode of point mutation. Taken together with the previously obtained results we have found that the Tx gene is not activated by gene rearrangement or gene amplification. Therefore, we consider that the Tx gene may have been activated by its interaction with other regulatory factors.

[A study on detecting the gene deletion or loss of heterozygosity by polymerase chain reaction].

The authors investigated the possibility of using PCR to dsetect the deletion of gene or loss of heterozygosity (LOH) of gene in biopsies. With some aids, such as selection of PCR cycle number, choice of internal control and screening of samples, the authors detected the deletion of GSH Clone 7 in nasophryngeal carcinoma (NPC) by PCR-Southern, and successfully found one case which partially deleted GSH Clone 7. This results was confirmed by image densitometry. Through this study, an available method to detect the deletion of gene by PCR is provided.

[Pre- and post-operative changes in serum levels of glycocholic acid and 3,5,3'-triiodothyronine and their clinical significance in patients with cholelithiasis].

Pre- and post-operative changes of serum glycine-conjugated cholic acid (CG) and 3,5,3'-triiodothyronine (T3) levels were observed in twenty-two patients with cholelithiasis. The increase of the levels of serum CG varied with the patients; the highest levels were seen in patients with cirrhosis. After operation, the levels of serum CG and T3 were decreased significantly in patients without cirrhosis. There was positive correlation between serum CG and T3 (r = 0.4667, P < 0.01). It is indicated that the lowering of serum CG after operation may be related to the lowering of serum T3; the significant increase of serum CG level is useful to the diagnosis of liver cirrhosis, which is more sensitive than Type B ultrasonography or routine examination of liver function.

In vivo angiogenic activity and hypoxia induction of heterodimers of placenta growth factor/vascular endothelial growth factor.

To investigate the in vivo angiogenic activity of placenta growth factor (PIGF) and its heterodimers with vascular endothelial growth factor (VEGF), the induction of neovascularization of these factors in the mouse cornea was studied. VEGF165 is sufficiently potent to stimulate new capillary growth from the limbal vessels. PIGF129/VEGF165 heterodimers also induce corneal neovascularization with a maximal vessel length similar to VEGF165, but with a marked decrease of vessel density. In contrast, PIGF129 has little or no effect in this in vivo angiogenesis assay. The expression of VEGF mRNA and protein is drastically up-regulated by hypoxia in choriocarcinoma cells, whereas expression of PIGF is not affected by the low concentration of oxygen. Up-regulation of VEGF production results in increased formation of PIGF/VEGF heterodimers in these tumor cells. Thus, hypoxia indirectly up-regulates expression levels of PIGF/VEGF heterodimers and modulates VEGF activity when these factors are co-expressed.

Kringle domains of human angiostatin. Characterization of the anti-proliferative activity on endothelial cells.

Recently we have identified angiostatin, an endogenous angiogenesis inhibitor of 38 kDa which specifically blocks the growth of endothelial cells (O'Reilly, M. S., Holmgren, L., Shing, Y., Chen, C. , Rosenthal, R. A., Moses, M., Lane, W. S., Cao, Y., Sage, E. H., and Folkman, J. (1994) Cell 79, 315-328; Folkman, J. (1995) Nat. Med. 1, 27-31). Angiostatin was shown to represent an internal fragment of plasminogen containing the first four kringle structures. We now report on the inhibitory effects of individual or combined kringle structures of angiostatin on capillary endothelial cell proliferation. Recombinant kringle 1 and kringle 3 exhibit potent inhibitory activity with half-maximal concentrations (ED50) of 320 nM and 460 nM, respectively. Also, recombinant kringle 2 displays a significant inhibition, although decreased compared with both kringle 1 and kringle 3. In contrast, kringle 4 is an ineffective inhibitor of basic fibroblast growth factor-stimulated endothelial cell proliferation. Among the tandem kringle arrays, the recombinant kringle 2-3 fragment exerts inhibitory activity similar to kringle 2 alone. However, relative to kringle 2-3, a marked enhancement in inhibition is observed when individual kringle 2 and kringle 3 are added together to endothelial cells. This implies that it is necessary to open the cystine bridge between kringle 2 and kringle 3 to obtain the maximal inhibitory effect of kringle 2-3. An increased (<2-fold) inhibitory activity is observed for the kringle 1-3 fragment (ED50 = 70 nM) compared with kringle 1-4 (ED50 = 135 nM). These data indicate that the anti-proliferative activity of angiostatin on endothelial cells is shared by kringle 1, kringle 2, and kringle 3, but probably not by kringle 4 and that more potent inhibition results when kringle 4 is removed from angiostatin. Thus, in view of the variable lysine affinity of the homologous domains, it would appear that lysine binding capability does not correlate with the relative inhibitory effects of the kringle-containing constructs. However, as we also demonstrate, appropriate folding of kringle structures is essential for angiostatin to maintain its full anti-endothelial activity.

Presence of mRNA for glutamic acid decarboxylase in both excitatory and inhibitory neurons.

Neurons in very low density hippocampal cultures that are physiologically identified as either GABAergic inhibitory or glutamatergic excitatory all contain mRNA for the gamma-aminobutyric acid (GABA) synthetic enzyme, glutamic acid decarboxylase (GAD), as detected by single cell mRNA amplification and PCR. However, consistent with the physiology, immunocytochemistry revealed that only a subset of the neurons stain for either GAD protein or GABA. A similar fraction hybridize with RNA probes for GAD65 and GAD67. Hippocampal CA1 pyramidal neurons in slice preparations, which are traditionally thought to be excitatory, also contain mRNA for GAD65 and GAD67. Hippocampal neurons in culture did not contain mRNA for two other neurotransmitter synthesizing enzymes, tyrosine hydroxylase, and choline acetyl transferase. These data suggest that in some neurons, presumably the excitatory neurons, GAD mRNA is selectively regulated at the level of translation. We propose that neurotransmitter phenotype may be posttranscriptionally regulated and neurons may exhibit transient phenotypic plasticity in response to environmental influences.

Spinal cord repair in adult paraplegic rats: partial restoration of hind limb function.

Complete spinal cord gaps in adult rats were bridged with multiple intercostal nerve grafts that redirected specific pathways from white to gray matter. The grafted area was stabilized with fibrin glue containing acidic fibroblast growth factor and by compressive wiring of posterior spinal processes. Hind limb function improved progressively during the first 6 months, as assessed by two scoring systems. The corticospinal tract regenerated through the grafted area to the lumbar enlargement, as did several bulbospinal pathways. These data suggest a possible repair strategy for spinal cord injury.
