Detection of p53 gene mutations in human leukemia by PCR-SSCP analysis and direct nucleotide sequencing.

OBJECTIVE: To look for mutations of the p53 gene in leukemic patients and study the relationship between abnormalities in p53 gene and leukemogenesis. METHODS: The peripheral blood and Bone Marrow Samples were collected from 36 patients with various leukemia types including 14 cases of lymphocytic leukemia [8 cases of acute lymphocytic leukemia (ALL), 4 cases of chronic lymphocytic leukemia (CLL), 2 cases of hairy cell leukemia (HCL)] and 22 cases of myelocytic leukemia [11 cases of acute nonlymphocytic leukemia (ANLL), 11 cases of chronic myelocytic leukemia (CML)]. DNA structures of exon 5-8 of the p53 gene were scanned by PCR-SSCP (single strand conformation polymorphism analysis of polymerase chain reaction products). The appropriate DNA fragments were amplified, Purified and sequenced directly. RESULTS: By PCR-SSCP analysis, shifts in electrophoretic mobility of the p53 gene were detected in 3 of 14 patients with lymphocytic leukemia (2 ALL and 1 CLL), but none in 22 patients with myelocytic leukemia including one in blastic crisis. Direct nucleotide sequencing in one patient with ALL showed transition of CTG to CAG at codon 257 of exon 7, resulting in a change of its encoded amino acids from aspartic acid to valine. To our knowledge, the mutation at this codon has not been previously reported hitherto. CONCLUSIONS: The p53 gene mutations are specifically associated with lymphocytic leukemia. Alternations of the p53 gene may play a certain role in leukemogenesis in some cases of lymphocytic leukemia.

[The characteristics of plasma--sprayed coatings of hydroxy apatite in vitro].

In order to investigate the stability of Hydroxyapatite (HA) coated material, the plasma-sprayed coatings of HA were divided into four groups: 1. Keeping in water vapour at 125 degrees C, with a pressure of 0.15 MPa for 6 hr; 2. Heating at 650 degrees C in air for half an hr; 3. Keeping in water vapour at 490 degrees C, with a pressure of 0.01 MPa for 2 hr; and 4. The control. The XRD, FTIR analysis and the dissolution test were carried out. The results showed: 1. The degree of crystalization in XRD analysis was 3. > 1. > 2. > 4.; 2. The ampitude of OH- peak in FTIR analysis was 3. > 1. > 2. > 4. and 3. The dissolution rate in tris-HCl buffer was 3. < 1. < 2. < 4. The conclusions were 1. The treating with water vapour could decrease the transformation temperature which was needed to convert the amorphous phase into cystalline phase; 2. Water vapour treatment could accelerate the transformation of Z-TCP, TCPM into crystalline HA; 3. Water vapour treatment could promote the structural integrity of plasma--sprayed coated of and HA and 4. Water vapour treatment could lower the dissolution rate of HA coated in Tris-HCl buffer.

[Effects of high frequency jet ventilation on respiratory airflow and gas exchange in dogs with inhalation injury].

OBJECTIVE: The experiment was to investigate the effects of respiratory airflow on gas exchange. METHOD: Eight healthy dogs were anesthetized with thiopentone. A 3-D partial dynamic analyzer was used to determine the inspiratory and expiratory flow velocity, and arterial blood samples were collected for blood-gas analysis during spontaneous respiration. Intravenous muscle relaxant was given quickly and the animals were kept apneic and then vapor (105 degrees C) was inhaled for 5 seconds. HFJV was carried out at random under 5 different conditions, each for 20 minutes. Changes in inspiratory velocity (Vi), expiratory velocity (Ve), Reynole's number, PaCO2, PaO2, and PIP were observed. RESULT: 1. During spontaneous respiration, airflow was in form of laminar flow when Re < 1000, and expiratory airflow was either laminar or turbulent when 1000 < Re < 1500. 2. During HFJV, both expiratory and inspiratory airflow were turbulent when Re > 1500. 3. The expiratory velocity with HFTJV increased significantly (P < 0.05). PaCO2 decreased significantly (P < 0.05) compared with HFJV. Obvious negative correlation was found between them (r = -0.9216, P < 0.05), meanwhile inspiratory velocity had an increasing tendency. CONCLUSION: 1. HFJV could maintain normal ventilation of dogs with inhalation injury. The mechanism might be associated with turbulent flow. 2. On the basis of HFJV, HFTJV increases inverse jet airflow, thus the respiratory airflow velocity may be increased and CO2 elimination improved.

Immunohistochemistry and reverse transcription-polymerase chain reaction for detecting adrenomedullin in the central nervous system.

OBJECTIVE: To demonstrate the presence of adrenomedullin in the central nervous system. METHODS: Using immunohistochemical method (ABC) and reverse transcription-polymerase chain reaction (RT-PCR) analysis, the distribution of immunoreactive (IR-) AdM and AdM mRNA was observed in the rat and the human brain. RESULTS: Immunoreactive (IR-) AdM and AdM mRNA were detectable in almost every region of the rat brain examined, including cerebral cortex, paraventricular tissues, hypothalamus, mesencephalon, medulla oblongata and cerebellum. The levels of AdM mRNA in paraventricular tissues and medulla oblongata were higher than those in the other brain regions. AdM mRNA was also detectable in the human brain. CONCLUSION: The present study shows that AdM is present in the central nervous system, suggesting that AdM might act as a neurotransmitter, neuromodulator or neurohormone in rats or humans.

[The experimental research of inner ear metabolism and electrical physiology of autoimmune sensorineural hearing loss].

Inquire into the mechanism of inner ear pathological physiology in autoimmune sensorineural hearing loss (ASHL). With the auditory electric-physiological techniques and enzyme-histochemical method, the change of inner ear hearing function and enzyme activity were observed. These animals, which threshold of auditory nerve compound active potential (CAP) and cochlear microphonic potential(CM) heightening evidently, showed that the amplitude of endolymphatic potential(EP) (include-EP) bring down in various degrees, which was related to the change of the active of Na(+)-K(+)-ATPase and SDH in vascularis stria and endolymphatic sac. The abnormality of enzymes metabolism in inner ear tissues, which following autoimmune inflammation damage, is the pathological foundation of hearing dysfunction.

[Cochlear and retrocochlear deafness induced by immunity with different inner ear tissue antigens].

OBJECTIVES: To inquire into whether the different inner ear tissue antigens could cause different kind of hearing loss, and to find out the disorder positions of auditory system. METHODS: The basilar membrane (BM), spiral ligament (SL), and spiral ganglion (SG) of guinea pigs were removed for making antigens, respectively. Then, we used these antigens to immune guinea pigs. The special humour and cellular immune reaction, hearing function, and inner ear histopathological changes were observed. RESULTS: In BM-antigen and SL-antigen immune group, the various degrees of cochlear microphonic potential disturbance, recruitment, and immune pathological inflammation in cochlear duct and stria vascularis were found. In SG-antigen immune group, auditory nerve compound action potential changes were prominent, and the inner ear pathological damage mainly existed in cochlear axis vessels or surround areas, and SG. CONCLUSIONS: It was confirmed that the inner ear antigens come from different part of auditory system, which could cause cochlear or retrocochlear autoimmune disease.

[Determination of linear sintering shrinkage rate of slip-casting infiltrated ceramic].

This study tested the linear sintering shrinkage (SS) rate of ceramic infiltrated with different glasses (Vita In-ceram, G I-I La, G I-II La, G I-II Ce), the results showed that the linear SS rate was 0.08%-0.32%, which occurred mainly in the firing stage of alumina (P < 0.05) and glass infiltration had no influences on the dimensional stability of ceramic(P > 0.05). It suggests that the effects of glass infiltration on the dimensional stability of GI glasses were comparable with that of Vita In-ceram.

[Assessment of marginal tissue around HA-coated titanium implants for orthodontic anchorage in dogs].

12 two-stage HA-coated titanium implants placed in dogs' mandibles were loaded with an orthodontic force of 150 g for 3 month. Gingival bleeding index, plague and calcucus index, mobility index, probing depth and marginal bone were investigated. The results showed that the implants were stable and peri-implant soft tissue displayed slight inflammation, but no marginal bone absorption and no difference of bone quantity between the tension side and compression side were found. It can be concluded that the clinical trials of marginal tissue around the HA-coated titanium implants are satisfactory when the implants are used as anchorage for short-term orthodontic treatment.
