The Gcn4p activation domain interacts specifically in vitro with RNA polymerase II holoenzyme, TFIID, and the Adap-Gcn5p coactivator complex.

The Gcn4p activation domain contains seven clusters of hydrophobic residues that make additive contributions to transcriptional activation in vivo. We observed efficient binding of a glutathione S-transferase (GST)-Gcn4p fusion protein to components of three different coactivator complexes in Saccharomyces cerevisiae cell extracts, including subunits of transcription factor IID (TFIID) (yeast TAFII20 [yTAFII20], yTAFII60, and yTAFII90), the holoenzyme mediator (Srb2p, Srb4p, and Srb7p), and the Adap-Gcn5p complex (Ada2p and Ada3p). The binding to these coactivator subunits was completely dependent on the hydrophobic clusters in the Gcn4p activation domain. Alanine substitutions in single clusters led to moderate reductions in binding, double-cluster substitutions generally led to greater reductions in binding than the corresponding single-cluster mutations, and mutations in four or more clusters reduced binding to all of the coactivator proteins to background levels. The additive effects of these mutations on binding of coactivator proteins correlated with their cumulative effects on transcriptional activation by Gcn4p in vivo, particularly with Ada3p, suggesting that recruitment of these coactivator complexes to the promoter is a cardinal function of the Gcn4p activation domain. As judged by immunoprecipitation analysis, components of the mediator were not associated with constituents of TFIID and Adap-Gcn5p in the extracts, implying that GST-Gcn4p interacted with the mediator independently of these other coactivators. Unexpectedly, a proportion of Ada2p coimmunoprecipitated with yTAFII90, and the yTAFII20, -60, and -90 proteins were coimmunoprecipitated with Ada3p, revealing a stable interaction between components of TFIID and the Adap-Gcn5p complex. Because GST-Gcn4p did not bind specifically to highly purified TFIID, Gcn4p may interact with TFIID via the Adap-Gcn5p complex or some other adapter proteins. The ability of Gcn4p to interact with several distinct coactivator complexes that are physically and genetically linked to TATA box-binding protein can provide an explanation for the observation that yTAFII proteins are dispensable for activation by Gcn4p in vivo.

Quality of commercial shelf-stable soymilk products.

Four brands of shelf-stable soymilks (three lots each) were evaluated for their quality. All samples were tested for trypsin inhibitor activity, microbial load (coliforms, aerobic mesophiles, and psychrotrophs), solids content, pH, color, and viscosity. Storage stability at 5 degrees C of the opened original containers was evaluated by microbiological analyses. Physical properties were consistent among three lots of the same brand, but varied among brands. Trypsin inhibitor activity ranged from 3.43 to 10.35 mg/g dried soymilk. The pH values of the soymilks varied between 6.41 and 7.34. Viscosity was measured between 16.5 and 26.4 mPa. Solids content varied considerably among soymilks, from 6.88 to 12.37% (wt/vol). Freshly opened commercial soymilk contained fewer than one microorganism per ml in all microbial counts. During storage at 5 degrees C, microbial counts increased sharply after 2 to 3 weeks. A single microorganism predominated in each stored sample. Shelf life (defined as the time during which aerobic mesophile counts remained below 10(6) CFU/ml) varied from 3 to 4 weeks. The appearance and pH of soymilks remained unchanged even after 4 weeks of storage. Five types of microorganisms were found in stored soymilk samples; none of them produced acid.

Enhancement of ADP-induced aggregation by 5-HT in rabbit platelets.

AIM: To study the enhanced effects of 5-hydroxytryptamine (5-HT) on ADP-induced aggregation. METHODS: Platelet aggregation was quantified by the light transmission, the cytosolic-free calcium ([Ca2+]i) was measured by digital fluorescent microscopy, and inositol 1,4,5-triphosphate (IP3) was determined by receptor binding assay. RESULTS: In rabbit platelet-rich plasma (PRP), 5-HT 0.03-3 mumol.L-1 induced a decrease in light transmission (DLT) in a concentration-dependent manner with centralization of granules, as revealed by electron microscopy. The DLT was accompanied with neither platelet aggregation nor a release reaction. In single washed platelets loaded with Fura-2, 5-HT caused a concentration-dependent elevation of [Ca2+]i, and IP3 level was also transiently increased in washed platelets at 15 s after stimulation by 5-HT. Adenosine diphosphate (ADP) also caused DLT transiently in PRP before its own aggregation without a release reaction. Pretreatment of PRP or washed platelets with 5-HT, the DLT by ADP was reduced concentration-dependently and ADP-induced aggregation and [Ca2+]i mobilization were enhanced. CONCLUSION: The enhancement of ADP-induced aggregation was attributed to the superimposition of the calcium release from the storage sites and calcium influx induced by ADP over the calcium release from the storage sites by 5-HT.

[The effects of nimodipine on cerebrovascular function in acute cerebral infarction].

OBJECTIVES: To use cerebrovascular hemodynamics indexes (CVDI) to evaluate the effects of nimodipine on cerebrovascular function in acute cerebral infarction (ACI), and to investigate the changes of blood pressure (BP), heart rate (HR), and electrocardiography (ECG). METHODS: 30 patients with ACI in internal carotid artery system within 72 h from the oneset of symptoms were given nimodipine (10 mg, 0.25-0.5 microgram/kg/min) by intravenous drip (i.v.) every day for 14 days. CVDI, BP, HR and ECG were examined at various times during treatment. RESULTS: Nimodipine improved cerebrovascular function significantly. Cerebral blood velocity and flow increased, cerebrovascular resistance and criticle pressure decreased, and cerebrovascular elasticity and autoregulation improved. BP dropped significantly at the third day, and mainteined stable afterward. HR dropped remarkably at the fourteen day (P = 0.011). It was reasonable that the treatment of nimodipine (i.v.) lasted 10 days or so. CONCLUSIONS: Nimodipine improves cerebrovascular function significantly, but monitoring of BP and ECG is necessary during its application.

Polymorphism of the glycogen synthase gene and non-insulin-dependent diabetes mellitus in Chinese population.

OBJECTIVE: To investigate the possible association between the Xbal polymorphism of the glycogen synthase gene and non-insulin-dependent diabetes mellitus (NIDDM) in Chinese population. METHODS: 216 NIDDM patients and 106 healthy controls were studied. DNA fragment containing Xbal restriction site was amplified by the polymerase chain reaction, digested by the Xbal enzyme and compared by gel electrophoresis with a positive control from Finland. RESULTS: The Xbal polymorphism was found in 18 of 216 Chinese patients (8.3%) and was also found in 10 of 106 controls (9.4%) (P > 0.05). CONCLUSIONS: The results suggest that the Xbal polymorphism of the glycogen synthase gene could not be used as a genetic marker for NIDDM in Chinese population of Henan Han nationality.

[Studies on red orpiment induction of NB4 and HL-60 cell apoptosis].

OBJECTIVE: To study the possible mechanism of red orpiment, which is main component of composite indigo naturalis tablets, in the treatment of acute promyelocytic leukemia(APL). METHODS: The effect of red orpiment on induction of APL cell line NB4 and HL-60 apoptosis were studied by cell morphology, DNA gel electrophoresis and flow cytometry assay. RESULTS: Red orpiment induced NB4 and HL-60 cell apoptosis. When treated with different concentration of red orpiment(25-200 micrograms/ml) for 16 hours, both NB4 and HL-60 cells showed typical apoptosis features. If decreased the concentration of red orpiment to 12.5 micrograms/ml, the NB4 cell still showed apoptosis features while the HL-60 cell did not when cultured for 72 hours. Arsenic disulfide(As2S2) had the same effect as red orpiment did under the same experiment condition. CONCLUSION: It is the main component, As2S2 of the red orpiment that can induces NB4 and HL-60 cell apoptosis. and the red orpiment is responsible for the high CR rate of APL induced by the composite indigo naturalis tablets.

[Validity of Sternberg dualtask in evaluating flight ability and effect of educational factor].

Validity of the Sternberg dualtask (tracking task and remember) for evaluating flight ability of military pilots, and the influencing factors were studied in 112 grade pilots with different educational levels, ages and kinds of planes. The evaluations of their flight ability were made by their leaders using the multiple-criterion evaluation system. The results showed: the Sternberg dualtask can be used to evaluate the abilities of rapid reaction of operating fighters; the influence of the educational level and age should be considered during evaluating flight ability. It suggests that the Sternberg dualtask is very effective in evaluating flight ability, and it shows that fighter pilots with high educational level, age from 25 to 29, are best candidates for astronauts in China.

[An experimental study on the changes of estrogen receptor in the condylar cartilages with DCC method in rats after functional mandibular protrusion].

OBJECTIVE: The purpose of the study was to evaluate the estrogen receptors (ER) in the condylar cartilages in relation to the hyperplasia, hypertrophy and functional adaptive remodelling after functional mandibular protrusion. METHODS: Dextran-coated charcoal (DCC) method was performed to quantitively detect the level of ER. Fifty female rats of 4-week-old were selected and simulated functional appliances were used to guide their mandibles. The rats were sacrificed after 3 days, 1 week, 2 week, 3 week and 4 week. RESULTS: The results showed ER do exist in the condylar cartilages of the rats, with the highest levels during their actively proliferative period and the amount of ER was increased significantly after functional mandibular protrusion. CONCLUSION: It is suggested that ER has a close relationship with the hyperplasia, hypertrophy of the condylar cartilages and plays a very important role in the adaptive remodelling mechanism after functional mandibular protrusion.

[A new saikosaponin from Bupleurum chinense DC].

Bupleurum chinense DC. is a well-known and very important traditional Chinese drug. It is often used to treat common cold with fever, alternating chill and fever, feeling of fullness and oppression in the chest. A new saikosaponin was isolated from Bupleurum chinense DC. and its structure was identified as 3-O-[beta-D-glucopyranosyl (1-->3)-beta-D-fucopyranosyl]-3 beta, 16 alpha, 23, 28-tetrahydroxy-olean-11, 13 (18)-dien-30-oic acid 30-O-[xylityl(1-->1)]-beta-D-glucopyranosyl-6-ester on the basis of chemical and spectral evidence. In addition, three known saikosaponins were isolated and characterized as saikosaponin 1, 2"-O-acetyl-saikosaponin b2 and 2"-O-acetyl-saikosaponin a. All the compounds were isolated from Bupleurum chinense DC. for the first time.

[Determination of Sb in the resin and products of PET by graphite furnace atomic absorption spectrometry].

The method of determing Sb in resin and products of PET by graphite furnace atomic absorption spectrometry was studied. The sample was pretreated by 4% acetic acid and determined by graphite furnace atomic absorption spectrometry. The sensitivity of this method was 30.5 pg/1% absorption, the relative standard deviation was 3.12% and the rate of recovery was 91.68-97.74%. The results obtained by this method agreed with those of malachite green spectrophotometry.

[Determination of paeoniflorin in Huoxuezhuyu powder].

Thin-layer chromatogram was used to separate paeoniflorin from other compositions in Huoxuezhuyu powder. The content was determined by UV spectrophotometry. The mean recovery was 98.74%, and CV was 1.43%. This method could be used for other compound preparations of traditional Chinese medicines.

Preventive effects of selenium-enriched spiruline (SESP) on radiation pneumonitis.

We studied the early effects of selenium-enriched spiruline (SESP) on radiation-induced pulmonary fibrosis using histopathology, cDNA-mRNA dot-blot hybridization, and a biochemical technique for quantifying the hydroxyproline content. We found marked lung injury in the irradiated group, whereas only slight hyperemia, hemorrhage, exudation, and thickness of the interalveolar wall in the lung in the selenium-treated group. The hydroxyproline (hyp) content in the lungs and the synthesis of type III collagen mRNA decreased in the SESP-treated group. Selenium-enriched spiruline may have a protective effect on the development of radiation pneumonitis.

Clinical treatment planning optimization by Powell's method for gamma unit treatment system.

PURPOSE: This article presents a new optimization method for stereotactic radiosurgery treatment planning for gamma unit treatment system. METHODS AND MATERIALS: The gamma unit has been utilized in stereotactic radiosurgery for about 30 years, but the usual procedure for a physician-physicist team to design a treatment plan is a trial-and-error approach. Isodose curves are viewed on two-dimensional computed tomography (CT) or magnetic resonance (MR) image planes, which is not only time consuming but also seldom achieves the optimal treatment plan, especially when the isocenter weights are regarded. We developed a treatment-planning system on a computer workstation in which Powell's optimization method is realized. The optimization process starts with the initial parameters (the number of isocenters as well as corresponding 3D isocenters' coordinates, collimator sizes, and weight factors) roughly determined by the physician-physicist team. The objective function can be changed to consider protection of sensitive tissues. RESULTS: We use the plan parameters given by a well-trained physician-physicist team, or ones that the author give roughly as the initial parameters for the optimization procedure. Dosimetric results of optimization show a better high dose-volume conformation to the target volume compared to the doctor's plan. CONCLUSION: This method converges quickly and is not sensitive to the initial parameters. It achieves an excellent conformation of the estimated isodose curves with the contours of the target volume. If the initial parameters are varied, there will be a little difference in parameters' configuration, but the dosimetric results proved almost to be the same.

Hydrogen exchange: the modern legacy of Linderstrøm-Lang.

This discussion, prepared for the Protein Society's symposium honoring the 100th anniversary of Kaj Linderstrøm-Lang, shows how hydrogen exchange approaches initially conceived and implemented by Lang and his colleagues some 50 years ago are contributing to current progress in structural biology. Examples are chosen from the active protein folding field. Hydrogen exchange methods now make it possible to define the structure of protein folding intermediates in various contexts: as tenuous molten globule forms at equilibrium under destabilizing conditions, in kinetic intermediates that exist for less than one second, and as infinitesimally populated excited state forms under native conditions. More generally, similar methods now find broad application in studies of protein structure, energetics, and interactions. This article considers the rise of these capabilities from their inception at the Carlsberg Labs to their contemporary role as a significant tool of modern structural biology.

Absence of a stable intermediate on the folding pathway of protein A.

The B-domain of protein A has one of the simplest protein topologies, a three-helix bundle. Its folding has been studied as a model for elementary steps in the folding of larger proteins. Earlier studies suggested that folding might occur by way of a helical hairpin intermediate. Equilibrium hydrogen exchange measurements indicate that the C-terminal helical hairpin could be a potential folding intermediate. Kinetic refolding experiments were performed using stopped-flow circular dichroism and NMR hydrogen-deuterium exchange pulse labeling. Folding of the entire molecule is essentially complete within the 6 ms dead time of the quench-flow apparatus, indicating that the intermediate, if formed, progresses rapidly to the final folded state. Site-directed mutagenesis of the isoleucine residue at position 16 was used to generate a variant protein containing tryptophan (the 116 W mutant). The formation of the putative folding intermediate was expected to be favored in this mutant at the expense of the native folded form, due to predicted unfavorable steric interactions of the bulky tryptophan side chain in the folded state. The 116 W mutant refolds completely within the dead time of a stopped-flow fluorescence experiment. No partly folded intermediate could be detected by either kinetic or equilibrium measurements. Studies of peptide fragments suggest that the protein A sequence has an intrinsic propensity to form a helix II/helix III hairpin. However, its stability appears to be marginal (of the order of 1/2 kT) and it could not be an obligatory intermediate on a defined folding pathway. These results explicitly demonstrate that the protein A B domain folds extremely rapidly by an apparent two-state mechanism without formation of stable partly folded intermediates. Similar mechanisms may also be involved in the rapid folding of subdomains of larger proteins to form the compact molten globule intermediates that often accumulate during the folding process.

The cure of acute Babesiasis perroncitoi in swine.

From June to August in 1988, a total of 64 pure-bred Landrace, Yorkshire (large white) and Inner Mongolian Black Swine from a farm in Huhehot were affected with a disease, 13 died. The pathogen was confirmed primarily to be Babesia perroncitoi. The drugs used for treatment were Berenil and Acaprin. The intramuscular dose for Berenil was 3 mg per kilogram weight. The subcutaneous dose for Acaprin was 0.8 mg per kilogram weight. All of which had satisfactory results. In the same season in the second year and the third year, the disease was also reported, but all sick animals were cured.

Study of the acid-catalyzed isomerization of dihydroveatchine.

A study of the acid-catalyzed isomerization of dihydroveatchine (5) resulted in the isolation of a single major compound, the aldehyde 6. Structure 6 has been derived from its spectral data. Reduction of 6 with NaBH4 gave compound 7, which was characterized through detailed NMR studies including 1D, 2D, and selective INEPT experiments, as well as preparation of its mono- and bis(p-nitrobenzoyl) derivatives 8 and 9. A plausible mechanism for the formation of 6, derived from the spectral data of the isomerized product obtained by deuterium labeling, is reported. Interestingly, the acid-catalyzed isomerization products of the allylic alcohols garryfoline (1) and dihydroveatchine (5) are different and appear to be dependent on the configurational orientation of the C(15) hydroxyl group. Unambiguous NMR chemical shift assignments for 5 are also reported.
