Transcriptional profiles of latent human immunodeficiency virus in infected individuals: effects of Tat on the host and reservoir.

The persistence of human immunodeficiency virus (HIV) in optimally treated infected individuals poses a major therapeutic problem. In latently infected cells, one of the observed phenotypes is absent elongation of viral transcription. Thus, the positive elongation factor b (P-TEFb), which is usually recruited by NF-kappaB or Tat, is not present on the HIV long terminal repeat (LTR). Although most attempts to activate these proviruses centered on NF-kappaB, we investigated effects of Tat. To this end, we generated transgenic mice, which secreted a chimera between Tat and the green fluorescent protein from beta cells of the pancreas. This extracellular Tat distributed widely, entered nuclei of resting cells, and specifically transactivated the HIV LTR. No deleterious side effects of Tat were found. Next, we determined that Tat can activate latent proviruses in optimally treated infected individuals. In their cells, T-cell activation or exogenous Tat could induce viral replication equivalently. Thus, P-TEFb could activate the majority of the latent HIV, in this case by Tat.

Role of TFF in healing of stress-induced gastric lesions.

AIM: To determine the changes of pS2 and ITF of TFF expression in gastric mucosa and the effect on ulcer healing of pS2, ITF to Water-immersion and restraint stress (WRS) in rats. METHODS: Wistar rats were exposed to single or repeated WRS for 4 h every other day for up to 6 days.Gastric mucosal blood flow (GMBF) was measured by LDF-3 flowmeter and the extent of gastric mucosal lesions were evaluated grossly and histologically. Expression of pS2 and ITF mRNA was determined by RT-PCR. Immunohistochemistry was used to further detect the expression of pS2 and ITF. RESULTS: WRS applied once produced numerous gastric mucosal erosions, but the number of these lesions gradually declined and GMBF restored at 2, 4, 8 h after stress. The area of gastric mucosal lesion was reduced by 64.9 % and GMBF was increased by 89.8 % at 8 h. The healing of stress-induced ulcerations was accompanied by increased expression of pS2 (0.51+/-0.14 vs 0.77+/-0.11, P<0.01) and ITF (0.022+/-0.001 vs 0.177+/-0.010, P<0.01). The results were demonstrated further by immunohistochemistry of pS2 (0.95+/-0.11 vs 1.41+/-0.04, P<0.01) and ITF (0.134+/-0.001 vs 0.253+/-0.01,P<0.01). With repeated WRS, adaptation to this WRS developed, the area of gastric mucosal lesions was reduced by 22.0 % after four consecutive WRS. This adaptation to WRS was accompanied by increased GMBF (being increased by 94.2 %), active cell proliferation in the neck region of gastric glands, and increased expression of pS2 (0.37+/-0.02 vs 0.77+/-0.01, P<0.01) and ITF (0.040+/- 0.001 vs 0.372+/-0.010, P<0.01). The result was demonstrated further by immunohistochemistry of pS2 (0.55+/-0.04 vs 2.46+/-0.08, P<0.01) and ITF (0.134+/-0.001 vs 0.354+/-0.070, P<0.01). CONCLUSION: TFF may not only participate in the early phase of epithelial repair known as restitution (maked by increased cell migration), but also play an important role in the subsequent, protracted phase of glandular renewal(made by cell proliferation).

[Pathological changes in rabbit brain after traumatic head injuries with seawater immersion].

OBJECTIVE: To observe the pathological impact of seawater on rabbit brain tissue with severe traumatic injury. METHODS: Modified rabbit models of severe brain injury was utilized, in which the damaged brain tissues were subjected to immersion with fresh seawater for 30 min. A control group was set up in which the trauma was induced without subsequent seawater immersion. Tissue sampling was performed at the brain injury sites at 0, 3, 8 h after seawater immersion and the pathological changes in the brain tissues were observed by means of HE staining. RESULTS: Severe traumatic brain edema occurred in both of the two groups, but the onset time of edema differed. In the control group, brain edema was obvious at 3 h after treatment and hardly aggravated at hour 8; while in seawater treatment group, severe brain edema occurred at 8 h after the treatment and aggravated progressively. CONCLUSION: Seawater immersion delays the onset and peak of traumatic brain edema following severe brain injuries, but can eventually aggravate the traumatic edema.

[Two phases bioremediation of oil contaminated soil from Liaohe oil field].

A treatment engineering of prepared bed was set up for the bioremediation of oil contaminated soil with oil in this study, soils contaminated with different type of oils were treated using composting process in the prepared bed and the treatment period was divided into 2 phases with total time of 210 days. When the concentration of total petroleum hydrocarbons (TPH), which consist of thin oil, high condensation oil, special viscous oil, and viscous oil, were in the range of 25.8-77.2 g.kg-1 dry soil, petroleum removal rates can reach 38.37%-56.74% by 53 days operation. In the second phase, total petroleum removal rates reached 66.59%-80.96% by the 156 days operation in the next year. The results showed that most hydrocarbon pollutants that are easier to be degraded were removed in the first phase, the remedying efficiency obviously dropped in the second phase.

Hydroxyethyl starch inhibits NF-kappaB activation and prevents the expression of inflammatory mediators in endotoxic rats.

Hydroxyethyl starch (HES) has been shown to be beneficial in several inflammatory situations, but the mechanisms are unclear. The present study tested the hypothesis that HES has effects on nuclear factor kappa B (NF-kappaB) activation and the expression of inflammatory mediators induced by lipopolysaccharide. Sepsis was induced in male Wistar rats by injection of lipopolysaccharide (LPS, 6 mg/kg, i.p.). At 1 min after the LPS challenge, HES was infused via the right external jugular vein at the following doses: 3.75, 7.5, 15, or 30 ml/kg. NF-kappaB activation in peripheral blood mononuclear cells and neutrophils, plasma concentrations of tumor necrosis factor (TNF)-alpha, cytokine-induced neutrophil chemoattractant (CINC), expression of CD11b on the blood neutrophil cell surface, and neutrophil sequestration in multiple organs were examined 2 or 4 hr after the LPS challenge. Treatment of rats with HES (3.75 and 7.5 ml/kg) prevented LPS-induced NF-kappaB activation, and inhibited, in a dose-related manner, LPS-induced TNF-alpha and CINC expression. The 4 graded doses of HES decreased CD11b expression in a dose-dependent manner. HES significantly reduced neutrophil sequestration in lung, heart, and liver. These results suggest that HES has an anti-inflammatory effect in endotoxic rats. This effect is mediated by inhibition in the production pathways for inflammatory mediators, including NF-kappaB activation.

[Surgical treatment for patients with late-stage thyroid carcinoma].

OBJECTIVE: To explore the value of surgical treatment for patients with late-stage thyroid carcinoma. METHOD: Eleven cases of late-stage thyroid carcinoma underwent radical neck lymphnode dicission of the affected side plus functional neck lymphnode dicission on the opposite side. Three of them received lobectomy of the affected sides and excission of the isthmus. Six cases received lobectomy of affected sides, excission of isthmus and subtotal thyroidectomy of the opposite side; 2 of them received total thyroidectomy and parathyroid transpiantation. RESULT: With 9 cases followed up, 8 cases survived without recurrence and advancing. One for 6 years, 1 for 5 years, 2 for 3 years, 3 for 2 years and 1 for 1 year. The other one died, suffered from papillary adenocarcinoma combined with metastatic adenocarcinoma. CONCLUSION: For patients with late-stage thyroid carcinoma, operation should be performed as possible. Postoperative thyroid hormone administration could inhibit the development of the primary and metastatic lesions effectively. Preservation of parathyroid glands could improve the patient's life quality.

A requirement for CARMA1 in TCR-induced NF-kappa B activation.

Stimulation of the T cell receptor (TCR) complex initiates multiple signaling cascades that lead to the activation of several transcription factors, including the NF-kappa B family members. Although various proximal signaling components of the TCR have been intensively studied, the distal components that mediate TCR-induced NF-kappa B activation remain largely unknown. Using a somatic mutagenesis approach, we cloned a CARMA1-deficient T cell line. Deficiency in CARMA1 (originally known as CARDII) resulted in selectively impaired activation of NF-kappa B induced by the TCR and a consequent defect in interleukin-2 (IL-2) production. Reconstitution of the CARMA1-deficient cells with CARMA1 fully rescued this signaling defect. Together, our results show that CARMA1 is an essential signaling component that mediates TCR-induced NF-kappa B activation.

P-TEFb containing cyclin K and Cdk9 can activate transcription via RNA.

Different positive transcription elongation factor b (P-TEFb) complexes isolated from mammalian cells contain a common catalytic subunit (Cdk9) and the unique regulatory cyclins CycT1, CycT2a, CycT2b, or CycK. The role of CycK as a transcriptional cyclin was demonstrated in this study. First, CycK activated transcription when tethered heterologously to RNA, which required the kinase activity of Cdk9. Although this P-TEFb could phosphorylate the C-terminal domain (CTD) of RNA polymerase II (RNAPII) in vitro, in contrast to CycT1 and CycT2, CycK did not activate transcription when tethered to DNA. Interestingly, when the C termini of CycT1 and CycT2 or only the histidine-rich stretch from positions 481 to 551 in CycT1 were added to CycK, the extended chimeras activated transcription equivalently via DNA. Moreover, these transcriptional effects required the CTD of RNAPII in cells. Thus, CycK functions as P-TEFb only via RNA, which suggests the presence of cellular RNA-bound activators that require CycK for their transcriptional activity.

Interaction between P-TEFb and the C-terminal domain of RNA polymerase II activates transcriptional elongation from sites upstream or downstream of target genes.

Transcriptional elongation by RNA polymerase II (RNAPII) is regulated by the positive transcription elongation factor b (P-TEFb). P-TEFb is composed of Cdk9 and C-type cyclin T1 (CycT1), CycT2a, CycT2b, or CycK. The role of the C-terminal region of CycT1 and CycT2 remains unknown. In this report, we demonstrate that these sequences are essential for the activation of transcription by P-TEFb via DNA, i.e., when CycT1 is tethered upstream or downstream of promoters and coding sequences. A histidine-rich stretch, which is conserved between CycT1 and CycT2 in this region, bound the C-terminal domain of RNAPII. This binding was required for the subsequent expression of full-length transcripts from target genes. Thus, P-TEFb could mediate effects of enhancers on the elongation of transcription.

Effect of mild hypothermia on partial pressure of oxygen in brain tissue and brain temperature in patients with severe head injury.

OBJECTIVE: To study the changes of partial pressure of oxygen in brain tissue (P(bt)O(2)) and brain temperature (BT) in patient s in acute phase of severe head injury, and to study the effect of mild hypothermia on P(bt)O(2) and BT. METHODS: The P(bt)O(2) and the BT of 18 patients with severe head injury were monitored, and the patients were treated with mild hypothermia within 20 hours after injury. The rectal temperature (RT) of the patients was kept on 31.5-34.9 degrees C for 1-7 days (57.7 hours+/-28.4 hours averagely), simultaneously, the indexes of P(bt)O(2) and BT were monitored for 1-5 days (with an average of 54.8 hours+/-27.0 hours). According to Glasgow Outcome Scale (GOS), the prognosis of the patients was evaluated at 6 months after injury. RESULTS: Within 24 hours after severe head injury, the P(bt)O(2) was significantly lower (9.6 mm Hg+/-6.8 mm Hg, 1 mm Hg=0.133 kPa) than the normal value (16-40 mm Hg). After treatment of mild hypothermia, the mean P(bt)O(2) increased to 28.7 mm Hg+/-8.8 mm Hg during the first 24 hours, and the P(bt)O(2) was still maintained within the range of normal value at 3 days after injury. The BT was higher than the RT in the patients in acute phase of severe head injury, and the difference between the BT and the RT significantly increased after treatment of mild hypothermia. Hyperventilation (the partial pressure of carbon dioxide in artery (P(a)CO(2)) approximately 25 mm Hg) decreased the high intracranial pressure (ICP) and significantly decreased the P(bt)O(2). CONCLUSIONS: This study demonstrates that P(pt)O(2) and BT monitoring is a safe, reliable and sensitive diagnostic method to follow cerebral oxygenation. It might become an important tool in our treatment regime for patients in the acute phase of severe head injury requiring hypothermia and hyperventilation.

[Clinical analysis of primary head and neck non-Hodgkin's ].

OBJECTIVE: To research the clinical features of primary head and neck non-Hodgkin's lymphoms (NHL). METHOD: 62 patients with primary head and neck NHL were analyzed in the positions of focuses, clinical aspects, diagnosis and treatment. RESULT: Tonsilla palatina(25.8%), cervical lymphonodi (19.4%), nasal cavity and nasal sinuses(12.9%) were the common place of the origin of NHL which had various clinical manifestations. High-grade malignant lymphomas represented 75.8% of these cases whose I E stage and II E stage were 38.7% and 24.2% respectively. The complete response, partial response, and response rate were 75.8%, 14.5%, and 90.3%, respectively. Some patients had been long-term survival without tumor. CONCLUSION: Head and neck NHL is generally high degree malignancy. It often originates from outside lymph node and easily metastasizes to lymph node. Accurate surgical biopsy linking immunohistochemical test will be advantageous to make a definite diagnosis. Combining Chemotherapy and radiotherapy or operation should be put into practice to improve prognosis.
