Identification of a novel point mutation of mouse proto-oncogene c-kit through N-ethyl-N-nitrosourea mutagenesis.

Manipulation of the mouse genome has emerged as an important approach for studying gene function and establishing human disease models. In this study, the mouse mutants were generated through N-ethyl-N-nitrosourea (ENU)-induced mutagenesis in C57BL/6J mice. The screening for dominant mutations yielded several mice with fur color abnormalities. One of them causes a phenotype similar to that shown by dominant-white spotting (W) allele mutants. This strain was named Wads because the homozygous mutant mice are white color, anemic, deaf, and sterile. The new mutation was mapped to 42 cM on chromosome five, where proto-oncogene c-kit resides. Sequence analysis of c-kit cDNA from Wads(m/m) revealed a unique T-to-C transition mutation that resulted in Phe-to-Ser substitution at amino acid 856 within a highly conserved tyrosine kinase domain. Compared with other c-kit mutants, Wads may present a novel loss-of-function or hypomorphic mutation. In addition to the examination of adult phenotypes in hearing loss, anemia, and mast cell deficiency, we also detected some early developmental defects during germ cell differentiation in the testis and ovary of neonatal Wads(m/m) mice. Therefore, the Wads mutant may serve as a new disease model of human piebaldism, anemia, deafness, sterility, and mast cell diseases.

[Cytotoxic effect of cisplatin with proteasome inhibitor on osteosarcoma cells].

OBJECTIVE: To observe the cytotoxic effect of cisplatin with proteasome inhibitor on osteosarcoma cells. METHODS: Cell survival was tested by MTT, apoptotic morphology was observed by electron microscopy, apoptotic rate was analyzed by flow cytometry, the transcription level of excision repair cross complementation-1 (ERCC-1) was tested by reverse transcription polymerase reaction. RESULTS: Compared with cells treated with cisplatin alone, cells treated with cisplatin and proteasome inhibitor showed a decreased survival rate, more typical apoptotic morphology, higher apoptotic rate [(14.37 +/-2.37)% vs. (50.93 +/-4.84)%, P<0.01)], and lower transcription level of excision repair cross complementation-1. CONCLUSION: Proteasome inhibitor could increase the cytotoxic effect of cisplatin on osteosarcoma cells and promote cisplatin-induced osteosarcoma apoptosis. These effects may be associated with the decreased transcription of excision repair cross complementation-1.

[Diagnostic role of anti-saccharomyces cerevisiae and antineutrophil cytoplasmic autoantibodies in inflammatory bowel disease].

OBJECTIVE: To determine the accuracy of the assay using perinuclear antineutrophil cytoplasmic antibody (pANCA) and anti-saccharomyces cerevisiae antibodies (ASCA) in diagnosing ulcerative colitis (UC) and Crohn's disease (CD) and whether the presence of ASCA and pANCA antibodies could differentiate either CD from UC, or inflammatory bowel disease (IBD) from normal controls. METHODS: Serum samples were obtained from 34 patients with CD and 29 with UC, and from 25 normal volunteers. Diagnosis was established on clinical findings, X-ray or endoscopy and histology. Determination of ASCA and pANCA antibodies was performed using indirect immunofluorescence technique. RESULTS: 47.1% patients with CD against 69.0% patients with UC expressed pANCA (P < 0.05). Vice versa 58.6% patients with UC against 11.8% patients with CD expressed ASCA (P < 0.05). The sensibility, specificity and positive predictive value of combination of positive ASCA and negative pANCA to diagnosis CD was 0, 89.7% and 0 respectively, and those of combination of positive pANCA and negative ASCA to diagnosis of UC was 20.7%, 64.7% and 33.3% respectively. CONCLUSIONS: The positive of either ASCA or pANCA are not enough sensible to screen the IBD, but useful to diagnosis IBD. The combination pANCA and ASCA can not be as a serum differential diagnosis marker for IBD.

[Antioxidative activity of water-soluble extracts from Fructus Polygoni Orientalis].

OBJECTIVE: To study the antioxidative activity of the water-soluble extracts from Fructus Polygoni Orientalis. METHODS: Malondialdehyde and superoxide dismutase were measured by thiobarbituric acid assay, nitrite method, respectively. The activity of glutathione peroxidase was determined by 5,5'-dithionbis (2-nitrobenzoic acid) photometric method. Lipofuscin was determined by Sohal method in the brain of senile model mice induced by D-galactose. The extracts of 0.8, 1.6 and 3.2 g.kg(-1).d(-1) were administered to the mice separately through oral gavage daily for 7 weeks. RESULTS: Malondialdehyde content increased, activities of superoxide dismutase and glutathione peroxidase in the heart, liver and kidney decreased. Levels of lipofuscin in the brain of senile model mice induced by D-galactose increased. The three doses of the extracts could inhibit the levels of lipofuscin in the brain, malondialdehyde in the heart, liver and kidney. These results showed that there was a negative dose-effect relationship. The extracts also up-regulated the levels of superoxide dismutase and glutathione peroxidase in the heart, liver and kidney with a positive dose-effect relationship. CONCLUSION: The water-soluble extracts exert its antioxidative activity by eliminating free hydroxyl radicals, increasing activities of superoxide dismutase and glutathione peroxidase.

[Effect of flue gas conditions on NO oxidation process by DC corona radical shower].

Using an air-H2O DC corona radical shower system, the influences of reside time of flue gas in the reactor, velocity of flue gas and NO concentration on NO oxidation process were studied. The results show that the increasing velocity of flue gas can restrain corona development and the increasing NO concentration can make discharge more easy. The reside time of flue gas in the reactor has less effect on the NO oxidation. The NO oxidation rate increased only from 54.5% to 57.6% at 2 W input power when the reside time of flue gas in the reactor increased from 8.5 s to 34.2 s. However, the velocity of flue gas has important effect on the NO oxidation. At 1.7 W x h/m3 energy density, when the velocity of flue gas increased from 1.4 cm/s to 6.3 cm/s, the NO oxidation rate dropped from 60.0% to 38.6% and the energy yield also falled from 20.8 g/(kW x h) to 13.3 g/(kW x h). Under the certain flux of humid air, NO initial concentration has a best value, which was about 100 x 10(-6) in this experiment.

[Establishment of transgenic mouse line skin-specifically expressing hCTLA4-Ig].

hCTLA4-Ig,which is capable of inhibiting T cell activation and rendering T cell to be anergy by blocking co-stimulatory signal pathway, is recognized as a potential therapeutic molecule to extend the survival of skin graft in burn medicine. To investigate whether the survival of skin graft can be further extended by skin-specific expression of hCTLA4-Ig, a skin-specific expression vector was constructed by pacing the encoding sequence of hCTLA4-Ig under the drive of K14 promoter, and a transgenic mouse line was established with the vector. RT-PCR and Northern blot assay indicated that hCTLA4-Ig was skin-specifically expressed in transgenic mice at a rather high level. Compared with the expression of GAPDH, the expression of hCTLA4-Ig remained constant through generations and over lifetime. The results of this paper indicated that a transgenic mouse line skin-specifically and constitutively expressing hCTLA4-Ig had been established.

Manganese(III) acetate-mediated free radical reactions of [60]fullerene with beta-dicarbonyl compounds.

[60]Fullerene reacted with various beta-dicarbonyl compounds in the presence of Mn(OAc)3*2H2O to generate dihydrofuran-fused [60]fullerene derivatives or 1,4-bisadducts. Dihydrofuran-fused [60]fullerene derivatives 2 could be formed by treatment of alpha-unsubstituted beta-diketones 1a-e or beta-ketoesters 1f and 1g with [60]fullerene in refluxing chlorobenzene in the presence of Mn(III). Solvent-participated unsymmetrical 1,4-bisadducts 3 were obtained through the reaction of [60]fullerene with dimethyl malonate 1h or alpha-substituted beta-dicarbonyl compounds 1i-1n in toluene. A possible reaction mechanism for the formation of different fullerene derivatives is proposed.

Effects of Zuogui Wan on neurocyte apoptosis and down-regulation of TGF-beta1 expression in nuclei of arcuate hypothalamus of monosodium glutamate -liver regeneration rats.

AIM: To inquire into the effects and mechanism of Zuogui Wan (Pills for Kidney Yin) on neurocyte apoptosis in nuclei of arcuate hypothalamus (ARN) of monosodium glutamate (MSG)-liver regeneration rats, and the mechanism of liver regeneration by using optic microscope, electron microscope and in situ end labeling technology to adjust nerve-endocrine-immunity network. METHODS: Neurocyte apoptosis in ARN of the experiment rats was observed by using optic microscope, electron microscope and in situ end labeling technology. Expression of TGF-beta1 in ARN was observed by using immunohistochemistry method. RESULTS: The expression of TGF-beta1 in rats of model group was increased with the increase of ARN neurocyte apoptosis index (AI) (t = 8.3097, 12.9884, P<0.01). As compared with the rats of model group, the expression of TGF-beta1 in rats of Zuogui Wan treatment group was decreased with the significant decrease of ARN neurocyte apoptosis (t = 4.5624, 11.1420, P<0.01). CONCLUSION: Brain neurocyte calcium ion overexertion and TGF-beta1 protein participate in the adjustment and control of ARN neurocyte apoptosis in MSG-liver regeneration-rats. Zuogui Wan can prevent ARN neurocyte apoptosis of MSG-liver regeneration in rats by down-regulating the expression of TGF-beta1, and influence liver regeneration through adjusting nerve-endocrine-immune network.

The diversity of LTR retrotransposons.

Eukaryotic genomes are full of long terminal repeat (LTR) retrotransposons. Although most LTR retrotransposons have common structural features and encode similar genes, there is nonetheless considerable diversity in their genomic organization, reflecting the different strategies they use to proliferate within the genomes of their hosts.

NF-kappaB activation is required for the development of cardiac hypertrophy in vivo.

In the present study, we examined whether NF-kappaB activation is required for cardiac hypertrophy in vivo. Cardiac hypertrophy in rats was induced by aortic banding for 1, 3, and 5 days and 1-6 wk, and age-matched sham-operated rats served as controls. In a separate group of rats, an IkappaB-alpha dominant negative mutant (IkappaB-alphaM), a superrepressor of NF-kappaB activation, or pyrrolidinedithiocarbamate (PDTC), an antioxidant that can inhibit NF-kappaB activation, was administered to aortic-banded rats for 3 wk. The heart weight-to-body weight ratio was significantly increased at 5 days after aortic banding, peaked at 4 wk, and remained elevated at 6 wk compared with age-matched sham controls. Atrial natriuretic peptide and brain natriuretic peptide mRNA expressions were significantly increased after 1 wk of aortic banding, reached a maximum between 2 and 3 wk, and remained increased at 6 wk compared with age-matched sham controls. NF-kappaB activity was significantly increased at 1 day, reached a peak at 3 wk, and remained elevated at 6 wk, and IKK-beta activity was significantly increased at 1 day, peaked at 5 days, and then decreased but remained elevated at 6 wk after aortic banding compared with age-matched sham controls. Inhibiting NF-kappaB activation in vivo by cardiac transfection of IkappaB-alphaM or by PDTC treatment significantly attenuated the development of cardiac hypertrophy in vivo with a concomitant decrease in NF-kappaB activity. Our results suggest that NF-kappaB activation is required for the development of cardiac hypertrophy in vivo and that NF-kappaB could be an important target for inhibiting the development of cardiac hypertrophy in vivo.

hNRAGE, a human neurotrophin receptor interacting MAGE homologue, regulates p53 transcriptional activity and inhibits cell proliferation.

hNRAGE, a neurotrophin receptor p75 interacting MAGE homologue, is cloned from a human placenta cDNA library. hNRAGE can inhibit the colony formation of and arrest cell proliferation at the G1/S and G2/M stages in hNRAGE overexpressing cells. Interestingly, hNRAGE also increases the p53 protein level as well as its phosphorylation (Ser392). Further studies demonstrated that hNRAGE does not affect the proliferation of mouse p53-/- embryonic fibroblasts, suggesting that p53 function is required for hNRAGE induced cell cycle arrest. Moreover, the cell cycle inhibiting protein p21(WAF) is induced by hNRAGE in a p53 dependent manner. The data provide original evidence that hNRAGE arrests cell growth through a p53 dependent pathway.

Modulating Toll-like receptor mediated signaling by (1-->3)-beta-D-glucan rapidly induces cardioprotection.

OBJECTIVE: Immune and inflammatory signaling pathways, initiated by the innate response, are involved in myocardial ischemia/reperfusion (I/R) injury. Toll-like receptor (TLR) mediated MyD88-dependent NFkappaB pathways play a role in the induction of innate immunity. We have reported that glucan phosphate (GP) improved survival in experimental sepsis, which correlated with decreased tissue NFkappaB activation. In the present study, we report that GP rapidly induced cardioprotection against I/R injury in vivo. METHODS: Sprague-Dawley rats were pretreated with GP (40 mg/kg, i.p) 1 h before 45 min of ligation of the left anterior descending coronary followed by reperfusion for 4 and 24 h. Infarction size was examined by triphenyltetrazolium chloride (TTC) staining. NFkappaB activation was analyzed by electrophoretic mobility shift assay (EMSA). IkappaB kinase-beta (IKKbeta), IL-1 receptor-associated kinase (IRAK) and Phosphoinositide 3-kinase (PI3K) activities were determined by kinase assay with appropriate substrates. Association of TLR4 with MyD88 or with PI3K p85 was assessed by immunoprecipitation with anti-TLR4 followed by immunoblotting with anti-MyD88 or anti-p85. RESULTS: GP treatment reduced infarct size by 47% in rat hearts subjected to reperfusion for 4 h and by 50% following reperfusion for 24 h. The same protective effect was observed when GP was administrated 5 min after initiation of ischemia. The mechanisms of GP induced cardioprotection involve decreased association of TLR4 with MyD88, inhibition of I/R induced IRAK and IKKbeta activity and decreased NFkappaB activity. In addition, GP increased TLR4 phosphotyrosine, resulting in increasing PI3K/Akt activity in the myocardium, which correlated with decreased cardiac myocyte apoptosis following I/R. CONCLUSION: The results suggest that activation of the TLR mediated MyD88-dependent NFkappaB signaling pathway may play an important role in myocardial I/R injury, while stimulation of the PI3K/Akt signaling could serve a protective role. The data indicates that GP treatment shifts the TLR mediated activation signal in I/R from a predominantly NFkappaB pathway to a predominant PI3K/Akt signaling pathway.

Improvement of growth and camptothecin yield by altering nitrogen source supply in cell suspension cultures of Camptotheca acuminata.

Nitrate at 70 mM gave the highest biomass of Camptotheca acuminata in suspension culture in MS medium, but a NH4+/NO3- molar ratio of 5:1 (giving a total of 40 mM N) gave the maximum camptothecin yield. A two-stage flask culture system was established to improve culture efficiency; cell dry weight, camptothecin content and yield was increased by 30%, 280% and 340%, respectively when compared with those of control, reaching up to 36 g l(-1), 0.36 mg g(-1), and 12.8 mg l(-1), respectively.

Phosphatase of regenerating liver-3 promotes motility and metastasis of mouse melanoma cells.

Recent reports suggested that phosphatase of regenerating liver (PRL)-3 might be involved in colorectal carcinoma metastasis with an unknown mechanism. Here we demonstrated that PRL-3 expression was up-regulated in human liver carcinoma compared with normal liver. PRL-3 was also highly expressed in metastatic melanoma B16-BL6 cells but not in its lowly metastatic parental cell line, B16 cells. B16 cells transfected with PRL-3 cDNA displayed morphological transformation from epithelial-like shape to fibroblast-like shape. PRL-3-overexpressed cells showed much higher migratory ability, which could be reversed by specific anti-sense oligodeoxynucleotide and the phosphatase inhibitors sodium orthovanadate or potassium bisperoxo oxovanadate V. Meanwhile, the expression of the catalytically inactive PRL-3 mutations (D72A or C104S) significantly reduced the cell migratory capability. In addition, PRL-3 transfectants demonstrated altered extracellular matrix adhesive property and up-regulated integrin-mediated cell spreading efficiency. Furthermore, we confirmed that PRL-3 could facilitate lung and liver metastasis of B16 cells in an experimental metastasis model in mice, consistent with accelerated proliferation and growth rate both in vitro and in vivo. Together, these observations provide convincing evidence that PRL-3 truly plays a causal role in tumor metastasis.

Atomic force microscopy studies on DNA structural changes induced by vincristine sulfate and aspirin.

We report that atomic force microscopy (AFM) studies on structural variations of a linear plasmid DNA interact with various concentrations of vincristine sulfate and aspirin. The different binding images show that vincrinstine sulfate binding DNA chains caused some loops and cleavages of the DNA fragments, whereas aspirin interaction caused the width changes and conformational transition of the DNA fragments. Two different DNA structural alternations could be explained by the different mechanisms of the interactions with these two components. Our work indicates that the AFM is a powerful tool in studying the interaction between DNA and small molecules.
