Synthesis and structure of an unexpected platinum pi complex formed from substituted 1,4-diamino ligands through an elimination process.

An unusual reactivity of cis-1,2-bis[(N,N-dimethylamino)methyl] cyclohexane with PtCl(2) was observed, resulting in the formation of a platinum(II) pi-olefin complex instead of the conventional square-planar cis Pt(II) coordination complex with the diamino ligand. This behavior was interpreted on the basis of the steric hindrance of the dimethylamino groups whose electron lone pairs are barely accessible to a platinum atom, which can make it difficult for both dimethylamino groups to bind platinum at the same time. This complex has been physically and spectroscopically characterized and its structure has been confirmed by using X-ray diffraction analyses on single crystals.

Synthesis, characterization and biological activity of trans-platinum(II) and trans-platinum(IV) complexes with 4-hydroxymethylpyridine.

The synthesis and chemical characterization of two new trans platinum complexes, trans-[PtCl(2)NH(3)(4-hydroxymethylpyridine)] (1) and trans-[PtCl(4)NH(3)(4-hydroxymethylpyridine)] (2) are described. Their ability to interact with 5'-GMP by themselves and in the presence of reducing agents in the case of trans-[PtCl(4)NH(3)(4-hydroxymethylpyridine)] were tested. Circular dichroism, electrophoretic mobility in agarose gel, and atomic force microscopy studies showed that the interaction of complex 1 with DNA is stronger than that of complex 2. Cytotoxicity tests against HL-60 tumor cells also showed higher activity for trans-[PtCl(2)NH(3)(4-hydroxymethylpyridine)] than for trans-[PtCl(4)NH(3)(4-hydroxymethylpyridine)]. Complex 1 presents similar behavior to cisplatin, but with a lower IC(50) at 24 h. Complex 1 also showed high apoptosis induction.

Kinetico-mechanistic studies of C-H bond activation on new Pd complexes containing N,N'-chelating ligands.

The hybrid imine/amine palladium(II) coordination complexes [PdX2(kappa2-N(imino),N(amino))](X = Cl, AcO; kappa2-N(imino),N(amino)= 4ClC6H4CHNCH2(CH2)nN(CH3)2, n= 1, 2) have been prepared in different isomeric forms which include E/Z arrangement around the C[double bond]N bond of the hybrid ligand and {Pd(kappa(2)-N(imino),N(amino))} ring conformation. The crystal structures of four of them, E-1AcO, Z-1AcO, E-2AcO and E-2Cl, have been determined and the solution behaviour in acetic acid, the common cyclometallating solvent, for all these systems studied. The complexes in acetic acid solution are shown to maintain the structure determined by X-ray crystallography, as they do in deuterated chloroform. Nevertheless, a partial opening equilibrium of the {Pd(kappa2-N(imino),N(amino))} ring is observed by NMR experiments. When the complexes are held in solution for longer periods the corresponding cyclometallated derivatives, 1AcO-CM, 2AcO-CM, 1Cl-CM and 2Cl-CM, containing the {Pd(kappa2-C,N(imino))} palladacycle are obtained, as characterized by 1H NMR spectroscopy. In these compounds the total opening of the N(amino) moiety of the ligand has occurred. The C-H bond activation process has been studied kinetico-mechanistically at different temperatures, pressures and acid concentrations; the results agree with the need of an opening of the chelate ring in [PdX2(kappa2-N(imino),N(amino))] prior to the proper cyclometallation reaction. The values of the enthalpies of activation are higher than those observed for known N-monodentated cyclometallating ligands, as should correspond to the contribution of a ligand dechelation pre-equilibrium. The entropies and volumes of activation are also indicative of this predissociation that include an important amount of contractive ordering. The presence of small amounts of triflic acid in the reaction medium accelerates the reaction to the value observed for N(imino)-monodentate systems, indicating that the full opening of the chelate ring has taken place. For the badly oriented isomeric forms of the ligand in the chelated complex (Z), the cyclometallation process is even more slow and corresponds directly to the reorganization of the ligand to its cyclopalladation-active (E) conformation.

The syntheses, structural characterization, and magnetic behavior of the three new polynuclear copper(II) complexes with formulas [Cu(4)(eta(2):mu-CH(3)COO)(2)(mu-OH)(2)(mu-OH(2))(mu-bdmap)(2)](ClO(4))(2).H(2)O (1), [Cu(8)(NCO)(2)(eta(1):mu-NCO)(4)(mu-OH)(2)(mu(3)-OH)(2)(mu-OH(2))(3)(mu-bdmap)(4)](ClO(4))(2)x2H(2)O (2), and [Cu(9)(eta(1):mu-NCO)(8)(mu(3)-OH)(4)(OH(2))(2)(mu-bdmap)(4)](ClO(4))(2).4H(2)O (3), in which bdmapH is 1,3-bis(dimethylamino)-2-propanol, are reported. Tetranuclear complex 1 crystallizes in the triclinic system, space group P, with unit cell parameters a = 12.160(1) A, b = 13.051(1) A, c = 13.235(1) A, alpha = 110.745(1) degrees , beta = 109.683(1) degrees , gamma = 97.014(1), and Z = 2. Octanuclear complex 2 crystallizes in the monoclinic system, space group C2/c, with unit cell parameters a = 26.609(1) A, b = 14.496(1) A, c = 16.652(1) A, beta = 97.814(1) degrees , and Z = 4, and nonanuclear complex 3 crystallizes in the monoclinic system, space group C2/c, with unit cell parameters a = 24.104(1) A, b = 13.542(1) A, c = 24.355(1) A, beta = 109.98(1) degrees , and Z = 4. The magnetic behavior of the three complexes has been checked showing strong antiferromagnetic coupling in all the cases.

Polynuclear and 1-D derivatives of 1,3-bis(dimethylamino)-2-propanolato ligand. Structure and magnetic characterization.

Several new Cu(II) derivatives of the 1,3-bis(dimethylamino)-2-propanolato (bdmap) ligand with formula [Cu(2)(bdmap)(acac)(NH(3))(3)(MeOH)](ClO(4))(2), [Cu(2)(bdmap)(NO(2))(3)(H(2)O)](4) and [Cu(2)(bdmap)(OH)(ox)(0.5)(H(2)O)(2)](n)(ClO(4))(n)xnH(2)O were synthesized and characterized both structurally and magnetically. Dinuclear compound crystallizes in the monoclinic system, space group P2(1)/c, octanuclear compound crystallizes in the triclinic space group P1 and the 1-D alternating system crystallizes in the monoclinic system, space group P2/n. Magnetic analysis indicates strong antiferromagnetic coupling for all derivatives, mainly due to the interaction through the alkoxo O-atom of the bdmap ligand. The effect on the magnetic behaviour of the additional bridging ligands is analysed.

A systematic evaluation of molecular recognition phenomena. 4. Selective binding of dicarboxylic acids to hexaazamacrocyclic ligands based on molecular flexibility.

The host-guest interaction between four hexaaza macrocyclic ligands (3,6,9,17,20,23-hexaazatricyclo[23.3.1.1]triaconta-1(29),11,13,15 (30),25(27)-hexaene (Bd), 3,6,9,16,19,22-hexaazatricyclo[22.2.2.2]triaconta-1(27),11(30),12,14(29),24(28),25-hexaene (P2), 3,7,11,19,23,27-hexaazatricyclo[27.3.1.1]tetratriaconta-1(33),13, 15,17(34),29,31-hexaene (Bn), 3,7,11,18,22,26-hexaazatricyclo[26.2.2.2]tetratriaconta-1(31),13(34),14,16(33),28(32),29-hexaene (P3)) and two dicarboxylic acids (oxalic acid, H2Ox; oxydiacetic acid, H2Od) have been investigated using potentiometric equilibrium methods. Ternary complexes are formed in aqueous solution as a result of hydrogen bond formation and Coulombic interactions between the host and the guest. In the [(H6P2)(Ox)]4+ complex those bonding interactions reach a maximum yielding a log KR6 of 6.08. This species has been further characterized by means of X-ray diffraction analysis showing that the oxalate guest molecule is situated inside the macrocyclic cavity of the P2 host. X-ray diffraction analysis has also been carried out for the complex [(H6Bn)(Od)2](Br)2.6H2O, where now the oxydiacetate is bonded to the host but outside the macrocyclic cavity. Competitive distribution diagrams and total species distribution diagrams are used to graphically illustrate the most salient features of these systems, which are the following: (a) The Bd and P2 ligands bind Ox significantly much more stronger than Od. This is clearly manifested for the P2:Ox:Od competitive system, where a selectivity of 92.5% in favor of the P2-Ox complexation against P2-Od is obtained at p[H] = 2.8. (b) No isomeric effect is found when comparing binding capacities of oxalate with two isomeric ligands such as P2 and Bd since their affinity to bind the substrate is relatively similar. (c) Bn and P3 ligands have a similar behavior as described in (a) for P2 and Bd except that due to the increase of cavity size the differentiation becomes smaller. (d) Less basic ligands containing two methylenic units Bd (log betaH6 = 40.42) and P2 (40.42) bind stronger to the substrates than those containing three methylenic units Bn (50.32) and P3 (50.64) even though their relative predominance depends on p[H].

Crystal structure and magnetic interactions in nickel(II) dibridged complexes formed by two azide groups or by both phenolate oxygen-azide, -thiocyanate, -carboxylate, or -cyanate groups.

Tridentate/tetradentate Schiff base ligands L(1) and L(2), derived from the condensation of o-vanillin or pyridine-2-aldehyde with N,N-dimethylethylenediammine, react with nickel acetate or perchlorate salt and azide, cyanate, or thiocyanate to give rise to a series of dinuclear complexes of formulas [Ni(L(1))(micro(1,1)-N(3))Ni(L(1))(N(3))(OH(2))].H(2)O (1), [[Ni(L(1))(micro(1,1)-NCS)Ni(L(1))(NCS)(OH(2))][Ni(L(1))(micro-CH(3)COO)Ni(L(1))( NCS) (OH(2))]] (2) [[2A][2B]], [Ni(L(1))(micro(1,1)-NCO)Ni(L(1))(NCO)(OH(2))].H(2)O (3), and [Ni(L(2)-OMe)(micro(1,1)-N(3))(N(3))](2) (4), where L(1) = Me(2)N(CH(2))(2)NCHC(6)H(3)(O(-))(OCH(3)) and L(2) = Me(2)N(CH(2))(2)NCHC(6)H(3)N. We have characterized these complexes by analytical, spectroscopic, and variable-temperature magnetic susceptibility measurements. The coordination geometry around all of the Ni(II) centers is a distorted octahedron with bridging azide, thiocyanate/acetate, or cyanate in a micro(1,1) mode and micro(2)-phenolate oxygen ion for 1-3, respectively, or with a double-bridging azide for 4. The magnetic properties of the complexes were studied by magnetic susceptibility (chi(M)) versus temperature measurements. The chi(M) nus T plot reveals that compounds 1 and 4 are strongly ferromagnetically coupled, 3 shows a weak ferromagnetic behavior, and 2 is very weakly antiferromagnetically coupled.

Intramolecular Pd-mediated processes of amino-tethered aryl halides and ketones: insight into the ketone alpha-arylation and carbonyl-addition dichotomy. A new class of four-membered azapalladacycles.

An exploration of the scope and limitations of Pd(0)-catalyzed intramolecular coupling reactions of amino-tethered aryl halides and ketones has been conducted. Two different and competitive reaction pathways starting from omega-(2-haloanilino) alkanones, enolate arylation and addition to the carbonyl group, have been observed, while omega-(2-halobenzylamino) alkanones exclusively underwent the enolate arylation process. The dichotomy between ketone alpha-arylation and carbonyl-addition in the reactions of omega-(2-haloanilino) alkanones has been rationalized by the intermediacy of unprecedented four-membered azapalladacycles, from which X-ray data and chemical behavior are reported.

Synthesis, equilibrium studies and structural characterisation of the Zn(II) complexes with trimethylene-N6,N6'-bisadenine.

The new compound trimethylene-N(6),N(6')-bisadenine (L), in which two adenine molecules are linked together by a trimethylene bridge that connects the N(6) atoms, has been prepared. Reaction of L with HgCl(2) and ZnCl(2) in concentrated HCl solution leads to crystalline solids. The X-ray characterisation of the Hg(II) complex (H(2)L)[HgCl(4)].3H(2)O reveals that it is an outer-sphere complex in which the ligand is protonated at N(1) and N(1'). In contrast, the structure of the complex [H(2)L(ZnCl(3))(2)].2H(2)O shows the ligand co-ordinated to two different Zn(II) ions through the N(7) of both adenine fragments, the protons being located on the N(1) atoms. The latter compound constitutes the first crystallographic evidence of an inner sphere complex with bis-adenines and, for this reason, an equilibrium study was carried out on the Zn(II)-L-H(+) system. Potentiometric studies indicate that L is protonated in aqueous solution to form HL(+) and H(2)L(2+) with logK(H) values of 4.42 and 3.35 (25 degrees C, 0.10 M KNO(3)). The data from potentiometric titrations in the presence of Zn(2+) can be analysed considering the formation of the species LZn(2+), HLZn(3+), LZn(2)(4+) and HLZn(2)(5+), whose stability constants exceed the value expected for a monodentate interaction of the metal ion with adenine and suggest the possibility of a polydentate behaviour of L in the pH range 2.5-5.0. In contrast, spectrophotometric titrations carried out under conditions similar to those used in the synthetic work (1 M HCl) can be fitted with a model involving exclusively the H(2)LZn(4+) and H(2)LZn(2)(6+) species with logK(M) values reasonable for the interaction of Zn(II) with the N(7) of the protonated adenine fragments. Despite the H(2)LZn(2)(6+) species has a low stability, the spectrophotometric results are in agreement with its formation under the conditions in which the solid complex was prepared.

N-chloroacetyl-beta-alanine.

The beta-alanine residue of the title compound, C(5)H(8)ClNO(3), has a ggt folded conformation, which is mainly stabilized through intermolecular N-H...O=C (amide-acid) and O-H...O=C (acid-amide) hydrogen bonds. In addition, a cis conformation is found for the Cl-CH(2)-C(=O)-NH torsion angle, which is associated with the presence of an intramolecular hydrogen bond.

Synthesis, resolution, and reactivity of benzylmesitylphenylphosphine: a new p-chiral bulky ligand.

The synthesis of P,P'-dimesityl-P,P'-diphenyldiphosphine and benzylmesitylphenylphosphine is described as well as the resolution of the latter ligand by means of homochiral organometallic complexes. The absolute configuration of the phosphine is assigned by NMR spectra, using the homochiral palladacycle as a reference point. The configuration has been confirmed by single crystal X-ray diffraction. Molecular mechanics calculations were performed in [PdCl-(R)-(+)-C10H6CH(Me)NH2(PBnMesPh)], and showed that the rotation around the Pd-P bond is restricted in this complex. [Pd(eta3-2-MeC3H4)Cl(PBnMesPh)] was obtained and used as a precursor in the catalytic hydrovinylation of styrene. Benzylmesitylphenylphosphine has a strong tendency to form phosphapalladacycles by activation of one of the ortho-methyl groups. The formation of this metallacycle from cyclopalladated N-donor derivatives by a ligand-exchange reaction is also described.

Preparation and characterization of platinum(II) and (IV) complexes of 1,3-diaminepropane and 1,4-diaminebutane: circumvention of cisplatin resistance and DNA interstrand cross-link formation in CH1cisR ovarian tumor cells.

The reaction of Pt(dimethyl sulfoxide)(2)CBDCA (CBDCA = 1,1-cyclobutanedicarboxylate) with 1,4-diaminebutane and 1,3-diaminepropane ligands yields, under certain conditions, new [Pt(diamine)(2)]CBDCA complexes (1a,b), where the CBDCA ligand has been removed from the coordination sphere of the platinum atom by the diamine ligand, instead of forming the expected [Pt(diamine)CBDCA] complexes (1'a,b). The structure of complexes 1a and 1'b was solved by X-ray diffraction. Complex 1a crystallizes in the orthorhombic system, in the noncentrosymmetric C222 space group, with unit cell parameters: a = 20.053(2) A; b = 8.655(2) A, c = 5.711(3) A; V = 991.2(6) A(3); delta (calcd) = 1.627 mg/m(3); and R = 0.050. The Pt atom displays an unexpected distorted tetrahedral coordination with a N-Pt-N inner bond angle equal to 81(2) degrees for N atoms of the same 1,3-propanediamine ligand and a N-Pt-N bond angle for different ligands equal to 135.4(9) degrees. Complex 1'b crystallizes in the monoclinic system, in the centrosymmetric P2(1)/c space group, with unit cell parameters: a = 6.007(2) A; b = 15.336(4) A, c = 13.232(5) A; beta = 101.90(3) degrees; V = 1192.8(7) A(3); delta (calcd) = 2.369 mg/m(3); and R = 0.067. Cytotoxicity data show that of all the synthesized compounds, only complexes 1'a and 1'b exhibit remarkable cytotoxic properties. Thus, in contrast with carboplatin (cis-diammine-1,1-cyclobutane dicarboxilatoplatinum(II)), compounds 1'a and 1'b, which also contain the CBDCA ligand, are able to circumvent cisplatin (cis-diamminedichloroplatinum(II)) resistance in several tumor cells. Moreover, after 24 h of incubation of CH1cisR ovarian tumor cells with 10 microM of compounds 1'a and 1'b, the level of DNA interstrand cross-links (ICLs) induced by compounds 1'a and 1'b is 3.3 and 3.8 times higher, respectively, than that of carboplatin and 3.5 and 4.0 times higher, respectively, than that of cisplatin. Interestingly, under the same conditions, the intracellular accumulation of compounds 1'a and 1'b is similar to that of carboplatin and cisplatin. However, the extent of binding to DNA of compounds 1'a and 1'b is similar to that of cisplatin but slightly higher than that of carboplatin. We propose that circumvention of cisplatin resistance in CH1cisR cells by compounds 1'a and 1'b might be related to its higher ability to form DNA ICLs relative to carboplatin and cisplatin.

Bis[mu-4-(ethylammoniomethyl)-3,5-dimethylpyrazolato-kappa2N1:N2]bis[(eta4-1,5-cyclooctadiene)rhodium(I)] dichloride dichloromethane methanol solvate.

In the title compound, [Rh(2)(C(8)H(15)N(3))(2)(C(8)H(12))(2)]Cl(2).CH(2)Cl(2).CH(3)OH, the dinuclear Rh(I) complex has C(2) symmetry and the two pyrazolato ligands act as mu-bridges. The coordination of each Rh(I) cation is completed by one cyclooctadiene (COD) ligand. It is shown that the average Rh-C(COD) distance is linearly dependent on the Rh-N(pyrazole) distance in this type of compound, and this is ascribed to the steric hindrance produced by the packing.
