[Diverse effects of glucocorticoids on diverse leucocytes apoptosis].

OBJECTIVE: To observe effects of glucocorticoids on the process of apoptosis in main inflammatory cells related to pulmonary inflammation. METHOD: Human peripheral blood neutrophil(PMN), eosinophil (EOS) and T lymphocyte(TLC) were cultured in vitro under glucocorticoids condition. In situ end-labeling technique of fragmented DNA was used to detect the dynamic fesatures of apoptosisin experimental collected cell. The peripheral PMN apoptosis in acute respiratory distress syndrome(ARDS) and EOS, TLC apoptosis in bronchial asthma were determined by the same method before and after treatment with glucocorticoids. RESULT: PMN, EOS and TLC showed signs of apoptosis spontaneously and no signs of surviving cells after 12 h/43 h, 15 h/65 h and 65 h/128 h of culture respectively. Dexamethasone accelerated EOS and TLC apoptosis, while inhibited PMN apoptosis. The PMN apoptosis in ARDS and EOS, TLC apoptosis in bronchial asthma were in various degreof inhibitory state. The above time of PMN, EOS and TLC apoptosis were 41.5 h/92.6 h,29.3 h/100 h,92.4 h/152.2 h respectively. The inhibitory state of PMN apoptosis was not influenced by glucocorticoids therapy, but the EOS and TLC apoptosis were relieved from their inhibitory state. CONCLUSION: Effects of glucocorticoids are different in diverse leucocyte apoptosis. This could have an differential significance in pulmonary inflammation.

[Separation and assay of bilirubin subspecies with capillary micellar electrokinetic chromatography].

With the use of high performance capillary electrophoretic system, the authors developed a micellar electrokinetic chromatographic method with direct sample injection to separate and assay serum or bile fluid bilirubin subspecies. The four bilirubins, alpha, beta, gamma and delta subspecies, in serum or in bile fluid were successfully separated within 20 min. Under the concentration of combined bilirubin (bilirubin ditaurite), there was a good relationship between the concentration and the peak area. The detection limit reached 1.5 mumol/L. Being compared with the results of the first run with a concentration of 255 mumol/L and migration time of 16 min, the results of the 50th run were 238 mumol/L and 16.7 min respectively.

[Relationship between lipometabolic disorder in stroke with TCM syndrome differentiation and typing].

OBJECTIVE: To analyse the relationship between lipometabolic disorder in stroke with TCM Syndrome Differentiation and Typing. METHODS: The serum lipid, lipoprotein and apo-lipoprotein level of TCM treated group (142 cases) and control group (100 cases) were determined. The patients were subdivided into three Type-groups according to TCM Syndrome Differentiation: Phlegm-Dampness Stagnation (PDS), Yin Deficiency with Yang Hyperactivity (YDYH) and Qi Deficiency with Blood Stasis (QDBS) group. RESULTS: The results showed that the level of TC, TG, HDL, LDL, apoA1, apoB and apoA1/apoB were significantly different in PDS and QDBS as compared with control group (P < 0.05, P < 0.01, respectively), while lipometabolic disorder of PDS was most serious in TCM Syndrome Differentiation of stroke. The level of TG, apoA1/apoB were significantly different in YDYH in comparing with control group (P < 0.05). While the level of TC, HDL, LDL and apoB were insignificantly different than those of control group (P > 0.05). CONCLUSIONS: This study showed that lipometabolic disorder of stroke were correlated with Syndrome Differentiation of TCM. Stroke with PDS and QSBS should give removing Phlegm-Dampness, dredging Meridian and removing blood stasis treatment besides routine treatment.

[Biomechanical characteristics of masticatory muscles of rat after using bite plate].

OBJECTIVE: The purpose of this study is to evaluate the changes on biomechanical characteristics of masticatory muscles of rat after using bite plate. METHODS: 30 growing rats were selected to wear the bite plate for four weeks. The changes of contractive force and time of masticatory muscles were measured by method and technique of biomechanics. RESULTS: The biomechanical characteristics of masticatory musclse changed significantly. The contractive tension and contraction time of masseter muscles increased shortened respectively, while that of the digastric muscles increased and prolonged. CONCLUSION: This suggests that the orthopedic mechanism of the bite plate is to alter the original masticatory muscle dynamics and finally to reach the goal of functional jaw orthopedics.

[An experimental study on mechanism of intraocular pressure decline after allogeneic transplantation of trabecular meshwork].

OBJECTIVE: To study the mechanism of intraocular pressure (IOP) decline after allogeneic transplantation of trabecular meshwork (TM) in primate experimental glaucoma. METHODS: The experimental open-angle glaucoma models were created in 8 rhesus monkey eyes by repeated pangoniophotocoagulation on functional TM using argon laser. After 2, 3, 5 months respectively, avidin-gold or self-erythrocytes, which acted as tracers of the aqueous humor outflow, were perfused into the anterior chamber under normal pressure and volume conditions. 30 minutes later, 2.5% glutaraldehyde was perfused into the anterior chamber under the same conditions. Allograft TMs and photocoagulated area (serving as the control) were observed by light, transmission, scanning electron microscopy and ultrahistochemistry study. RESULTS: The IOP of 5 eyes of 6 experimental eyes became normal. A blood reflux into Schlemm's canal was seen by gonioscopy in 4 eyes. The surface of TMs from graft showed the normal uveal TM cords and intertrabecular spaces. Erythrocytes were found in the intertrabecular spaces and Schlemm's canal. Avidin-gold and vacuolar containing gold were observed in the cells of TM and Schlemm's canal. The TMs of photocoagulated area appeared to be fewer and narrower intertrabecular spaces. The uveal cords of this zone were confused. The content of acid mucopolysaccharides in the grafts was closer to normal than that in the photocoagulated areas. CONCLUSION: TM of allogeneic transplantation not only can survive but also function as an aqueous humor outflow pathway. We consider that this operative procedure can be used for treatment of some cases with glaucoma cautiously.

[A study of Na(+)-K(+)-ATP ase activity in erythrocyte membrane from diabetic retinopathy patients].

OBJECTIVE: To study the changes of Na(+)-K(+)-ATPase activity in erythrocyte membrane and its significance. METHODS: Na(+)-K(+)-ATPase activity in erythrocyte membrane was investigated with malachite green colorimetric analysis in 40 patients with diabetic retinopathy (DR), 20 diabetic patients with non DR (NDR), and 20 sex and age metched normal control subjects. The relations of Na(+)-K(+)-ATPase activity with content of Na(+) and K(+) erythrocyte deformability index (EDI), levels of fasting blood glucose (FBG), glycosylated hemoglobin (HbA(1)c), and the course of disease were also studied. RESULTS: Na(+)-K(+)-ATPase activity in erythrocyte membrane was significantly decreased in diabetic patients (P < 0.01). The decrease was most significantly in patients with DR (P < 0.01). The decreased Na(+)-K(+)-ATPase activity was related to the content of Na(+) and K(+) in erythrocyte, the changes of EDI, FBG, HbA(1)c, and the course of disease (P < 0.01). CONCLUSIONS: The decreased Na(+)-K(+)-ATPase activity in erythrocyte membrane affects the deformability of erythrocyte. It may contribute to the occurrence and development of DR.

[Effect of heparinoid on the proliferation of rabbit vascular smooth muscle cells in vitro].

Proliferation of vascular smooth muscle cells(VSMCs) represents an important event in vascular lesion formation. In the present study, we investigated whether heparinoid abstracted from porcine duodenum inhibits the proliferation of rabbit aortic VSMCs in vitro, using the method of colorimetric MTT (tetrazolium). Our results showed that heparinoid at 1.6-0.05 mg.ml-1 significantly inhibits VSMCs proliferation induced by fetal calf serum(FCS, 10%), basic fibroblast growth factor(bFGF, 50 ng.ml-1) or interleukin-1(IL-1, 50 u.ml-1). In inducing with bFGF, the inhibitive effect of heparinoid was more potent than that of heparin, while in inducing with FCS or IL-1, the inhibitive effect of heparin was more potent than that of heparinoid. Preincubation with heparinoid at 0.2-0.05 mg.ml-1, the vascular smooth muscle cell proliferation induced by FCS was also inhibited. Our results imply that heparinoid may be useful to protect the atherosclerosis and angioplasty restenosis.

[Simultaneous determination of the enantiomers of verapamil and norverapamil in plasma by high performance capillary electrophoresis].

An analytical method has been developed to determine simultaneously plasma concentration of the enantiomers of verapamil and its major metabolite, norverapamil, using capillary zone electrophoresis. Trimethyl-beta-cyclodextrin was selected as chiral selector. Good separation was achieved under the conditions as below: Running electrolyte was pH 2.5, 60 mmol.L-1 phosphate buffer containing 60 mmol.L-1 trimethyl-beta-cyclodextrin; capillary: 30 cm x 75 microns (ID), coated; setpoint of capillary temperature: 20 degrees C; detector: UV 200 nm; injection: electromigration at 12 kV for 7 s; running voltage: 14 kV. Solvent of sample was doubly deionized water. The migration times of enantiomers were less than 12 min. The good linear range was from 300.0 ng.ml-1 to 2.50 ng.ml-1. Relative standard deviations of assay accuracy and precision were less than 12%. It fits for routine therapeutic monitoring as it is highly efficient, rapid, sensitive, with less solvent and sample consumption and automation.

[Configuration in solution and activity mechanism studies on the anticancer Fe(II)-pingyangmycin (PYM) complex].

In order to elucidate the micro-configuration and mechanism of anticancer activity of Fe(II)-PYM complex in solution, the coordination of high spin paramagnetic ferrous ion, Fe(II), and Pingyangmycin, (PYM) was studied in D2O solution by using the paramagnetic ion probe principle and PMR technique and compared with the inactive complexes, e.g., Cu(II)-PYM and Fe(III)-PYM. The basic configuration parameter of the complexes were determined. When the pyrimidine methyl proton was close to the central ion, 6.5 x 10(-10) m was taken as standard, the absolute distance between the central ion and other protons or ligand could be calculated. Results showed that, in Fe(III) solution, the proton of pyrimidine methyl and the terminal acyl group of the sugar moiety were closer to the central ion, but other protons were relaxed and the distance was between 7.0 x 10(-10) m and 15 x 10(-10) m. In the case of Fe(II) complex, the distance of the central ion and the ligand group was between 4.91 x 10(-10) m and 8.85 x 10(-10) m. The distinctive structure diversity with those of inactive complexes was observed and the relationship of the activity mechanism was postulated.

[Electrophoretic analysis of soluble antigens of a pathogenic isolate of Trichomonas vaginalis].

AIM: To analyze the parasite soluble antigens of a pathogenic isolate of Trichomonas vaginalis. METHODS: The soluble antigens of a pathogenic isolate of T. vaginalis isolated from a patient suffered from trichomonad vaginitis was studied by means of SDS-PAGE followed by densitometric analysis and immunoblotting. RESULTS: A total of 14 distinct protein bands was demonstrated using a 10% resolution gel concentration. Densitometrogram indicated that the soluble antigens with MWs of equal to and below 72 kDa accounted for 89.5% of the total volume with the dominant fractions of 48-51 kDa which accounted for 19.2%. Having MWs over 100 kDa accounted only for 7.4%, while the lowest M. W. fraction demonstrated was found to be at the site of 13 kDa. By immunoblotting, the specific anti-T. vaginalis antibodies raised in mice recognized a broad spectrum of molecular size range of these antigenic fraction. However, no immunological reaction was seen at the 86 kDa site. CONCLUSION: The proteins of MWs 48-51 kDa possess a larger protein content and a higher immunological reactivity.

[The influence of basic fibroblast growth factor on the adherence of endothelial cell to polyurethane].

This study addressed the problem of how to promote the adherence of endothelial cells (EC) to polyurethane (PU). The influence of basic fibroblast growth factor (bFGF) on the adherence of EC to PU was observed by using the fluorescent method. The results showed that the attachment rates for two bFGF groups(PU precoated with fibronectin and PU precoated with collagen IV) were 71% and 68.5%, which were significantly higher than those (62.5% and 55.25%) for two control groups, respectively. These demonstrate that bFGF can promote EC attachment to PU, and suggest that bFGF can be used as a supplement factor in media for achievement of endothelialization on artificial heart and blood vessels.
