[Study on preventive and therapeutic effect of radix salviae miltiorrhizae on recurrence and metastasis of liver cancer].

OBJECTIVE: To study the inhibitory effect of Radix Salviae Miltiorrhizae (RSM) on adhesive and invasive ability of SMMC-7721 liver cancer cells, and on the metastasis and recurrence on post-hepatectomy liver cancer in nude mice. METHODS: Effect of RSM on SMMC-7721 cell membrane intercellular adhesive molecule-1 (ICAM-1) expression was observed by immunofluorescence flow cytometry; effect on invasive ability and of SMMC-7721 cell and the detachment of which attached to fibronectin (FN) by cell migration experiment; effcet on adhesion of SMMC-7721 cell with FN by MTT method and effect on adhesion between 7721-7721, 7721-lymphocyte and 7721-endothelial cell by cell adhesion experiment. LCI-D20 human liver cancer metastasis model, after hepatectomy, was used to observe the effect of RSM on recurrence and metastasis of liver carcinoma in nude mice. RESULTS: ICAM-1 expression in SMMC-7721 cells incubated with RSM was significantly lower than that in cells did not treated with RSM. RSM could inhibit the invasive ability of SMMC-7721 cell and made the cells already attached to FN exfoliated. It could also inhibit the adhesion of 7721-7721, 7721-lymphocyte and 7721-endothelial cell. And RSM showed preventive and therapeutic effect on intrahepatic and remote metastasis/recurrence of early and late human post-hepatectomy liver cacer in nude mice. CONCLUSION: RSM could inhibit the invasion and adhesion of SMMC-7721 cell and could also prevent and inhibit metastasis and recurrence of human liver cancer after hepatectomy in nude mice.

[Expression and significance of MXR7 mRNA in human hepatocellular carcinoma].

OBJECTIVE: To find out the expression and significance of MXR7 mRNA in human hepatocellular carcinoma. METHODS: The (32)P labeled and purified PCR product of MXR7 cDNA was taken as probe. MXR7 mRNA expression was detected by Northern blot analysis in 30 samples of preoperatively untreated HCC and corresponding surrounding non-cancerous hepatic tissue, and 12 normal liver tissues confirmed pathologically. RESULTS: The frequency of MXR7 mRNA expression in HCC, the corresponding surrounding noncancerous hepatic, and normal liver tissues was 76.6%, 13.3% and 0, respectively. The frequency of MXR7 mRNA expression in HCC without serum AFP elevation and in HCC < 5 cm was 9/10 and 5/6 respectively. CONCLUSIONS: MXR7 mRNA over-expression in human HCC is common and MXR7 mRNA may be expressed at early stage of HCC and can serve as tumor biomarker for HCC. The detection of MXR7 mRNA expression in liver puncture tissues may find small HCC without serum AFP elevation in the subclinical stage.

[A study on pathology and immunoglobulin of orbital tissues from cases with Grave's ophthalmopathy].

OBJECTIVE: To investigate pathological changes and the expression, localization of immunoglobulins (IgA & IgE) in orbital tissues from patients with Grave's ophthalmopathy. METHODS: Orbital tissues were obtained at surgery from 19 patients with Grave's ophthalmopathy and 17 control subjects. Immunohistochemical studies were carried out by using anti-IgA, anti-IgE antibody and a streptavidin-biotin-peroxidase detection system. In addition, hematoxylin-eosin, alcian blue and Masson trichrome stains were employed to demonstrate the pathologic changes in the tissues. RESULTS: 12 of 17 samples from patients exhibited IgA positive staining for the connective tissue associated with the extraocular muscles and orbicularis oculi, however only 2 control muscles reacted minimally. IgE positive material was found in association with the majority of leukocytes and with muscle fibers in 13 patients, but only 4 control subjects displayed mild reactions. Significant expression of IgE was more often demonstrated in patients with eye disease of short course than in those of longer course. CONCLUSION: These results support the notion that the eye muscle fiber and fibroblast are important targets of the orbital autoimmune reaction in Graves' ophthalmopathy in which IgA and IgE both play important roles.

[Genetic polymorphism and susceptibility to occupational chronic manganism: a case-control study].

OBJECTIVE: In a case-control study, the possible genetic factors relevance to occupational chronic manganism were investigated. METHODS: Forty-nine manganisms who were welders and ferromanganese smelters occupationally exposed to manganese dust and fume from three metallurgical industries, and fifty unrelated healthy control subjects who were working same workshops were recruited. The subjects were matched for sex, age, cigarette and alcohol intake. The manganese exposure duration was also matched in this case-control study. Genetic polymorphism of cytochrome P450 2D6L (CYP2D6L) gene and NAD(P)H: quinone oxidreductase (NQO1) genes from all subjects were investigated. The mutations of CYP2D6L gene located exon 6 were analyzed by a polymerase chain reaction (PCR)-based DNA amplification combined with Hha I restriction fragment length polymorphism (RFLP). The substitution of 609(C-T) at exon 6 of DT-diaphroase gene was analyzed by PCR combined Hinf I RFLP. RESULTS: The frequency of polymorphic allels, a mutation of CYP2D6, was significantly lower in patients with manganism (16.3%) than that of the controls (29.0%). A significant association was also found between the homozygote variant of CYP2D6L gene and occupational manganism. These results suggest that the CYP2D6 gene might be one of the susceptibility genes for Mn-induced neurotoxicity. The allele and genotype frequencies of NQO1 gene were similar in the manganism cases and control subjects. CONCLUSION: It is possible that CYP2D6 gene may be a valuable susceptibility biomarker responded to Mn-induced central nervous system disorders in workers exposed to manganese.

[Analysis for spatial distribution of Oncomelania snail in mainland China by geographic information system(GIS) database].

OBJECTIVE: To analyze spatial distribution of Oncomelania snail populations in mainland China by geographic information system (GIS) database. METHODS: Genetic variation experiment data and experimental data of susceptibility of 34 snail populations were collected from nine provinces in China, to set up a database. A world digital map was linked with the database in a software ArcView Release 3.0 a to be used to divide zones with ArcView spatial analyze function and GIS overlaying function. AVHRR NDVI satellite images were collected to form a new one for prevalent season, which were overlaid with distribution maps of heterozygous indices of snail populations, polymorphic locus percentage and infection rates of snail population with schistosome for classifying the images. RESULTS: Spatial analysis showed that distribution maps of genetic heterozygous indices of snail, percentage of polymophic loci and infection rate of snail can be divided into two large zones, i.e., east zone and west zone, both with varied pictures. Analysis for overlaying three-stratum distribution map with AVHRR NDVI satellite image showed four distinctly different spatial zones, including west Sichuan zone, west Yunnan zone, middle zone of river, lake and marshland, and southeast coastal zone. CONCLUSION: It is the first time using GIS to analyze data of Oncomelania population genetics and confirmed the population structure of Oncomelania snails presented discrete sub-population model, which supports the theory of sub-species of Oncomelania spp. Exited in mainland China.

[mRNA and protein expression of apoptosis-regulating gene bcl-x in lymphomas].

OBJECTIVE: To observe mRNA and protein expression of bcl-x and explore the role of bcl-x in the regulation of apoptosis and oncogenesis and their value in differential diagnosis of lymphomas. METHODS: Reverse transcription-polymerase chain reaction (RT-PCR) and citric acid-microwave-ABC immunohistochemistry (ICH) were used to observe mRNA transcription forms and protein distributions of bcl-x in 30 frozen tissues and 109 paraffin embedded tissues (both including reactive hyperplasia and common types of lymphoma). RESULTS: RT-PCR revealed that all 30 frozen cases showed bcl-x transcription, of which 29 had bcl-x1 band and 13 had bcl-xs band. Bcl-x1 band was always stronger than bcl-xs band. IHC found that 97/109 cases had fine positive bcl-x granules in the cytoplasm. In lymphoid reactive hyperplasia, bcl-x exhibited weak immunoactivity in cells located in the germinal centers and interfollicular regions of the nodes but not in the mantle zone lymphocytes which surround germinal centers. In lymphomas, bcl-x immunoactivity was always strong and diffuse. The staining fractions in different types of lymphomas were significantly higher than those in lymphoid reactive hyperplasias (P > 0.05), but no significant difference was found between the positive rate and staining fraction of different types of lymphomas. CONCLUSION: Bcl-x shows active mRNA transcription and high protein expression in lymphomas. The long isoform of bcl-x (bcl-xl) is the predominant expression. The characteristics of bcl-x protein expression may be helpful in differential diagnosis of reactive hyperplasia and lymphoma.

[Changes of neurobehavioral functions in workers exposed to manganese].

The aim of this study was to assess the neurotoxic effects associated with the level of airborne manganese in the work place. A cumulative exposure index(CEI) was calculated for each subject. The neurobehavioral test battery, in terms of choosing reaction time, finger tapping, digit symbol, stylus in hole, Benton visual retention and Santa Ana dexterity was applied in the study. The manganese exposed workers had slower motor and response speed, poorer visuomotor coordination and steadiness. There was a dose-response relationship tendency between the cumulative exposure index and the behavioral function. Hence the neurobehavioral test may be a sensitive method in detecting early changes in motor and cognitive function among manganese-exposed workers. The study also demonstrates that the cumulative exposure of manganese about 1 mg/m3 per year may induce subclinical signs of intoxication.

[The boundary design of mandibular model by means of the three-dimensional finite element method].

OBJECTIVE: To construct the mandibular model and simulate the boundary conditions by means of the three-dimensional finite element method. METHODS: A juvenile mandible, including TMJ, was scanned transversely by CT, then a three-dimensional finite element model of mandible was constructed. At the same time, the functions of stomatologic muscles and ligaments and the functional actions of temporomandibular joint (TMJ) were simulated by means of cable elements and compressive gap elements respectively. RESULTS: The cable elements were consistent with the functions of stomatologic muscles and ligaments, and the gap elements were similar to the boundary conditions of the TMJ and teeth. CONCLUSION: Cable and gap elements are reasonable boundary conditions of the finite element model of mandible, which will improve the similarity of the model.

[Separation and selection of salivary adhesion receptors of Streptococcus mutans to tooth surface].

OBJECTIVE: To select and purify salivary receptors of Streptococcus mutans from experimental salivary acquired pellicle. METHODS: Experimental salivary acquired pellicle (SAP) was performed by coating hydroxyapatite (HA) with whole saliva. Then SAP was washed from HA by 1 mol/L NaCl and 0.5 mol/L phosphate buffer sequentially. The proteins were further separated by chromatography of Sephadex G75 and DEAE-Sephadex A25. Receptors of Streptococcus mutans were selected by bacterial adhesion test and competitive inhibition adhesion test. Identification was performed by PAGE, SDS-PAGE, IP-PAGE and detection of amylase activity and inmunodifusion test. RESULTS: IgA degraded fragments, a protein of 13 kD and amylases were the receptors of S. mutans. The first two only promote the adhesion but the amylases can both promote and inhibit S. mutans adhesion. CONCLUSION: The adhesion of S. mutans to tooth surface is a result of interaction between adhesins of S. mutans and multiple salivary receptors.
